Abstract:Water-soluble cupric oxide nanoparticles are fabricated via a quick-precipitation method and used as peroxidase mimetics for ultrasensitive detection of hydrogen peroxide and glucose. The water-soluble CuO nanoparticles show much higher catalytic activity than that of commercial CuO nanoparticles due to their higher affinity to hydrogen peroxide. In addition, the as-prepared CuO nanoparticles are stable over a wide range of pH and temperature. This excellent stability in the form of aqueous colloidal suspensions makes the application of the water-soluble CuO nanoparticles easier in aqueous systems. A colorimetric assay for hydrogen peroxide and glucose has been established based on the catalytic oxidation of phenol coupled with 4-amino-atipyrine by the action of hydrogen peroxide. This analytical platform not only confirms the intrinsic peroxidase-like activity of the water-soluble cupric oxide nanoparticles, but also shows its great potential applications in environmental chemistry, biotechnology and medicine.
Fluorescent hydrogen peroxide sensor based on cupric oxide nanoparticles and its application for glucose and L-lactate detection.

Abstract:A novel fluorescent hydrogen peroxide sensor was developed based on the peroxidase-like activity of cupric oxide nanoparticles. Cupric oxide nanoparticles effectively catalyzed the decomposition of hydrogen peroxide into hydroxyl radicals. Then terephthalic acid was oxidized by hydroxyl radical to form a highly fluorescent product. The linear range of hydrogen peroxide estimated to be 5.0 × 10(-6)-2.0 × 10(-4)M with a detection limit of 3.4 × 10(-7)M. Moreover, this detection system enabled the sensing of analytes which can enzymatically generate hydrogen peroxide. By coupling the oxidation of glucose or L-lactate catalyzed by their corresponding oxidase enzymes with terephthalic acid oxidation catalyzed by cupric oxide nanoparticles, sensitive assays of glucose and l-lactate with detection limits of 1.0 × 10(-6) and 4.5 × 10(-8)M were realized. The successful applications of this approach in human serum samples have also been demonstrated.
Colorimetric Detection of Urea, Urease, and Urease Inhibitor Based on the Peroxidase-Like Activity of Gold Nanoparticles.

Abstract:Herein, we reported for the first time that gold nanoparticles-catalyzed 3,3',5,5'-tetramethylbenzidine-H2O2 system can serve as an ultrasensitive colorimetric pH indicator. Gold nanoparticles acted as a catalyst and imitated the function of horseradish peroxidase. The absorbance at 450 nm of the yellow-color product in the catalytic reaction exhibited a linear fashion over the pH range of 6.40-6.60. On the basis of this property, we constructed a novel sensing platform for the determination of urea, urease, and urease inhibitor. The limit of detection for urea and urease was 5 μM and 1.8 U/L, respectively. The half-maximal inhibition value IC50 of acetohydroxamic acid was found to be 0.05 mM. Urea in human urine and urease in soil were detected with satisfied results.
Preparation of copper nanoparticles fluorescent probes and detection of hydrogen peroxide and glucose

Abstract:Fluorescent copper nanoparticles (CuNPs) was synthesized by one-step chemical reduction method using ascorbic acid (AA) and copper sulfate (CuSO4⋅5H2O) as raw materials, which had good water solubility and fluorescence properties. A green, simple and safe CuNPs@Fe2+ fluorescence probe was developed for the detection of hydrogen peroxide and glucose using Fe2+ as a bridge. The prepared CuNPs could obtain the maximum fluorescence emission wavelength at 440 nm when the excitation wavelength was 360 nm. The average particle size of CuNPs was 10 nm, which had good photobleach resistance, stability and salt tolerance. The fluorescence intensity was quenched due to electron transfer (ET) process when hydrogen peroxide was added to CuNPs@Fe2+ system. This result was mainly because Fenton reaction occured between hydrogen peroxide and Fe2+, producing hydroxyl free radicals (OH) and Fe3+. Since glucose could be catalyzed by specific glucose oxidase (GOX) to produce H2O2 and corresponding oxidation products, the quantitative analysis of glucose was realized when glucose oxidase was introduced into the CuNPs@Fe2+ sensor system. Therefore, a novel CuNPs@Fe2+ fluorescent probe sensor study was constructed to further achieve quantitative detection of H2O2 and glucose. Under the optimized experimental conditions, the linear ranges for H2O2 and glucose were 28.219-171.562 μM and 1.237-75.771 μM, respectively. And the detection limits for H2O2 and glucose were 7.169 μM and 0.540 μM, respectively. In addition, the mechanism of fluorescence probe quenching caused by the interaction between H2O2 and CuNPs@Fe2+ was also discussed. The proposed sensing system had been applied successfully to the detection of glucose in human serum samples.

Abstract:The composite of copper iodide (CuI) nanomaterials and CuI‐isopropanolamine complexes was prepared by dissolving CuI in C3H9NO and subsequent annealing. It exhibited exceptional sensing properties to 0.15 (mole fraction) ammonia: a high response of 7.5×105, a rapid response time of 5 s, and exceptional selectivity, indicating promising potential for practical application in ammonia detection. With the increasing awareness of the hazards associated with ammonia (NH3) contamination and the potential application of NH3 detection in physical health monitoring, the exploration of high‐performance NH3 sensors has attracted significant attention in recent years. In this paper, we prepared copper iodide (CuI) nanomaterials combined with CuI‐isopropanolamine (C3H9NO) complexes by dissolving CuI in C3H9NO and subsequent annealing. The resulting product demonstrated exceptional sensing performance to 0.15 (mole fraction) NH3, exhibiting a maximum response of 7.5×105 with a response time of 5 s. The achieved detection limit was 10 ppm. Our findings provide a valuable reference for further research on NH3 sensors utilizing complexes formed by copper compounds and organic amines.

Abstract:Based on the intermediate states of metal ions in metal oxide nanomaterials (NMs) that acted as the primary active species, the design of high-performance nanozymes has greatly stimulated current research in diverse biomedical applications. Herein, Cu2O nanocubes-grafted highly dense Au nanoparticles (NPs) was developed as an appealing nanozyme for H2O2 colorimetric sensor and antioxidant detections. The obtained Au/Cu2O heterostructures show efficient electron-transfer from metallic NPs to Cu2O nanocubes owing to the difference of Fermi energy between two components. The modulated electronic structure of Au/Cu2O hybrids enables them to possess enhanced peroxidase catalytic activity for the oxidation of 3, 3', 5, 5'-tetramethylbenzidine (TMB) in the presence of H2O2, which is about 32% higher than that of pristine Cu2O nanocubes. Then, an excellent H2O2 colorimetric sensor was established by using Au/Cu2O heterostructures with a low limit of detection (LOD) of 0.054 μM, which is much lower than the H2O2 allowance level of US FDA regulations (ca.15 μM, 0.05 wt%). The obtained Au/Cu2O nanoproducts exhibit pronounced long-time stability with 95% peroxidase activity maintained after keeping them for 30 days, while residual 64.5% via Cu2O nanocubes. Furthermore, we assessed the anti-oxidative behavior of three natural antioxidants (tannic acid, gallic acid, tartaric acid) with the LODs as low as 0.039, 0.16 and 1.55 μM, respectively, and the antioxidant capacity in the following order: tannic acid > gallic acid > tartaric acid. Therefore, it is believed that the as-prepared Au/Cu2O nanozymes have promising potential applications in fields of biomedicine and food safety.

Abstract:CuO nanoparticles with good water solubility and uniform particle size were successfully prepared. Interestingly, the oxidase-like activity of CuO NPs was continuously enhanced by the addition of thiourea (TU), and the enzyme activity was further enhanced by the addition of aluminum ion (Al3+). By systematically exploring and optimizing the experimental conditions, including the key parameters such as temperature, reaction time, and pH, a fluorescence-colorimetric dual-mode sensing system based on CuO nanoparticles was constructed. The detection range of TU and Al3+ were 1-100 µM and 1-100 µM, respectively, and the selectivity and precision of detection were further improved. In addition, the catalytic mechanism of CuO NPs as oxidase-like catalysts and the specific process in the reaction were investigated. Finally, the nano-sensing system was successfully applied to the analysis of three real environmental samples, namely, tap water, lake water and river water, which provided an effective new strategy for the future development of nano-sensing technology for TU and Al3+.

Abstract:Luminescent copper nanoclusters (CuNCs) constitute a very active research topic due to their unique properties and lower cost than gold and silver NCs. In this study, we report a new, facile, and rapid top-down etching method for synthesizing luminescent CuNCs, using Cu nanoparticles (CuNPs) as the precursor and ammonia (NH3) as the etchant. The etching mechanism is systematically investigated and the optical and structural properties of the obtained CuNCs are carefully studied. The NH3-triggered etching process is very fast and the newly generated CuNCs can emit strong green fluorescence with a high quantum yield. Moreover, by coupling the urease-catalyzed hydrolysis of urea with the NH3-induced etching of CuNPs, we developed a novel fluorescence turn-on assay for urea. The linear range for urea detection is from 0.25 to 5 mM, and the limit of detection is 0.01 mM. This novel sensing approach, with good sensitivity and excellent selectivity, is then successfully utilized to detect urea in human serum samples, demonstrating its great potential in clinical diagnosis. In addition, the proposed coupling method can be extended to monitor other analytes that influence the size-focusing etching process, allowing metal NCs to be used to construct diverse chemosensors and biosensors.
Double Enzyme Mimetic Activities of Multifunctional Ag Nanoparticles Decorated Co3v2o8 Hollow Hexagonal Prismatic Pencils for Application in Colorimetric Sensors and Disinfection

Abstract:Since the catalytic activity of most nanozymes is still far lower than the corresponding natural enzymes, there is urgent need to discover novel highly efficient enzyme-like materials, especially multifunctional artificial enzyme. In this work, Co3V2O8 with hollow hexagonal prismatic pencil structures were prepared as novel artificial enzyme mimics, which was then decorated by photo-depositing Ag nanoparticles (Ag NPs) on the surface to further improve its catalytic activities. The Ag NPs decorated Co3V2O8 (ACVPs) showed both excellent oxidase- and peroxidase-like catalytic activities, which can oxidize the colorless 3,3',5,5'-tetramethylbenzidine rapidly to induce a blue change. The enhanced enzyme mimetic activities can be attributed to the surface plasma resonance (SPR) effect of Ag NPs as well as the synergistic catalytic effect between Ag NPs and Co3V2O8. On this basis, ACVPs were applied in constructing colorimetric sensor for detection of Hg2+ and Cd2+, validating the occurrence of Fenton reaction, and disinfection reaction, presenting favorable catalytic performances. Furthermore, the enzyme-like catalytic mechanism was studied, indicating the chief role of ×O2- radicals in the catalytic process. This work not only discover a novel functional material with double enzyme mimetic activities but also provides a new insight into exploiting artificial enzyme mimics with highly efficient catalytic abilities.
A nano-sized Cu-MOF with high peroxidase-like activity and its potential application in colorimetric detection of H2O2 and glucose

Abstract:Peroxidase widely exists in nature and can be applied for the diagnosis and detection of H2O2, glucose, ascorbic acid and other aspects. However, the natural peroxidase has low stability and its catalytic efficiency is easily affected by external conditions. In this work, a copper-based metal-organic framework (Cu-MOF) was prepared by hydrothermal method, and characterized by means of XRD, SEM, FT-IR and EDS. The synthesized Cu-MOF material showed high peroxidase-like activity and could be utilized to catalyze the oxidation of o-phenylenediamine (OPDA) and 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2. The steady-state kinetics experiments of the oxidation of OPDA and TMB catalyzed by Cu-MOF were performed, and the kinetic parameters were obtained by linear least-squares fitting to Lineweaver-Burk plot. The results indicated that the affinity of Cu-MOF towards TMB and OPDA was close to that of the natural horseradish peroxidase (HRP). The as-prepared Cu-MOF can be applied for colorimetric detection of H2O2 and glucose with wide linear ranges of 5 to 300 μM and 50 to 500 μM for H2O2 and glucose, respectively. Furthermore, the specificity of detection of glucose was compared with other sugar species interference such as sucrose, lactose and maltose. In addition, the detection of ascorbic acid and sodium thiosulfate was also performed upon the inhibition of TMB oxidation. Based on the high catalytic activity, affinity and wide linear range, the as-prepared Cu-MOF may be used for artificial enzyme mimics in the fields of catalysis, biosensors, medicines and food industry.

Abstract:This work reports the synthesis, characterization, and application of bifunctional semiconducting CuO nanotubes (NTs) electrode for innovative synergized cathodic photoelectrochemical (PEC) enzymatic bioanalysis. Specifically, CuO NTs electrode was fabricated by surface oxidation of the copper foil in an alkaline aqueous solution with (NH4)2S2O8 and then annealed in air at 200 °C. After the subsequent coupling with the model enzyme of xanthine oxidase (XOD), the resulted photocathodic enzyme biosensor exhibited good analytical performance of rapid response, high stability, and good sensitivity. Especially, due to the unique catalytic property of CuO toward H2O2, a novel enzymatic cascade design between biological catalyst (XOD as natural enzyme) and biomimetic catalyst (CuO as the peroxidase mimetics) was constructed, and the dual-catalyst system with special synergy effect could achieve the cathodic PEC guanine bioanalysis with enhanced efficiency. In the determination, the cathodic photocurrent was found to be proportional to the guanine concentration, which was different from the commonly observed O2-dependent suppression of the photocurrent. In all, such a bifunctional CuO NTs-based PEC bioassay format has not been reported. The success of this work can offer great chances for further development and implementation of novel CuO-based PEC bioanalytical systems. More importantly, the strategy proposed here could contribute to the development of an original prototype for general PEC enzymatic bioanalysis.
Iron-carbon dots embedded in molybdenum single-atom nanoflowers as multifunctional nanozyme for dual-mode detection of hydrogen peroxide and uric acid

Abstract:Single-atom catalysts (SACs) have attracted extensive attention in the field of catalysis due to their excellent catalytic ability and enhanced atomic utilization, but the multi-mode single-atom nanozymes for biosensors remain a challenging issue. In this work, iron-doped carbon dots (Fe CDs) were loaded onto the edges and pores of Mo SACs with nanoflower morphology; accordingly, a composite material Fe CDs/Mo SACs was prepared successfully, which improves the catalytic performance and develops a fluorescence mode without changing the original morphology. The steady-state kinetic data indicates that the material prepared have better affinity for substrates and faster reaction rates under optimized conditions. The specific kinetic parameters Km and Vmax were calculated as 0.39 mM and 7.502×10-7 M·s-1 respectively. The excellent peroxidase-like activity of Fe CDs/Mo SACs allows H2O2 to decompose into •OH, which in turn oxidizes colorless o-phenylenediamine (OPD) to yellow 2,3-diaminophenazine (DAP). At the same time, the fluorescence signal of Fe CDs/Mo SACs quenches obviously by DAP at 460 nm through internal filtration effect (IFE), while the characteristic fluorescence response of DAP gradually increases at 590 nm. Based on this sensing mechanism, a sensitive and accurate dual-mode (colorimetric and ratiometric fluorescent) sensor was constructed to detect H2O2 and uric acid, and the rate of recovery and linearity were acceptable for the detection of UA in human serum and urine samples. This method provides a new strategy for rapid and sensitive detection of UA, and also broadens the development of SACs in the field of biosensors.
Cu3(HITP)2 with peroxidase- and ascorbic acid oxidase-like catalytic activity for fluorescence/chemiluminescence sensing of ascorbic acid

Abstract:Nanomaterials with intrinsic enzyme mimicking activity have achieved widespread application. However, developing novel nanomaterials with multienzyme mimicry activity remains challenging. Herein, Cu3(HITP)2 (HITP = 2,3,6,7,10,11-hexaiminotriphenylene) with ascorbic acid oxidase (AAO)- and peroxidase (POD)-like activity are successfully synthesized. Cu3(HITP)2 exhibits excellent AAO-like activity and can specifically catalyze the oxidation reaction of ascorbic acid (AA). Dehydroascorbic acid (DHAA) obtained from the oxidation of AA is allowed to react with nonfluorescent o-phenylenediamine (OPD) to form 3-(1,2-dihydrox-yethyl) furo[3,4-b]quinoxaline-1-one (DFQ) with strong fluorescence emission. Moreover, Cu3(HITP)2 is able to catalyze the chemiluminescence (CL) reaction of ABEI-H2O2 to generate a strong and glow-type emission based on its POD activity. Inspired by the multienzyme mimicry activity of Cu3(HITP)2, the simple and sensitive fluorescence and chemiluminescence sensing platforms are successfully constructed and applied for the detection of AA. The sensors show high sensitivity and excellent selectivity. We believe that this multienzyme mimicry activity nanomaterial not only can be used to construct the multiple-mode biosensing platform, but also enables the extensive applications in the fields of biomedicine, energy, and environment.
Enhanced peroxidase‐like activity of MnFe2O4 nanoparticles on halloysite nanotubes for uric acid detection

Abstract:Nanozymes have significantly advanced sensing assays by replicating native enzyme functions. However, designing nanozymes with high catalytic activity and easy recyclability remains challenging. The study presented here has resulted in the development of a highly efficient and sensitive colorimetric system for the detection of uric acid, utilizing MnFe2O4@HNTs—a novel composite material consisting of MnFe2O4 loaded onto halloysite nanotubes. These nanocomposites exhibited outstanding peroxidase‐like activity and attractive magnetic properties. The catalytic efficiency of the MnFe2O4@HNTs in the oxidation of 3,3′,5,5′‐tetramethylbenzidine, in the presence of H2O2, was remarkable, leading to a distinct color change from colorless to blue. A linear relationship was observed between absorbance and UA concentration in the range of 1–20 μM, with a detection limit as low as 52 nM. Mechanistic investigations revealed that reactive oxygen species (ROS), specifically singlet oxygen (1O2), were generated through the decomposition of H2O2, which is responsible for the peroxidase‐like activity demonstrated by the MnFe2O4@HNTs. The method showed minimal interference from serum substances and high selectivity. Magnetic MnFe2O4 allowed easy separation and maintained over 95% activity after seven reuse cycles. The developed assay was successfully applied to the detection of uric acid in human serum, achieving recoveries greater than 98.60%. This research significantly advances the design of recyclable high‐performance nanozymes and establishes an effective colorimetric sensing platform for UA detection in clinical samples, potentially improving diagnostic tools for healthcare applications.

Abstract:Light-activated oxidase mimics have a wide application prospect in environment, detection, medicine and other fields because of its stability, environmental friendliness and high activity. Here a S-scheme heterojunction formed by NiO nanosheets modified with carbon quantum dots (CQDs) is prepared as a light-activated oxidase mimic. The modification of CQDs expands the light absorption range of NiO, while NiO as a matrix improves the dispersion of CQDs. Moreover, at the contact interface a S-scheme heterojunction forms, accelerating the separation of photoexcited electrons and holes under the built-in electric field and band bending. The unique transfer of photoexcited carriers maintains high oxidation and reduction activity of NiO@CQDs, improving its photocatalytic activity. NiO@CQDs can oxidize o-phenylenediamine (OPD) to yellow under light, and when Cu 2+ was present in the solution, this colorimetric reaction was inhibited. Therefore, NiO@CQDs could be used for colorimetric detection of Cu 2+ , with a wide linear range of detection (0–0.1 μM, 0.1–1 μM, 1–10 μM, 10–100 μM), low detection limit (0.024 μM), good selectivity and stability.

Abstract:The construction of simple and efficient artificial enzymes is of great importance in the fields of sensing and catalysis. In this work, peptide nanotubes (CA-PNTs) co-assembled from two short peptides were non-covalently combined with hemin to obtain a series of CA-PNTs/hemin composites as biomimetic peroxidase materials. Due to the introduction of histidine and arginine in the short peptides simulating the microenvironment near the hydrophobic cavity in the activity center of the natural enzyme, CA-PNTs/hemin shows enhanced peroxidase activity compared with free hemin and the single short peptide self-assembled peptide nanotubes-supported hemin complex, and the enzyme activity of the material could be regulated by simply adjusting the proportion of co-assembled short peptides. Further investigation of the catalytic mechanism demonstrates that CA-PNTs/hemin can effectively bind to H 2 O 2 to form active intermediates and catalyze the decomposition of H 2 O 2 to generate superoxide radicals (O 2 •− ), thus promoting the whole enzymatic reaction. Combined with an enzyme cascade reaction and enzyme catalytic chromogenic system, a colorimetric method based on CA-PNTs/hemin for uric acid (UA) sensing was constructed. The strategy shows a linear range of 0.5–80 μM with a low limit of detection (0.2 μM) and good selectivity, and has been successfully applied to the determination of UA in human urine. A portable platform for UA detection was further developed by conjugating the strategy with smartphones.

Abstract:Mimicking enzyme have significantly advanced sensing assays by replicating native enzyme functions, yet achieving both high catalytic activity and easy recyclability remains a challenge. In this study, Ag-doped MnFe2O4/halloysite nanotubes (HNTs) were rationally designed as a novel nanozyme by depositing in-situ Ag and MnFe2O4 nanoparticles onto HNTs. The resulting nanocomposite exhibited excellent peroxidase-like activity along with magnetic properties. Leveraging these features, a highly efficient and sensitive colorimetric system for detecting uric acid (UA) was developed. The Ag-doped MnFe2O4/HNTs catalyzed the oxidation of 3,3',5,5'-tetramethylbenzidine in the presence of H2O2, causing a color change from colorless to blue. The system showed a linear absorbance response to UA concentrations ranging from 1 to 20 μM, with a detection limit of 59 nM. Mechanistic studies revealed that reactive oxygen species intermediates (1O2) were generated through the decomposition of H2O2, leading to peroxidase-like activity in the Ag-doped MnFe2O4/HNTs. The assay was successfully applied to detect UA in human serum with recoveries over 99.68 %. This study indicates the successful application of Ag-doped MnFe2O4/HNTs for colorimetric UA detection in human serum. This research introduces a novel approach for designing recyclable, high-performance mimicking enzyme and establishes an effective colorimetric sensing platform for UA detection in human serum.

Abstract:BackgroundThe detection of hydrogen peroxide (H2O2) and nitrite ion (NO2-) is of great important in various fields including clinic, food, pharmaceutical and environmental analyses. Compared with many methods that have been developed for the determination of them, the electrochemical detection method has attracted much attention. In recent years, with the development of nanotechnology, many kinds of micro/nano-scale materials have been used in the construction of electrochemical biosensors because of their unique and particular properties. Among these catalysts, copper oxide (CuO), as a well known p-type semiconductor, has gained increasing attention not only for its unique properties but also for its applications in many fields such as gas sensors, photocatalyst and electrochemistry sensors. Continuing our previous investigations on transition-metal oxide including cuprous oxide and α-Fe2O3 modified electrode, in the present paper we examine the electrochemical and electrocatalytical behavior of flower like copper oxide modified glass carbon electrodes (CuO/GCE).ResultsFlower like copper oxide (CuO) composed of many nanoflake was synthesized by a simple hydrothermal reaction and characterized using field-emission scanning electron microscopy (FE-SEM) and X-ray diffraction (XRD). CuO modified glass carbon electrode (CuO/GCE) was fabricated and characterized electrochemically. A highly sensitive method for the rapid amperometric detection of hydrogen peroxide (H2O2) and nitrite (NO2-) was reported.ConclusionsDue to the large specific surface area and inner characteristic of the flower like CuO, the resulting electrode show excellent electrocatalytic reduction for H2O2 and oxidation of NO2-. Its sensitivity, low detection limit, fast response time and simplicity are satisfactory. Furthermore, this synthetic approach can also be applied for the synthesis of other inorganic oxides with improved performances and they can also be extended to construct other micro/nano-structured functional surfaces.

Abstract:Urea oxidation reaction (UOR) has been known as a viable method for renal/liver disease diagnostic detection. Here, we reported a three-dimensional (3D) nickel oxide nanoparticles dressed carbonized eggshell membrane (3D NiO/c-ESM) as a modified electrode toward urea detection. Several common physical measurements were employed to confirm its structural and morphological information. NiO/c-ESM modified electrode exhibits an outstanding performance for urea determination with a linear range from 0.05 to 2.5 mM, and limit detection of ~20 μM (3σ). This work offered a green approach for introducing 3D nanostructure through employing biowaste ESMs as templates, providing a typical example for producing new value-added nanomaterials with urea detection.
