Abstract:An asymmetric hydrogenation reaction of benzoxazinones has been accomplished via a relay iron/chiral Brønsted acid catalysis. This approach provides a variety of chiral dihydrobenzoxazinones in good to high yields (75-96%) and enantioselectivities (up to 98:2 er). It is noteworthy that challenging 3-alkyl-substituted substrates underwent highly enantioselective reduction. A key to success is the utilization of a nonchiral phosphine ligand to reduce disadvantageous background reactions through tuning the catalytic activity of Fe3(CO)12.

The pyrazole moiety played an important role in providing H-bond acceptor sites, which is critical for achieving high reactivities and enantioselectivities.

Abstract:A subtle change in the ligand structure, replacing the carbonyl oxygen with sulfur in simple alpha-amino acid amides, resulted in a dramatic activity and selectivity improvement in the rhodium- or ruthenium-catalyzed reduction of ketones under hydrogen transfer conditions. In addition, in most cases, a switch of the product's absolute configuration was observed on going from amides to the corresponding thioamides. Under optimized conditions, we obtained the secondary alcohol products in high yield and enantioselectivity (up to 97% ee) using only 0.25 mol % catalyst loading. [structure: see text]

Abstract:The asymmetric transfer hydrogenation of 3‐substituted 2H‐1,4‐benzoxazines with an azeotropic mixture of HCO2H/NEt3 (5/2) using tethered Cp*Rh(III)‐diamine catalysis has been realized. This process allows access to a broad range of chiral 3,4‐dihydro‐2H‐1,4‐benzoxazines in high yields with up to 99% ee, and tolerates a variety of functional groups. The enantiocontrol is achieved by the judicious choice of catalyst and hydrogen source. This reaction proceeds with unexpected reversal of enantioselectivity, which is attributed to the acidic reaction conditions and the hydrogen bond between the N−H of the rhodium species and the O atom in the substrate.

Abstract:Pnictogen-bonding catalysis based on σ-hole interactions has recently attracted the attention of synthetic chemists. As a proof-of-concept for asymmetric pnictogen-bonding catalysis, we report herein an enantioselective transfer hydrogenation of benzoxazines catalyzed by a novel chiral antimony cation/anion pair. The chiral pnictogen catalyst library could be rapidly accessed from triarylstibine with readily available mandelic acid analogues, and the catalyst displays remarkable efficiency and enantiocontrol potency even at 0.05 mol % loading. Abstract:The highly enantioselective transfer hydrogenation of benzoxazinones with chiral phosphoric acids under H2 was successfully achieved, where boranes promoted the hydrogenation of phenanthridine for the regeneration of dihydrophenanthridine as the hydrogen donor. A variety of dihydrobenzoxazinones were obtained in high yields with up to 99% ee. The current work provides a promising solution to unreactive substrates for frustrated Lewis pair-catalyzed asymmetric hydrogenation.

Abstract:An enantioselective reduction of simple carbenium ions with cyclohexadienes containing a hydridic C-H bond at an asymmetrically substituted carbon atom is disclosed. The net reaction is a transfer hydrogenation of alkenes (styrenes) only employing chiral cyclohexadienes as dihydrogen surrogates. The trityl cation is used to initiate a Brønsted acid-promoted process, in which a delicate intermolecular capture of a carbenium-ion intermediate by the aforementioned chiral hydride source is enantioselectivity determining. Exclusively non-covalent interactions are rendering one of the transition states energetically more favored, giving the reduction products in good enantiomeric ratios. The computed reaction mechanism supports the present findings as well as previous results obtained from studies on other transfer-hydrogenation methods involving the cyclohexadiene platform.

Abstract:A highly chemo- and enantioselective hydrogenation of β-diketones was achieved by using [Ru(benzene)(S)-SunPhosCl]Cl for consistency in THF. The neighboring heteroatoms played important roles in guaranteeing the reactivity and controlling the chemoselectivity. These results suggested a potential approach for the clean and facile synthesis of functionalized chiral β-hydroxy ketones, which could otherwise be prepared through much less step-economic transformations.

Abstract:A series of 3-oxoglutaric acid derivatives have been hydrogenated in different solvents in the presence of [RuCl(benzene)(S)-SunPhos]Cl (SunPhos=(2,2,2',2'-tetramethyl-[4,4'-bibenzo[d][1,3]dioxole]-5,5'-diyl)bis(diphenylphosphine)). Unlike simple beta-keto acid derivatives, these advanced analogues can be readily hydrogenated in uncommon solvents such as THF, CH2Cl2, acetone, and dioxane with high enantioselectivities. Two possible catalytic cycles have been proposed to explain the different reactivities of these 1,3,5-tricarbonyl substrates in the tested solvents. The C-2 and C-4 substituents had notable but irregular influence on the reactivity and enantioselectivity of the reactions. More pronounced solvent effects were observed: the ee values increased from around 20?% in EtOH or THF to 90?% in acetone. Inversion of the product configuration was observed when the solvent was changed from EtOH to THF or acetone, and a mixed solvent system can lead to better enantioselectivity than a single solvent.

Abstract:A sustainable and simple catalytic system for the atom-economical alkynylation of benzamides with low nickel loadings is described. No organic or metallic oxidants and expensive ligands are required. A broad range of benzamides and bromoalkynes bearing various synthetically useful functional groups are compatible with this reaction. The versatility of this operationally simple protocol has been further demonstrated by the controllable mono-and di-alkynylation. Importantly, substrate/catalyst ratios of up to 200, and a turnover number of 196 were achieved, highlighting the potential of this protocol for synthetic applications.

Abstract:The transfer hydrogenation of alkenes was realized by using a simple transition-metal compound (FeBr 3 ) and 1,4-cyclohexadiene (1,4-CHD) as the hydrogen donor. The conversion of a number of di- and trisubstituted alkenes was investigated, and even a tetrasubstituted alkene was successfully converted. Compared with previously published work with the more expensive InBr 3 , the reaction times were considerably reduced and significantly milder reaction conditions could be applied. Interestingly, a transformation that was catalytic in 1,4-CHD, with molecular hydrogen as a stoichiometric reducing agent at 1 bar, was also accomplished. Abstract:Chiral tetrahydroquinoxalines and dihydroquinoxalinones represent the core structure of many bioactive molecules. Herein, a simple and efficient Rh-thiourea-catalyzed asymmetric hydrogenation for enantiopure tetrahydroquinoxalines and dihydroquinoxalinones was developed under 1 MPa H pressure at room temperature. The reaction was magnified to the gram scale furnishing the desired products with undamaged yield and enantioselectivity. Application of this methodology was also conducted successfully under continuous flow conditions. In addition, H NMR experiments revealed that the introduction of a strong Brønsted acid, HCl, not only activated the substrate but also established anion binding between the substrate and the ligand. More importantly, the chloride ion facilitated heterolytic cleavage of dihydrogen to regenerate the active dihydride species and HCl, which was computed to be the rate-determining step. Further deuterium labeling experiments and density functional theory (DFT) calculations demonstrated that this reaction underwent a plausible outer-sphere mechanism in this new catalytic transformation.

Abstract:An enantioselective hydrogenation of hydrazones derived from heterocyclic ketones was developed with up to 85% ee. The enantiomeric purity was enriched to >99% ee by crystallization from EtOAc in >80% yield. Optimization studies have revealed a notable solvent effect that resulted in inversion of enantioselectivity from 85% ee in MeOH to −27% ee in DCE. The hydrazone geometry and possible hydrogenation via endocyclic alkene were examined as possible factors for the inversion of enantioselectivity.

Abstract:A chiral nickel complex that catalyzed asymmetric transfer hydrogenation of α-substituted acrylic acids under mild conditions and avoided the use of high-pressure hydrogen gas was developed. The products included chiral β-amino acids and α-methyl carboxylic acids such as three nonsteroidal anti-inflammatory profens. Deuterium-labeling experiments and DFT studies pointed to an unconventional protonation of a metalacyclopropane complex formed by α-phenylacrylic acid which was hydrogen bonded with formic acid. An alternative conventional pathway of nickel hydride insertion cannot explain selective deuteration at β-position of α-phenylacrylic acid when HCO2D was used.

Abstract:The regioselective hydrogenation of a, b-unsaturated carbonyl compounds is an extremely important transformation. However, its development is hampered by a lack of precise and efficient catalysts. In this work, triggered by the steric hindrance and confinement capacity of the catalyst, we designed a novel catalyst FLP/MIL-101(Cr), with the bulky frustrated Lewis pair (FLP) of PPh3 and B(C6F5)3 immobilized in NH2-MIL-101(Cr). In the well-tailored FLP/MIL-101(Cr), the promoted selective reduction behaviors were realized, with cinnamyl alcohol selectivity up to 99 % and the complete substrate conversion in cinnamyl aldehyde reduction. Experimental characterizations and theoretical calculations indicated that the C=N bond and p-p stacking interaction between FLP and the organic ligand of MIL-101(Cr) implemented and reinforced a rigid structure. Compared with homogeneous FLP, the special nano-space in FLP/MIL-101(Cr) catalyst benefits the selective reduction behavior towards the unsaturated bond with the smaller steric hindrance in a,b-unsaturated carbonyl compounds. As expected, FLP/MIL-101(Cr) showed good stabilities of skeleton structure and catalytic performance. This contribution provides an ingenious strategy to design FLP heterogeneous catalysts and represents its utilization possibility in selective reduction. (c) 2023 Elsevier Inc. All rights reserved.
