Abstract:Emergence of endogenous nanoparticles in thermally processed food has aroused much attention due to their unique properties and potential biological impact. The aim of this study was to investigate the presence of fluorescence nanoparticles in roasted chicken breasts, elemental composition, physicochemical properties, and their molecular interaction with human serum albumin (HSA). Transmission electron microscopy analysis revealed that the foodborne nanoparticles from roasted chicken were nearly spherical with an average particle size of 1.7 +/- 0.4 nm. The elemental analysis of X-ray photoelectron spectroscopy showed the composition of nanoparticles as 47.4% C, 25.8% O, and 26.1% N. The fluorescence of HSA was quenched by the nanoparticles following a static mode, and the molecular interaction of nanoparticles with HSA was spontaneous (Delta G(0) < 0), where hydrogen bonding and van der Waals forces played an important role during HSA-nanoparticles complex stabilization through thermodynamic analysis by isothermal titration calorimetry. The principal location of the nanoparticles binding site on HSA was primarily in site I as determined by site-specific marker competition. The conformational of HSA was also changed and alpha-helical structure decreased in the presence of nanoparticles. Our studies revealed that fluorescent nanoparticles were produced after roasting of chicken breast at 230 C for 30 min for the first time. The obtained nanoparticles can interact with HSA in a spontaneous manner, thus providing valuable insight into foodborne NPs as well as their effects to human albumin protein.

Abstract:A new fluorescent nanoparticle was synthesized by chemical crosslinking of polyacrylic acid using 4-aminoethanol-N-hydroxyethanyl-1,8-naphthalimide as the crosslinker. The particle possesses excellent characteristics, including low cytotoxicity, water solubility and good fluorescence properties. The interaction of the particle with bovine serum albumin was investigated by absorbance, fluorescence and circular dichroism spectroscopic methods. The interaction mechanisms, binding model and reciprocal effects on structure and fluorescence between nanoparticle and protein are discussed. The spectral data indicated that the nanoparticle could spontaneously form a reversible complex with bovine serum albumin in solution used mainly by hydrogen bonds and van der Waals forces. Fluorescence resonance energy transfer from tryptophan to naphthalimide in the complex was demonstrated and employed to determine their relative separation distance. During the interaction process, unfolding of polypeptide chains in the protein occurred but no adjustments of local polarities around the tryptophan and tyrosine residues were observed. In addition, it was found that the presence of protein induced a notable enhancement in nanoparticle fluorescence and a blue-shift in the emission maximum. These results will be useful for further applications of the developed nanoparticle in biomedical areas.
Fabrication of Fluorescent Silica Nanoparticles Hybridized with AIE Luminogens and Exploration of Their Applications As Nanobiosensors in Intracellular Imaging

Abstract:Highly emissive inorganic-organic nanoparticles with core-shell structures are fabricated by a one-pot, surfactant-free hybridization process. The surfactant-free sol-gel reactions of tetraphenylethene- (TPE) and silole-functionalized siloxanes followed by reactions with tetraethoxysilane afford fluorescent silica nanoparticles FSNP-1 and FSNP-2, respectively. The FSNPs are uniformly sized, surface-charged and colloidally stable. The diameters of the FSNPs are tunable in the range of 45-295 nm by changing the reaction conditions. Whereas their TPE and silole precursors are non-emissive, the FSNPs strongly emit in the visible vision, as a result of the novel aggregation-induced emission (AIE) characteristics of the TPE and silole aggregates in the hybrid nanoparticles. The FSNPs pose no toxicity to living cells and can be utilized to selectively image cytoplasm of HeLa cells.
Using Chicken Embryos to Identify the Key Determinants of Nanoparticles for the Crossing of Air-Blood Barriers.

Abstract:Fine particulates (FPs) are a major class of airborne pollutants. In mammals, FPs may reach the alveoli through the respiratory system, cross the air-blood barrier, spread into other organs, and induce hazardous effects. Although birds have much higher respiratory risks to FPs than mammals, the biological fate of inhaled FPs in birds has rarely been explored. Herein, we attempted to disclose the key properties that dictate the lung penetration of nanoparticles (NPs) by visualizing a library of 27 fluorescent nanoparticles (FNPs) in chicken embryos. The FNP library was prepared by combinational chemistry to tune their compositions, morphologies, sizes, and surface charges. These NPs were injected into the lungs of chicken embryos for dynamic imaging of their distributions by IVIS Spectrum. FNPs with diameters <16 nm could cross the air-blood barrier in 20 min, spread into the blood, and accumulate in the yolk sac. In contrast, large FNPs (>30 nm) were mainly retained in the lungs and rarely detected in other tissues/organs. In addition to size, surface charge was the secondary determinant for NPs to cross the air-blood barrier. Compared to cationic and anionic particles, neutrally charged FNPs showed the fastest lung penetration. A predictive model was therefore developed to rank the lung penetration capability of FNPs by in silico analysis. The in silico predictions could be well validated in chicks by oropharyngeal exposure to six FNPs. Overall, our study discovered the key properties of NPs that are responsible for their lung penetration and established a predictive model that will greatly facilitate respiratory risk assessments of nanoproducts.

Abstract:The presence of nanoparticles in food has drawn much attention in recent years. Fluorescent carbon nanoparticles are a new class of nanostructures; however, the distribution and physicochemical properties of such nanoparticles in food remain unclear. Herein, the presence of fluorescent carbon nanopartides in baked lamb was confirmed, and their physicochemical properties were investigated. The fluorescent carbon nanoparticles from baked lamb emit strong blue fluorescence under ultraviolet light with a 10% fluorescent quantum yield. The nanoparticles are roughly spherical in appearance with a diameter of around 2.0 nm. Hydroxyl, amino, and carboxyl groups exist on the surface of nanoparticles. In addition, the nanoparticles could serve as a fluorescence sensor for glucose detection through an oxidation reduction reaction. This work is the first report on fluorescent carbon nanoparticles present in baked lamb, which provides valuable insight into the physicochemical properties of such nanoparticles and their potential application in sensors.

Abstract:Biocompatible fluorescent carbon dots (CDs) were prepared via a simple and green route using duck breasts as a natural carbon source. The CDs from duck breasts were well dispersed, and their mean particle size decreased from 2.59 to 1.95 nm when the roasting temperature increased from 200 to 300 degrees C. Abundant functional groups such as -OH, -COOH, and -NH2 were observed on the surface of the CDs, providing the CDs with good water solubility. These CDs emitted strong fluorescence under ultraviolet light irradiation and exhibited superior photostability. The absolute fluorescence quantum yield of CDs rose from 10.53% to 38.05% when the relative nitrogen content of CDs increased from 7.18% to 12.73%. The CDs showed low toxicity to PC12 cells for prolonged exposure. Therefore, the duck CDs were successfully developed as fluorescent probes for in vitro PC12 cells and in vivo Caenorhabditis elegans imaging. These results indicated that the CDs derived from roast duck were biocompatible and can potentially be used as probes in bio-imaging.

Abstract:Food-borne nanoparticles that are generated during the thermal processing of various consumed foods are of great concern due to their unique properties. In this study, the presence of fluorescent nanoparticles (FNPs) in pizza, their biodistribution and cytotoxicity were investigated. The spherical FNPs have a diameter of about 3.33 nm. X-ray photoelectron spectroscopy and Fourier transform infrared spectroscopy analysis revealed that they contained 68.21% C, 27.44% O, 2.75% N and 1.60% S, and the functional groups on their surface included -OH, -COOH, C[double bond, length as m-dash]C, -NH2 and C[double bond, length as m-dash]O. In vitro and in vivo biodistribution of pizza FNPs was evaluated using normal rat kidney (NRK) cells, onion epidermal cells, Caenorhabditis elegans and mice. The fluorescence microscopy images clearly indicate that the pizza FNPs appear to be localized within the cytoplasm. However, the FNPs remained restricted to the extracellular space of the onion epithelium and did not enter the onion cell cytoplasm because of the cell wall. The FNPs were swallowed by the Caenorhabditis elegans worms when exposed to food OP50 and distributed within the pharynx, intestine and anus. Obvious fluorescence of the FNPs in the stomach, intestine, liver, lung and kidney was observed for the FNPs in mouse organs, but not the brain, heart, and spleen. Furthermore, the produced FNPs were found to cause cell cycle arrest at the G0/G1 phase in NRK cells, and resulted in cell apoptosis at high doses. The outcome of this research offers an important insight into the nature of thermal processing-induced nanoparticles and their in vivo and in vitro biological effects.

Abstract:Fluorescent nanoparticles (FNPs) have been widely used in chemistry and medicine for decades, but their employment in biology is relatively recent. Past reviews on FNPs have focused on chemical, physical or medical uses, making the extrapolation to biological applications difficult. In biology, FNPs have largely been used for biosensing and molecular tracking. However, concerns over toxicity in early types of FNPs, such as cadmium-containing quantum dots (QDs), may have prevented wide adoption. Recent developments, especially in non-Cd-containing FNPs, have alleviated toxicity problems, facilitating the use of FNPs for addressing ecological, physiological and molecule-level processes in biological research. Standardised protocols from synthesis to application and interdisciplinary approaches are critical for establishing FNPs in the biologists' tool kit. Here, we present an introduction to FNPs, summarise their use in biological applications, and discuss technical issues such as data reliability and biocompatibility. We assess whether biological research can benefit from FNPs and suggest ways in which FNPs can be applied to answer questions in biology. We conclude that FNPs have a great potential for studying various biological processes, especially tracking, sensing and imaging in physiology and ecology.

Abstract:Food-derived colloidal nanoparticles (CNPs) have been found in many food cooking processes, and their specific effects on human health need to be further explored. Here, we report on the successful isolation of CNPs from duck soup. The hydrodynamic diameters of the obtained CNPs were 255.23 ± 12.77 nm, which comprised lipids (51.2%), protein (30.8%), and carbohydrates (7.9%). As indicated by the tests of free radical scavenging and ferric reducing capacities, the CNPs possessed remarkable antioxidant activity. Macrophages and enterocytes are essential for intestinal homeostasis. Therefore, RAW 264.7 and Caco-2 were applied to establish an oxidative stress model to investigate the antioxidant characteristics of the CNPs. The results showed that the CNPs from duck soup could be engulfed by these two cell lines, and could significantly alleviate 2,2′-Azobis(2-methylpropionamidine) dihydrochloride (AAPH)-induced oxidative damage. It indicates that the intake of duck soup is beneficial for intestinal health. These data contribute to revealing the underlying functional mechanism of Chinese traditional duck soup and the development of food-derived functional components.

Abstract:Fluorescent organic small molecular nanomaterials, due to their excellence performance in organic electronics and biological sensing, have attracted more and more attention in the fields of chemistry, materials, biology, and so on. In the past years, significant advances in both the experimental and theoretical fronts have been made for the nanomaterials and nanotechnology. However, compared with fluorescent inorganic quantum dots or polymer nanoparticles, fluorescent organic nanoparticles (FONs) which based on pi-conjugated systems, are becoming more and more important to bio-imaging, drug carriers and optoelectronic, since the wider variability and flexibility in materials synthesis and nanoparticles preparation of organic molecules. This paper systematically introduces the recent research developments of the design, synthesis, photophysical properties and application of fluorescent organic small molecular nanomaterials. The highlights of this paper are the preparation methods of fluorescent organic small molecular nanomaterials, such as reprecipitation method, ion association method, self-assembly method, micro-emulsion method, laser fabrication method, adsorbent assisted physical vapor deposition method. We briefly conclude the advantages and disadvantages of the above-mentioned synthetic methods. In addition, we also compare the properties of the fluorescent organic small molecular nanomaterials synthesized by different method, and give a brief introduction to their application on organic optoelectronic devices, chemical and biological sensing, bio-imaging and other fields.

Abstract:Nanoparticles (NPs) are small-sized, with high surface activity and antibacterial and antioxidant properties. As a result, some NPs are used as functional ingredients in food additives, food packaging materials, nutrient delivery, nanopesticides, animal feeds, and fertilizers to improve the bioavailability, quality, and performance complement or upgrade. However, the widespread use of NPs in the industry increases the exposure risk of NPs to humans due to their migration from the environment to food. Nevertheless, some NPs, such as carbon dots, NPs found in various thermally processed foods, are also naturally produced from the food during food processing. Given their excellent ability to penetrate biopermeable barriers, the potential safety hazards of NPs on human health have attracted increased attention. Herein, three emerging NPs are introduced including carbon-based NPs (e.g., CNTs), nanoselenium NPs (SeNPs), and rare earth oxide NPs (e.g., CeO2 NPs). In addition, their applications in the food industry, absorption pathways into the human body, and potential risk mechanisms are discussed. Challenges and prospects for the use of NPs in food are also proposed.

Abstract:A prerequisite for in vivo fluorescence imaging is the safety of fluorescent probes. Among all fluorescent probes, fluorescent proteins (FPs) might be the safest ones, which have been widely used in biological sciences at the gene level. But FPs have not been used in vivo in the purified form yet due to the instability of proteins. Here, we individually coat near-infrared (NIR) FPs (NIRFPs) with a silica nanoshell, resulting in NIRFP@silica, one of the safest and brightest NIR fluorescent nanoprobes with a quantum yield of 0.33 for in vivo imaging. The silica shell not only protects NIRFPs from denaturation and metabolic digestion, but also enhances the quantum yield and photostability of the coated NIRFPs. When injected via the tail vein, NIRFP@silica NPs can distribute all over the mouse body, and then can be efficiently eliminated through urine in 24 h, demonstrating its potential applications as a safe and robust NIR fluorescence probe for whole body imaging.
Fluorescent Labeling of Human Serum Albumin by Thiol-Cyanimide Addition and Its Application in the Fluorescence Quenching Method for Nanoparticle–Protein Interactions

Abstract:A boron-dipyrromethene (BODIPY)-based fluorescent probe, BDP-CN, was synthesized in this work. It had a fluorescence emission maximum at 512 nm and a high quantum yield (48%). As evidenced by agarose gel electrophoresis and liquid chromatography-mass spectrometry, it could realize the fluorescent labeling of human serum albumin (HSA) through a thiol-cyanimide addition. Interestingly, f-HSA, defined as HSA labeled by BDP-CN, had an even higher quantum yield (77%). In addition, BDP-CN would not affect the secondary structure of HSA. Based on the successful formation of f-HSA, it was further applied to study the interactions with nanoparticles. The fluorescence quenching of f-HSA by dihydrolipoic acid-coated gold nanoclusters (DHLA-AuNCs) obeyed a dynamic mechanism, consistent with the intrinsic fluorescence quenching of HSA by DHLA-AuNCs. The association constant Ka between f-HSA and DHLA-AuNCs at 298 K was 1.5 × 105 M-1, which was the same order of magnitude as that between HSA and DHLA-AuNCs. Moreover, the interactions of f-HSA with glutathione-coated gold nanoclusters confirmed that the labeled fluorescence could replace the intrinsic fluorescence to monitor the interactions between proteins and nanoparticles. By this method, strong fluorescence ensures better stability and reproducibility, excitation at a longer wavelength reduces the damage to the proteins, and covalent conjugation with cysteine residues eliminates the inner filter effects to a great extent. Therefore, the strategy for the fluorescent labeling of HSA can be expanded to investigate a broad class of nanoparticle-protein interactions and inspire even more fluorescent labeling methods with organic dyes.

Abstract:A key aspect of biochip and biosensor preparation is optimization of the optical or electrochemical techniques that combine high sensitivity and specificity. Among them, optical techniques such as the use of fluorescent polymeric nanoparticles have resulted in dramatic progress in the field of diagnostics due to their range of advantages. We herein report a facile approach for the development of novel fluorescein polymeric nanoparticles (FPNPs) with immobilization of specific biomolecules for application in a highly sensitive optical biosensor. A series of three amphiphilic fluorescein polymers (poly(FMA-r-NAS-r-MA)), comprising hydrophobic fluorescein O-methacrylate (FMA), hydrophilic N-acryloxysuccinimide (NAS), and methacrylic acid (MA) monomers were synthesized through radical polymerization. In an aqueous environment, these fluorescein polymers self-assembled into spherical shaped nanoparticles with a well-defined particle size, narrow particle size distribution, and enhanced fluorescence properties. The bio-immobilization properties of the FPNPs were also tunable by control of the activated N-hydroxysuccinimide ester group in the polymer series. Furthermore, the fluorescence sensitivity of bovine serum albumin detection by the FPNPs indicates that the limit of detection and sensitivity were improved compared to conventional fluorescence dye-labelled proteins. These novel FPNPs therefore represent a suitable technology for disease diagnosis and biomarker detection to ultimately improve the sensitivity of existing analytical methodologies in a facile and cost-effective manner.

Abstract:Fluorescent organic nanoparticles (FONs) have been regarded as the promising candidates for biomedical applications owing to their well adjustment of chemical structure and optical properties and good biological properties. However, the preparation of FONs from the natural derived polymers has been rarely reported thus far. In current work, we reported a novel strategy for preparation of FONs based on the self-polymerization of starch-dopamine conjugates and polyethyleneimine in rather mild experimental conditions, including air atmosphere, aqueous solution, absent catalysts and at room temperature. The morphology, chemical structure and optical properties of the resultant starch-based FONs were investigated by different characterization techniques. Biological evaluation results demonstrated that these starch-based FONs possess good biocompatibility and fluorescent imaging performance. More importantly, the novel strategy might also be extended for the preparation of many other carbohydrate polymers based FONs with different structure and functions. Therefore, this work opens a new avenue for the preparation and biomedical applications of luminescent carbohydrate polymers.

Abstract:Noble metal materials, especially platinum nanoparticles (Pt NPs), have immense potential in nanomedicine as therapeutic agents on account of their high electron density and their high surface area. Intravenous injection is proposed as the best mode to deliver the product to patients. However, our understanding of the reaction of nanoparticles with blood components, especially proteins, is far behind the explosive development of these agents. Using synchrotron radiation circular dichroism (SRCD), we investigated the structural and stability changes of human serum albumin (HSA) upon interaction with PEG-OH coated Pt NPs at nanomolar concentrations, conditions potentially encountered for intravenous injection. There is no strong complexation found between HSA and Pt NPs. However, for the highest molar ratio of NP:HSA of 1:1, an increase of 18°C in the thermal unfolding of HSA was observed, which is attributed to increased thermal stability of HSA generated by preferential hydration. This work proposes a new and fast method to probe the potential toxicity of nanoparticles intended for clinical use with intravenous injection.

A Novel Fluorescent Label Based on Biological Fluorescent Nanoparticles and Its Application in Cell Recognition

Abstract:Uniform-sized fluorescent nanoparticles have been prepared by employing silica as the shell and a highly luminescent dye complex of ruthenium ion and bipyridyl, tris(2,2 ’-bipyridyl) dichlororuthenium( II) hexahydrate as the core of the nanoparticles. A novel fluorescent label method is proposed, which is based on the biological fluorescent nanoparticles on the foundation of nanotechnology, biotechnology and fluorescent label technology. In comparison with the conventional fluorophores as fluorescent labels such as fluorescein isothiocyanate (FITC) label, this new label shows more superiority in photochemical stability, detection sensitivity and application scope for the biomedicine research. SmIgG+ B lymphocytes isolated from the circulating blood of human beings can be easily recognized by using this new fluorescent label.

Abstract:Novel fluorescent nanoparticle (FNP)-based bioprobes are expected to generate new medical diagnostic techniques in biomedical and biological areas for their superior brightness and photostability compared with conventional molecular probes including small organic dyes and fluorescent proteins. Potentially interesting nanoscale platforms for various biomedical applications are greatly attractive due to the potential to avoid exposure of human tissues to toxic drugs, enhancing delivery of hydrophobic therapeutics and fabricating multifunctional imaging, targeting and delivery system. In this review, recent progress in the area of novel aggregation induced emission (AIE)-based FNPs is summarized over the past few years (2007-2013), and the reported fabrication methodologies of these fluorescent systems including non-covalent and covalent strategies are mainly discussed, and the biomedical applications of AIE-based FNPs are also summarized.
