Abstract:In order to find out the respective influences of Cu ,Zn and their joint effects on goldfish ,this article evaluates the acute toxicities of Cu2+ and Zn2+ on Mactra veneriformis Reeve and compares the differences of toxicity among the heavy metals ,and carries out the experiments according to toxicological methods .T he re-sults show that with the increase of heavy metal ion concentration ,the fish acute toxicity effects gradually in-crease ;under the same concentration ,Cu2+ effects are heavier than Zn2+ on goldfish .According to the results of joint toxicity test ,Cu and Zn concentration in the 1 :1 ratio cause synergistic effect on goldfish .

Abstract:The effects of cadmium, hydroxylated multi-walled carbon nanotubes, and their mixture on metal accumulation and antioxidant defenses were studied using the goldfish Carassius auratus as the test organism. The fish were exposed to 0.1 mg/L Cd, 0.5 mg/L OH-MWCNTs, or 0.1 mg/L Cd+0.5 mg/L OH-MWCNTs for 3 and 12 days. Then, the Cd concentration was determined in the gill, liver and muscle. Moreover, hepatic antioxidant enzyme activity (superoxide dismutase, catalase and glutathione peroxidase), glutathione level and malondialdehyde content were also measured. A continuous accumulation of Cd was observed throughout the experimental period. Cd accumulation in tissues occurred in the following order: gill>liver>muscle at 3 days and liver>gill>muscle at 12 days. The concentrations of Cd in the livers of fish exposed to the combination of Cd+OH-MWCNTs were significantly higher than those in fish exposed to either single chemical after 12 d of exposure. Meanwhile, the mixture evoked severe oxidative stress in the exposed fish, as indicated by significant inhibition of SOD, CAT and GPx activity, a remarkable decrease in GSH level, and simultaneous elevation of MDA content. These results suggested that the effect of the combined factors on metal accumulation and oxidative stress biomarkers was more obvious than that of single factors at longer exposure durations.
Toxic Effect of Combined Exposure of Microplastics and Copper on Goldfish (Carassius auratus): Insight from Oxidative Stress, Inflammation, Apoptosis and Autophagy in Hepatopancreas and Intestine

Abstract:The enhancement of the toxic effect of microplastics (MPs) on heavy metals and its mechanism needs more in-depth and systematic research. In this study, the copper (Cu) accumulation, histological injury, and expression of genes involved in oxidative stress, inflammation, apoptosis, and autophagy of goldfish after single or combined exposure of MPs (1 mg/L) and Cu2+ (0.1 mg/L) for 7 days were determined. The results demonstrated that MPs enhanced the Cu accumulation in hepatopancreas and intestine of goldfish and induced more severe oxidative stress in the hepatopancreas and intestine of goldfish. Additionally, combined exposure of MPs and Cu induced inflammation, excessive apoptosis and insufficient autophagy in the hepatopancreas. Contrary, the inflammation and apoptosis were depressed in the intestine after combined exposure of MPs and Cu, which still requires further exploration. Hence, these findings provide further evidence for the threat of MPs and its adsorbed heavy metals.
Tin and Lead Compounds in Fish and Crustacean from Aquaculture Ponds in Zhejiang Province of East China: Accumulation and Health Risk Assessment

Abstract:mu Aquaculture industries supply half of the global production, leading to an increasing concern about health risk of the aquaculture products because of widespread distribution of heavy metals in the environment. Considering high dependence of toxicity and bioaccumulation of one metal not merely upon total content but mainly upon its chemical forms, recent investigations into risk assessment of aquaculture products based on total contents of heavy metals may be unreasonable and unfair. In this study, we conducted reasonable assessment to bioaccumulation and health risk of aquaculture fish and crustacean collected from eight aquaculture ponds in Zhejiang Province of East China in terms of chemical forms of two model heavy metals (Sn(II), Pb(II), four organotin compounds, trimethyl lead -TML and triethyl lead -TEL). Although all metal compounds were presented below 0.2-4.1 mu g kg(-1) in aquatic food, they were at moderate contents in environmental matrices following an order of sediment > feedstuff > aquatic food > water for most of the target compounds (i.e. 54-376 mu g kg(-1) Pb(II) in sediment, 14-94 mu g kg(-1) Pb(II) in feedstuff and organotin compounds at pg L-1 levels in water), indicating feedstuff as an important exposure source apart from water adsorption. Bioaccumulation factors, biota-sediment accumulation factors and biomagnification factors demonstrated low bioaccumulation probability although the bioaccumulation level followed an order of organotin and organolead > inorganic forms > total metal. In addition, estimated daily intakes, target hazard quotients and cancer risk demonstrated no health risk for all tin compounds and no carcinogenic risks for all lead compounds but moderate non-carcinogenic risks for TML and TEL. Overall, the accumulation level and health risk were in a closer relationship with the chemical forms of Sn and Pb than their total contents, demonstrating a high necessity to risk assessment in terms of chemical forms.

Abstract:In this study, laboratory experiments were conducted to investigate the combined effect of zinc and pH on metal accumulation and oxidative stress biomarkers in Carassius auratus. Fish were exposed to 0.1 and 1.0mg Zn/L at three pH values (5.0, 7.25, 9.0) for 3, 12, and 30 d. After each exposure, the contents of three trace elements (Zn, Fe and Cu) were determined in liver. Generally, longer exposure to zinc (12d and 30 d) increased hepatic Zn and Cu deposition, but decreased Fe content. Increasing accumulation of Zn in the tissue was also observed with increasing zinc concentration in the exposure medium. Moreover, hepatic antioxidant enzyme activities including superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), together with the level of glutathione (GSH) were measured to evaluate the oxidative stress status. The decreases in the four measured biochemical parameters after 3d exposure might reflect the failure of the antioxidant defense system in neutralizing the ROS generated during the metabolic process, while the recovery of the antioxidants at days 12 and 30 suggested a possible shift toward a detoxification mechanism. With regard to the influence of pH on zinc toxicity, the general observation was that the living environment became more stressful when the water conditions changed from an acidic state toward a near-neutral or alkaline state.
Effects on heavy metal accumulation in freshwater fishes: species, tissues, and sizes

Abstract:Three fish species ( Carassius auratus , Pelteobagrus fulvidraco , and Squaliobarbus curriculus ) were collected from Xiang River near Changsha City, Southern China. The concentrations of heavy metals including arsenic (As), cadmium (Cd), copper (Cu), iron (Fe), manganese (Mn), lead (Pb), and zinc (Zn) in the muscle, gill, and liver of three species were determined by the inductively coupled plasma mass spectrometry method. One-way analysis of variance (ANOVA) was introduced to determine the significant variations ( p < 0.05) of heavy metals. Livers were found to accumulate Cd and Cu due to the metallothionein proteins. High levels of Mn and Pb in the gills indicated that the main uptake pathway of these heavy metals was from the water. The carnivorous species, P. fulvidraco , was found to accumulate the highest levels of toxic elements (As, Cd, and Pb), while relatively high concentrations of nutrient elements (Cu, Fe, Mn, and Zn) were accumulated in omnivorous species ( C. auratus and S. curriculus ). According to the results of Pearson’s correlation analysis, there were few significant relationships at p < 0.01 level between the concentrations of the analyzed elements and the fish sizes. The results of risk assessment indicated that exposure to the toxic heavy metals from fish muscle consumption posed no non-carcinogenic health risk to local inhabitants.

Abstract:In order to assess the condition of heavy metal pollution in the Yellow River, Lanzhou section, China, and to quantify heavy metal (copper, lead, zinc, and cadmium) contents in tissues (liver, kidney, gills, and muscles) of two fish species (Triplophysa pappenheimi and Gobio hwanghensis), levels of these four metals in the water body, sediment, and tissues of the two fish were measured using inductively coupled plasma-atomic emission spectrometry. The metal levels from this study were compared with the threshold values in the guidelines of water, sediment, and food given by the National Environmental Protection Agency of China, the National Oceanic and Atmospheric Administration of America, and the National Standards Management Department of China. We found the mean concentrations of Cu, Pb, Zn, and Cd in THE water body, sediment, and muscles of two fish species were far below the values in guidelines. We also found that the type of metals present and their concentrations varied in different tissues and species. The results suggested that (1) Cu, Pb, Zn, and Cd did not contaminate the aquatic ecosystem severely and did not threaten the safety of human consumption in the Yellow River, Lanzhou section, and (2) organs that are sensitive to accumulating heavy metals may be useful to develop bioindicators for monitoring metal contamination. Considering environmental variables, further study is necessary before deciding which fish species or tissue could be the ideal bioindicators for aquatic pollution.

Abstract:Zinc Oxide Nanoparticles (ZnO NPs) have attracted increasing concerns because of their widespread use and toxic potential. In this study, Zn accumulations in different tissues (gills, liver, muscle, and gut) of goldfish (Carassius auratus) after exposure to ZnO NPs were studied in comparison with bulk ZnO and Zn2+. And the technique of subcellular partitioning was firstly used on the liver of goldfish to study the hepatic accumulation of ZnO NPs. The results showed that at sublethal Zn concentration (2 mg/L), bioaccumulation in goldfish was tissue-specific and dependent on the exposure materials. Compared with Zn2+, the particles of bulk ZnO and the ZnO NPs appeared to aggregate in the environmentally contacted tissues (gills and gut), rather than transport to the internal tissues (liver and muscle). The subcellular distributions of liver differed for the three exposure treatments. After ZnO NPs exposure, Zn percentage in metal-rich granule (MRG) increased significantly, and after Zn2+ exposure, it increased significantly in the organelles. Metallothionein-like proteins (MTLP) were the main target for Zn2+, while MRG played dominant role for ZnO NPs. The different results of subcellular distributions revealed that metal detoxification mechanisms of liver for ZnO NPs, bulk ZnO, and Zn2+ were different. Overall, subcellular partitioning provided an interesting start to better understanding of the toxicity of nano-and conventional materials.

Abstract:This comprehensive review provides a thorough exploration of a mounting environmental issue: heavy metal contamination in aquatic ecosystems and its far-reaching impacts on fish populations and human health. Starting with the generalized sources of different heavy metals that lead to their entry into the aquatic environment, this review then considers each of the major heavy metals (copper, cadmium, chromium, arsenic, nickel, lead, zinc, and mercury) and ventures deep into the intricate mechanisms governing the uptake, bioaccumulation, and toxicity of heavy metals in fish, shedding light on the profound consequences these processes have for fish health and behavior. A critical aspect emphasized in this article is the activation of antioxidant defense mechanisms and involvement of metallothionein in fish as an adaptive response aimed at mitigating the pervasive oxidative stress triggered by heavy metal exposure. Of utmost concern is the trophic transfer of heavy metals from contaminated fish to humans through consumption, which poses a direct threat to human health regarding various physiological functions. The article underscores the urgency of addressing this issue comprehensively. Given the concerning discoveries at hand, this review fervently supports the enactment of rigorous regulations, the embracing of sustainable management techniques, and the stringent enforcement of pollution containment strategies.

ACCUMULATIONS OF LEAD AND CADMIUM IN GOLDFISH, CARASSIUS AURATUS

Abstract:Accumulations of lead and cadmium in gills, blood, liver, kidney, gut and muscle of goldfish, Carnssius auratus var. exposed to various concentrations of either lead or cadmium were studied. The Langmuir isotherms were employed to describe the gill surface binding activity of lead or cadmium. The binding capacities(B_(max)) and conditional eduilibrium constants (log K) were 0.9424 mmol/ kg dry wt and 7.80 for lead, 1.7662 #mu#mol/ kg dry wt and 6.38 for cadmium, respectively. The binding cooperativities(H_(n)) were very closed to unit (1.012 for lead and 1.175 for cadmium) indicating independent binding to the sites with one apparent affinity. There were positive correladons of metal contents between gills and blood and between blood and kidney or liver. There were more metal accumulated in kidney than in other organs.

Abstract:Concentrations of Cu, Zn, Cr, As, Cd, Pb and Hg were determined in 153 samples covering 18 fish species collected from Heilongjiang River, China, and the potential health risks to local residents through fish consumption were estimated. Results revealed that all metals were detected with Cd and Pb having considerably lower detection rate. There were 28.6 % of Lethenteron camtschaticum samples exceeding the limit of inorganic As content established by Chinese legislation. Metal bioaccumulation in fish were poorly correlated with fish sizes, and generally tended to increase with trophic levels. The hazard quotient values of all fish species were far below 1 for general population and fishermen. The carcinogenic risk of As in L. camtschaticum for the local fishermen exceeded the acceptable level of 10−4 rise in cancer rates, but was considered safe given the migratory habits of the fish.

Abstract:Heavy metals (HMs) in an aquatic environment mainly affects fish, and thus, fish are convenient pollution bio-indicators. In this study, the toxic effects of HM mixture (chromium (Cr), cadmium (Cd), copper (Cu)) in 0 mg/L to 3.2 mg/L concentration range was investigated in Cyprinus carpio (28 days). HM accumulation, histopathology, oxidative stress, and gut microbial changes were evaluated. HMs accumulated in the order of Cr > Cu > Cd, primarily in the kidneys and finally scales. Reactive oxygen species generation increased in all exposure groups up to day 14, with maximum generation at 3.2 mg/L mixture, which later decreased on day 28 in all. Malondialdehydeand and superoxide dismutase levels increased from day 7 to 28 with increased HM concentrations, while total protein showed an inverse trend. Gill histopathology showed major changes such as uplifted and disintegrated primary lamella, and secondary lamella shortening. The kidneys were characterized by glomerular necrosis, Bowman's capsule expansion, and tubular space dilatation. Proteobacteria and Firmicutes abundance increased up to 59.4% and 99.16% in 0.8 mg/L and 3.2 mg/L treatment groups, respectively. This study provided a better understanding on the physiology and gut microbiota alteration in C. carpio under multiple HM stress.
Water Hardness Improves the Antioxidant Response of Zinc-Exposed Goldfish (Carassius auratus)

Abstract:Zinc (Zn), a heavy metal, is an essential element in fish; however, exposure to high concentrations causes oxidative stress. Water hardness reduces oxidative stress reactions caused by heavy metals. To confirm the effect of water hardness on oxidative stress caused by Zn, goldfish were exposed to various Zn concentrations (1.0, 2.0, and 5.0 mg/L) and water hardness (soft (S), hard (H), and very hard (V)). The activity of superoxide dismutase (SOD) and catalase (CAT) in plasma increased with 1.0, 2.0, and 5.0 mg/L of Zn, and decreased with H and V water hardness. The levels of H2O2 and lipid peroxide (LPO) increased with Zn above 1.0 mg/L and decreased with H and V of water hardness. Caspase-9 mRNA expression in the liver increased after 7 and 14 days of Zn exposure and decreased with H and V water hardness. It was confirmed that DNA damage was less dependent on H and V water hardness. Based on the results of this study, at least 1.0 mg/L Zn causes oxidative stress in goldfish, and a high level of apoptosis occurs when exposed for more than 7 days. It appears that the oxidative stress generated by Zn can be alleviated by water hardness of at least 270 mg/L CaCO3. This study provides information on the relationship between the antioxidant response caused by heavy metals and water hardness in fish.

Abstract:AbstractSeabass (Lateolabraxjaponicus) is a major aquacultured fish species worldwide. The bioconcentration, bioaccumulation, and biomagnification of metals in water, sediments, and commercial feed were investigated in L. japonicus from an aquaculture pond in the Pearl River Delta of South China. Aluminum (Al), chromium (Cr), manganese (Mn), iron (Fe), nickel (Ni), copper (Cu), zinc (Zn), arsenic (As), cadmium (Cd), and lead (Pb) were determined in the dorsal muscle, viscera, backbone, gill, and stomach contents of L. japonicus. The gill and stomach contents had higher levels of bioconcentration of most metals than other parts of fish. Based on the bioaccumulation factor, the gill and backbone exhibited the highest accumulation of Zn, while the viscera had the highest capacity to accumulate Cu. The mean biomagnification factor values exceeded for As in dorsal muscle, for Cu in the viscera, for Cr and Pb in the gill, and for Al, Cr, Fe, Cu, and Pb in the stomach contents, indicating efficient bioaccumulation from commercial feed and their habitat. Non-metric multidimensional scaling analysis revealed two groups that resulted from the accumulation of metals in various parts of L. japonicus. Moreover, health risk assessment indicated that no notable adverse health effects occurred from the ingestion of L. japonicus.

Differential response of heavy metal accumulation in freshwater aquatic organisms to organic matter pathway (δ13C) and trophic level (δ15N)

Abstract:The migration of heavy metals into aquatic ecosystems is a concern for the safety of aquatic organisms and human health. However, the migration of heavy metals from habitats to the food chain in freshwater ecosystems requires extensive exploration. We extensively investigated the levels of heavy metals in multiple media of freshwater ecosystems and explored their migration from freshwater habitats to the food chain. The results showed that the concentrations of Cr, Cu, Cd, Zn, Sb, and Pb in sediments, Cr in mollusks, and Cd in clams exceeded their standard limit values. Feeding habits, species, and body length considerably affected heavy metal levels in fish, whereas regional differences, body length, and weight considerably affected heavy metal levels in mollusks. The bioconcentration capacity (improved biotawater (sediment) accumulation factors) of the muscles was higher than that of the gills and visceral mass. Mollusks were strongly enriched in Cu, Cd, Zn, and Mn in their habitats, whereas fish were more favourable for the accumulation of Cu, Zn, and Cr. Heavy metals in sediments pose a higher risk to aquatic organisms than those in the overlying water. Trophic level (TL) (δ15N) considerably diluted levels of most metals, whereas Cu and Zn levels increased along the organic matter pathway (δ13C) in the entire aquatic food chain. The levels of typical pollutants, such as Cu, Zn, As, Cd, Pb, and Cr changed considerably along the organic matter pathway in mollusks. The levels of heavy metals in fish were not affected by TL or organic matter pathway, except for Cu. The potential risk of consuming aquatic organisms from Poyang Lake was ranked as clams > snails > fish for adults and children and As in aquatic organisms was the primary contributor to health risk.

Abstract:Metal content was determined in two fish species (Misgurnus anguillicaudatus and Monopterus albus) inhabiting rice paddies along the Middle Reach of the Yangtze River. Both are species important food fish in East Asia. The levels of eight metals (Fe, Mn, Zn, Cu, Cr, As, Cd, Pb) in skin, muscle and intestinal tissues were measured and correlated with fish size. Metal distribution among the tissues was examined and the potential risk of fish consumption was evaluated. Correlations between metal concentration and fish size were not significant. Concentrations of essential metals varied significantly among tissues, but the differences were not significant for non-essential metals. Correlations among metals in skin tissue were stronger than in intestinal and muscle tissues. Metal concentrations in the edible tissues of both fish met permissible levels for consumption, except Pb in loach skin and Cr loach in muscle. The potential risk caused by Pb from skin and Cr from muscle of loach consumption should be considered in future risk assessmeents.

Abstract:The migration of heavy metals into aquatic ecosystems is a concern for the safety of aquatic organisms and human health. However, the migration of heavy metals from habitats to the food chain in freshwater ecosystems requires extensive exploration. We extensively investigated the levels of heavy metals in multiple media of freshwater ecosystems and explored their migration from freshwater habitats to the food chain. The results showed that the concentrations of Cr, Cu, Cd, Zn, Sb, and Pb in sediments, Cr in mollusks, and Cd in clams exceeded their standard limit values. Feeding habits, species, and body length considerably affected heavy metal levels in fish, whereas regional differences, body length, and weight considerably affected heavy metal levels in mollusks. The bioconcentration capacity (improved biotawater (sediment) accumulation factors) of the muscles was higher than that of the gills and visceral mass. Mollusks were strongly enriched in Cu, Cd, Zn, and Mn in their habitats, whereas fish were more favourable for the accumulation of Cu, Zn, and Cr. Heavy metals in sediments pose a higher risk to aquatic organisms than those in the overlying water. Trophic level (TL) (δ 15 N) considerably diluted levels of most metals, whereas Cu and Zn levels increased along the organic matter pathway (δ 13 C) in the entire aquatic food chain. The levels of typical pollutants, such as Cu, Zn, As, Cd, Pb, and Cr changed considerably along the organic matter pathway in mollusks. The levels of heavy metals in fish were not affected by TL or organic matter pathway, except for Cu. The potential risk of consuming aquatic organisms from Poyang Lake was ranked as clams > snails > fish for adults and children and As in aquatic organisms was the primary contributor to health risk.

Abstract:Heavy metals can cause a significant damage to submerged macrophytes and affect its periphyton biofilms in aquatic environments. This study investigated the effects of heavy metals such as copper (Cu), lead (Pb), cadmium (Cd) and their mixture on physiological and biochemical responses and ultrastructure characteristics of Vallisneria natans (V. natans). Furthermore, differences in structures of microbial communities were observed in biofilms. The results showed that Cu2+, Pb2+, Cd2+ and their mixture could destroy cell structure and photosynthetic system, and directly caused oxidative damage to submerged macrophyte and induced antioxidant enzyme system. In general, biomass and total chlorophyll content of V. natans noticeably decreased, while the activities of superoxide dismutase, peroxidase and catalase were enhanced by heavy metal stress inducement in restricted range, and the malondialdehyde content increased with the aggravation of the damage. The single heavy metal stress played a negative impact, however, the combined stress was not always synergistic effects on plants. High-throughput sequencing analysis suggested that heavy metals changed the abundance and structure of the microbial biofilm community. Proteobacteria and Bacteroidete were the dominant bacteria under heavy metal stress and other species and abundance of bacteria such as Firmicute, Cyanobacteria, Chloroflexi, Actinobacteria, Verrucomicrobia, Acidobacteria, Deinococcus-Thermus, Chlamydiae were also present. These findings provided useful information for further understanding about submerged macrophytes and periphyton biofilms responsed to heavy metal stress in aquatic environments in the future.

Abstract:With the rapid industrialization and urbanization of coastal areas, marine pollution (such as heavy metals) is increasingly contaminating the environment, posing significant public health risks. Eastern Guangdong, a key aquaculture and fisheries hub in China, has a growing market for aquatic products. Heavy metals persist in the environment and are difficult to degrade and bioaccumulate in marine organisms through the food web, presenting carcinogenic and mutagenic risks to humans, as top predators. This study analyzed 10 key species commonly consumed by residents of eastern Guangdong (bivalves, crustaceans, and fish), measuring the concentrations of six heavy metals (As, Cd, Cr, Cu, Ni, Pb) using inductively coupled plasma mass spectrometry (ICP-MS). Pollution levels were assessed using the pollution index (Pi), and dietary exposure risks were evaluated via the target hazard quotient (THQ) for different age groups. Results showed that Pi values for all metals were within normal background levels, but bivalves had a high capacity for Cd accumulation, with pollution severity ranking as bivalves > crustaceans > fish. The THQ values for both adults and teenagers were <1 across all samples, indicating no risk to the health of residents. However, the TTHQ for Sanguinolaria sp. exceeded 1, indicating potential health risks. This study highlights the health risks of consuming heavy metal-contaminated aquatic products, particularly bivalves. Reducing the consumption of these high-metal species could help lower dietary exposure and associated risks. Our findings provide essential data for the quality assessment of aquatic products and offer dietary recommendations for residents in eastern Guangdong.
