Abstract:A new fluorescent nanoparticle was synthesized by chemical crosslinking of polyacrylic acid using 4-aminoethanol-N-hydroxyethanyl-1,8-naphthalimide as the crosslinker. The particle possesses excellent characteristics, including low cytotoxicity, water solubility and good fluorescence properties. The interaction of the particle with bovine serum albumin was investigated by absorbance, fluorescence and circular dichroism spectroscopic methods. The interaction mechanisms, binding model and reciprocal effects on structure and fluorescence between nanoparticle and protein are discussed. The spectral data indicated that the nanoparticle could spontaneously form a reversible complex with bovine serum albumin in solution used mainly by hydrogen bonds and van der Waals forces. Fluorescence resonance energy transfer from tryptophan to naphthalimide in the complex was demonstrated and employed to determine their relative separation distance. During the interaction process, unfolding of polypeptide chains in the protein occurred but no adjustments of local polarities around the tryptophan and tyrosine residues were observed. In addition, it was found that the presence of protein induced a notable enhancement in nanoparticle fluorescence and a blue-shift in the emission maximum. These results will be useful for further applications of the developed nanoparticle in biomedical areas.

Abstract:Regarding the importance of the biological effects of nanomaterials, there is still limited knowledge about the binding structure and stability of the protein corona on nanomaterials and the subsequent impacts. Here we designed a hard serum albumin protein corona (BSA) on CTAB-coated gold nanorods (AuNRs) and captured the structure of protein adsorption using synchrotron radiation X-ray absorption spectroscopy, microbeam X-ray fluorescent spectroscopy, and circular dichroism in combination with molecular dynamics simulations. The protein adsorption is attributed to at least 12 Au-S bonds and the stable corona reduced the cytotoxicity of CTAB/AuNRs. These combined strategies using physical, chemical, and biological approaches will improve our understanding of the protective effects of protein coronas against the toxicity of nanomaterials. These findings have shed light on a new strategy for studying interactions between proteins and nanomaterials, and this information will help further guide the rational design of nanomaterials for safe and effective biomedical applications.

Abstract:Understanding protein adsorption on the surface of nanoparticles (NPs) is crucial for determining their behavior in biological environments. Early research in this field faced challenges in producing high-quality NPs. Advancements in NP fabrication now allow for precise modifications of specific parameters, such as zeta potential. However, creating a series of NPs where only one parameter, such as surface charge, is independently varied remains challenging due to concurrent alterations in other properties. In this study, we address these challenges using the ferritin nanocage (Ftn) as a model system for NPs. By modifying only a few amino acids on the outer surface of Ftn, we produce NPs with highly defined properties, focusing solely on variations in surface charge. This approach enables us to generate a controlled series of protein-based nanocages, labeled with fluorophores inside the nanocage. We utilize fluorescent correlation spectroscopy (FCS) to investigate the adsorption of bovine serum albumin (BSA) on these NPs, analyzing the dependence of BSA binding on surface charge. This fundamental study enhances our understanding of the driving forces behind protein adsorption, contributing valuable insights into the design of NPs for biomedical applications.

Abstract:Nanoparticles (NPs) can directly or indirectly enter into the body because of their small size; then they tend to alter the conformation and function of proteins upon interaction with them. Thus, it is crucial to understand the impact of NPs in a biological medium. Recently, niobium pentoxide nanoparticles (Nb2O5 NPs) are finding increasing applications in the biological system, for example, bone tissue and dental material, matrix for biosensing of proteins, etc. In all such applications, the Nb2O5 NP interacts with proteins and other biomolecules. Hence, the study of such interactions is of considerable importance. Here in this work, we present the impact of Nb2O5 NP on the structure, stability and activity of blood proteins, bovine serum albumin (BSA) and human serum albumin (HSA) by means of various spectroscopic approaches. Steady-state fluorescence studies indicated that intrinsic fluorescence intensities of both serum albumin proteins got quenched upon their interaction with NP. The nature of the quenching was elucidated by time-resolved fluorescence and absorption measurements. Using circular dichroism (CD) and synchronous fluorescence spectroscopy (SFS), the structural perturbations of the protein molecules after interaction with NP were investigated. Moreover, the role of temperature on protein stability upon complexation with NP was also explored. In addition, the effect of NP on protein functionality was probed by esterase-like activity assays.Communicated by Ramaswamy H. Sarma.
Core-Shell Nanoparticles Fabricated By Tri-Capillary Coaxial Electrospray And Template Removal Method For Protein Delivery

Abstract:A coaxial tri-capillary electrospray and template removal method was proposed to produce nanosized core-shell particles for encapsulating of proteins. Polyethylene glycol (PEG) and poly (D, L-lactide-co-glycolide) (PLGA) were used as corona and shell materials, while Bovine serum albumin (BSA), as a model protein,was entrapped in the core. Microparticles with smooth surfaces and distinct core-shell-corona structures were obtained by coaxial tri-capillary electrospray and nanoparticles with diameter about 100 nm were achieved by removing the corona PEG template from core-shell-corona microparticles. Moreover, the nanoparticle size could be modulated by adjusting the feed rate of shell fluid, and the diameter of nanoparticles could be reduced to (68 +/- 8) nm when the shell fluid feed rate was decreased to 0.5 mL/h. The drug loading content and entrapment efficiency of BSA were 3. 7% and 78. 3%, respectively. It was also found that BSA released from nanoparticles in a typical bi-phased release profile and the process may last for 60 h. The second order structure of BSA released from particles was monitored with circular dichroism spectrum. The results showed that the electrospray process didn ' t affect the second order structure of proteins, implying that the denaturation of the protein didn ' t occur during the process of electrospraying encapsulation.

Abstract:Serum albumin is a popular macromolecule for studying the effect of proteins on the colloidal stability of nanoparticle (NP) dispersions, as well as the protein–nanoparticle interaction and protein corona formation. In this work, we analyze the specific conformation-dependent phase, redox, and fatty acid delivery properties of bovine albumin in the presence of shungite carbon (ShC) molecular graphenes stabilized in aqueous dispersions in the form of NPs in order to reveal the features of NP bioactivity. The formation of NP complexes with proteins (protein corona around NP) affects the transport properties of albumin for the delivery of fatty acids. Being acceptors of electrons and ligands, ShC NPs are capable of exhibiting both their own biological activity and significantly affecting conformational and phase transformations in protein systems.

Functional Nanoparticles with Magnetic 3D Covalent Organic Framework for the Specific Recognition and Separation of Bovine Serum Albumin

Abstract:Glutathione functionalized magnetic 3D covalent organic frameworks combined with molecularly imprinted polymer (magnetic 3D COF–GSH MIPs) were developed for the selective recognition and separation of bovine serum albumin (BSA). Ultrasonication was used to prepare magnetic 3D COFs with high porosity (~1 nm) and a large surface area (373 m2 g−1). The magnetic 3D COF–GSH MIP nanoparticles had an imprinting factor of 4.79, absorption capacity of 429 mg g−1, magnetic susceptibility of 32 emu g−1, and five adsorption–desorption cycles of stability. The proposed method has the advantages of a shorter equilibrium absorption time (1.5 h), higher magnetic susceptibility (32 emu g−1), and larger imprinting factor (4.79) compared with those reported from other studies. The magnetic 3D COF–GSH MIPs used with BSA had selectivity factors of 3.68, 2.76, and 3.30 for lysozyme, ovalbumin, and cytochrome C, respectively. The successful recognition and separation of BSA in a real sample analysis verified the capability of the magnetic 3D COF–GSH MIP nanoparticles.

Abstract:Nanoparticles (NPs) have been used in numerous applications as anticancer, antibacterial and antioxidant agents. Artificial engineering of protein interactions with NPs in biological systems is crucial to develop potential NPs for drug delivery and cancer nanotherapy. The protein corona (PC) on the NP surface, displays an interface between biomacromolecules and NPs, governing their pharmacokinetics and pharmacodynamics. Upon interaction of proteins with the NPs, their surface features are modified and they can easily be removed from the circulation by the mononuclear phagocytic system (MPS). PC properties heavily depend on the biological microenvironment and NP physicochemical parameters. Based on this context, we have surveyed different approaches that have been used for artificial engineering of the PC composition on NP surfaces. We discussed the effects of NP size, shape, surface modifications (PEGylation, self-peptide, other polymers), and protein pre-coating on the PC properties. Additionally, other factors including protein source and structure, intravenous injection and the subsequent shear flow, plasma protein gradients, temperature and local heat transfer, and washing media were considered in the context of their effects on the PC properties and overall target cellular effects. Moreover, the effects of NP-PC complexes on cancer cells based on cellular interactions, organization of intracellular PC (IPC), targeted drug delivery (TDD) and regulation of burst drug release profile of nanoplatforms, enhanced biocompatibility, and clinical applications were discussed followed by challenges and future perspective of the field. In conclusion, this paper can provide useful information to manipulate PC properties on the NP surface, thus trying to provide a literature survey to shorten their shipping from preclinical to clinical trials and to lay the basis for a personalized PC.

Abstract:Nanotechnology is developing at a rapid pace with promises of a brilliant socio-economic future. The apprehensions of vivid future involvement with nano technology make nanoobjects ubiquitous in the macroscopic world of humans. Nanotechnology helps us to visualize the new mysterious horizons in engineering, sophisticated electronics, environmental remediation, biosensing, and nanomedicine. In all these hotspots, cobalt ferrite (CoFe) nanoparticles (NPs) are outstanding contestants because of their astonishing controllable physicochemical and magnetic properties with ease of synthesis methods. The extensive use of CoFe NPs may result in CoFe NPs easily penetrating the human body unintentionally by ingestion, inhalation, adsorption, etc. and intentionally being instilled into the human body during biomedical diagnostics and treatment. After being housed in the human body, it might induce oxidative stress, cytotoxicity, genotoxicity, inflammation, apoptosis, and developmental, metabolic and hormonal abnormalities. In this review, we compiled the toxicity knowledge of CoFe NPs aimed to provide the safe usage of this breed of nanomaterials.
Comparative functional dynamics studies on the enzyme nano-bio interface

Abstract:Introduction: Biomedical applications of nanoparticles (NPs) as enzyme inhibitors have recently come to light. Oxides of metals native to the physiological environment (eg, Fe, Zn, Mg, etc.) are of particular interest-especially the functional consequences of their enzyme interaction. Materials and methods: Here, Fe2O3, zinc oxide (ZnO), magnesium oxide (MgO) and nickel oxide (NiO) NPs are compared to copper (Cu) and boron carbide (B4C) NPs. The functional impact of NP interaction to the model enzyme luciferase is determined by 2-dimensional fluorescence difference spectroscopy (2-D FDS) and 2-dimensional photoluminescence difference spectroscopy (2-D PLDS). By 2-D FDS analysis, the change in maximal intensity and in 2-D FDS area under the curve (AUC) is in the order Cu~B4C>ZnO>NiO>>Fe2O3>MgO. The induced changes in protein conformation are confirmed by tryptic digests and gel electrophoresis. Results: Analysis of possible trypsin cleavage sites suggest that cleavage mostly occurs in the range of residues 112-155 and 372-439, giving a major 45 kDa band. By 2-D PLDS, it is found that B4C NPs completely ablate bioluminescence, while Cu and Fe2O3 NPs yield a unique bimodal negative decay rate, -7.67×103 and -3.50×101 relative light units respectively. Cu NPs, in particular, give a remarkable 271% change in enzyme activity. Molecular dynamics simulations in water predicted that the surfaces of metal oxide NPs become capped with metal hydroxide groups under physiological conditions, while the surface of B4C becomes populated with boronic acid or borinic acid groups. These predictions are supported by the experimentally determined zeta potential. Thin layer chromatography patterns further support this conception of the NP surfaces, where stabilizing interactions were in the order ionic>polar>non-polar for the series tested. Conclusion: Overall the results suggest that B4C and Cu NP functional dynamics on enzyme biochemistry are unique and should be examined further for potential ramifications on other model, physiological or disease-relevant enzymes.
Insight into the Preformed Albumin Corona on in Vitro and in Vivo Performances of Albumin-Selective Nanoparticles

Abstract:Preformed albumin corona of albumin-nonselective nanoparticles (NPs) is widely exploited to inhibit the unavoidable protein adsorption upon intravenous administration. However, very few studies have concerned the preformed albumin corona of albumin-selective NPs. Herein, we report a novel type of albumin-selective NPs by decorating 6-maleimidocaproyl polyethylene glycol stearate (SA) onto PLGA NPs (SP NPs) surface, taking albumin-nonselective PLGA NPs as control. PLGA NPs and SP NPs were prepared by emulsion-solvent evaporation method and the resultant NPs were in spherical shape with an average diameter around 180 nm. The corresponding albumin-coating PLGA NPs (PLGA@BSA NPs) and albumin-coating SP NPs (SP@BSA NPs) were formulated by incubating SP NPs or PLGA NPs with bovine serum albumin solution, respectively. The impact of albumin corona on particle characteristics, stability, photothermal effect, cytotoxicity, cell uptake, spheroid penetration and pharmacokinetics was investigated. In line with previous findings of preformed albumin coating, PLGA@BSA NPs exhibited higher stability, cytotoxicity, cell internalization and spheroid penetration performances in vitro, and longer blood circulation time in vivo than those of albumin-nonselective PLGA NPs, but albumin-selective SP NPs is capable of achieving a comparable in vitro and in vivo performances with both SP@BSA NPs and PLGA@BSA NPs. Our results demonstrate that SA decorated albumin-selective NPs pave a versatile avenue for optimizing nanoparticulate delivery without preformed albumin corona.
The Effects of Serum Albumin Pre-Adsorption of Nanoparticles on Protein Corona and Membrane Interaction: A Molecular Simulation Study

Abstract:As a platform to deliver imaging and therapeutic agents to targeted sites in vivo , nanoparticles (NPs) have widespread applications in diagnosis and treatment of cancer. However, the poor in vivo delivery efficiency of nanoparticles limits its potential for further application. Once enter the physiological environment, nanoparticles immediately interact with proteins and form protein corona, which changes the physicochemical properties of nanoparticle surface and further affects their transport. In this study, we performed molecular dynamics simulations to study the adsorption mechanism of nanoparticles with various surface modifications and different proteins ( e.g. , human serum albumin, complement protein C3b), and their interactions with cell membrane. The results show that protein human serum albumin prefers to interact with hydrophobic and positively charged nanoparticles, while the protein C3b prefers the hydrophobic and charged nanoparticles. The pre-adsorption of human serum albumin on the nanoparticle surface obviously decreases the interaction of nanoparticle with C3b. Furthermore, the high amount of protein pre-adsorption could decrease the probability of nanoparticle-membrane interaction. These results indicate that appropriate modification of nanoparticles with protein provides nanoparticles with better capability of targeting, which could be used to guide nanoparticle design and improve transport efficiency.

Abstract:Magnetic nanoparticles are now used in magnetic-mediated hyperthermia for tumor treatment.Magnetic Fe3O4 nanoparticles with a mean diameter of 10 ～15nm,in the range of superparamagnetism,are normally prepared by the traditional method of co precipitation from ferrous and ferric electrolyte solutions.This work presents a novel and efficient way to prepare a stable aqueous nanomagnetite dispersion.A stable ferrofluid containing Fe3O4 nanoparticles coated by carboxyl groups was synthesized via improved coprecipitation in the presence of polyacrylic acid.Furthermore,Bovine Serum Albumin(BSA) used as the model targeting molecule was conjugated with magnetic nanoparticles via activation by N-ethyl-N'-(3-dimethylamino-propyl) carbodiimide hydrochloride(EDC) and N-hydroxysuccinimide(NHS).The results indicate that the average diameter of PAA oligomers coated Fe3O4 particles was about 10nm,and had a good dispersion in aqueous phase.In addition,both quantitative analysis and fluorescent microscopy observation confirmed that BSA can be stably conjugated with carboxyl-Fe3O4 particles via EDC and NHS activation.

Abstract:Anisotropic self-assembly of nanoparticles (NPs) stems from the fine-tuning of their surface functionality and NP interaction. Strategies involving ligand interaction, protein interaction, and external stimulus have been developed. However, robust construction of monodispersed magnetic NPs to tens of microns of anisotropically aligned colloidal assembly triggered by adsorbed protein intermolecular interaction is yet to be elucidated. Here, we present the NP-protein interaction, magnetic force, and protein corona intermolecular interaction serially but independently induced path-dependent self-assembly of 100 nm Fe3O4@SiO2 nanocomposites. Dynamic formation of the micron-sized anisotropic magnetic assembly was reproducibly realized in a continuous medium in a controllable manner. Formation of the primary globular clusters upon the unique NP-protein complexes with the help of ions acts as the prerequisite for the anisotropic colloidal assembly, followed by the magnetic force-driven pre-organization and protein intermolecular electrostatic interaction-mediated elongation. The protein concentration rather than the protein original structure plays a more pivotal role in the NP-protein interaction and subsequent colloidal assembly process. Two typical serum proteins fibrinogen and bovine serum albumin enable formation of the anisotropic colloidal assembly but with a different subtle morphology. Furthermore, the obtained micron-sized magnetic colloidal assembly can be dissociated rapidly by adding a negative electrolyte in the medium due to the interference in the NP-protein interaction. However, the self-assembly process can be recycled based on the dissociated colloidal assembly.

Abstract:Recently, small (<5 nm diameter) nanoparticles (NPs) have shown improved in vivo biocompatibility compared to that of larger (>10 nm) NPs. However, the fate of small NPs under physiological conditions is poorly understood and remains unexplored. Here, the long-term aggregation behavior of gold nanoparticles (AuNPs) exposed to serum proteins in a near-physiological setup is studied using continuous photon correlation spectroscopy and computer simulations. It is found that the medium, temperature, and NP concentration affect the aggregation of AuNPs, but the observed aggregates are much smaller than previously reported. Simulations show that a single layer of albumin is deposited on the NP surface, but the properties of the aggregates (size, shape, and internal structure) depend critically on the charge distribution on the proteins, which changes with the conditions of the solution. These results explain the seemingly conflicting data reported in the literature regarding the size of aggregates and the morphology of the albumin corona. The simulations suggest that controlling the concentration of NPs as well as the pH and ionic strength of the solution prior to intravenous administration may help to preserve properties of the functionalized NPs in the bloodstream.
Bovine Serum Albumin as a Versatile Platform for Cancer Imaging and Therapy

Abstract:Background: Due to the good biocompatibility, biodegradability, facile surface functionalization and high water solubility, Bovine serum albumin has gain increasing attention in the nanomedicine. Objective: Despite there are many reviews on albumin based nanoparticles, most of them focus on one aspect of the albumin functionality, e.g., drug delivery, cancer theranostics or half-life extension in vivo. This review aims to comprehensively summary bovine serum albumin as a versatile platform in the applications of cancer imaging and therapy. Methods: We review the extensive applications of bovine serum albumin in drug carrier, surface engineering and biomimetic synthesis for cancer imaging and therapy. Conclusion: Based on the studies reviewed, variety of in vitro and in vivo studies show good performance of bovine serum albumin as the drug carrier, surface modification agent and biomimetic template in cancer imaging and therapy. Nevertheless, there are still some issues to be solved, e.g., the technological parameters for enhancing the drug loading efficiency and controlling drug release, optimizing surface modification process to provide more stable nanoagents, investigation of the biomimetic mechanism, in-depth study of their toxicity, further exploring their bioapplications, etc.
Polycarboxylate functionalized magnetic nanoparticles Fe3O4@SiO2@CS-COOH: Preparation, characterization, and immobilization of bovine serum albumin

Abstract:Magnetic nanoparticles with increasing superparamagnetism and magnetic targeting have found widespread application in fields such as food and medicine. In this study, polycarboxylated magnetic nanoparticles (Fe3O4@SiO2@CS-COOH) were prepared by surface functionalizing iron tetraoxide (Fe3O4) nanoparticles with ethylenediaminetetraacetic acid (EDTA) as a modifier. The appropriate degree of functionalization modification was obtained by adjusting the EDTA concentration and the ratio of cross-linking agents. The prepared magnetic nanoparticles were analyzed with structural and property characterization. The results showed that the Fe3O4@SiO2@CS-COOH magnetic nanoparticles prepared with 4 % EDTA and cross-linking agents at a molar ratio of 3:4 were uniform in particle size, with an average size of roughly 7 nm, and possessed an abundant carboxylate content (310.8064 μmol/g) and a high magnetization intensity (35.05 emu/g). As a model protein, bovine serum albumin (BSA) was immobilized on the surface of magnetic particles. The largest amount of immobilized protein was 500.4376 mg BSA/g at pH 4.0 and no extra salt ions. According to molecular docking simulations, its immobilization was due to the interaction of amino and carboxyl groups at the Fe3O4@SiO2@CS-COOH/BSA interface. Fe3O4@SiO2@CS-COOH possesses a large number of carboxyl groups, strong protein immobilization, and magnetic responsiveness, which may have potential applications in biomedical and food fields.

Abstract:The interaction of human proteins and metal species, both ions and nanoparticles, is poorly understood despite their growing importance. These materials are the by-products of corrosion processes and are of relevance for food and drug manufacturing, nanomedicine, and biomedical implant corrosion. Here, we study the interaction of Cr(III) ions and chromium oxide nanoparticles with bovine serum albumin in physiological conditions. This study combined electrophoretic mobility measurements, spectroscopy, and time-of-flight secondary ion mass spectrometry with principal component analysis. It was determined that neither metal species resulted in global albumin unfolding. The Cr(III) ions interacted strongly with amino acids found in previously discovered metal binding sites, but also were most strongly implicated in the interaction with negatively charged acid residues, suggesting an electrostatic interaction. Bovine serum albumin (BSA) was found to bind to the Cr2O3 nanoparticles in a preferential orientation, due to electrostatic interactions between the positive amino acid residues and the negative chromium oxide nanoparticle surface. These findings ameliorate our understanding of the interaction between trivalent chromium ions and nanoparticles, and biological macromolecules.
