Abstract:Municipal wastewater discharge is threatening the ecological security of the local water environment. This study investigated the field process performance and microorganism characteristics of enhanced biological phosphorus removal (EBPR) in municipal wastewater treatment plants (WWTPs) in China. The results showed that three WWTPs met the required criteria of phosphorus for discharge (≤1 mg/L), but with a low level of Accumulibacter (6.4 to 3.8%, on average) and a relatively high level of Competibacter (3.2 to 9.1%) in sludge. The phosphorus release and uptake rates were varied from 0.224 to 7.770 mg/gVSS·h and 0.386 to 7.901 mg/gVSS·h, respectively. Denitrifying polyphosphate‐accumulating organisms were estimated to be 28.2% of the polyphosphate‐accumulating organisms. Sludge characteristics (phosphorus release and uptake rates) were positively correlated with the abundance of Accumulibacter and negatively correlated with the proportion of Competibacter. Moreover, the lower the ratio of anaerobic phosphorus/acetate (e.g., 0.496) is, the more abundant the Competibacter would be. Further discussion on an improvement strategy for these WWTPs for EBPR should be comprehensively based on the data of periodic investigations on field operation, sludge activities, and microbial populations.
Characterization of phosphorus-releasing bacteria in a small eutrophic shallow lake, Eastern China.

Abstract:Phosphorus contents and phosphorus-releasing bacteria were characterized in a small eutrophic lake, Eastern China. Total phosphorus and water soluable inorganic phosphate (WSIP) in water bodies were as high as the levels of hyper-eutrophic lakes. Calcium bound phosphate and organic phosphorus were 2 major forms of phosphorus existing in the sediments. Enumeration of inorganic phosphate-solubilizing bacteria (IPB) and organic phosphorus-mineralizing bacteria (OPB) with culture-dependent method showed that these bacterial groups were not very rich in the ecosystem. Molecular identification and phylogenetic analysis of the predominant IPB and OPB strains indicated that there existed various kinds of bacteria participating in the phosphorus release. Laboratory tests on phosphorus release abilities showed IPB strains could liberate more than 50μgml−1, while OPB strains produced less than 2μgml−1 WSIP every day.

Abstract:Abstract To reduce aquatic eutrophication, measurements of bioavailable phosphorus (BAP) rather than total phosphorus (TP) are deemed critical. However, current methods require much time to separate sediments from river water, which limits the routine measurement of BAP in rivers. Therefore, in this study, a simultaneous multisample ultrasonic extraction method is proposed to directly measure total BAP (TBAP) in river water without the separation of sediment and water. Spike-and-recovery assessments showed that at least three extractions are required to maintain efficiency. A process including 2-min extraction time and three extractions was suggested. The concentrations of TBAP extracted by this process showed no significant differences with the spike calculations. Furthermore, river water TBAP was quantified using the conventional and proposed method to examine the practicality of using the proposed method for simultaneous multisample ultrasonic extraction and to evaluate its adaptability to actual river water analysis. The extracted concentrations matched those obtained using the conventional method, in which total BAP is calculated as the sum of dissolved BAP and particulate BAP; no significant difference was observed between the concentrations. Ultrasonic extraction was considerably less time-consuming than the conventional method because more samples could be analyzed during a single run. Therefore, the simultaneous multisample ultrasonic extraction method proposed in this study can be used to directly quantify total BAP in river water.

Abstract:The light microscope is widely used to count algae, however, there are some disadvantages associated with this method, such as time consuming and laborious. In this study, a qPCR-based method was established for quantifying seven phyla of common algae in freshwater, including Cyanophyta, Chlorophyta, Euglenophyta, Bacillariophyta, Dinophyta, Cryptophyta, and Chrysophyta. The accuracy of qPCR in estimating algal cells was confirmed by comparing it with the microscopic counting. The qPCR was used to detect the cell concentration of seven phyla of algae in Longhu Reservoir, showing that green algal blooms occurred during the monitoring period. The intensity of algal blooms was further evaluated according to the classification standard, which suggested that the grade of this bloom was mild. An early warning system was proposed to early warn the occurrence of algal blooms in two water sources, Longhu Reservoir and Dongzhang Reservoir. The qPCR method developed in this study could be a useful tool in the monitoring of algae. The early warning system reported here will have important implications for the effective warning of algal blooms.

Abstract:Considering the impact of the high salinity and high turbidity of coastal seawater on phosphorus forms, a new method was proposed to determine bioavailable inorganic phosphorus (BIP). The phosphorus most relevant to eutrophication is BIP, and traditional analysis methods may underestimate the degree of eutrophication. In this study, a microelectrode of multigold (AuμE) was fabricated for direct voltammetric determination of BIP without filtration, and BIP environmental characteristics including distribution and correlation relationships with environmental factors in typical coastal seawater of Northern China were analyzed. The proposed AuμE showed a low detection limit of 0.03 μM. The surface and bottom BIP concentrations ranged from 1.00 to 2.13 and from 0.88 to 2.05 μM, respectively. BIP dominated the total P (TP) accounting for 48.5–67.5 % in the surface layer samples, and 32.6–92.7 % in the bottom layer samples, respectively. The concentrations of BIP were obviously higher than those of DIP, indicating that DIP may underestimate the probability of eutrophication occurring. And BIP was positively correlated with dissolved oxygen (DO) ( P < 0.05). BIP may be a promising indicator of eutrophication potential in coastal areas with high salinity and high turbidity. The proposed reliable voltammetry method provides a new indicator for environmental assessment and represents a significant step in the comprehensive analysis of P species.

Abstract:Phosphorus (P) fractions and phosphatase activities were measured in 22 coastal wetland soils with typical vegetation successions in Yancheng National Nature Reserve, China. P forms and phosphatase activities varied greatly from site to site even under the same vegetation cover. NH4Cl-P, bicarbonate/dithionite extracted P and NaOH-P were remarkably higher (p < 0.05) in soils With exotic invasive plants, Spartina alterniflora, than in soils with the native species Suaeda salsa, Scirpus mango etc and Phragmites australis. HCl-P and refractory P showed little variation. No significant differences were detected for either alkaline phosphatase (ALAP) or acid phosphatase (ACAP) among the soils. All of the above properties were much higher in soils with plant growth compared to bare flat soils. Regression analysis demonstrated that organic matter (OM), Al, Ca, Fe and total P (TP) were able to explain more than 70% of the variations in the P fractions (except 29% of NH4Cl-P), and OM was the most important contributing factor. ALAP and ACAP were irrelevant to P but were significantly related to TOC, suggesting that carbon was a limiting factor for P mineralization in this area. Owing to its huge biomass and densities, Spartina alterniflora displayed great potential for carbon input, thus facilitating P mineralization and cycling. The results enhance our understanding of P availability differences in this area covered by invasive and native vegetation. (C) 2015 Elsevier Ltd. All rights reserved.

Abstract:The increasing prevalence of phosphorus limitation in coastal waters has drawn attention to the bioavailability of cellular surface-adsorbed phosphorus (SP) as a reservoir of phosphorus in phytoplankton. This study examined the storage, utilization, and regulation of SP in the coastal waters of the East China Sea, as well as three cultivated algal bloom species ( Skeletonema marinoi , Prorocentrum shikokuense , and Karenia mikimotoi ) prevalent in the area. SP accounted for 14.3%–45.5% of particulate phosphorus in the field and laboratory species. After the depletion of external phosphate, the studied species can rapidly transport SP within 3–24 h. The storage of SP is regulated by both external phosphate conditions and the internal growth stage of cells, but it is not influenced by the various cellular surface structures of the studied species. This study highlights the significance of SP as a crucial phosphorus reservoir and the potential use of the SP level as an indicator of phosphorus deficiency in phytoplankton.

Abstract:The speciation of phosphorus (P) in freshwater affects its bioavailability. Analytical detection methods were compared for different colloidal and dissolved organic forms of phosphorus (P) in relation to the potential P bioavailability to Pseudokirchneriella subcapitata, determined with a 14 day growth response to P. Growth on these P-forms was referenced to supplies of inorganic P (Pi) as operational definition of the relative potential bioavailability. The bioavailability of ten organic P molecules ranged 1–70% of Pi while these forms were generally not detected by colorimetric method (CM, malachite green) or ion chromatography (IC). The bioavailability of P associated with Fe- and Al oxides ranged 55–85% of Pi and these forms were completely detected by CM and partially by IC. The bioavailability of total dissolved P in the environmental samples ranged 7–85% (mean 43%) of Pi. The P detected by IC underestimates bioavailable P while CM and total dissolved P (inductively coupled plasma, ICP) overestimate P bioavailability by, on average 44% (CM) or 57% (ICP) in the environmental samples. We conclude that CM is the best index among the three tested for predicting long-term availability of environmental dissolved P in which colloidal P contributes more importantly than organic P.

Abstract:Sediment phosphorus (P) release in shallow eutrophic lakes is a major contributor of P to algal blooms. This research proposes an innovative notion in which the P diffusive fluxes at the sediment-water interface (SWI) of shallow lakes are controlled by the P adsorption-desorption equilibria, with pH as the major regulating factor. The P equilibrium concentration (Ce) at SWI was conceptualized into a dependent variable responding to two factor-dependent variables, the algae/cyanobacteria-available P pools of the SWI and the pH in the water column, resulting in the empirical equation Ce(pH) = Cm/[1 + e-k(pH-pH1/2)]. Cm is the maximum P equilibrium concentration when all algae/cyanobacteria-available P in sediments is released, and the value relies on the thickness of the oxygen and pH transition layer that contains iron/aluminium (hydr)oxide-adsorbed P. The parameters in the empirical equation are accessible from P desorption tests conducted on a set of sediment samples with different P pollution levels. This research provides a quantitative approach for determining the sediment P criteria of shallow lakes, with sediment iron/aluminium (hydr)oxide-adsorbed P and water depth as two main indicators with ecological implications. A decrease in water depth would proportionally increase the P concentration at a similar sediment P releasing flux and increase algae/cyanobacteria-available P pools that are ready to equilibrate with the water column by increasing hydrodynamic disturbance of the SWI.

Abstract:This review summarizes the main species of polyphosphate accumulating organisms (PAOs) and algae, illustrates their pathways and key enzymes, discusses biological phosphorous (P) recovery from dilute waters, and identifies research avenues to encourage adoption and implementation. Phylogenic analysis indicates that the Proteobacteria phylum plays an important role in enhanced biological phosphorus removal (EBPR). The use of meta-transcriptome analysis and single cell-based techniques to help overcome the challenges associated with non-PAO competition was discussed. For algae capable of luxury phosphorus uptake, fundamental research is needed to illustrate the phosphorus regulation process and key proteins involved. Emerging technologies and processes have great potential to further advance phosphorus recovery, including combined PAO/algae reactors, bioelectrochemical systems, and biosorption by phosphorus binding proteins. As the paradigm shifts toward holistic resource recovery, research is needed to explore P+ recovery with other resources (e.g., metals from sludge), using a combination of biological and chemical approaches.

Abstract:Phosphorus(P) recycling from waste streams is crucial to mitigate the P depletion crisis. P forms and contents in organic waste are critical for determining the recycling method and efficiency. We constructed an approach to characterize P forms in seven organic waste by combining chemical sequential extraction, enzymatic hydrolysis, and nuclear magnetic resonance(NMR). Livestock manure and straw exhibited a higher active P(H 2 O-P&NaHCO 3 -P)(70.54%-84.40% and 65.78%-85.26% of total P) than sewage sludge(18.22%) and food waste(43.90%). Enzymatic hydrolysis revealed over 10% P in the so-called active P of corn(11.30%) and rice straw(13.32%) was phytate-like P, which is not bioavailable. These findings indicate the chemical sequential extraction inaccurately gauges bioavailable-P and underscores the need to convert phytate into plant-available P in recycling processes(biogas, composting), especially for crop straws and chicken manure. This work introduces a novel methodological framework for assessing P potential bioavailability in organic waste, providing fundamental knowledge for the P recycling process optimization.

Abstract:A new method using polyphosphate kinase (PPK) probe was developed in this study to measure the ratio of denitrifying phosphate-accumulating organisms (DNPAOs) to bacteria. Advantages and disadvantages of this method and previously published methods were evaluated. Biochemical characteristics and metabolic mechanisms of the bacterial community in activated sludge were analyzed. The result indicated that PPK probe targeting at the gene of polyphosphate kinase in bacterial cells could marked more PAOs than traditional PAO probes. The ratio of DNPAOs/bacteria reached 60.30% in activated sludge. The whole bacterial community in activated sludge, at the end of A/A stage, presented significant non-fermentative characteristics although the activated sludge still contained some fermentative bacteria. Study on the metabolic mechanisms suggested that intracellular PHB served as the major carbon source and energy source of bacterial community in activated sludge.

Abstract:The bioavailability of sedimentary P was investigated under repeated sediment resuspension with and without the addition of algae. Results show that the release of P as dissolved total P and dissolved inorganic P depends on the reduction in Fe/Al–P and Ca–P in the sediments. The reduction rates can be attributed to sediment resuspension and algal assimilation. However, under a low algal concentration, the assimilation of algae in P is lower than the readsorption of reoxidized Fe or other P-binding matter, such as clays and CaCO3 particles. This result was confirmed by the increase in algal available P (AAP) under sediment resuspension conditions with and without the addition of algae. Although measuring the formation and reduction rates of AAP is difficult, AAP is a good predictor of the bioavailability of sedimentary P. The bioavailable P in the sediments can be estimated using the sum of NH4Cl-P, AAP, and %Ca–P at certain concentrations of algae given the assimilation of Ca–P under sediment resuspension conditions with the addition of algae.

Abstract:Phosphorus (P) characteristics in eutrophic lakes change during algal growth. Furthermore, algae have a significant relationship with the microbial communities of lake sediments. This study addressed the influence of algal growth and soluble reactive phosphorus (SRP) concentrations on P characteristics within the sediment-water-algal (SWA) system. Results indicated that the SWA system simulating a high algal bloom level (SWA-HAB) had a correspondingly high SRP concentration (258.9 μg L −1 ), and that algal growth promoted a high SRP concentration in the overlying water. The high SRP concentration in overlying water could support algal growth, resulting in a high chlorophyll a (Chl-a) concentration (285.23 mg L −1 ). During algal growth, the P release flux was high in sediments from the high-SRP SWA system, with the highest P release flux measured at 0.982 mg m −2 day −1 . Furthermore, microbial community abundance had a significant relationship with Chl-a concentrations in overlying water ( p < 0.05) and increases with algae growth.

Abstract:Characteristics and transformation of organic phosphorus in water are vital to biogeochemical cycling of phosphorus and support of blooms of phytoplankton and cyanobacteria. Using solution 31P nuclear magnetic resonance (NMR), combined with field surveys and lab analyses, composition and structural characteristics of dissolved phosphorus (DP), particulate phosphorus (PP) and organic P in algae were studied in two eutrophic lakes in China, Tai Lake and Chao Lake. Factors influencing migration and transformation of these constituents in lake ecosystems were also investigated. A method was developed to extract, flocculate and concentrate DP and PP from lake water samples. Results showed that orthophosphate (Ortho-P) constituted 32.4%–81.3% of DP and 43.7%–54.9% of PP, respectively; while monoester phosphorus (Mono-P) was 13.2%–54.0% of DP and 32.9%–43.7% of PP, respectively. Phosphorus in algae was mostly organic P, especially Mono-P, which was ≥50% of TP. Environmental factors and water quality parameters such as temperature (T), electrical conductivity (EC), pH, secchi depth (SD), dissolved oxygen (DO), chemical oxygen demand (CODcr), chlorophyll-a (Chl-a), affected the absolute and relative concentrations of various P components in the two lakes. Increased temperature promoted bioavailable P (Ortho-P and Mono-P) release to the lake waters. The results can provide an important theoretical basis for the mutual conversion process of organic P components between various media in the lake water environment.

Abstract:It is well-accepted that phosphorus, particularly orthophosphate, is a determinant factor in aquatic eutrophication. However, numerous kinds of phosphorus sources exist in real world scenario, and limited studies have characterized the pairwise relationships among abundant different phosphorus sources and the physiological behaviour of algae. The present study developed a high-throughput assay to investigate the effects of 59 different phosphorus sources (equal initial concentration of total phosphorus) on the growth and alkaline phosphatase (AKP) activities of Microcystis aeruginosa, a model cyanobacteria whose predominance holds sway in lake eutrophication. M. aeruginosa cultivated with nucleoside monophosphates (NMPs) had higher growth, relative AKP activities and residual orthophosphate, which were positively intercorrelated. Oppositely, non-NMPs cultivation of M. aeruginosa led to negative relationships between the relative AKP activities and their growth or residual orthophosphate. These results indicated distinct mechanisms for M. aeruginosa to utilize different phosphorus sources in real-world scenario, and both phosphorus source and content are determinant factors on the growth and physiological behaviour of M. aeruginosa. Given the complicated and vast phosphorus pool in the natural environment, phosphorus resources might significantly alter the abundance and physiological behaviour of M. aeruginosa and other bloom-forming algae, then influence the phytoplanktonic community structure and affect the possibility and intensity of algal bloom. Our work hints the underestimation of the restriction factors in lake eutrophication and provides a new tool to study the driven forces of phytoplanktonic community dynamics as phosphorus from both internal and external sources.
Phosphorus bioavailability and the diversity of microbial community in sediment in response to modified calcium peroxide ceramsite capping

A 62-day experiment was conducted by capping with modified calcium peroxide ceramsite (MCPC) at different addition dosages (from 1:1 to 1:4, the ratio of mobile P in sediment to MCPC). P disappearance and release were expressed by the dissolved inorganic P (DIP) in overlying water. The results show that the proportion of disappeared P in released P was reduced sharply from 44% (Control) to 2% (1:4) under the capping with MCPC. Under the capping, the concentrations of DIP and Fe(Ⅱ) in pore water decreased markedly compared with the control, the removal up to 71.6% and 59.3% (mean, P < 0.05), respectively. The bioavailable P (BAP) presented the obvious decline under the capping with MCPC, which indicates the inhibition on the algae growth. The algae available P (AAP), water soluble P (WSP) and readily desorbable P (RDP) were reduced obviously at the 1:2 ratio compared with the other ratios. On the contrary, the biggest increase of Olsen-P in sediment was at the 1:2 ratio. This may be the explanation why the disappearance of DIP in the pore water was found. The result of microbial community structures in sediment shows that the relative abundance of Proteobacteria and Nitrospirae increased under the MCPC capping. It is analyzed that the microbial diversity is related negatively with the BAP in the sediments under the capping with MCPC, suggesting that microbial diversity is the key to control the BAP.
Sustainable and eco-friendly approach for phosphorus recovery from wastewater by hydrothermally carbonized microalgae: Study on spent bio-char as fertilizer

Abstract:This study focuses on phosphorus recovery from simulated aqueous environment by hydrothermally carbonized microalgae biomass. A detailed study on parameters influencing adsorption such as time (10–60 min), temperature (10–60 °C), agitation speed (50–250 rpm) and bio-char concentration (0.3–1.5 g/L) was carried out. Batch studies showed a maximum 90 % phosphorus removal for a concentration of bio-char of 1.5 g/L, agitation speed of 150 rpm, temperature of 60 °C and at 60 min. The adsorption experimental data fitted well with Pseudo-first order kinetics and Langmuir isotherm models with correlation coefficient (R2) value of 0.99 and 0.97 respectively. According to Langmuir model, phosphorus and ammoniacal nitrogen adsorption capacity of bio-char was 2505 and 6.7 × e4 mg/g with least error. To avoid leaching of ammonia and phosphorus to atmosphere, the spent bio-char was activated with ammonia and phosphorus solubilizing bacteria and tested as fertilizer. Pot experiments performed with tomato plant showed that spent bio-char is an excellent fertilizer and comparable to commercial di-ammonium phosphate (DAP). The plants cultivated with spent bio-char fertilizer have reached an average height of 22 ± 0.5 cm and their chlorophyll contents were 24.5 ± 0.42 mg/g. This study provides a proof of concept and it can be extended to real time wastewater in future. This study is an example of sustainable approach towards recovery of valuable phosphorus from aqueous environment and using it as fertile soil amendment.

Abstract:Organic phosphorus mineralization is one of the important pathways for phosphorus mobilization. However, the level of organic phosphorus mineralization in rice paddies under the coupling of water-saving irrigation and biochar has yet to be determined. To address this knowledge gap, we investigated the interplay between available phosphorus fractions, alkaline phosphomonoesterase activity, and bacterial communities encoding alkaline phosphomonoesterase in water-saving irrigated paddy fields subjected to different biochar applicated rates (0 t ha −1 , 20 t ha −1 , and 40 t ha −1 ), compared with a control (flooding irrigation with 0 t ha −1 biochar application). The findings revealed that changes in alkaline phosphomonoesterase activity were linked to variations in phoD gene numbers and the composition of phoD microbial communities. Water-saving irrigation, as opposed to flooding irrigation, notably enhanced the capacity of organic phosphorus mineralization in paddy fields. This enhancement was supported by a 27.94%–45.84% increase in alkaline phosphomonoesterase activity and a 23.00%–36.06% decrease in enzyme-extractable phosphorus in water-saving irrigated paddy fields vs. flooding irrigated paddy fields. Simultaneously, the important role of Firmicutes among phoD -harboring microorganism in water-saving irrigated rice paddies was detected. However, the increased capacity of microbes-mediated organic phosphorus mineralization under the condition of water-saving irrigation alone was insufficient to offset the resulting phosphorus depletion. Biochar could improve the available phosphorus content, which promoted competition among phoD -carrying microorganisms, raising the risk of phosphorus loss. The combined approach of water-saving irrigation and 40 t ha −1 biochar application enhanced the availability of phosphorus and simultaneously mitigated the risk of phosphorus loss, which is proved to be a more suitable field management strategy for paddy fields in southern China.
