Abstract:The structure-property relationship of carborane-modified iridium(III) complexes was investigated. Firstly, an efficient approach for the synthesis of o-carborane-containing pyridine ligands a-f in high yields was developed by utilizing stable and cheap B10H10(Et4N)(2) as the starting material. By using these ligands, iridium(III) complexes I-VII were efficiently prepared. In combination with DFT calculations, the photophysical and electrochemical properties of these complexes were studied. The hydrophilic nido-o-carborane-based iridium(III) complex VII showed the highest phosphorescence efficiency (abs. phi P =0.48) among known water-soluble homoleptic cyclometalated iridium(III) complexes and long emission lifetime (=1.24s) in aqueous solution. Both of them are sensitive to O-2, and thus endocellular hypoxia imaging of complex VII was realized by time-resolved luminescence imaging (TRLI). This is the first example of applying TRLI in endocellular oxygen detection with a water-soluble nido-carborane functionalized iridium(III) complex.

Abstract:Many luminescent probes have been developed for intracellular imaging and sensing. During cellular luminescence sensing, it is difficult to distinguish species generated inside cells from those internalized from extracellular environments since they are chemically the same and lead to the same luminescence response of the probes. Considering that endogenous species usually give more information about the physiological and pathological parameters of the cells while internalized species often reflect the extracellular environmental conditions, we herein reported a series of cyclometalated iridium(iii) complexes as phosphorescent probes that are partially retained in the cell membrane during their cellular uptake. The utilization of the probes for sensing and distinguishing between exogenous and endogenous analytes has been demonstrated using hypoxia and hypochlorite as two examples of target analytes. The endogenous analytes lead to the luminescence response of the intracellular probes while the exogenous analytes are reported by the probes retained in the cell membrane during their internalization.

Abstract:A series of phosphorescent zwitterionic iridium(III) complexes, with 4-carboxy-2, 2'-bipyridine-4'-carboxylate (Hdcbpy) as ancillary ligand, Ir(C(wedge)N)(2)(Hdcbpy) (C(wedge)N = 1-phenylpyrazole (ppz), 1-phenyl-pyridine (ppy), 2-(4',6'-difluoro-phenyl)pyridine (dfppy), 1-phenyl-isoquinoline (piq), dibenzo[f,h]quinoxaline (dbq)), were prepared and characterized. Their photophysical properties were studied, and intense luminescence emissions were observed based on metal-to-ligand-charge-transfer ((3)MLCT), ligand-to-ligand charge-transfer ((3)LLCT), ligand-centered transitions ((3)LC, i.e., (3)pi --> pi*), or intraligand-charge-transfer ((3)ILCT) excited states, which were confirmed by theoretical calculations. The quantum yield of Ir(dfppy)(2)(Hdcbpy) is as high as 0.106 in aqueous solution. With Hdcbpy as a hydrophilic part, their amphiphilic structures as further confirmed by X-ray single crystal data endow them with different solubilities in phosphate buffer solution (PBS, pH 7.0). The compounds were successfully applied as luminescent dyes for cell imaging in aqueous solution. Their different stain ability in cell imaging was fairly well supported by the experimental data based on the measurement of oil/water partition coefficients and encapsulation/release with liposomes.
Dual-Phosphorescent Iridium(III) Complexes Extending Oxygen Sensing from Hypoxia to Hyperoxia.

Abstract:Hypoxia and hyperoxia, referring to states of biological tissues in which oxygen supply is in sufficient and excessive, respectively, are often pathological conditions. Many luminescent oxygen probes have been developed for imaging intracellular and in vivo hypoxia, but their sensitivity toward hyperoxia becomes very low. Here we report a series of iridium(III) complexes in which limited internal conversion between two excited states results in dual phosphorescence from two different excited states upon excitation at a single wavelength. Structural manipulation of the complexes allows rational tuning of the dual-phosphorescence properties and the spectral profile response of the complexes toward oxygen. By manipulating the efficiency of internal conversion between the two emissive states, we obtained a complex exhibiting naked-eye distinguishable green, orange, and red emission in aqueous buffer solution under an atmosphere of N2, air, and O2, respectively. This complex is used for intracellular and in vivo oxygen sensing not only in the hypoxic region but also in normoxic and hyperoxic intervals. To the best of our knowledge, this is the first example of using a molecular probe for simultaneous bioimaging of hypoxia and hyperoxia.

Abstract:Hypoxia is an important characteristic of malignant solid tumors and is considered as a possible causative factor for serious resistance to chemo-and radiotherapy. The exploration of novel fluorescent probes capable of detecting hypoxia in solid tumors will aid tumor diagnosis and treatment. In this study, we reported the design and synthesis of a series of "off-on" phosphorescence probes for hypoxia detection in adherent and three-dimensional multicellular spheroid models. All of the iridium(III) complexes incorporate an azo group as an azo-reductase reactive moiety to detect hypoxia. Reduction of non-phosphorescent probes Ir1-Ir8 by reductases under hypoxic conditions resulted in the generation of highly phosphorescent corresponding amines for detection of hypoxic regions. Moreover, these probes can penetrate into 3D multicellular spheroids over 100 mu m and image the hypoxic regions. Most importantly, these probes display a high selectivity for the detection of hypoxia in 2D cells and 3D multicellular spheroids.

Abstract:It is well known that hypoxia is a critical parameter with respect to several pathologies, like solid tumors. Hence, it is necessary to realize highly sensitive and selective monitoring of oxygen levels in biological systems. Phosphorescent iridium(III) complexes are an excellent class of oxygen probe through the energy transfer from triplet excitons to oxygen molecules. Current research mainly focuses on the neutral and cationic complexes, while the anionic ones received few attentions. Herein, we designed a series of anionic iridium(III) complexes with different CISI ligands and investigated their oxygen-0sensing capability in solution and living cells. Importantly, based on the metathesis reactions of the anionic iridium(III) complexes with oxygen-0insensitive cationic polyfluorene, water-soluble polymer soft salt P1 was synthesized successfully. Thus, ratiometric luminescent and reversible polymer hypoxia probe can be obtained because P1 can give dual emission with oxygen-sensitive phosphorescence from the iridium(III) complexes and reference fluorescence from polyfluorene, respectively. Furthermore, the photoluminescence lifetime imaging at different oxygen levels exhibited that P1 is an excellent oxygen probe utilizing the sensitive phosphorescent lifetime to oxygen level. To our knowledge, this is the first report about the application of anionic iridium(III) complexes and their conjugated polymer soft salts in biosensing and bioimaging fields. (C) 2018 Elsevier B.V. Abstract:<p>Hypoxia is considered as a key microenvironmental feature of solid tumors. Luminescent transition metal complexes particularly those based on iridium and ruthenium have shown remarkable potentials for constructing sensitive oxygen‐sensing probes due to their unique oxygen quenching pathway. However, the low aqueous solubility of these complexes largely retards their sensing applications in biological media. Moreover, it remains difficult so far to use the existing complexes typically possessing only one luminescent domain to quantitatively detect the intratumoral hypoxia degree. Herein, an Ir(III) complex showing red emissions is designed and synthesized, and innovatively encapsulated within a hydrophobic pocket of Cyanine7‐modified cyclodextrin. The Ir(III) complex enables the oxygen detection, while the cyclodextrin is used not only for improving the water solubility and suppressing the luminescence quenching effect of the surrounding aqueous media, but also for carrying Cyanine7 to establish a ratiometric oxygen fluorescence probe. 2D nuclear magnetic resonance is carried out to confirm the host–guest structure. The oxygen‐responsive ability of the resulting ratiometric probe is evaluated through in vitro cell and multicellular experiments. Further animal studies about tumor oxygen level mapping demonstrate that the probe can be successfully used for quantitatively visualizing tumor hypoxia in vivo.</p>

Abstract:AbstractIridium complex, a promising organic light-emitting diode for next generation television displays, emits phosphorescence. Phosphorescence is quenched by oxygen. We used this oxygen-quenching feature for imaging tumor hypoxia. Red light-emitting iridium complex Ir(btp)~2~(acac) (BTP) presented hypoxia-dependent light emission in culture cell lines, whose intensity was in parallel with HIF-1[alpha] expression. BTP was further applied to imaging five tumors (four from human origin and one from mouse origin) transplanted in athymic mice. All tumors presented a bright BTP-emitting image even 5 min after the injection. The BTP-dependent tumor image peaked at 1 to 2 h after the injection, and was then cleared from tumors within 24 h. The minimal BTP image recognition size was 3 to 4 mm in diameter. Compared with ^18^F-FDG/PET images, BTP delineated a clearer image for a tumor profile. We suggest that iridium complex has a vast potential for imaging hypoxic lesions such as tumor tissues.

Abstract:Oxygen plays a crucial role in many biological processes. Accurate monitoring of oxygen level is important for diagnosis and treatment of diseases. Autofluorescence is an unavoidable interference in luminescent bioimaging, so that an amount of research work has been devoted to reducing background autofluorescence. Herein, a phosphorescent iridium(III) complex-modified nanoprobe is developed, which can monitor oxygen concentration and also reduce autofluorescence under both downconversion and upconversion channels. The nanoprobe is designed based on the mesoporous silica coated lanthanide-doped upconversion nanoparticles, which contains oxygen-sensitive iridium(III) complex in the outer silica shell. To image intracellular hypoxia without the interferences of autofluorescence, time-resolved luminescent imaging technology and near-infrared light excitation, both of which can reduce autofluorescence effectively, are adopted in this work. Moreover, gradient O2 concentration can be detected clearly through confocal microscopy luminescence intensity imaging, phosphorescence lifetime imaging microscopy, and time-gated imaging, which is meaningful to oxygen sensing in tissues with nonuniform oxygen distribution.
Iridium(Iii) Anthraquinone Complexes As Two-Photon Phosphorescence Probes for Mitochondria Imaging and Tracking under Hypoxia

Abstract:In the present study, four mitochondria-specific and two-photon phosphorescence iridium(III) complexes, Ir1-Ir4, were developed for mitochondria imaging in hypoxic tumor cells. The iridium(III) complex has two anthraquinone groups that are hypoxia-sensitive moieties. The phosphorescence of the iridium(III) complex was quenched by the functions of the intramolecular quinone unit, and it was restored through two-electron bioreduction under hypoxia. When the probes were reduced by reductase to hydroquinone derivative products under hypoxia, a significant enhancement in phosphorescence intensity was observed under one-(lambda = 405 nm) and two-photon (lambda = 720 nm) excitation, with a two-photon absorption cross section of 76153 GM at lambda = 720 nm. More importantly, these probes possessed excellent specificity for mitochondria, which allowed imaging and tracking of the mitochondrial morphological changes in a hypoxic environment over a long period of time. Moreover, the probes can visualize hypoxic mitochondria in 3D multicellular spheroids and living zebrafish through two-photon phosphorescence imaging.

One of them, BTPHSA, depicted clear imaging from tumors transplanted 6 to 7 mm deep from the skin surface. Abstract:The early detection of cancer shows great promise for the control and prevention of cancer. For early detection, one of the challenges that still exists is searching for methods that can illuminate tumors with high sensitivity. Here, acidity and hypoxia, two typical features that exist universally in a solid tumor microenvironment, were focused on to attain synergistic imaging with an enhanced signal-to-noise ratio. This was realized using an iridium(iii) based optical probe (Ir-1) that could sense acidity and hypoxia simultaneously and synergistically. Through the synergistic sensing of acidic pH and hypoxia, stronger emission signals or larger lifetime changes can be obtained than if a single factor (acidity or hypoxia) is used to induce variations. Furthermore, its potential for biological applications was confirmed by employing Ir-1 for phosphorescence synergistic intensity and lifetime imaging of acidity and hypoxia in live monolayer cells and 3D multicellular spheroids.

Abstract:Real-time monitoring of the contents of molecular oxygen (O2) in tumor cells is of great significance in early diagnosis of cancer. At the same time, the photodynamic therapy (PDT) could be realized by highly toxic singlet oxygen (1O2) generated in situ during the O2 sensing, making it one of the most promising methods for cancer therapy. Herein, the iridium(III) complex cored hyperbranched phosphorescent conjugated polymer dots with the negative charges for hypoxia imaging and highly efficient PDT was rationally designed and synthesized. The incomplete energy transfer between the polyfluorene and the iridium(III) complexes realized the ratiometric sensing of O2 for the accurate measurements. Furthermore, the O2-dependent emission lifetimes are also used in photoluminescence lifetime imaging and time-gated luminescence imaging for eliminating the autofluorescence remarkably to enhance the signal-to-noise ratio of imaging. Notably, the polymer dots designed could generate the 1O2 effectively in aqueous solution, and the image-guided PDT of the cancer cells was successfully realized and investigated in detail by confocal laser scanning microscope. To the best of our knowledge, this represents the first example of the iridium(III) complex cored hyperbranched conjugated polymer dots with the negative charges for both hypoxia imaging and PDT of cancer cells simultaneously.

Abstract:Mitochondria are one of the major sources of cellular reactive oxygen species (ROS) and the hypochlorite ion (ClO-) is one kind of highly ROS involved in many important biological events. Herein, we present a novel class of mitochondrial ClO- probes. A series of iridium(iii) complexes, Ir1-Ir4, were synthesized which exhibited a highly specific response of their phosphorescence signal toward ClO- over other ROS. By changing the cyclometalated ligands, the turn-on phosphorescent emission colours can be tuned from green to red. ICP-MS results and Mito-tracker co-staining experiments revealed that Ir1-Ir4 localized specifically in mitochondria. Finally, complexes Ir1-Ir4 were successfully applied in subcellular imaging for detection of endogenous ClO- in mitochondria.

Abstract:In this work, two new biscyclometalated Ir(III) complexes with diimine ligands were synthesized. The compounds were characterized by 1D and 2D NMR spectroscopy and HR ESI mass spectrometry. All complexes exhibit efficient phosphorescence with pronounced sensitivity to the presence of oxygen. The photophysical properties of the obtained compounds, including absorption, emission spectra, lifetimes and quantum yields of phosphorescence were measured in various aqueous and model biological media with variations in pH, temperature and at various O2 concentrations. The complexes exhibit a very good sensory response to the changes in oxygen concentration, resulting in an increase in quantum yields and lifetimes by 2.8–5.5 times upon oxygen removal from aerated aqueous solutions. Biological tests with CHO‐K1 cell line have shown that these compounds display low toxicity, rapid internalization into cells and localization predominantly in lysosomes. For the most promising complex the phosphorescence lifetime imaging experiments were conducted, revealing that this sensor markedly changes the phosphorescence lifetime values in cells from 1.8 to 4.1 μs upon transition from normoxia to simulated hypoxia. The obtained results indicate that this type of iridium chromophores can be effectively used for assessing the oxygen status of living objects by using time‐resolved luminescent microscopy.

Abstract:Chemiluminescence (CL) probes that possess near-infrared (NIR) emission are highly desirable for in vivo imaging due to their deeper tissue penetration ability and intrinsically high sensitivity. Herein, a novel iridium-based CL probe () with direct NIR emission was reported as the result of hypochlorous acid (HClO)-initiated oxidative deoximation. To improve its biocompatibility and extend the CL time for in vivo imaging applications, this was prepared as a CL nanoparticle probe (s) through encapsulation by an amphiphilic polymer Pluronic F127 (F127). All results demonstrate that the have good selectivity and sensitivity for visualization of HClO even at the depth of 1.2 cm. Owing to these advantages, the CL imaging of exogenous and endogenous HClO in mice was achieved. This study could provide new insights into the construction of new NIR emission CL probes and expand their applications in biomedical imaging.

Abstract:Phosphorescent iridium complexes ranged from far-red to NIR have attracted great attention as oxygen probes or photosensitizers recently. In this work, a far-red phosphorescent iridium complex ((DPQ)2Ir(acac)) was adopted to prepare biocompatible nanoparticles (Ir-NPs) for both phosphorescence imaging and photodynamic therapy of living cells. The iridium complex was highly sensitive to oxygen in organic solvent, but became less insensitive after being incorporated into NPs, though the particle matrix was highly permeable to oxygen. Moreover, the Ir-NPs exhibited a fast rate of singlet oxygen generation under 660 nm light irradiation. Taking advantage of these Ir-NPs, cellular imaging in the far-red range was realized; meanwhile, in vitro PDT was successfully performed. These results suggested that the Ir-NPs can function as both bio-imaging agents and nano-photosensitizers that work in the far red range.

Abstract:A new phosphorescent dinuclear cationic iridium(III) complex (Ir1) with a donor-acceptor-pi-bridge-acceptor-donor (D-A-pi-A-D)-conjugated oligomer (L1) as a N<^>N ligand and a triarylboron compound as a C<^>N ligand has been synthesized. The photophysical and excited-state properties of Ir1 and L1 were investigated by UV/Vis absorption spectroscopy, photoluminescence spectroscopy, and molecular-orbital calculations, and they were compared with those of the mononuclear iridium(III) complex [Ir(Bpq)(2)(bpy)]+PF6- (Ir0). Compared with Ir0, complex Ir1 shows a more-intense optical-absorption capability, especially in the visible-light region. For example, complex Ir1 shows an intense absorption band that is centered at lambda=448 nm with a molar extinction coefficient (epsilon) of about 10(4), which is rarely observed for iridium(III) complexes. Complex Ir1 displays highly efficient orange-red phosphorescent emission with an emission wavelength of 606 nm and a quantum efficiency of 0.13 at room temperature. We also investigated the two-photon-absorption properties of complexes Ir0, Ir1, and L1. The free ligand (L1) has a relatively small two-photon absorption cross-section (delta(max)=195 GM), but, when complexed with iridium(III) to afford dinuclear complex Ir1, it exhibits a higher two-photon-absorption cross-section than ligand L1 in the near-infrared region and an intense two-photon-excited phosphorescent emission. The maximum two-photon-absorption cross-section of Ir1 is 481 GM, which is also significantly larger than that of Ir0. In addition, because the strong B-F interaction between the dimesitylboryl groups and F- ions interrupts the extended pi-conjugation, complex Ir1 can be used as an excellent one-and two-photon-excited "ON-OFF" phosphorescent probe for F- ions.
