Abstract:Highly enantioselective carbonyl-ene reaction of glyoxal derivatives and glyoxylate with various alkenes was accomplished using a novel nickel (II)-N,N'-dioxide complex under mild conditions, which facilitated the asymmetric synthesis of biologically interesting a-hydroxy carbonyl compounds. Various aromatic, aliphatic, and heteroaromatic glyoxal derivatives, as well as glyoxylate, could be tolerated in this system and afforded the corresponding adducts in excellent enantioselectivities (97->99% ee) with high yields. Moreover, the catalyst loading could even be decreased to 1 mol%, while the enantioselectivity was basically maintained.

Abstract:AbstractThe transition‐metal‐catalyzed C−N cross‐coupling has revolutionized the construction of amines. Despite the innovations of multiple generations of ligands to modulate the reactivity of the metal center, ligands for the low‐temperature enantioselective amination of aryl halides remain a coveted target of catalyst engineering. Designs that promote one elementary reaction often create bottlenecks at other steps. We here report an unprecedented low‐temperature (as low as −50 °C), enantioselective Ni‐catalyzed C−N cross‐coupling of aryl chlorides with sterically hindered secondary amines via a kinetic resolution process (s factor up to >300). A bulky yet flexible chiral N‐heterocyclic carbene (NHC) ligand is leveraged to drive both oxidative addition and reductive elimination with low barriers and control the enantioselectivity. Computational studies indicate that the rotations of multiple σ‐bonds on the C2‐symmetric chiral ligand adapt to the changing needs of catalytic processes. We expect this design would be widely applicable to diverse transition states to achieve other challenging metal‐catalyzed asymmetric cross‐coupling reactions.

Abstract:The chemical synthesis of organic molecules involves, at its very essence, the creation of carbon–carbon bonds. In this context, the aldol reaction is among the most important synthetic methods, and a wide variety of catalytic and stereoselective versions have been reported. However, aldolizations yielding tertiary aldols, which result from the reaction of an enolate with a ketone, are challenging and only a few catalytic asymmetric Mukaiyama aldol reactions with ketones as electrophiles have been described. These methods typically require relatively high catalyst loadings, deliver substandard enantioselectivity or need special reagents or additives. We now report extremely potent catalysts that readily enable the reaction of silyl ketene acetals with a diverse set of ketones to furnish the corresponding tertiary aldol products in excellent yields and enantioselectivities. Parts per million (ppm) levels of catalyst loadings can be routinely used and provide fast and quantitative product formation in high enantiopurity. In situ spectroscopic studies and acidity measurements suggest a silylium ion based, asymmetric counteranion-directed Lewis acid catalysis mechanism.

Abstract:Catalytic enantioselective conjugate additions with easily accessible alkenylboronic acid pinacol esters as nucleophiles promoted by chiral copper complexes of N-heterocyclic carbenes are presented. These processes constitute an unprecedented instance of conjugate additions of a variety of functionalized alkenyl groups and afford desired products that are otherwise difficult to access in up to 98% yield and 99.5:0.5 enantiomeric ratio. The origins of ligand-controlled enantioselectivity are elucidated with density functional theory (DFT) calculations.

Abstract:An approach to the α,β-unsaturated acyl azolium has been developed that exploits N-heterocyclic carbenes (NHCs) and acyl fluorides, without additional oxidants, bases, or preactivated pro-nucleophiles. These conditions have been applied in four classes of NHC-catalyzed reaction. Abstract:A series of C-2' modified cinchonine-derived phase-transfer catalysts were synthesized and used in the enantioselective photo-organocatalytic aerobic oxidation of β-dicarbonyl compounds with excellent yields (up to 97%) and high enantioselectivities (up to 90% ee). Furthermore, the reaction was carried out in a flow photomicroreactor, in which the heterogeneous gas-liquid-liquid asymmetric photocatalytic oxidation reaction was performed affording good yields (up to 97%) and enantioselectivities (up to 86% ee) within 0.89 min.

Abstract:All-carbon quaternary stereocenters constitute a significant portion of natural products and bioactive compounds. Here we disclose a nickel and chiral amine cooperative catalysis to enable a highly enantioselective allylic alkylation reaction between α-branched aldehydes and a wide scope of allyl esters, allowing the all-carbon quaternary stereocenter to be accessed with excellent enantioselectivity (up to 98% ee) and structural diversity. Abstract:Combinatorial coordination chemistry strategy combined with high-throughput screening techniques has been successfully applied to engineering practical enantioselective catalysts for asymmetric hetero-Diels-Alder reaction. The reaction of Danishefsky's diene with a variety of aldehydes can be carried out with 0.1-0.005 mol % of H4-BINOL/Ti/H4-BINOL or H4-BINOL/Ti/H8-BINOL catalysts at room temperature under solvent- and MS-free conditions to afford dihydropyrone derivatives with up to quantitative yield and 99.8% ee.

Abstract:Asymmetric hydrogenation is one of the most powerful methods for the preparation of single enantiomer compounds. However, the chemo- and enantioselective hydrogenation of the relatively inert unsaturated group in substrates possessing multiple unsaturated bonds remains a challenge. We herein report a protocol for the highly chemo- and enantioselective hydrogenation of conjugated enynes while keeping the alkynyl bond intact. Mechanism studies indicate that the accompanying Zn 2+ generated from zinc reduction of the Co II complex plays a critical role to initiate a plausible Co I /Co III catalytic cycle. This approach allows for the highly efficient generation of chiral propargylamines (up to 99.9% ee and 2000 S/C) and further useful chemical transformations.

Abstract:Despite the fact that metal-catalyzed asymmetric alkylative cross-couplings have been well-established, enantioselective alkylative substitution of an unactivated C–O bond remains a challenge due to the lack of strategies to cleave the C–O bond and suppress β-H elimination as well as control stereochemistry simultaneously. Herein, the enantioselective alkylative activation of an unactivated C–O bond with β-H-containing alkylating reagents was described using a chiral nickel catalyst, and versatile axially chiral biaryls bearing alkyl moieties with different chain lengths were delivered in good yields and with high ee. Control experiments demonstrated the significant role of the solvent tetrahydrofuran to facilitate this transformation. DFT calculations revealed that the coordination of THF to Mg(II) is pivotal for suppressing β-H elimination during reductive elimination, thus unlocking how the solvent molecule affects the competing β-H elimination and reductive elimination dynamics in transition-metal-catalyzed alkylative cross-coupling reactions.

Abstract:Multifunctionalization from the interception of active intermediates is an attractive synthetic strategy for the efficient construction of complex molecular scaffolds in an atom and step economic fashion. However, the design of reactions involving metal carbynoids that exhibit carbene/carbocation behavior is currently limited, and developing catalyst-controlled highly enantioselective versions poses significant challenges. In this study, we present the first asymmetric trifunctionalization reactions with rhodium carbynoids. This reaction unveils the distinctive reactivity of the carbynoid precursor, enabling it to react with simultaneously two nucleophiles and one electrophile. This process involves the formation of two distinct carbene ylides with the alcohol/carbamate and the trapping of one ylide with the imine, resulting in the formation of three new bonds. Furthermore, this strategy allows for the divergent synthesis of a wide array of β-amino esters in high yields and exceptional enantioselectivity.

Abstract:A catalytic asymmetric carbonyl-ene reaction of beta, gamma-unsaturated alpha-ketoesters with 5-methyleneoxazolines was accomplished. The process was based on the utilization of a chiral N, N '-dioxide/Mg-II catalyst, providing the desired products with excellent outcomes (up to 99% yield, >99% ee) under mild reaction conditions. Based on the experimental investigations and previous reports, a possible transition state was proposed.
