Abstract:With the rising awareness of a healthy lifestyle, natural functional foods have gained much interest as promising alternatives to synthetic functional drugs. Recently, wild Agaricus bisporus (Lange) Sing. Chaidam has been found and artificially cultivated for its thick fresh body and excellent taste, with its antioxidant and anti-hypoxic abilities unknown. In this work, the antioxidant potential of its methanolic, 55% ethanolic, aqueous extracts and crude polysaccharide was evaluated in different systems. The results showed that polysaccharide was the most effective in scavenging ability on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radicals, metal chelating activity and reducing power, with EC50 values of 0.02, 2.79, 1.29, and 1.82 mg/mL, respectively. Therefore, we further studied the anti-hypoxic activity of crude polysaccharide. The results turned out that polysaccharide (300 mg/kg) prolonged the survival time, decreased the blood urea nitrogen and lactic acid content as well as increased the liver glycogen significantly, compared with the blank control and the commercialized product Hongjingtian (p < 0.05). With such excellent activities, we purified the polysaccharide and analyzed its molecular weight (120 kDa) as well as monosaccharide components (glucose, fructose and mannose). This study indicated that wild Agaricus bisporus (Lange) Sing. Chaidam had strong potential to be exploited as an effective natural functional food to relieve oxidative and hypoxia stresses.

Abstract:Three kinds of new water-soluble polysaccharides (FA, FB and FC) were isolated from wild mushroom Agaricus bitorquis (Quél.) Sacc. Chaidam by the classical method "water extraction and alcohol precipitation" and purified by column chromatography. The Mw of FA, FB and FC ranged from 5690 Da to 38,340 Da. The three polysaccharide fractions in the fruiting body were mainly composed of 4 kinds of monosaccharides, including glucose, galactose, mannose, and arabinose, among which glucose and galactose were the major monosaccharides. The FTIR and NMR spectroscopy indicated that the skeleton of three fractions composed of a (1→4)-α-D-glycosidic backbone containing α-D-mannopyranose. In vitro anti-hypoxia activity data showed that three polysaccharide fractions possessed a significant effect on inhibiting PASM cells apoptosis under hypoxia. Among them, FC at the concentration of 200 µg/mL revealed a significant anti-hypoxia effect. These results revealed that the intracellular polysaccharides possessed potent anti-hypoxic activity, which might be related to inhibiting LDH and NADPH oxidase expression and promoting the formation of 5-hydroxytryptamine, dopamine, endothelins, acetylcholine. More importantly, FC showed good performance inducing KV1.5 expression and prohibiting KIR6.2 formation at protein level.
Chemical characterization, in vitro antioxidant, anti-cancer and enzyme inhibition activities of three edible mushroom species

Abstract:This study aimed to investigate HPLC-DAD based chemical compositions, total phenolic compounds, antioxidant, enzyme inhibition and anti-cancer activities of three edible mushrooms: Clavariadelphus truncatus Donk, Craterellus tubaeformis (Fr.) Quél. and Hygrophorus pudorinus (Fr.) Fr. p -Hydroxy benzoic acid was specified as the main compound in C. truncatus methanol extract (67.40 ± 0.18 μg/g), trans -aconitic acid in C. tubaeformis water extract (129.50 ± 0.11 μg/g), fumaric acid in other mushroom extracts (138.80 ± 0.25–203.40 ± 0.36 μg/g). C. truncatus methanol extract (DPPH • : IC 50 : 133.99 ± 0.30 μg/mL; ABTS •+ : IC 50 : 120.95 ± 0.93 μg/mL; CUPRAC: A 0.50 : 108.10 ± 0.42 μg/mL; PRAP: A 0.50 : 116.81 ± 1.18 μg/mL; metal chelating: IC 50 : 143.18 ± 0.22 μg/mL) was the most active in antioxidant activity with the highest level of total phenolic compounds (29.18 ± 0.70 μg GAEs/mg extract). When C. tubaeformis water extract (42.13 ± 0.51%) was the highest active in AChE inhibition activity, C. tubaeformis hexane extract (29.14 ± 0.04%) was the highest active in BChE inhibition activity at 200 μg/mL. All hexane extracts displayed the high inhibition on α-amylase (47.82 ± 1.16–81.06 ± 0.54% inhibition at 0.50 mg/mL) and α-glucosidase (IC 50 : 0.11 ± 0.01–0.13 ± 0.02 mg/mL). C. truncatus water (IC 50 : 36.19 ± 0.95 μg/mL) and C. tubaeformis methanol (IC 50 : 48.26 ± 0.51 μg/mL) extracts indicated the highest anti-cancer activity on HT-29. Also, C. tubaeformis methanol extract (IC 50 : 68.93 ± 0.87 μg/mL) was also found to be best anti-cancer active in HeLa. Also, the significant correlation was observed between the chemical compounds and bioactivities. This research affords a scientific basis for the exploration of novel and natural antioxidant, enzyme inhibition and anti-cancer sources with the chemical contents. All studied mushrooms could be used in food, pharmaceutical, and cosmetic applications.

Abstract:Pueraria lobata , an edible food and medicinal plant, is a rich source of bioactive components. In this study, a polyphenol-rich extract was isolated from P. lobata . Puerarin was identified, and the high antioxidant bioactivity of the P. lobata extract was evaluated using the methods of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS), and hydroxyl free radical scavenging ratio. Additionally, the IC 50 values of DPPH, ABTS, and hydroxyl radical scavenging activities were 50.8, 13.9, and 100.4 μg/ml, respectively. Then, the P. lobata extract was administered to C57Bl/6J mice and confirmed to have a superior effect on enhancing the antioxidant status including improving superoxide dismutase activity, glutathione peroxidase peroxide activity, total antioxidant capacity activity, and malondialdehyde contents in vivo . Furthermore, the P. lobata extract had beneficial and prebiotic effects on the composition and structure of gut microbiota. Results showed that the P. lobata extract significantly increased the abundance of beneficial bacteria, involving Lactobacillaceae and Bacteroidetes , and decreased the abundance of Ruminococcaceae , Prevotellaceae , and Burkholderiaceae . Overall, our results provided a basis for using the P. lobata extract as a promising and potential functional ingredient for the food industry.

Abstract:Phellinus baumii, a fungus that grows on mulberry trees and is used in traditional Chinese medicine, exerts therapeutic effects against various diseases, including cancer. Polyphenols, generally considered to be antioxidants, have antitumor and proapoptotic effects. In this study, we identified the composition of Phellinus baumii polyphenol (PBP) and characterized its 17 chemical components by UPLC-ESI-QTOF-MS. Furthermore, to clarify the potential mechanism of PBP against Lung Cancer Cells, network pharmacology and experimental verification were combined. Molecular docking elucidated the binding conformation and mechanism of the primary active components (Osmundacetone and hispidin) to the core targets CASP3, PARP1 and TP53. In addition, potential molecular mechanisms of PBP predicted by network pharmacology analysis were validated in vitro. PBP significantly inhibited the human lung cancer A549 cells and showed typical apoptotic characteristics, without significant cytotoxicity to normal human embryonic kidney (HEK293) cells. Analysis using flow cytometry and western blot indicated that PBP caused apoptosis, cell cycle arrest, reactive oxygen species (ROS) accumulation, and mitochondrial membrane potential (MMP) depression in A549 cells to exercise its antitumor effects. These results reveal that PBP has great potential for use as an active ingredient for antitumor therapy.

Abstract:BACKGROUND Sanghuang mushrooms are medicinal fungi widely used in Eastern Asia. In this study, antioxidant and anti-inflammatory activities of a novel extracellular polysaccharopeptide (SePSP) was investigated. SePSP was purified from the submerged fermentation broth of a Sanghuang mycelium, Sanghuangporus lonicericola stain CBS17 that was isolated from a wild Sanghuang fruiting body. RESULTS The SePSP was extracted using an ethanol precipitation procedure, followed by a DEAE anion-exchange and size-exclusion chromatography. The mass ratio of the polysaccharide and peptide components in the purified SePSP was approximately 4.87:1. By determining its free radical scavenging abilities using DPPH, the hydroxyl free radical, and the superoxide anion free radical, as well as its total reducing power, SePSP had strong concentration-dependent antioxidant activity in vitro. Further, SePSP effectively alleviated dextran sodium sulfate (DSS)-induced ulcerative colitis (UC) in mice. Administration of 200 mg/kg SePSP by gavage for 7 days prevented body weight loss; significantly reduced the mRNA levels of proinflammatory cytokines including, TNF-α and IL-1β; increased mRNA level of the anti-inflammatory cytokine IL-10 in the colon; and decreased the malondialdehyde concentration from 6.42 to 4.82 μmol/L in the blood in UC mice. CONCLUSION The SePSP had strong concentration-dependent antioxidant activity in vitro and effectively alleviated DSS-induced UC in mice. The in vivo therapeutic efficacy in DSS-induced UC may be mediated through modulating the expression of inflammatory cytokines and inhibiting oxidative stress. The findings provide a scientific rational to utilize bioactive nutraceuticals from Sanghuang mushrooms to develop functional foods for the prevention and treatment of UC. Abstract:BACKGROUND: Lentinus edodes is a medicinal mushroom widely used in Asian countries for protecting people against some types of cancer and other diseases.OBJECTIVE: The objective of the present study was to investigate the direct antiproliferation activity and the antitumor mechanisms of water-extracted polysaccharide (WEP1) purified from L. edodes in H22 cells and H22-bearing mice.DESIGN: The extraction, isolation, purification, and structure determination of the water-soluted L. edodes polysaccharide WEP1 were performed. The growth inhibitory effects of WEP1 on H22 cells and H22-bearing mice were determined by 3-(4,5-Dimethylthiazol-2-Yl)-2,5-Diphenyltetrazolium Bromide (MTT) method and animal studies. Flow cytometry, scanning electron microscopy, and laser scanning confocal microscopy were used to observe the morphological characteristics of apoptotic cells. The levels of intracellular reactive oxygen species (ROS) were detected by flow cytometry using 2',7'-dichlorofluorescein-3',6'-diacetate (DCFH-DA). Western blot was used to determine the expressions of cell cycle proteins and apoptosis-related proteins.RESULTS: Results showed that WEP1 with a molecular weight of 662.1 kDa exhibited direct antiproliferation activity on H22 cells in a dose-dependent manner. In vivo, WEP1 significantly inhibited the growth of tumor at different doses (50, 100, and 200 mg/kg) and the inhibition rates were 28.27, 35.17, and 51.72%, respectively. Furthermore, morphological changes of apoptosis and ROS overproduction were observed in H22 cells by WEP1 treatment. Cell cycle assay and western blot analyses indicated that the apoptosis induction activity of WEP1 was associated with arresting cell cycle at G2/M phase and activating mitochondrial-apoptotic pathway. Besides, WEP1 disrupted the microtubule network accompanied by alteration of cellular morphology.CONCLUSION: Results suggested that the antitumor mechanisms of WEP1 might be related to arresting cell cycle at G2/M phase, inhibiting tubulin polymerization and inducing mitochondrial apoptosis. Therefore, WEP1 possibly could be used as a promising functional food for preventing or treating liver cancer.

Abstract:A carboxymethyl polysaccharide CMP33 (15.23 × 104 Da) was isolated from edible and pharmaceutical mushroom Poria cocos using alkaline extraction followed by DEAE-52 and Saphadex-G200 + Saphadex-G150 column chromatographies. The structure analysis showed that CMP33 was composed of glucosyl residues containing a backbone chain of (1 → 3)-linked glucose residues and side chains of (1 → 6) and (1 → 2)-linked glucose residues, and possessed triple-helix structure. Bioassay results revealed that CMP33 displayed a dose-dependent inhibition on 5 cancer cells (HepG-2, MCF-7, SGC-7901, A549) in the range of 31.25-1000 μg/mL, but low cytotoxicity on normal liver cells L-O2. Moreover, CMP33 stimulated NO release and cytokine secretion (IL-1β, IL-6 and TNF-α), and also inhibited LPS-stimulated overproduction of NO, IL-6, TNF-α and IL-1β, in RAW264.7 cells. These results suggested that CMP33 possessed anticancer, anti-inflammation and immune-stimulation activities, and potential for developing as a bioactive ingredient in functional foods.

Abstract:The immunoglobulin (Ig) protein domain is widespread in nature having a well-recognized role in proteins of the immune system. In this review, we describe the proteins containing Ig-like domains in Escherichia coli and enterobacteria, reporting their structural and functional properties, protein folding, and diverse biological roles. In addition, we cover the expression of heterologous Ig domains in E. coli owing to its biotechnological application for expression and selection of antibody fragments and full-length IgG molecules. Ig-like domains in E. coli and enterobacteria are frequently found in cell surface proteins and fimbrial organelles playing important functions during host cell adhesion and invasion of pathogenic strains, being structural components of pilus and nonpilus fimbrial systems and members of the intimin/invasin family of outer membrane (OM) adhesins. Ig-like domains are also found in periplasmic chaperones and OM usher proteins assembling fimbriae, in oxidoreductases and hydrolytic enzymes, ATP-binding cassette transporters, sugar-binding and metal-resistance proteins. The folding of most E. coli Ig-like domains is assisted by periplasmic chaperones, peptidyl-prolyl cis/trans isomerases and disulfide bond catalysts that also participate in the folding of antibodies expressed in this bacterium. The technologies for expression and selection of recombinant antibodies in E. coli are described along with their biotechnological potential.
Antioxidant and Anti-tumor Activities of Extracts from Lepista irina

Abstract:The dried powder of fruit body of Lepista irina (Fr.) Bigelow was extracted with petroleum ether (PE), ethyl acetate (EA), ethanol (Eth) and polysaccharides were prepared. The yield of various extracts was 7.284% (Eth-portion), 2.284% (PE-portion), 0.858% (EA-portion) and 4.107% (polysaccharides), respectively. The antioxidant and antitumor properties of solvent extracts were studied by free radical scavenging assay and MTT assay. The results showed that all extracts and polysaccharides provided various antioxidant activities against radicals of DPPH and hydroxyl and EA and Eth extracts possessed the highest effect. MTT data suggest the varying antitumor activities of extracts and the EA extract showed the highest antiproliferative effect (82.7%). The polysaccharide followed the second and showed higher antitumor activity equivalent to cisplatin. In summary, the mushroom is a promising and potential source for screening natural antioxidants and antitumor agents.

Abstract:Due to climate change and economic difficulties, food production and supply in the world is getting harder and harder. The importance of edible mushrooms is increasing day by day due to their consumption as food, nutritional value and health benefits. Many types of mushrooms are cultivated to overcome the difficulties of obtaining them naturally. Among the mushrooms cultivated in the world, the Pleurotus species ranks second after Agaricus spp. The aim of the current research was to determine the antimicrobial, antioxidant, cytotoxic and deoxyribonucleic acid protective effects of Pleurotus pulmonarius (Fr.) Quel. grown on some local cellulosic wastes. The antimicrobial activity and the minimum inhibitory concentration (MIC) tests carried out by the disk diffusion method and macro-broth dilution techniques. Antioxidant activity was ascertained by the radical scavenging capacity method of 2.2-diphenyl-1-picrylhydrazil. Total antioxidant status and total oxidant status of P. pulmonarius extracts were determined with rel assay kits. The DNA protective effect was evaluated using plasmid pBR322 DNA treated with ultraviolet (UV) and H 2 O 2 , and also the cytotoxic effects on the human lung cancer cell (A549) and human breast cancer cell (MDA-MB-231) lines were evaluated using the 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay method. It was observed to be highly active against all pathogen bacteria (except S. mutans ) and dermatophytes ( Epidermophyton sp. and Trichophyton sp.) (14.0–29.3 mm) compared to the control group (10.0–32.7 mm), but low against Candida tropicalis . It also showed significant antioxidant potential, the highest TAS (1.33 mmol/L) and TOS (6.74 μmol/L) values were obtained in the mixture of Medicago sativa + Populus nigra (MS-PS (1:1)), on Medicago sativa (MS) medium, and also DPPH radical scavenging effect was more effective at 25 mg (68.09%) on MS-PS (1:1) medium. The methanol extract of P. pulmonarius cultured on MS-PS (1:1) at a concentration of 400 μg/ml significantly reduced the percent viability in MDA-MB-231 cell lines (1.41%). However, it was determined that P. pulmonarius did not have a DNA protective effect. As a result, it was observed that P. pulmonarius had strong antimicrobial, antioxidant and cytotoxic activities, but did not have a DNA protective effect.

Abstract:Early mushrooms were categorized under functional food/ nutritional food. However, later on, an increasing number of scientific studies strongly support their role in disease prevention and suppression or remission of a diseased state. Mycotherapy of cancer, a new promising field, deals with anticancerogenic agents derived from mushrooms. Rich in nutritive value, undemanding cultivation conditions, and an army of health-promoting bioactive molecules, collectively make Pleurotus mushroom ideal for a researcher to investigate its anti-cancer potential. The present review is a literature survey via. electronic search by two database search engines: Science Direct and PubMed, on the anti-cancer potential of Pleurotus species till 2021. This data profiling will -track information regarding the in vitro and in vivo anti-cancer activity of different Pleurotus species, along with identifying the bioactive molecules responsible for anti-cancer potential with their underlying mechanism of action. Resultant findings from the electronic search revealed that till 2021, nearly 13 species of Pleurotus mushroom had been studied for anti-cancer potential. The biomarkers responsible for activity are mostly higher molecular weight molecules, comprising polysaccharides, polypeptides, proteins, lectins, and enzymes. The lower molecular weight molecules like terpenoids, sterols, and phenolic compounds are less explored and untapped. The main underlying mechanism for the anticancer activity of Pleurotus mushrooms is the induction of the mitochondria intrinsic pathway. For anticancer, Pleurotus mushrooms are widely formulated as metal nanoparticles. Altogether, this detailed insight may give the researcher a path to be addressed, thus driving mass attention to identifying novel,effective, and safer anticancer potential biomarkers.

Abstract:Fungi generate different metabolites some of which are intrinsically bioactive and could therefore serve as templates for drug development. In the current study, six endophytic fungi namely Aspergillus flavus, Aspergillus tubigenesis, Aspergillus oryzae, Penicillium oxalicum, Aspergillus niger, and Aspergillus brasiliensis were isolated and identified from the medicinal plant, Silybum marianum. These endophytic fungi were identified through intra transcribed sequence (ITS) gene sequencing. The bioactive potentials of fungal extracts were investigated using several bioassays such as antibacterial activity by well-diffusion, MIC, MBC, anti-biofilm, antioxidant, and haemolysis. The Pseudomonas aeruginosa PAO1 was used to determine the antibiofilm activity. The ethyl acetate extract of Aspergillus flavus showed strong to moderate efficacy against Staphylococcus aureus, Escherichia coli, P. aeruginosa, and Bacillus spizizenii. Aspergillus flavus and Aspergillus brasiliensis exhibited significant antibiofilm activity with IC50 at 4.02 and 3.63 mg/ml, while A. flavus exhibited maximum antioxidant activity of 50.8%. Based on HPLC, LC-MS, and NMR experiments kojic acid (1) and carbamic acid (methylene-4, 1-phenylene) bis-dimethyl ester (2) were identified from A. flavus. Kojic acid exhibited DPPH free radical scavenging activity with an IC50 value of 99.3 μg/ml and moderate activity against ovarian teratocarcinoma (CH1), colon carcinoma (SW480), and non-small cell lung cancer (A549) cell lines. These findings suggest that endophytic fungi are able to produce promising bioactive compounds which deserve further investigation.
Extraction of Natural Antioxidants from the Thelephora ganbajun Mushroom by an Ultrasound-Assisted Extraction Technique and Evaluation of Antiproliferative Activity of the Extract against Human Cancer Cells

Abstract:The Thelephora ganbajun mushroom has been found to be a potential rich source of natural antioxidants. In this study, an ultrasound-assisted extraction (UAE) technique together with GRAS (generally recognized as safe) solvents (ethanol and water) was used to maximize the extraction of antioxidants from Thelephora ganbajun. Five extraction parameters (ethanol concentration, solvent to solid ratio, extraction time, temperature and ultrasound power) were investigated by single-factor experiments, and then a central composite rotatable design was employed to study interaction of three key extraction parameters. The optimum conditions were as follows: 57.38% ethanol, 70.15 mL/g solvent to solid ratio, 10.58 min extraction time, 40 °C extraction temperature and 500 W ultrasound power. Under the optimum conditions, the antioxidant activity obtained was 346.98 ± 12.19 µmol Trolox/g DW, in accordance with the predicted value of 344.67 µmol Trolox/g DW. Comparison of UAE with conventional maceration and Soxhlet extraction, the UAE method showed stronger extract efficiency in a shorter extraction time. These results showed that UAE was an effective technique to extract antioxidants from Thelephora ganbajun. Furthermore, the extracts obtained under the optimized conditions exhibited antiproliferative activities toward human lung (A549), breast (MCF-7), liver (HepG2) and colon (HT-29) cancer cells, especially for liver and lung cancer cells. In addition, rutin, 2-hydrocinnamic acid and epicatechin were identified in the extract, which might contribute to antioxidant and antiproliferative activities.

Abstract:Eight polyprenylphenol derivatives were isolated from the wild edible mushroom Suillus granulatus, including seven novel compounds, named suillin F‒L (2‒8), and one previously identified compound (1). The structures of the new compounds were elucidated using HR-ESI-MS and 1D and 2D NMR data. The absolute configuration of compound 8 were assigned based on the comparison of the experimental and calculated ECD data. All isolated compounds were evaluated for their cytotoxicity against HepG2 cancer cell lines. Compounds 1 and 3‒6 demonstrated significant antitumor activity compared to the positive control (cisplatin), with IC50 values ranging from 8.19 to 13.97 μM. Furthermore, DARTS assay and LC-MS/MS analysis was used to identify HSP90AA1 as the direct target of compound 5, and the interaction between compound 5 and HSP90AA1 were verified by molecular docking.

Abstract:Hericium erinaceus (HE), a widely utilized natural remedy and dietary source, has garnered significant attention for its therapeutic potential in various diseases. In this study, we employed supercritical fluid extraction (SFE) technology to isolate the bioactive compounds from HE's fruiting body. Comprehensive assessments of the antioxidant and antibacterial activities were conducted, along with in vitro investigations on the human colon cancer cell line (HCT-8). The SFE rate served as the evaluation metric, while the variables of extraction time, pressure, and temperature were systematically examined. By integrating the response surface center composite design, we successfully optimized the extraction process, yielding optimal parameters of 80 min, 30 MPa, and 35 °C, thus resulting in an extraction rate of 2.51%. These optimized conditions exhibited considerable antioxidant capacity, anticancer activity, and antibacterial potential. Furthermore, we employed graded alcohol extraction to refine the crude extracts, thereby confirming superior anticancer effects under a 70% alcohol precipitation. To elucidate the composition, Fourier-transform infrared spectroscopy (FT-IR) and gas chromatography-mass spectrometry (GC-MS) were employed to analyze the crude extracts and isolates of HE, facilitating a comparative analysis of six HE varieties. Our findings suggest that sterol derivatives hold promise as the active component against the colon cancer HCT-8 cell line. In conclusion, this study underscores the potential of HE SFE in the development of functional foods or alternative drugs for colon cancer treatment, thus opening new avenues for therapeutic interventions.
