Abstract:We here present a Förster resonance energy transfer (FRET)-based and environment-sensitive fluorescent probe VG-1 for vicinal-dithiol-containing proteins (VDPs). VG-1 uniquely contains two sites sensitive to the protein environment (SPE), thus it shows weak fluorescence in both blue and green channels (a low FRET efficiency) in solution. After specifically binding with VDPs, its fluorescence in the green channel increases, while that in the blue channel disappears, achieving the specific detection of VDPs. The obvious signal changes in fluorescence may be attributed to that the increased rigidity of the molecular skeletons causes the enhanced FRET efficiency. The probe also achieved the cell super-resolution imaging of VDPs and the confocal imaging of VDPs in zebrafish.

Abstract:Vicinal dithiol proteins (VDPs) facilitate cellular redox homeostasis, modulate protein synthesis and participate in post-translational modifications through the dynamic equilibrium of dithiol and disulfide bonds. Herein, an activatable red emitting fluorescent probe, VDP-red, is developed for detecting VDPs. With the aid of this probe, we have discovered for the first time a reduction in the levels of reduced VDPs in a stroke mouse model. This work provides a fresh viewpoint for understanding stroke mechanisms.

Abstract:A novel selective fluorescent chemosensor 1 has been synthesized with a phenanthrene-fused dipyrromethene structure. Selective binding of Cu2+ by 1 results in a complex that displays high selectivity and sensitivity for H2S. The signal transduction occurs via reversible formation-separation of the Cu-1 complex and CuS. Abstract:Environmentally sensitive fluorescent probes (including AIEgens) play pivotal roles in numerous biological studies. Many of these functional materials are developed based on the twisted intramolecular charge transfer (TICT) mechanism. However, the TICT tendency of dialkylated amino groups in biocompatible main-stream fluorophores (i.e., coumarins and rhodamines) is weak, limiting their sensitivities. Herein, by replacing dialkylated amino donors with an N-methylpyrrole group to enhance TICT, a simple and general method to engineer highly environmentally sensitive fluorescent probes is reported. This method yields a platter of colorful fluorescent probes that demonstrates outstanding polarity and viscosity sensitivity with large turn-on ratios (up to 191 times for polarity and 14 times for viscosity), as well as distinct aggregation-induced emission (AIE) characteristics. The utilities of these probes in both wash-free bioimaging and protein detections are also successfully demonstrated. It is expected that this molecular design strategy will inspire the creation of many environmentally sensitive probes.

Abstract:As an important sulfur-containing amino acid, aberrant levels of cysteine are closely related with an array of dieases. Although many Cys-specific fluorescent probes have been designed, most of them wouldn't be able to detect Cys in buffer solution, which limits the application of the probe in biological system. In this work, a novel near-infrared fluorescent probe Cys-WR with a large Stokes shift (about 110 nm) was developed for the detection of Cys by conjugating the fluorophore WR-4 with crotonate moiety. The probe exhibited off-on response to Cys with wide linear range of 0-100 mu M, as well as the color of the probe solution changes from deep pink to brown observed by naked eyes. Moreover, the probe showed little cytotoxicity. Imaging of Cys using this probe in living A549 cells and zebrafish had also been well demonstrated.

Abstract:Fluorescent probes have emerged as indispensable chemical tools to the field of chemical biology and medicine. The ability to detect intracellular species and monitor physiological processes has not only advanced our knowledge in biology but has provided new approaches towards disease diagnosis. In this review, we detail the design criteria and strategies for some recently reported fluorescent probes that can detect a wide range of biologically important species in cells and in vivo. In doing so, we highlight the importance of each biological species and their role in biological systems and for disease progression. We then discuss the current problems and challenges of existing technologies and provide our perspective on the future directions of the research area. Overall, we hope this review will provide inspiration for researchers and prove as useful guide for the development of the next generation of fluorescent probes.

Abstract:Development of multi-input fluorescent probes would facilitate the monitoring of multiple biomolecular events leading to a common disease pathology. Here, we reported a two input fluorescent probe(QME-N-3) based on quinoline scaffold with two distinct reactive sites, the alpha,beta-unsaturated carbonyl at 2 site and 6-azido group, which were specific acceptors of RSH and H2S respectively. The turn-on fluorescent probe could react with the two molecules via the Michael addition at the double bond for RSH and the reduction of the azide by H2S without mutual interference, to generate the intensively fluorescent product, which could act as a two-input AND logic gate. Furthermore, QME-N-3 has very low cytotoxicity and excellent stability, and the resulting sensing product has a high two-photon absorption cross section. Consequently, detection of intracellular thiols and H2S can be achieved by cell fluorescence imaging under both one- and two-photon excitations.

Abstract:The interactions between proteins and small molecules play an important role in the regulation of various cellular processes and modern drug discovery. Herein, we have developed a novel strategy for real-time monitoring the protein-ligand interaction based on vibration-induced emission (VIE) mechanism. These results demonstrated that the probe DPAC-DB could directly visualize the binding process in biotin-avidin system, which was undisturbed to other environmental stimuli, such as pH, interference species and temperature. Furthermore, the unique VIE effect caused by dramatic and reversible intramolecular vibrations enabled our strategy possible to build up the bio-logic gate. This method to efficiently control the emission of VIE-based probe through the reversible noncovalent protein-small molecules interactions opens new avenues for the development of potent protein pharmaceuticals and small molecule drugs.

Abstract:Detection of silver ions (Ag+) in living cells has becoming more and more attractive due to the important biological impact of Ag+ on cellular functions. Here, we put forward a new approach to realize the in situ fluorescence imaging and detection of Ag+ in single cells via an ultrasensitive Ag+-responsive probe, 3',6'-bis (diethylamino)-2-(2-iodoethyl) spiro[isoindoline-1,9'-xanthen]-3-one (BDISIX). In the presence of Ag+, the fluorescence of the probe can be turned 'on', generating strong red fluorescence. Using breast cancer cells (MCF-7) as the example, we successfully realize the imaging of intracellular Ag+ through one-step incubation of the probe, which is especially convenient and fast for the in situ intact detection of Ag+ in living cells.

Abstract:A novel seminaphthorhodafluor-based fluorescent probe was designed and prepared. It exhibited an outstanding off-on fluorescence response (about 45-fold enhancement) to Cu2+ because the structure changed from a spirocyclic form to a ring-open state. It showed satisfactory detection performances for Cu2+ with fast response speed, high selectivity and anti-interference, and a low detection limit (22.9 nM) due to its long emission wavelength and off-on fluorescence response mode. This probe was also satisfactorily applied for the monitoring of Cu2+ in living cells and zebrafish.

Abstract:Lipid droplets (LDs) and lysosomes are vital organelles that play crucial roles in various physiological and pathological processes. However, simultaneous two-color visualization of these two organelles using a single probe for cell homeostasis monitoring remains a challenge due to the lack of rational design strategies. To address this issue, we have developed an aggregation-induced emission (AIE) fluorescent probe named TPE-NDI-Mor with an electron donor (D)–acceptor (A) structure, which can stain both LDs and lysosomes with high selectivity through green and red fluorescence imaging, respectively. A detailed mechanism study revealed that TPE-NDI-Mor , with a twisted intramolecular charge transfer (TICT) effect, shows a high affinity for a polar microenvironment. Additionally, the probe also demonstrates good stability, high anti-interference performance and a large Stokes shift, making it suitable for visualizing cell homeostasis and further disease diagnosis by tracking the dynamic changes of LDs and lysosomes.

Abstract:Pathophysiological micro-environments such as viscosity, polarity and hypoxia are tightly correlated with regulating diffusion, transportation, and intermolecular interactions in living cells. However, the interplay and crosstalk among these key factors have not been clearly explored yet due to the lack of practical tools. In this work, based on logic-gate, we developed a versatile fluorescent probe (VPHPP) for simultaneously detecting viscosity, polarity and Nitroreductases (NTRs) in living cells. This probe was able to distinguish cancer cells from normal cells with signals in blue channel (lambda em = 540 nm) for NTRs and red channel (lambda em > 747 nm) for viscosity/polarity, which seemed more accurate (or more information at least) to define the cell types than testing only one factor. Meanwhile, the dynamic changes of viscosity, polarity and NTRs were simultaneously observed under hypoxic conditions in living cells for the first time. Furthermore, the detection system could image the simultaneous down-regulation of NTRs and the elevation of viscosity level (decreasing polarity degree) induced by dicoumarin in living cells under a confocal microscopy. Last but not least, VPHPP could achieve the imaging in tumor-bearing mouse model. In brief, VPHPP could be used as a promising visualizing agent in image-guided surgery.

Abstract:AML with eosinophilia belongs to the morphologic cytogenetic entity M1/M2 t(8;21) with the involvement of the genes AML1/ETO. These eosinophils differ only slightly from normal eosinophils with one rare exception i.e. Auer rods in eosinophils which has only been found on peroxidase staining: This subtype belongs to the good prognosis group of AML. AML with inv(16) (mostly M4Eo) per se is a morphologic-cytogenetic entity with inv(16),--rarely t(16;16), and the genes MYL 11/CBF beta involved. The eosinophils show special abnormalities. AML with inv(16) also belongs to the good prognosis group of AML.
