Abstract:Polygonatum sibiricum is one of the most widely used traditional Chinese medicine in China. Polygonatum sibiricum polysaccharide (PSP) is the main functional component of Polygonatum sibiricum . In this study, a water-soluble polysaccharide (PSP-1) was first isolated from Polygonatum sibiricum with a molecular weight of 38.65 kDa. Structural analysis was performed via methylation and FT-IR spectroscopy analyses, which in combination with NMR spectroscopy, revealed that PSP-1 has a → 4-α-D-Glcp-1 → backbone with the substitution at O-6 with the β-D-Glcp-1 → residues. Furthermore, PSP-1 exhibited potent and concentration-dependent anticancer effects, inducing HepG2 cell apoptosis and arresting the cell cycle at the G1 phase. Moreover, PSP-1 also decreased the mitochondrial membrane potential, damaged the nucleus of HepG2 cells, and increased the activity of caspase-9 and−3 in the intrinsic apoptotic pathways to induce HepG2 cell apoptosis. To conclude, PSP-1 might be a good candidate for the treatment of liver cancer, and this work provides important information for understanding the relationship between structure and antitumor activity of PSP-1, which is relevant for the treatment of hepatocellular carcinoma in clinic.

Abstract:Traditional Chinese Medicine (TCM) has been widely used in China and is regarded as the most important therapeutic. <i>Polygonatum sibiricum</i> (PS), a natural plant used in traditional Chinese medicine, has various functions associated with a number of its components. There are many compositions in PS including polysaccharides, steroids, anthraquinone, alkaloids, cardiac glycosides, lignin, vitamins, various acids, and so on. Of these, polysaccharides play a significant role in PS-based therapeutics. This article summarizes <i>Polygonatum sibiricum</i> polysaccharides (PSP) have many pharmacological applications and biological activities, such as their antioxidant activity, anti-aging activity, an anti-fatigue effect, immunity enhancement effect, antibacterial effect, anti-inflammatory effect, hypolipidemic and antiatherosclerotic effects, anti-osteoporosis effect, liver protection, treatment of diabetes mellitus (DM), anti-cancer effect, and may help prevent Alzheimer's disease, and so on. This review summarized the extraction method, purification method, compositions, pharmacological applications, biological activities, biosynthesis, and prospects of PSP, providing a basis for further study of PS and PSP.

Abstract:Polygonatum sibiricum contains copious polysaccharides that have pharmacological effects, including antioxidant, immune regulation, anti‐tumor, hypoglycemic activities. The mechanism of action has been influenced on structure‐activity relation. Polygonatum sibiricum is a medicinal and edible homologous plant of Polygonatum in Liliaceae, which contains many components, such as polysaccharides, proteins, alkaloids, lignans, and amino acids. A large number of studies have shown that Polygonatum sibiricum polysaccharide is the main active component of Polygonatum sibiricum, which has pharmacological effects, such as antioxidant, immune regulation, antitumor, hypoglycemic, and so on. In this article, the research on bioactivity and mechanism of Polygonatum sibiricum polysaccharide in recent 10 years is summarized, and the effect of structure–activity relationship of Polygonatum sibiricum polysaccharide on the mechanism of action is discussed. The research and application prospect of Polygonatum sibiricum polysaccharide in order is prospected to provide reference for the development of related drugs and healthy food.
Potential antitumor effect of polysaccharides extracted from Polygonatum sibiricum on human prostate cancer PC‐3 cells

Abstract:Polygonatum sibiricum polysaccharides (PSP) are a traditional herbal medicine component with potential therapeutic effects on several diseases. The present study aimed to assess the role of PSP in the treatment of human prostate cancer using a PC‐3 cell line by Cell CK‐8, transwell and wound healing assays, then elucidate the potential underlying mechanisms by western blot and quantitative Real‐time RT‐PCR. Different concentrations of PSP were applied to PC‐3 cells, and the proliferation, invasion and migration of PC‐3 cells were demonstrated to be significantly inhibited with increasing concentrations of PSP. Additionally, cell apoptosis rate and expression of caspase‐3 increased with higher PSP concentrations, and the cell cycle was arrested in the S phase. Furthermore, it was demonstrated that the expression of the multidrug resistance‐1 gene and its encoded protein P‐glycoprotein in PC‐3 cells decreased following PSP treatment, suggesting that PSP may have the potential to reverse multidrug resistance in PC‐3 cells. The present study also evaluated the possible mechanism of PSP action on PC‐3 cells. The results revealed that phosphorylated P65, PI3K and AKT decreased in a concentration‐dependent manner. As key molecules in the NF‐κB and PI3K/Akt signaling pathways, this finding suggests that the potential mechanism of the effect of PSP on prostate cancer cells may involve simultaneous mediation of the PI3K/Akt and NF‐κB signaling pathways. The present study demonstrated that PSP inhibit the proliferation, invasion and migration of PC‐3 cells in vitro , as well as reverse MDR in these cells. The underlying mechanism may involve the simultaneous regulation of the PI3K/Akt and NF‐κB signaling pathways.

Abstract:Plants serve as an important source of medicinal compounds, and their use in various diseases dates back hundreds of years. One such plant-based compound and the discovery of the antimalarial drug, artemisinin, has significantly brought phytomedicines into the spotlight and has enhanced the understanding and, consequently, the application of formulations derived from plants. Traditional Chinese Medicine (TCM), which is based primarily on plants, holds immense potential and is an unexplored source for modern medicine. Many herbs, including Polygonatum sibiricum (PS), are used in Chinese medicine to treat various diseases. Polygonatum sibiricum (PS) belongs to the Liliaceae family and is used not only as a medicinal but also dietary supplement. PS has a highly diverse composition of bioactive compounds such as flavones, homoisoflavanone, alkaloids, lignins, steroid saponins, triterpenoid saponins, polysaccharides, etc. Because of such diverse composition, PS has been used as an anti-aging, anti-inflammatory, anti-osteoporotic agent, as well as an immunity booster, sleep enhancer, etc. Therefore, in this article, we review the therapeutic effect of bioactive compounds such as polysaccharides, saponins, and PS extract in various diseases and their biological activities in fatigue, immunity, sleep, anti-aging, etc.

Abstract:Polysaccharide is one of the main active components of Polygonatum sibiricum. For this study, P. sibiricum polysaccharides (PSP) were obtained through purification using DEAE-cellulose52 and Sephacryl G-150 column chromatography. The obtained samples were named PSP1, PSP2 and PSP3. The PSP1 sample was found to have the highest content and the best solubility, and a subsequent. So, its structure and characterization were analyzed. The main sugar residue linkages were found to be □ → 1)-β-D-Fruf-(2 → 1)-β-D-Fruf-(2 → 1), 1 → -β-D-Fruf-(2 → 6)α-D Glcp (1→, →4)-β-D-Manp-(1 → 4)-β-D-Manp-(1→ and →6)-β-D-Glcp-(1 → 4)-β- D-Manp -(1→ link existed. Branch chain analysis indicated →1,6)-β-D-Fruf-(2→, β-D-Fruf-(2→, →1,6)-β-D-Fruf-(2→, →6)-β-D- Fruf-(2→ link existed, and the link site was at position C-6. In vitro antioxidant activity tests showed that PSP1 had a certain scavenging effect on DPPH, hydroxyl radical, superoxide anion radical and a particular effect on the chelating ability of ferrous. This suggested that P. sibiricum polysaccharides may be a potential antioxidant.

Abstract:Ethnopharmacological relevance Polygonatum sibiricum , commonly known as Siberian Solomon's seal, is a traditional herb widely used in various traditional medical systems, especially in East Asia. In ancient China, the use of polygonatum sibiricum in medicine and food was mentioned in Li Shizhen's Bencao Gangmu of traditional Chinese medicine (TCM). It was also used in history of India in Vedic medicine. The plant is rich in bioactive substances such as polysaccharides, saponins, flavonoid and alkaloids. Aim of the review The aim of this review is to understand the pharmacological and pharmacokinetics research progress of the major components of polygonatum sibiricum , and to prospect its potential application and development in the treatment of various diseases. Materials and methods We conducted a systematic literature search against major online databases on the Web, including PubMed, ancient books, patents, PubMed, Wiley, Google Scholar, Web of Science, and others. We select the pharmacological process and mechanism of the main components of polygonatum sibiricum in a variety of diseases, and make a strict but careful supplement and in-depth elaboration to this review. Results Several studies have demonstrated the strong antioxidant properties of polygonatum extract, which can be attributed to the presence of flavonoids and other polyphenol compounds; for diabetes and other metabolic-related diseases, polygonatum saponins have particular advantages in regulating intestinal flora and lipoprotein concentration in organisms. In addition, the polysaccharides extracted from this plant have a strong anti-inflammatory effect, which is related to its ability to regulate proinflammatory cytokine and mediators. In the aspect of anti-tumor effect, polygonatum derivatives can induce cancer cell apoptosis mainly by adjusting the cell membrane potential and cell cycle. It is worth noting that the combined action of the main components of polygonatum also offers promising solutions for the treatment of the disease. Conclusion Polygonatum polysaccharide has therapeutic effects on many diseases by adjusting cell signal pathways, polygonatum sibiricum have significant advantages in regulating intestinal flora, inducing apoptosis of tumor cells, activating antioxidant processes, etc. Further research and basic exploration are needed to prove the function and mechanisms of the main components of polygonatum sibiricum on related diseases. The study on the immunomodulatory properties of polygonatum revealed its potentiality of enhancing immune function, which made it an interesting subject for further exploration in the field of immunotherapy.

Abstract:Four polysaccharides (AEPA, AEPB, AEPC and AEPD) isolated and purified from the roots of Actinidia eriantha using gel filtration were subjected to composition analysis and valuated for the antitumor and immunomodulatory activity. The predominant neutral monosaccharides in four fractions were identified as galactose, arabinose and fucose, while the composition and ratio of the monosaccharides were different from one another. All four polysaccharides could significantly not only inhibit the growth of transplantable S180 sarcoma in mice, but promote splenocytes proliferation, natural killer cells activity, interleukin-2 production from splenocytes and serum tumor antigen-specific antibody levels in tumor-bearing mice, indicating that their antitumor activity might be achieved by improving immune response. Among four polysaccharides, AEPC and AEPD showed the higher antitumor and immunomodulatory activity. Taken together, the chemical composition of these polysaccharides could affect their antitumor and immunomodulatory activity, and AEPC and AEPD could be explored as antitumor agent with immunomodulatory activity.
Antitumor Effects and Related Mechanisms of Ethyl Acetate Extracts of Polygonum Perfoliatum L.

Abstract:Polygonum perfoliatum L. belongs to the genus Polygonaceae and has a long history to be used as a Chinese medicinal herb to reduce swelling, control body temperature, and promote detoxification. However, its anticancer activity and mechanisms of action have not been evaluated yet. In the present study, we used several cell lines and xenograft models from different cancers to demonstrate the broad-spectrum anticancer activity of P. perfoliatum L as well as its underlying mechanisms of action in vitro and in vivo. The ethyl acetate extract of P. perfoliatum L showed good anticancer activity and was further fractioned to obtain five active components, including PEA to PEE. Among these fractions, PEC showed the strongest cytotoxicities against various cancer cell lines. It was further observed that PEC inhibited cancer cell growth, arrested cells at G2 phase, and induced apoptosis in vitro and suppressed tumor growth and angiogenesis in vivo in a dose- and time-dependent manner. Furthermore, PEC decreased the expression of vascular endothelial growth factor (VEGF) and micro-vascular density (MVD) in tumor tissues in vivo. It also promoted the proliferation of T and B lymphocytes, increased the activities of natural killer (NK) cells and cytotoxic T lymphocytes (CTLs), enhanced the secretion of interleukin 2 (IL-2) by spleen cells, and raised the levels of IgG, IgG2a, and IgG2b antibodies in tumor-bearing mice in vivo, which were at least partially responsible for the anticancer activity of PEC. In summary, PEC has shown broad-spectrum anticancer activities without causing any host toxicity in vitro and in vivo and may be developed as a preventive and therapeutic agent against human cancer. Further studies are urgently needed to determine the anticancer compounds in PEC and their detailed molecular mechanisms.
Structural characterization and hypoglycemic effect of polysaccharides of Polygonatum sibiricum

Abstract:Polygonatum sibiricum polysaccharide (PSP) was extracted and purified from raw material obtained from P. sibiricum. The structural features of PSP were investigated by Congo red, circular dichroism spectrum, high-performance gel permeation chromatography, scanning electron microscope, atomic force microscope, ultraviolet spectroscopy, and Fourier transform infrared spectroscopy analysis. In vitro simulations were conducted to investigate the kinetics of PSP enzyme inhibition. Moreover, a type II diabetes mouse model (T2DM) with streptozotocin-induced insulin resistance was established, and the indexes of lipid quadruple, insulin resistance index, oral glucose tolerance (OGTT), organ index, and pancreatic morphology of model mice were measured. The results showed that PSP mainly consists of monosaccharides, such as mannose, glucose, galactose, xylose, and arabinose. It also has a β-glycosidic bond of a pyranose ring and an irregular reticulated aggregated structure with a triple helix. In vitro enzyme inhibition assays revealed that PSP acts as a reversible competitive inhibitor of α-glucosidase and α-amylase. Furthermore, PSP was found to reduce insulin resistance index, increase OGTT and serum insulin levels, decrease free fatty acid content to improve lipid metabolism, and lower glycated serum protein content to enhance glucose metabolism in T2DM mice, thereby leading to a reduction in blood glucose concentration. Additionally, PSP exhibited reparative effects on the damaged liver tissue cells and pancreatic tissue in T2DM mice. The experiment results provide a preliminary basis for the therapeutic mechanism of PSP about type II diabetes and a theoretical reference for application in food and pharmaceutical development.

Abstract:Polysaccharides extracted from Asian traditional food source have been demonstrated to possess different antitumor activities mostly without side effect. In this paper, we reviewed many kinds of polysaccharides from different Asian food source and their antitumor activities. Some are common food such as different mushroom with more research. Some are special e.g., Ginseng, Salvia, Astragalus, Lycium barbarum etc. with relatively fewer research. This review mainly focused on their structure, derivatives, antitumor activities and their mechanism of action in the last decades. It aimed to bridge traditional Asian ingredients with tumor and cancer curation in order to avoid side effect of traditional treatment. Practical applicationsThere are abundant resources of Asian food. And polysaccharides from these resources have been showed good antitumor activities and immunopotentiating activity. This review introduced the advance of the polysaccharides and their antitumor activities, which will promote the development antitumor medicine derived from Asian food source, or their applications as Adjuvant therapy of traditional chemotherapy and radiotherapy. Due to their multiple antitumor activities, enhancing immunity potential, and non-toxic side-effects, it might be utilized for the treatment of multiple tumors and improve the health and the life quality of patients whether as anti-tumor drugs or as adjuvant therapy method. Furthermore, traditional Asian food source is rich. In the near future, more and more efficient polysaccharides with antitumor activities of Asian food source will be discovered. There will be broad application market for the polysaccharides.

Abstract:Cancer, as one of the most life-threatening diseases, has attracted the attention of researchers to develop drugs with minimal side effects. The bioactive macromolecules, such as the polysaccharides, are considered the potential candidates against cancer due to their anti-tumor activities and non-toxic characteristics. The present review provides an overview on polysaccharides' extraction, isolation, purification, mechanisms for their anti-tumor activities, structure-activity relationships, absorption and metabolism of polysaccharides, and the applications of polysaccharides in anti-tumor therapy. Numerous research showed extraction methods of polysaccharides had a significant influence on their activities. Additionally, the anti-tumor activities of the polysaccharides are closely related to their structure, while molecular modification and high bioavailability may enhance the anti-tumor activity. Moreover, most of the polysaccharides exerted an anti-tumor activity mainly through the cell cycle arrest, anti-angiogenesis, apoptosis, and immunomodulation mechanisms. Also, recommendations were made to utilize the polysaccharides against cancer.

Abstract:Tumor is a serious threat to human health, with extremely high morbidity and mortality rates. However, tumor treatment is challenging, and the development of antitumor drugs has always been a significant research focus. Plant polysaccharides are known to possess various biological activities. They have many pharmacological properties such as immunomodulation, antitumor, antiviral, antioxidative, antithrombotic, and antiradiation effects, reduction of blood pressure and blood sugar levels, and protection from liver injury. Among these effects, the antitumor effect of plant polysaccharides has been widely studied. Plant polysaccharides can inhibit tumor proliferation and growth by inhibiting tumor cell invasion and metastasis, inducing cell apoptosis, affecting the cell cycle, and regulating the tumor microenvironment. They also have the characteristics of safety, high efficiency, and low toxicity, which can alleviate, to a certain extent, the adverse reactions caused by traditional tumor treatment methods such as surgery, radiotherapy, and chemotherapy. Therefore, this paper systematically summarizes the direct antitumor effects of plant polysaccharides, their regulatory effects on the tumor microenvironment, and intervening many common high-incidence tumors in other ways. It also provides data support for the administration of plant polysaccharides in modern tumor drug therapy, enabling the identification of new targets and development of new drugs for tumor therapy.

Abstract:Two polysaccharides from Crocus sativus petals (PCSPs), PCSPA and PCSPB have been previously reported to possess the immunopotentiation activity and improve innate immunity in mice. In this study, PCSPB was evaluated for the anti-tumor activity and explored its immunological mechanisms based on tumor microenvironment (TME) using S180 sarcoma-bearing mice. Although PCSPB showed the lower toxicity to a series of tumor cells, it significantly and dose-dependently suppressed the growth of S180 sarcomas transplanted in mice. HE staining, immunohistochemical analysis, and TUNEL assay revealed that PCSPB significantly induced tumor cell necrosis, apoptosis, and vessel disruption in sarcoma tissues. Meanwhile, PCSPB markedly decreased the levels of inflammatory factors TGF-β, IFN-γ, IL-10 and TNF-α and down-regulated the mRNA expression levels of TGF-β and TNF-α in tumor tissues. Flow cytometric analysis showed that PCSPB significantly increased the proportion of CD8+ T cells and NK cells, but decreased that of regulatory T cells (Tregs), total myeloid-derived suppressor cells (MDSCs), and tumor-associated macrophages (TAMs) in sarcoma tissues. Furthermore, immunofluorescence assay demonstrated that PCSPB noteworthily reprogrammed TAMs from a tumorigenic M2 towards an antitumorigenic M1 phenotype in S180 tissues. These findings demonstrated that PCSPB might exert the anti-tumor activity by reconstructing TME and could act as an anti-tumor candidate with low toxicity.

In this study, six extraction methods, including hot water extraction (HWE), alkali extraction (AAE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), microwave-assisted extraction (MAE), and freeze–thaw-assisted extraction (FAE) were used to extract PSPs, and their structure–activity relationships were analyzed. The results showed that all six PSPs had similar functional group compositions, thermal stability, and glycosidic bond compositions. PSP-As (PSPs extracted by AAE) exhibited better rheological properties due to their higher molecular weight (Mw). PSP-Es (PSPs extracted by EAE) and PSP-Fs (PSPs extracted by FAE) had better lipid-lowering activity due to their lower Mw. PSP-Es and PSP-Ms (PSPs extracted by MAE), which do not contain uronic acid and have a moderate Mw, had better 1,1-diphenyl-2-picrylhydrazyl (DPPH)-radical-scavenging activity. On the contrary, PSP-Hs (PSPs extracted by HWE) and PSP-Fs, with the Mw of uronic acid, had the best OH-radical-scavenging activity. The high-Mw PSP-As had the best Fe2+-chelating ability. In addition, mannose (Man) may play an important role in the immunomodulatory activity. These results indicate that different extraction methods affect the structure and biological activity of polysaccharides to varying degrees, and these results are helpful for understanding the structure–activity relationship of PSPs.

Characterization and protective effect of Polygonatum sibiricum polysaccharide against cyclophosphamide-induced immunosuppression in Balb/c mice

Abstract:In this study, the polysaccharide from Polygonatum sibiricum (PSP) was evaluated for the immunomodulatory activity by the cyclophosphamide (Cy)-induced immunosuppressed-model in vivo. The PSP has been analyzed in order to identify a variety of chemical properties such as monosaccharide compositions and structural confirmation. The results show that the main components of PSP were galactose and rhamnose. The PSP could significantly stimulate neutral red phagocytosis of RAW264.7 macrophages. Compared with the cyclophosphamide group, PSP accelerated recovery of spleen and thymus indexes, and enhanced T cell and B cell proliferation responses as well as peritoneal macrophage phagocytosis. In addition, PSP treatment restored the levels of IL-2, TNF-α, IL-8 and IL-10 in the serum of the Cy-treated mice in a dose-dependent manner. Therefore, PSP played an important role in the protection against immunosuppression in the Cy-treated mice and could be used as a potential immunostimulant agent.

Abstract:AimsNatural polysaccharides are emerging as a new class of immunomodulatory agents due to their potent immunostimulatory effects and suitable biocompatibility. The aim of this study was to identify potent and selective anticancer activity of a bioactive polysaccharide.Materials and methodsIn vitro, viability assay was performed to screen 16 of bioactive polysaccharides in a panel of normal and cancer cell lines. Foci formation, soft agar, BrdU incorporation, cell cycle analyses, and β-galactosidase staining were performed to validate the screening results. In vivo, both murine gastric cancer syngeneic and a human gastric tumor xenografts models were applied. Tumor histology, immunohistochemical staining, cytokine array and flow cytometry analyses were assayed.Key findingsBSP (bamboo shaving polysaccharides) was identified as the most selective polysaccharide for inhibiting the growth of six gastric cancer cell lines while having no toxic effect on normal gastric mucosal cells. Similarly, BSP had more potent killing effect on a subset of human stomach cancer cells than liver or lung cancer cells. In vivo, BSP significantly inhibited tumor growth and prolonged the survival of mice bearing a gastric tumor; these effects are mediated by tumor cell apoptosis and remodeling of the tumor microenvironment by boosting both immune cell subpopulations and cytokine production in murine gastric cancer syngeneic model. A significant decrease of F4/80-positive tumor-associated macrophages was also observed.SignificanceThe findings of this study suggest that the potent and selective anti-tumor activity of bioactive polysaccharides such as BSP warrants clinical testing for the treatment of gastric cancer.

Abstract:A homogeneous polysaccharide (PPB) was purified from fruiting bodies of Phellinus baumii. And in vitro antitumor and immunomodulation activities were investigated on HeLa, SGC-7901 and RAW264.7 cell lines. PPB inhibited the proliferation of HeLa and SGC-7901 cells significantly, and flow cytometric studies revealed that PPB could mediate the cell cycle in the G0/G1 and S phases. Furthermore, PPB could promote the growth and phagocytosis of RAW264.7 cells, activate the secretion of cytokines such as TNF-α and IL-6, which indicated that PPB had low toxicity. The results make PPB as a candidate adjuvant in cancer therapy.
