Abstract:Good sense: A highly sensitive and selective ratiometric fluorescent probe for Fe2+ was developed by linking an Fe2+-insensitive fluorophore to an Fe2+-sensitive reporter–fluorophore fragment. The fluorescence intensity ratio increased linearly with the concentration of Fe2+ (see scheme). The probe was applied to the ratiometric imaging of intracellular and adscititious fluctuations of Fe2+ in live HL-7702 and HepG2 cells.

Abstract:A simple ratiometric sensor (L-1) for the cascade detection of Zn2+ and H2PO4- with high selectivity was reported based on the intermolecular charge transfer (ICT) mechanism. This new sensor could distinguish Zn2+ from Cd2+, and features high fluorescence quantum yield with L-1 (0.45) and L-1-Zn2+ (0.27). There is a good linear relationship between the fluorescence ratio I-525 nm/I-430 nm upon addition of Zn2+ and the limit of detection (LOD) was evaluated to be 41.0 nM. Furthermore, the in situ prepared L-1-Zn2+ complex also displayed good selectivity and ratiometric response for H2PO4- and the detection limit of L-1-Zn2+ was found to be 49.0 nM. L-1 exhibits permeability and can been applied to the detection of intracellular Zn2+ and H2PO4- in HepG-2 cells.

Abstract:A multisignal sensor based on rhodamine B with a ferrocene substituent (FD7) has been developed as a highly sensitive chemosensor for Cr3+. A prominent fluorescence enhancement was found in the presence of Cr3+, which was accompanied by changes in the absorption spectrum and the electrochemical parameters. Furthermore, by means of confocal laser scanning microscopy experiments, it has been demonstrated that FD7 can be used as a fluorescent probe for monitoring Cr3+ in living cells.

Abstract:A ratiometric two-photon fluorescent probe (RN3) for Pd2+ was developed based on 7-substituted coumarin and rahodamine B through FRET mechanism. The probe exhibited highly sensitive and selective ratiometric fluorescent detection signal (575 nm/515 nm) for Pd2+ in aqueous solution. The two-photon absorption cross-section of the probe under adding Pd2+ was measured as 165 GM at 760 nm. RN3 could detect Pd2+ in living cells with low cytotoxicity under two-photon excitation (lambda(ex) = 760 nm). The bio-imaging results indicated that RN3 could work in deep tissue (similar to 130 mu m) and in five-days-old zebrafish with ratiometric fluorescent signal. (C) 2017 Elsevier B.V. Abstract:Colorimetric fluorescence probe B was synthesized from 2, 3, 3-trimethyl-3H-benzo [e] indole and p-dimethylaminobenzaldehyde using acidic solvent as catalyst and ethanol as solvent. In the mixed system (pH = 7. 4) of V(EtOH) V(HEPES) = 9 1, the color of solution B changed from pale yellow to purple-red after added Cr3+ to probe B solution, indicating that probe B can recognize Cr3+ with naked-eye. Fluorescence emission spectra shows that probe B had good selectivity of Cr3+, high sensitivity and good recognition ability with EDTA. The binding constant(K-a) of probe B on Cr3+ was 0.28 x 102 mol/L and the detection limit was 1. 90x 10(-8) mol/L. The detection limit was lower than the maximum Cr3+ content (9. 60 x 10(-8) mol/L) in drinking water stipulated by WHO. In addition, the concentration of Cr3+ in actual water samples was quantitatively detected by fluorescence emission spectra. Probe B has been applied to the detection of Cr3+ in living cells, which has a good application prospect and practical value.

Abstract:Carbon monoxide (CO) is an important gas signaling molecule and has been widely involved in regulating important life processes. Effective monitoring of CO in living systems is critical. Combined with the accuracy of ratio detection and the advantages of two-photon imaging, a simple ratiometric two-photon fluorescent probe RTFP was rationally designed and synthesized using 7-(diethylamino)-4-hydroxycoumarin as a two-photon fluorophore and allyl carbonate as the reactive unit. Probe RTFP exhibited excellent selectivity and sensitivity towards CO, and was successfully applied to image endogenous CO in living cells and zebrafish.

Abstract:A water-soluble rhodamine-based fluorescent probe (RD2) was designed and synthesized. Compared with the precursor RD1, RD2 shows excellent selectivity for Fe3+ over Cr3+ during the detection. The enhanced selectivity was achieved by incorporating another coordination moiety to the chelating moiety of the precursor RD1. Furthermore, bioimaging studies indicated that RD2 was cell permeable and suitable for the real-time imaging of Fe3+ in the living cells by confocal microscopy. (C) 2014 Elsevier B.V. Abstract:A new fluorescent probe, 3-((4-([2,2':6',2 ''-terpyridin]-4'-y1)phenyl)ethynyl)-7-methoxy-2H-chromen-2-one (ZC-F7) composed of coumarin as the fluorophore and terpyridine as the receptor is designed and synthesized. Based on the intramolecular charge transfer (ICT) effect, the probe exhibits significant variation on emission wavelengths with shifts more than 100 nm after combined with Zn2+, in accordance with the conversion of emission colors from blue to green. Good selectivity and sensitivity of this probe towards Zn2+ can be found even on the ppb level in aqueous solution. The Job's plot test suggests a 1:1 stoichiometry between ZC-F7 and Zn2+. The application of the fluorescence probe in bio-imaging is also demonstrated, proving its potential usage in fields such as environment protection, water treatment and safety inspection. Abstract:Isatin-3-(7′-Methoxychromone-3′-methylidene) hydrazone ( L ) was synthesized based on chromone schiff base, and used to construct a novel sensor to detect Cr 3+ . Fluorescence detection experiments were carried out for a range of different concentrations of Cr 3+ in aqueous solutions. A concentration calculation model was built on the basis of eliminating interference of excitation spectrum in the fluorescence spectra with mathematical method. Results showed that probe L displayed a 70 -fold fluorescence enhancement upon the addition of Cr 3+ due to the photo-induced electron transfer (PET) effect. On the other hand, metal ions except Cr 3+ did not cause significant change in either the absorption or the fluorescence spectrum of L. In addition, L showed a good selectivity to Cr 3+ over other metal cations, especially Al 3+ and Cu 2+ . The probe L can detect Cr 3+ highly and selectively by the direct chelation enhanced fluorescence with a detection limit of 3.14 × 10 −6 M. Furthermore, benefiting from their good water solubility and biocompatibility, cell imaging and real-time monitoring of Cr 3+ in living HepG2 cells were successfully achieved.

Abstract:A novel photoconvertible fluorescent probe, which can be activated by intracellular thiols, has been synthesized. Such a molecular probe comprises three parts: a 7-aminocoumarin phototrigger, a thiol-removable energy acceptor, and a caged fluorescein scaffold with intracellular thiols reactivity as the fluorescent reporter. Extracellularly, the energy acceptor blocks the emission of the coumarin that regulates the photocleavage and photoactivation of the fluorescein. Intracelluarly, the high concentration of thiols releases the energy acceptor, thus activating the S1 state of the phototrigger, which emits coumarin blue fluorescence for pre-visualization and liberates the caged green-fluorescent fluorescein to highlight the specific cell upon illumination. Compared to traditional photoactivated organic dyes, the intracellular thiols activated probe requires double activations: one by intracellular thiols and the other by light activation. The dual activations restrict fluorescence precisely inside live cells and at the particular spatial region of light activation, thus a probe with precise spatial accuracy in live cells.

Abstract:Ratiometric fluorescent probes based on FRET mechanism have attracted great attention due to their large pseudo-Stokes shifts and built-in correction for environmental effects. However, most donors failed to meet the requirement that the emission of the donor must overlap well with the absorption of the acceptor. Therefore, searching for new fluorophore to construct FRET system is in great need. In this paper, a new fluorescent dye pyrazolo[1,5-a]pyridine was synthesized and used as a donor in the FRET system for ratiometric sensing of Cu2+. The probe is based on FRET and PET mechanism. It shows high selectivity and sensitivity toward Cu2+ (detection limit 30 nM). Furthermore, it was successfully used to detect Cu2+ in Glioma cells.

One can observe that the emission colour changed from green to red. A Job's plot test suggested a 1 : 1 stoichiometry between ZC-F4 and Zn(2+), and the theoretical calculation based on density functional theory has been carried out to gain an insight into the sensing mechanism. Furthermore, imaging of Zn(2+) in cells was also performed to test its feasibility in biology. This fluorescence probe should be a promising candidate for applications in cell-imaging, environment protection, water treatment and safety inspection.

Abstract:We report a novel ratiometric and reversible fluorescent probe for Cd(2+) detection utilizing a 6-(dimethylamino)quinaldine derivative as the fluorophore and a 2-hydrazinopyridine derivative as Cd(2+) chelator. This ratiometric fluorescent probe possesses favorable photophysical properties. It shows a large (55 nm) red-shift from 515 nm to 570 nm in the emission spectrum. Moreover, this probe also exhibits an excellent linear relationship of fluorescence intensity ratio (F570/F515) (R(2)=0.989) vs. Cd(2+) concentration in the range of 0-10 µM at physiological pH, which can serve as a "quantitative detecting" probe for Cd(2+). Utilizing this sensitive and selective probe, we have successfully detected Cd(2+) in living cells.
