Abstract:A carboxymethyl polysaccharide CMP33 (15.23 × 104 Da) was isolated from edible and pharmaceutical mushroom Poria cocos using alkaline extraction followed by DEAE-52 and Saphadex-G200 + Saphadex-G150 column chromatographies. The structure analysis showed that CMP33 was composed of glucosyl residues containing a backbone chain of (1 → 3)-linked glucose residues and side chains of (1 → 6) and (1 → 2)-linked glucose residues, and possessed triple-helix structure. Bioassay results revealed that CMP33 displayed a dose-dependent inhibition on 5 cancer cells (HepG-2, MCF-7, SGC-7901, A549) in the range of 31.25-1000 μg/mL, but low cytotoxicity on normal liver cells L-O2. Moreover, CMP33 stimulated NO release and cytokine secretion (IL-1β, IL-6 and TNF-α), and also inhibited LPS-stimulated overproduction of NO, IL-6, TNF-α and IL-1β, in RAW264.7 cells. These results suggested that CMP33 possessed anticancer, anti-inflammation and immune-stimulation activities, and potential for developing as a bioactive ingredient in functional foods.

Abstract:In the last three decades, numerous polysaccharides and polysaccharide-protein complexes have been isolated from mushrooms and used as a source of therapeutic agents. The most promising biopharmacological activities of these biopolymers are their immunomodulation and anti-cancer effects. They are mainly present as glucans with different types of glycosidic linkages such as (1-3), (1-6)-b-glucans and (1-3)-a-glucans, and as true herteroglycans, while others mostly bind to protein residues as polysaccharide-protein complexes. Three antitumor mushroom polysaccharides, i.e. lentinan, schizophyllan and protein-bound polysaccharide (PSK, Krestin), isolated respectively, from Lentinus edodes, Schizophyllum commune and Coriolus versicolor, have become large market items in Japan. Lentinan and schizophyllan are pure b-glucans, whereas PSK is a protein-bound b-glucan. A polysaccharide peptide (PSP), isolated from a strain of Coriolus versicolor in China, has also been widely used as an anti-cancer and immunomodulatory agent. Although the mechansim of their antitumor action is still not completely clear, these polysaccharides and polysaccharide-protein complexes are suggested to enhance cell-mediated immune responses in vivo and in vitro and act as biological response modifiers. Potentiation of the host defense system may result in the activation of many kinds of immune cells that are vitally important for the maintenance of homeostasis. Polysaccharides or polysaccharide-protein complexes are considered as multi-cytokine inducers that are able to induce gene expression of vaious immunomodulatory cytokines and cytokine receptors. Some interesting studies focus on investigation of the relationship between their structure and antitumor activity, elucidation of their antitumor mechanism at the molecular level, and improvement of their various biological activities by chemical modifications.

Abstract:Poria cocos polysaccharides (PCPs) are fungal polysaccharides derived from the traditional Chinese medicine Poria cocos . They are considered an important active ingredient for their pharmacological activity. Herein, the extraction, separation and purification, structure, and application of PCPs are reviewed. Additional research is necessary to fully understand the advanced structure of PCPs, which has implications for their structure–activity relationship. Their application mostly involves the medical industry, with less involvement in other fields. This article highlights the current research status on PCPs in the above-mentioned areas and some problems that need to be solved in future research. Additionally, it points the way for further studies on PCPs in the hopes that they will be more widely and realistically used in various industries.

Abstract:The activity of polysaccharides is usually related to molecular weight. The molecular weight of polysaccharides is critical to their immunological effect in cancer therapy. Herein, the Codonopsis polysaccharides of different molecular weights were isolated using ultrafiltration membranes of 60- and 100-wDa molecular weight cut-off to determine the relationship between molecular weight and antitumor activities. First, three water-soluble polysaccharides CPPS-I ( 100 wDa) from Codonopsis were isolated and purified using a combination of macroporous adsorption resin chromatography and ultrafiltration. Their structural characteristics were determined through chemical derivatization, GPC, HPLC, FT–IR, and NMR techniques. In vitro experiments indicated that all Codonopsis polysaccharides exhibited significant antitumor activities, with the tumor inhibition rate in the following order: CPPS-II > CPPS-I > CPPS-III. The treatment of CPPS-II exhibited the highest inhibition rate at a high concentration among all groups, which was almost as efficient as that of the DOX·HCL (10 μg/mL) group at 125 μg/mL concentration. Notably, CPPS-II demonstrated the ability to enhance NO secretion and the antitumor ability of macrophages relative to the other two groups of polysaccharides. Finally, in vivo experiments revealed that CPPS-II increased the M1/M2 ratio in immune system regulation and that the tumor inhibition effect of CPPS-II + DOX was superior to that of DOX monotherapy, implying that CPPS-II + DOX played a synergistic role in regulating the immune system function and the direct tumor-killing ability of DOX. Therefore, CPPS-II is expected to be applied as an effective cancer treatment or adjuvant therapy.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Poria cocos F.A. Wolf is an edible fungus with forming sclerotia, which has the effects of promoting diuresis, exuding dampness, invigorating the spleen, and regulating the stomach. P. cocos has a high application in the clinic of traditional Chinese medicine, and some studies have indicated that P. cocos has a good effect on tumor diseases. According to ancient records and modern studies, P. cocos wine offers beneficial effects in terms of strengthening tendons and bones and anti-tumor effects.AIM OF THE STUDY: To understand the substance composition of P. cocos ethanol-soluble extract (PESE) and then further study the effect and potential mechanism of PESE components on gastric cancer.MATERIALS AND METHODS: In vitro and in vivo experiments were performed to detect the cell activity and apoptotic condition. Differential expression analysis and pathway enrichment were performed based on transcriptomics and were verified by real-time polymerase chain reaction and western blotting. The mice of the stomach cancer tumor model were randomly categorized into three groups. The weight and tumor volume of the mice were measured, and the pathological characteristics of tumor tissue and immunohistochemical changes were determined. Then, the main active components of PESE were detected by MKN45 cell fishing.RESULTS: In vitro experiments showed that PESE inhibited the proliferation of MKN45 cells, but it did not induce apoptosis. Based on the transcriptome and western blotting results, the inhibition of MKN45 proliferation by PESE may be influenced by mitogen-activated protein kinase (MAPK) and phosphoinositide-3-kinase-protein kinase B (PI3K-Akt) signaling pathways. In vivo experiments showed that PESE inhibited tumor growth in mice and caused partial necrosis of tumor cells but had no toxic effect on mice. Cell fishing identified nine triterpenoids of P. cocos as the major active components of PESE.CONCLUSIONS: The results indicated that PESE has a significant inhibitory effect on stomach cancer, and its mechanism probably commonly affects the MAPK and PI3K-Akt signaling pathways, which could be due to the triterpenoid components.

Abstract:Poria cocos (PC) refers to a fungal species which is also known as "Fuling" in China. For >2000 years, PC has demonstrated its therapeutic values as a kind of traditional medicine. It is believed that the various biological benefits created by PCs highly rely on the Poria cocos polysaccharide (PCP). This review recapitulates the recent progress made in PCP in four aspects: i) the methods of extraction, separation, and purification, ii) structural characterization and identification, iii) the related bioactivities and mechanism of action, and iv) structure-activity relationships. Through discussion about the objective as mentioned above, it can be found out that PCP is categorized into water-soluble polysaccharide (WPCP) and alkaline-soluble polysaccharide (APCP), which are totally different in structure and bioactivity. The structures of WPCP are multiplicity whose backbone can be (1,6)-α-galactan and (1,3)-β-mannoglucan etc. to perform various bioactivities including anti-tumor effect, anti-depressant effect, anti-Alzheimer effect, anti-atherosclerosis effect, hepatoprotection etc. The structures of APCP are much more single with backbone of (1,3)-β-D-glucan and the studies of activity concentrate on anti-tumor effect, anti-inflammatory effect and immunomodulation. Besides, the future opportunities of WPCP are primary structure identification. For APCP, scholars can focus on the conformation of polysaccharide and its relationship with activity.

Poria Acid, Triterpenoids Extracted from Poria cocos, Inhibits the Invasion and Metastasis of Gastric Cancer Cells

Abstract:Background: Poria cocos (P. cocos) is an important medicinal fungus in traditional Chinese medicine. Poria acid (PA), a triterpenoid compound, is an effective component of traditional Chinese medicine P. cocos. This experiment investigated the anti-gastric cancer biological activity of PA in vitro. Methods: The effect of PA on the viability of gastric cancer cells was detected by the thiazolyl blue (MTT) assay. Cell adhesion assays were used to detect changes in the adhesion of cells treated after PA (0, 20, 40, and 80 μmol/L). The ability of cell invasion and migration were detected by Transwell assays and wound healing assays. A high-content imaging system was used to dynamically record the motility of the gastric cancer cells after PA (0, 20, 40, and 80 μmol/L) treatment. Western blotting was used to detect the expression of epithelial–mesenchymal transformation (EMT), invasion and migration related proteins. Results: The MTT assay showed that the proliferation of gastric cancer cells was significantly inhibited after PA treatment. Cell adhesion experiments showed that the adhesion of gastric cancer cells was significantly decreased after PA treatment. Compared with the control group, the wound healing area of the gastric cancer cells treated with different concentrations of PA decreased. The Transwell assay showed that the number of gastric cancer cells passing through the cell membrane were significantly reduced after PA treatment. In addition, after PA treatment, the cells' movement distance and average movement speed were significantly lower than those of the control group. Finally, PA can significantly alter the expression of EMT-related proteins E-cadherin, N-cadherin, and Vimentin and decreased the expressions of metastasis-related proteins matrix metalloproteinase (MMP) 2, MMP-9 and tissue inhibition of matrix metalloproteinase (TIMP)1 in the gastric cancer cells. Conclusions: Triterpenoids from P. cocos have significant biological activity against gastric cancer, and the mechanism may be involved in the process of epithelial–mesenchymal transformation.

Death and Intracellular Degradation of Mycobacterium leprae after Exposure in vitro to Enzymic Free-Radical Generators

Abstract:An increase in the resistance to sulphones and the prohibitive cost of rifampicin necessitate the development of new chemotherapeutic agents against leprosy. The 02dependent myeloperoxidase-mediated bactericidal system of Klebanoff ( 1975) and analogous free-radical and/or singlet-02 generators are suitable as first-choice candidates for this purpose. Such generators proved highly and readily effective in vitro against a variety of infectious agents (Klebanoff, 1975) including, among intralysosomal parasites, the three mycobacteria causing tuberculosis in man (Demoulin-Brahy et al., 1975) and Mycobacterium lepraemurium (R. J. W. Rees & P. J. Jacques, unpublished work; Avila et al., 1976). The approach and techniques were essentially those previously described (Delville et a[., 1976). Fresh suspensions of M . leprae were exposed in vitro at pH5 and 36°C in aerobic conditions to a mixture of fungal glucose oxidase, glucose, plant peroxidase and iodide or to a similar mixture lacking peroxidase and iodide, for either a few seconds or 1 h. The controls were incubated for 1 h in the same conditions, except that iodide and both enzymes were omitted. After washing the microbial preparation in the cold and resuspension, hind foot-pads of normal mice were inoculated with this resuspended preparation. At various times, ranging from 180 to 424 days after inoculation, the foot-pads were collected and processed for enumeration of total acidfast bacteria and for differentiation between granular and solid-stained bacteria (Shepard, 1960; Rees & Valentine, 1962). As shown in Table 1 , contact in vitro of M . leprae with the complete oxidase mixture for only a few seconds resulted in a considerable decrease of the number of solid-stained (live) and granular (dead) mycobacteria that could be observed in mouse foot-pads up to 14 months after the treated microbial suspension had been transferred to that growth system in vivo. Since not a single acid-fast bacterium could be detected when treatment with the complete oxidase mixture had been applied for 1 h , we conclude that a true bactericidal action of the oxidative ‘cocktail’ has been observed. Abstract:The aim of this study is to evaluate the air pollution due to smoking in an office where no smoking control measures have been adopted. The concentration of suspended particulates (SP) at 8 points in this office were measured and the change of SP concentrations at 3 points were recorded for 24 hours. The results show that the main source of SP is tobacco smoke in the office. Smoking control measures should be promoted in such offices where smokers and non-smokers work together. The effectiveness of a ventilation system and practical smoking control measures are discussed.

Abstract:Polyporaceae is an important fungal family that has been a source of natural products with a range of pharmaceutical activities in China. In our previous study, two polysaccharides, PCWPW and PCWPS, with significant antioxidant and antidepressant activity were obtained from Poria cocos. In this study, we evaluated their potential molecular mechanisms in the immunomodulation of macrophages. PCWPW and PCWPS were characterized by GC–MS analysis to contain 1,3-linked Glcp. ELISA assays results demonstrated that the secretion of TNF-α was significantly enhanced by PCWPW/PCWPS. RNA-seq data demonstrated that PCWPS treatment modulated the expression of immune-related genes in macrophages, which was further confirmed by RT-qPCR assays. The activation of TNF-α secretion was found to be mannose receptor (MR) dependent and suppressed by MR inhibitor pretreatment. Moreover, the amount of TNF-α cytokine secretion in PCWPW/PCWPS-induced RAW264.7 cells was decreased when pretreated with NF-κB or MAPK signaling pathway inhibitors. Collectively, our results suggested that PCWPW and PCWPS possessed immunomodulatory activity that regulates TNF-α expression through the NF-κB/MAPK signaling pathway by binding to mannose receptors. Therefore, PCWPW and PCWPS isolated from Poria cocos have potential as drug candidates for immune-related disease treatment.

Abstract:Polygonatum sibiricum is one of the most widely used traditional Chinese medicine in China. Polygonatum sibiricum polysaccharide (PSP) is the main functional component of Polygonatum sibiricum . In this study, a water-soluble polysaccharide (PSP-1) was first isolated from Polygonatum sibiricum with a molecular weight of 38.65 kDa. Structural analysis was performed via methylation and FT-IR spectroscopy analyses, which in combination with NMR spectroscopy, revealed that PSP-1 has a → 4-α-D-Glcp-1 → backbone with the substitution at O-6 with the β-D-Glcp-1 → residues. Furthermore, PSP-1 exhibited potent and concentration-dependent anticancer effects, inducing HepG2 cell apoptosis and arresting the cell cycle at the G1 phase. Moreover, PSP-1 also decreased the mitochondrial membrane potential, damaged the nucleus of HepG2 cells, and increased the activity of caspase-9 and−3 in the intrinsic apoptotic pathways to induce HepG2 cell apoptosis. To conclude, PSP-1 might be a good candidate for the treatment of liver cancer, and this work provides important information for understanding the relationship between structure and antitumor activity of PSP-1, which is relevant for the treatment of hepatocellular carcinoma in clinic.

Abstract:Poria cocos is the dried sclerotium of Wolfiporia cocos (F.A. Wolf) Ryvarden & Gilb., which was the current accepted name and was formerly known as Macrohyporia cocos (Schwein.) I. Johans. & Ryvarden, Pachyma cocos (Schwein.) Fr., Poria cocos F.A. Wolf and Sclerotium cocos Schwein. It is one of the most important crude drugs in traditional Chinese medicine, with a wide range of applications in ameliorating phlegm and edema, relieving nephrosis and chronic gastritis and improving uneasiness of minds. Its extensive pharmacological effects have attracted considerable attention in recent years. However, there is no systematic review focusing on the chemical compounds and pharmacological activities of Poria cocos. Therefore, this review aimed to provide the latest information on the chemical compounds and pharmacological effects of Poria cocos, exploring the therapeutic potential of these compounds. We obtained the information of Poria cocos from electronic databases such as SCI finder, PubMed, Web of Science, CNKI, WanFang DATA and Google Scholar. Up to now, two main active ingredients, triterpenes and polysaccharides of Poria cocos, have been identified from Poria cocos. It has been reported that they have pharmacological effects on anti-tumor, anti-bacterial, anti-oxidant, anti-inflammatory, immunomodulation, and liver and kidney protection. The review summarizes the phytochemistry and pharmacological properties of Poria cocos, which suggest that researchers should focus on the development of new drugs about Poria cocos to make them exert greater therapeutic potential.
The development of a passive dosimeter for airborne aniline vapors.

Abstract:A passive diffusion dosimeter has been suggested as an economical and accurate means of sampling airborne concentrations of gases and vapors in the working environment. The dosimeter utilizes molecular diffusion through a tube of known geometry to obtain a quantitative determination of airborne concentrations of the contaminant of interest. A passive sampling dosimeter was designed to measure concentrations of aniline vapor in air. Diffusion tubes of 1.5, 3.0, and 4.5 cm lengths were tested under controlled conditions of relative humidity, air temperature, and vapor concentrations. The results indicate that the use of a passive diffusion dosimeter for determining time-weighted average concentrations of aniline is feasible. It was shown that a 1 cm diameter 3.0 cm length diffusion tube gave the most consistent results. It is suggested that further investigations be conducted in the development of passive diffusion dosimeters for other gaseous contaminants.
Chemotaxigenesis and activation of the alternative complement pathway by encapsulated and non-encapsulated Cryptococcus neoformans

Abstract:In the presence of serum, whole cells of encapsulated and non-encapsulated Cryptococcus neoformans generated a chemotactic response by neutrophils. Heat inactivation of serum ablated all chemotactic activity. Cryptococcal polysaccharide was not chemotaxigenic. Assays for alternative complement pathway activation such as depletion of alternative complement pathway factor B or electrophoretic conversion of factor B closely paralleled chemotaxis assays. Cells of encapsulated and non-encapsulated C. neoformans activated the alternative complement pathway, whereas cryptococcal polysaccharide was inactive. Failure of the capsular material to activate the alternative pathway was not due to serotype specificity because polysaccharide of several serotypes failed to achieve activation. The results suggest that chemotaxigenesis and alternative complement pathway activation are functions of the yeast cell wall. The results support our proposal that the cryptococcal capsul does not prevent potential opsonins from reaching binding and activation sites at the yeast cell wall or the release of biologically active soluble cleavage products into the surrounding medium; however, cell wall-bound cleavage products remain bound to the cell wall beneath the capsule. Therefore, they are unable to participate as opsonins in phagocytosis.
