The L-LYS content with HPLC method consisted with that with GB8245-87 method.

Abstract:The high-yielde-poly-L-lysine-producing Streptomyces violaceusniger strain isolated and preserved in our laboratory was cultured to producee-poly-L-lysine in this study.The mycelia-free supernatant obtained after the centrifugal separation of the fermentation broth was ultrafiltrated through 5 kD(larger than the molecular weight of e-poly-L-lysine) polysulfone membrane and the filtrate was collected.The static adsorption and desorption characteristics of the filtrate on three different types of ion exchange resins were compared,and the dynamic adsorption and desorption of the filtrate on the selected resin were studied with respect to the effect of operation conditions on adsorption quantity and recovery.Based on the results from static adsorption and desorption,weakly acidic cation exchange resin HD-2 had excellent separation performance one-poly-L-lysine.The best purification process for e-poly-L-lysine was that the sample to be separated was applied to a 22 mm×300 mm HD-2 column with a flow rate of 3 mL/min,followed by 0.1 mol/L NaCl elution with the same flow rate.Ae-poly-L-lysine recovery of 92.0% was achieved using the optimized purification process.
Determination of lysine content based on an in situ pretreatment and headspace gas chromatographic measurement technique

Abstract:This work reports on a simple method for the determination of lysine content by an in situ sample pretreatment and headspace gas chromatographic measurement (HS-GC) technique, based on carbon dioxide (CO2) formation from the pretreatment reaction (between lysine and ninhydrin solution) in a closed vial. It was observed that complete lysine conversion to CO2 could be achieved within 60 min at 60 °C in a phosphate buffer medium (pH = 4.0), with a minimum molar ratio of ninhydrin/lysine of 16. The results showed that the method had a good precision (RSD < 5.23%) and accuracy (within 6.80%), compared to the results measured by a reference method (ninhydrin spectroscopic method). Due to the feature of in situ sample pretreatment and headspace measurement, the present method becomes very simple and particularly suitable to be used for batch sample analysis in lysine-related research and applications. Abstract:Lysophosphatidic acid (LPA) is a class of lipids that play multiple biological functions. Several reports show that they are potential biomarkers for diagnosing ovarian cancer. Therefore, it is necessary to accurately quantify their levels in biological samples. Here we report a high throughput LC/ESI/MS/MS (liquid chromatography electrospray tandem mass spectrometry) method employing a reversed phase C18 column to quantify LPA. In this method, a [13C16] labeled 16:0 LPA is used as the internal standard and the lipids are extracted out from biological samples using Bligh–Dyer method under highly acidic condition. The total run time is 8min. The detection limits of the assay reach fmol level and the CV% of the assay are within 10%. Using this method, we quantify the levels of six LPA species (16:0, 18:2, 18:1, 18:0, 20:4, and 22:6 LPA) in plasma samples. We find that some unknown compounds present in plasma can interfere with the quantification of LPA if they are not well separated from LPA. These unknown compounds are more hydrophobic than LPA and can be removed by thin-layer chromatography (TLC). We also find that the levels of LPA species in human plasma generally follow the order: 18:2 LPA>16:0 LPA, 20:4 LPA>18:1 LPA, 22:6 LPA, and 18:0 LPA.
Determination of lycorine and galantamine contents in Lycoris aurea from different altitudes in Xuefeng Mountain by high performance liquid chromatography

Abstract:A high performance liquid chromatographic(HPLC)method was developed to determine the contents of lycorine and galantamine in the bulb of Lycoris aurea,and analyze the contents of them in Lycoris aurea populations from different altitudes in Xuefeng Mountain. The chromatographic conditions was as following: 20μL of samples or standards were injected into the Agilent Eclipse XDB-C 18(150 mm × 4. 6 mm,5 μm)column at 25 oC and eluted with mobile phase of water(including 0. 1% phosphoric acid) 65 : methanol 35 at flow rate of 1. 0 mL / min,and the eluents were detected by absorption at 288 nm. The results showed that the contents of lycorine and galantamine of the populations from the different altitudes were different,among which,the contents of them of the population at altitude 678 m were all the highest,about 445. 23μg·g-1and 75. 71μg·g-1respectively. The difference was significant compared with the others. This method was simple,rapid and highly. It was more suitable for cultivating Lycoris aurea in the region of altitude about 678 m in order to extract lycorine and galantamine from the bulb.
Determination of α-lactalbumin in milk products by high performance liquid chromatography

Abstract:A method for the determination ofα-lactalbumin in milk products by high performance liquid chromatography has been established.The chromatography conditions as follow: Alltima-C18 conlum(4.6 mm×200 mm,5 μm),UV detection wavelengs 215 nm.This method consisted of a linear gradient of the two mobile phases of 0.1% trifluoroacetic acid in water and 0.5% trifluoroacetic acid and 80% acetonitrile in water at a flow rate of 0.5 mL/min,The conlum temperature was 25 ℃.The result showed that for α-lactalbumin the calibration curve was linear in the range of 50 μg/mL～1 000 μg/mL(r=0.990 9).The average recovery percent was 97.27,the RSD was 0.76%(n=5).The methods is suitable for contents analysis of α-lactalbumin in milk products.

Abstract:Isotope dilution ultra-performance liquid chromatography-electrospray tandem mass spectrometry with derivatization by 9-fluorenylmethyl chloroformate was successfully applied to quantify N(ε) -(carboxymethyl)lysine (CML) and N(ε) -(carboxyethyl)lysine (CEL) in processed foods. We demonstrate that this analytical method is well validated for the determination of CML and CEL contents in processed foods. Relative standard deviations (RSD) indicate repeatability (RSD < 6% for CML and CEL) and reproducibility (RSD < 6% for CML and < 7% for CEL) in this method. Percent recovery is also good. We obtain recoveries of 102% to 112% for CML and 86% to 114% for CEL. CML levels detected in the samples vary from 2.29 to 480 mg/kg food, whereas CEL is detected in significantly lower concentrations ranging from 0.56 to 107 mg/kg food. These data could help consumers make better food choices by monitoring intake of advanced glycation end-products, which may pose a risk to human health.

Abstract:An HPLC method coupled to pre-column derivatization with 2,4-dinitrofluorobenzene (DNFB) was developed for simultaneous determination of 19 amino acids in tea infusion. The amino acid-derived samples were separated on a reversed-phase C18 column (4.6 × 250 mm) at 35 °C and monitored by an ultraviolet detector at 360 nm. Separation analysis showed a good resolution (Rs > 1.5) for most amino acids with a gradient elution of mobile phases at a flow rate of 1 ml min -1 . Precision analysis revealed low repeatability errors with relative standard deviation (RSD) ranging from 0.11 to 1.53 % for retention times and 0.60 to 2.84 % for peak areas, and recovery rate ranged from 70.60 to 108.80 %. Linear calibration curves were well established at the concentrations ranging from 0.57 to 57.38 μg ml −1 , with correlation coefficients R 2 ≥ 0.9918 for 19 detected amino acids. The superior stability of the amino acid derivatives at room temperature gave an advantage to simultaneous preparation of many samples and continuous analysis using auto sampler. The established method was successfully applied to separate and quantify amino acids in three tea cultivars, and the results indicated that the albino tea cultivars contained higher contents of total amino acid, theanine, γ-aminobutyric acid (GABA), glutamic acid (Glu), aspartic acid (Asp), and serine (Ser) than normal tea cultivar.
[Lysine production by Brevibacterium divaricatum NTU-2 and its recovery from the fermentation broth]

Abstract:An accumulation of L-lysine of about 42 g/l (as L-lysine-HCl) was obtained by cultivating Brevibacterium divaricatum NTU-2 in a medium containing 10.0% glucose, 4.0% (NH4)2 SO4, 0.1% KH2 PO4, 0.04% Mg SO4.7H2 O, 30 ml/l soybean meal hydrolysate, 50 mg/l DL-methionine, 100 micrograms/l d-biotin, 100 micrograms/l thiamine-HCl and 5% CaCO3 at pH 7.0. The yield was about 48.8% based on consumed glucose. The L-lysine accumulated in the broth was recovered and purified by simply using a strong cation-exchange resin, Diaion SK1B. The absorbed L-lysine was eluted with 4% ammonia water, and the average adsorption and desorption rates estimated (by 10 repeated experiments) were found to be 96.1% and 97.5%, respectively. The eluate was concentrated to about 50% by volume and adjusted to pH 4.0 with 8 N HCl. The solution was concentrated again to one half volume and followed by adding three volumes of 95% ethanol. The crude crystals of L-lysine-HCl in purity of 89.5% were obtained by filtration. The recovery was 93.5%. The resulting crude crystals were then dissolved in water, and subjected to decolorization with 0.5% active carbon. The decolorized solution thus obtained was recrystalized by concentration and 95% ethanol addition. After filtration and drying at 80 degrees C for 3 hr, white crystalline L-lysine with purity of 99.2% was finally obtained. The overall recovery of L-lysine was up to 86.7%.
Pre-column Derivatization-Hplc Method for Determination of 18 Amino Acids in Enteral Nutrition

Abstract:AIM To develop a pre-column derivatization-HPLC method for the determination of proline in enteral nutrition.METHODS An Agilent C_(18) column(150 mm×4.6 mm,5.0μm) was used with a mobile phase consisting of acetonitrile-sodium acetate(pH 4.2,50 mmol·L~(-1)) for gradient elution analysis.The flow rate was 1.0 mL·min~(-1). The column temperature was 30℃.Theλ_(ex) of FLD was 263 nm and theλ_(em) was 315 nm.The derivatization reagent was 9-fluorenylmethyl chloroformate(FMOC-Cl).RESULTS The linear range of proline was 0.1-1 mg·L~(-1)(r= 0.999 6).The range of average recoveries was 109.42%-112.72%.Both the inter-day RSD and the intra-day RSD were less than 7%.The sample solvent was stable in 24 h.CONCLUSION The method is simple,sensitive and specific which can be applied for determination of proline in enteral nutrition and for quality control of the preparation.

Abstract:An assay is presented for simultaneously determining 5 biogenic amines and the major non-protein amino acids: the toxin beta-N-oxalyl-L-alpha,beta-diaminopropanoic acid (beta-ODAP), its isomer alpha-ODAP and homoarginine in Lathyrus sativus extracts using the HPLC system after derivatization with para-nitrobenzyloxycarbonyl chloride (PNZ-Cl). However, it is more worthy of noting that this paper also describes a new extraction method using 0.2 mol/L HClO4. The new method has some advantages: shorter extraction-time, simultaneous extraction of free amino acids and polyamines, better inhibiting the isomerization of beta-ODAP to alpha-ODAP, and so on.

Abstract:ε-Poly-L-lysine (ε-PL) is a novel food biopreservative with broad antimicrobial activity. In the present study, single-factor experiments were applied to screen optimal carbon and nitrogen sources that affect the yield of a ε-PL-producing strain, Streptomyces violaceusniger, which was obtained in our laboratory recently. Box-Behnken design and response surface methodology (RSM) were adopted to derive a statistical model for optimizing the composition of the fermentation medium. The results showed that the main factors that affect ɛ-PL production were glucose, carbon source; peptone, organic nitrogen source; and (NH(4))(2)SO(4), inorganic nitrogen source. The optimum fermentation medium for ɛ-PL production of S. violaceusniger consisted of (g/L) glucose, 62.49; peptone, 5.71; (NH(4))(2)SO(4), 11.19; MgSO(4), 0.5; FeSO(4), 0.03; ZnSO(4), 0.04; KH(2)PO(4), 1.36; and Na(2)HPO(4).12H(2)O, 3.58. When cultured in the optimum medium, the ε-PL production was an average of 0.349 ± 0.025 mg/mL, which was 1.75 times higher than the initial medium.
Determination of Nε-(1-Carboxymethyl)-L-Lysine in brewing soy sauce by ultrahigh performance liquid chromatography coupled with tandem mass spectrometry

Abstract:A method for the determination of Nε-(1-Carboxymethyl)-L-Lysine (CML) in brewing soy sauce was developed using ultrahigh performance liquid chromatography-tandem mass spectrometry. A mixed cation exchange solid phase extraction cartridge was used for purification, while gradient elution of a hydrophilic interaction chromatography column was performed with a 0.1% formic acid aqueous solution-acetonitrile. The adding standard recovery rate of this method ranged from 86.94 to 92.61%, and the relative standard deviation (RSD) was between 3.69 and 6.18%. The limit of detection (LOD) was 0.038 ng/mL, and the limit of quantification was 0.127 ng/mL. The RSD values of the repeatability test and the parallel test were 3.44 and 6.56%, respectively. Using this method, the CML contents determined in eight brewing soy sauce samples were 3.16 ± 0.18 – 15.10 ± 1.19 μg/mL. This method is simple, accurate, sensitive, and precise, and it is an effective method for monitoring the CML content in brewing soy sauce.

Abstract:In this experiment,1-deoxynojirimycin(DNJ) in the silkworm larvae powder and mulberry leaf powder was extracted with 0.05 mol/L HCl,then it was derived by 9-fluorenylmethyl(FMOC) in pH 8.5 borate buffer solution.DNJ-FMOC came into being complex compound,which can be detected by ultraviolet.We separated and detected it by UV-RP-HPLC.The result indicated that the product was separated entirely.There are the most positive relevance between the peak area of DNJ-FMOC and the concentration of DNJ.The average recovery ratio of adding sample is 100.7%.The precision experiment,stability experiment,repeatability experiment and recovery rate experiment showed that the method is credible,so it can be used as an efficient method to detect DNJ content in the silkworm,mulberry and some other related medicines.

Abstract:A new simple and sensitive method for the simultaneous detection of free DL-amino acids (DL-AA) in natto was established by UPLC-fluorescence (FL) using the newly developed fluorescence derivatization reagent DBD-trans-2-methyl-L-proline (DBD-M-Pro) by precolumn derivatization. DBD-M-Pro reacts with DL-amino acids at 60 °C for 60 min. Using BEH C18 (2.1 mm × 100 mm, 1.7 μm) column gradient analysis for 20 min, the resolution of diastereomers after the derivatization of four DL-amino acids was 1.65–3.50. The detection limit for amino acids was 0.25–2.5 pmol. The average recovery (%) of DL-amino acids was 86.65–118.70%. The method was applied to the detection of chiral amino acids in natto beans and mucus, and four chiral amino acids, DL-Gln, DL-Asp, DL-Glu, and DL-Ala, were simultaneously detected. At the same time, the DL-amino acid content and D/L ratio of three different manufacturers of natto were compared quantitatively.

Abstract:OBJECTIVE: To eatablish a HPLC method for the determination of aminolevulinic acid and its related substances in aminolevulinic acid and aminolevulinic acid powder. METHODS: DIKMA Intersil ODS-3 column was used with acetonitrile-ion pair buffer solution (18:82) as the mobile phase. The buffer was prepared by dissolving 1.15 g ammonium biphosphate and 2.16 g octane-sulfonic acid sodium in 800 mL water and adjusted to pH 2.0 with phosphoric acid, diluted to 1 000 mL with water. The detection wavelength was at 205 nm. RESULTS: A satisfactory separation among aminolevulinic acid, related impurities and excipients was obtained. The calibration curve was linear in the range of 0.2007-1.8063 g·L-1 for aminolevulinic acid. The average recovery of aminolevulinic acid was 99.8% (RSD = 0.9%, n = 9). CONCLUSION: The method is simple, sensitive and accurate. It could be used for quality control of aminolevulinic acid and aminolevulinic acid.

Abstract:The paper proposes a new method to extract β-N-oxalyl-l-α,β-diaminopropionic acid (β-ODAP), α-N-oxalyl-l-α,β-diaminopropionic acid (α-ODAP), homoarginine and polyamines using 0.2M HClO4 for only 1h at 0–4°C. Based on the proposed extraction, an assay is presented for simultaneously determining polyamines, the neurotoxin β-ODAP, its non-neurotoxic isomer α-ODAP and homoarginine in Lathyrus sativus extracts using a liquid chromatography (LC) system after derivatization with para-nitrobenzyloxycarbonyl chloride (PNZ-Cl). The method exhibits some prominent advantages, which include: shorter extraction-time, simultaneous extraction of non-protein amino acids and polyamines and inhibition of the isomerization of β-ODAP to α-ODAP. The assay proved satisfactory for the quantitative determination of L. sativus extracts of seeds and seedlings.

Abstract:Determination of the L- and D-amino acid composition in proteins is important for monitoring process-induced racemization, and thereby protein quality loss, in food and feed. Such analysis has so far been challenging due to the need for sample hydrolysis, which generates racemization, thereby leading to an overestimation of D-amino acids. Here, validation of an LC–MS/MS-based method for the simultaneous determination of L- and D-amino acids in complex biological matrixes, like food and feed, was performed in combination with deuterated HCl hydrolysis. This approach eliminated a racemization-induced bias in the L- and D-amino acid ratios. The LC–MS/MS method was applied for the analysis of 18 free amino acids, with a quantification limit of either 12.5 or 62 ng/mL, except for D-phenylalanine, for which quantification was impaired by background interference from the derivatization agent. For hydrolyzed samples, the composition of 10 L- and D-amino acids pairs could be determined in protein. The average relative standard deviation was 5.5% and 6.1%, depending on the type of hydrolysis tubes. The method was applied on a green protein isolate (lucerne), which contained an average of 0.3% D-amino acids. In conclusion, this method allows for an unbiased analysis of L- and D-amino acid ratios in complex protein samples, such as food and feed.

Determination of Lycopene Concentration in Dog Plasma by RP-HPLC

Abstract:Aim A method for determination of lycopene concentration in dog plasma was established. Methods RP-HPLC was used; the mobile phase wavelength of detection was 472 nm, the column temperature was ambient temperature, and the flow rate was 1.0 mL·min-1. Results The standard curve was linear in the range from 0.012 4 to 0.496 μg·mL-1 with r=0.999 2. The average extraction recovery was 97.6%±4.2%. The intra-day and inter-day RSD were 1.52%-4.95% and 2.31%-7.38%, respectively. Conclusion This method is sensitive, rapid, reproducible, and of good selectivity for the analysis of lycopene in dog plasma.

Abstract:The excessive accumulation of antibiotic residue in animal-derived foods pose a threat to our heath. Therefore, it is necessary to establish a rapid, simple, effective, safe and highly sensitive lincomycin (LIN) residues detection method by high-performance liquid chromatography equipped with ultraviolet. Here, due to the weak UV absorption of LIN, the p -Toluenesulfonyl isocyanate (PTSI, has a strong UV absorption) was binded to the hydroxyl group of LINs by using pre-column derivatization method, improving the UV absorption of LIN. This 9-min-run analytical method detects and quantifies LIN residues with acceptable validation performance parameters in terms of sensitivity, selectivity, linearity, the limit of quantification, accuracy, and precision.Our result showed that the limits of detection and quantification for LIN in samples were 25–40 μg/kg and 40–60 μg/kg, respectively, under the optimized conditions. Furthermore, the LIN recoveries ranged from 72.4 % to 96.7 %; the determination coefficients were 0.9912–0.9996, and the relative standard deviations were 5.6 %-12.7 %. Therefore, the HPLC-UVD method for detecting LIN residues in animal-derived food products has been established.

Abstract:Objective To establish a quantitative method for the quality control of Banlangen Granule.Method A pre-column derivatization HPLC method, in which the analyte was eluted in a phenomen ODS C 18 column (10 μm, 4.6 mm×250 mm) with 0.1 mol/L NaAc buffer (pH6.4) -acetonitrile (86∶14) in the flow rate of 1.0 ml/min, was applied to analyze the arginine content of different factories under the wave length of 360 nm. Results Arginine content in Banlangen Granule was successfully determined, and it was found that the content of arginine in Banlangen Granule from different sources varied greatly.Conclusions The method is simple, reproducible and accurate. It can used as a quality control method for Banlangen Granule.
