Abstract:A novel mitochondria-targeted ratiometric two-photon fluorescent probe (Mito-MQ) for detecting intracellular cysteine (Cys) and homocysteine (Hcy) has been designed. Mito-MQ showed the ratiometric fluorescent detection signal (the green-to-blue emissionfrom 517nm to 460nm) to cysteine (Cys) and homocysteine (Hcy) over glutathione (GSH), along with the fast response rate (10min). The detection mechanism was illustrated by 1H NMR, ESI-MS and theoretical calculation. The co-localization coefficient of 0.87 between Mito-MQ and MitoTracker Red revealed that the probe was predominantly present in mitochondria, therefore, Mito-MQ was successfully applied to detect mitochondrial oxidative stress by detecting the change of Cys/Hcy. Moreover, imaging in fresh tissue slices indicated that Mito-MQ could work in deep tissue (ca. 130µm) under two-photon excitation. Furthermore, the measurement of Cys/Hcy detection in zebrafish showed that probe can be used in determination of biothiols in vivo.

Abstract:A novel one- and two-photon fluorescent probe CB1 has been developed for discriminating Cys and Hcy in a successive manner with high selectivity. The discrete time-dependent fluorescent responses enable us to sequentially detect Cys and Hcy in different time windows. Two-step reaction and kinetic modes were used to explain the sensing mechanism. As a promising biosensor for cell imaging, CB1 has been confirmed to exhibit membrane permeability to intact cells, low cytotoxicity to viable cells and photostability to ultraviolet light excitation. Furthermore, the results from the control assay have shown that the one- and two-photon fluorescence of CB1 within cells is associated with intracellular mercapto biomolecules but yet there is little interference with physiological pH value, viscosity and common bioanalytes. Finally one- and two-photon fluorescent images of CB1 within living SiHa cells have been presented.

Abstract:A fluorescence ratiometric probe 1 for cysteine (Cys) and homocysteine (Hcy) has been rationally constructed based on intramolecular charge transfer (ICT) mechanism. Upon treatment with Cys/Hcy, probe 1 exhibited a fluorescence ratiometric response, with the emission wavelength displaying a large shift (from 526 nm to 446 nm). When 90 μM Cys were added, the emission ratios (I446/I526) of the probe changed dramatically from 0.01797 to 4.65472. The detection limit was also measured to be 0.18 μM (S/N = 3). The theoretical calculations have confirmed that the ratiometric response of probe 1 to Cys/Hcy is due to the inhibition of ICT process upon the reaction of probe 1 with Cys/Hcy. Furthermore, the fluorescence imaging experiments in living cell demonstrated that probe 1 was favourable for intracellular Cys/Hcy imaging.

Abstract:A two-photon fluorescent probe (NQ) for cysteine (Cys) and homocysteine (Hcy) was developed based on twisted-intramolecular-charge-transfer (TICT) mechanism. NQshowed excellent "OFF-ON" fluorescence (at 540 nm) detection signal on addition of Cys/Hcy with high selectivity. The detection mechanism was successfully proved by theoretical calculations, viscosity dependent fluorescence and time resolved fluorescence spectra. NQ exhibits large two-photon absorption (TPA) cross-section with Cys/Hcy (580 and 710 GM) at nearly 800 nm, respectively. Cell-culture results indicated that NQ was cell-permeable and could be used to detect Cys/Hcy with low cytotoxicity under two-photon excitation in living cells. To our best knowledge, NQ is the first TICT based two-photon fluorescent probe for Cys/Hcy. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:4-(1H-phenanthro[9,10-d]imidazol-2-yl)benzaldehyde 1 was rationally designed as a novel ratiometric fluorescent probe for cysteine and homocysteine. Upon addition of cysteine or homocysteine, notably, the probe displayed a very large (125 nm) hypsochromic shift in emission due to switching off intramolecular charge transfer. This large emission wavelength shift may allow probe 1 to be employed for quantitatively detecting Cys/Hcy.

Abstract:Novel reactive probes (C1 and C2) towards homocysteine/cysteine (Hcy/Cys) were designed and synthesized, based on the unique nucleophilic nature of bio-thiols. In the presence of Hcy/Cys, probe Cl displayed remarkable fluorescence enhancement. Meanwhile, ratiometric fluorescent probe C2 was designed through subtle structure adjustment. Differently, compound C2 displayed dramatic blue-shift in both fluorescence (100 nm) and absorption (95 mn) spectra upon the addition of Hcy/Cys. By virtue of the specific nucleophilic reaction, probe C2 had outstanding selectivity towards Hcy over Cys, GSH and other amino acids. The detection limit of probe C2 was calculated to be as low as 2.8 x 10(-7) mol/L. Moreover, C2 was successfully applied to microscopic imaging for the detection of Hcv in HeLa cells with ratiometric fluorescent methods.

Abstract:The ratiometric detection of cysteine (Cys) and homocysteine (Hcy) is very challenging because of their highly similar chemical structures and properties. By introducing the phenylethynyl group into a coumarin dye as the sensing group, the ratiometric fluorescent probe CP was developed to selectively and rapidly discriminate between Cys and Hcy. With a single-wavelength excitation, the presence of Cys or Hcy induced the probe to produce distinct ratiometric fluorescence changes: from red (lambda(em)(max) = 608 nm) to blue (lambda(em)(max) = 485 nm) toward Cys and from red to mixed red/blue toward Hcy. Moreover, the probe was capable of visualizing and discriminating between intracellular Cys and Hcy in HeLa cells and zebrafish by exhibiting different ratiometric fluorescence signals.

Abstract:A novel quinoline-fused Rhodamine dye is designed and synthesized to achieve near-infrared fluorescent emission by expanding the pi-conjugated system. Ratiometric fluorescent probe (RhQ) is further constructed by introducing the 3,5-ditrifluormethylthiophenol moiety into dye through a thioester bond to take advantage of spimcyclization open-ring states of Rhodamine platform. The probe exhibits remarkable dual-channel fluorescence response toward Cys/Hcy in aqueous solutions between near-infrared and visible emission, arising from the formation of closed spimlactam configuration disrupting molecular pi-conjugation system. RhQ is highly selective and sensitive to Cys/Hcy with detection limits of 0.12 mu M and 0.25 mu M for Cys and Hcy, respectively. Moreover, imaging in cellular experiment indicates that probe displays excellent mitochondrial targeting ability, lower cytotoxicity and ratiometric responses with two well-defined emissions to ensure exactly comparison and quantitative analysis of Cys/Hcy changes in the cell.

Abstract:A novel ratiometric mitochondrial cysteine (Cys)-selective two-photon fluorescence probe has been developed on the basis of a merocyanine as the fluorophore and an acrylate moiety as the biothiol reaction site. The biocompatible and photostable acrylate-functionalized merocyanine probe shows not only a mitochondria-targeting property but also highly selective detection and monitoring of Cys over other biothiols such as homocysteine (Hcy) and glutathione (GSH) and hydrogen sulfide (H2S) in live cells. In addition, this probe exhibits ratiometric fluorescence emission characteristics (F518/F452), which are linearly proportional to Cys concentrations in the range of 0.5-40 μM. More importantly, the probe and its released fluorophore, merocyanine, exhibit strong two-photon excited fluorescence (TPEF) with two-photon action cross-section (Φσmax) of 65.2 GM at 740 nm and 72.6 GM at 760 nm in aqueous medium, respectively, which is highly desirable for high contrast and brightness ratiometric two-photon fluorescence imaging of the living samples. The probe has been successfully applied to ratiometrically image and detect mitochondrial Cys in live cells and intact tissues down to a depth of 150 μm by two-photon fluorescence microscopy. Thus, this ratiometric two-photon fluorescent probe is practically useful for an investigation of Cys in living biological systems.

Abstract:Cysteine is associated with various diseases in the organism. Monitoring Cys levels is important for the study of related diseases. Here, a ratiometric fluorescent probe based on a novel fluorophore was designed for detecting Cys. The fluorophore exhibits a large Stokes shift and good photostability. The probe QL‐A shows high specificity and has been used to visualize Cys in living cells. It is a potential tool for exploring the role of Cys in biological processes and diseases. As a biothiol, cysteine (Cys) is essential to both physiological and pathological processes and has been associated with many diseases, including neurological disorders, rheumatoid arthritis, and renal dysfunction. Therefore, the development of a high‐performance probe for detecting Cys levels can help prevent and diagnose disease. In this study, a ratiometric fluorescent probe based on a novel fluorophore was developed for detecting Cys, and it showed high specificity and a rapid response time toward Cys. This probe demonstrates excellent biocompatibility and has been utilized effectively for the imaging of Cys in living cells.

Abstract:Cysteine is an essential amino acid for humans and can be generated from the conversion of methionine or the interconversions with cystine. The detection of cysteine in the living system is of great importance for the diagnosis of related diseases. Based on the classical cysteine response mechanism, the sulfhydryl group of cysteine undergoes Michael addition with the acrylate to rapidly respond to form a seven-membered ring that specifically recognizes cysteine. In the present work, we designed a novel ratiometric fluorescent probe N-1, which contained the benzo[cd]indol fluorophore and the response group acrylate. As the concentration of cysteine increased, the fluorescence of I485 was significantly enhanced while the fluorescence intensity at 635 nm was weakened. Moreover, probe N-1 showed excellent sensitivity and selectivity to Cys with the calculated LOD of 4.0 × 10-9 M. Probe N-1 was successfully used for imaging cysteine in the living Hela cells with low cytotoxicity, indicating that this ratiometric fluorescent probe would be a novel and useful tool to monitor cysteine in the physiological or pathological processes.

Abstract:The very close structural similarities between cysteine and homocysteine present a great challenge to achieve their selective detection using regular fluorescent probes, limiting the biological and pathological studies of these two amino thiols. A coumarin-based fluorescent probe was designed featuring pH-promoted distinct turn-on followed by ratiometric fluorescence responses for Cys and turn-on fluorescence response for Hcy through two different reaction paths. These specific responses demonstrate the activity differences between Cys and Hcy qualitatively for the first time. The probe could also be used for Cys and Hcy imaging in living cells.

Abstract:A ratiometric fluorescent probe DPCA based on coumarin and pyrazoline was designed and synthesized for sensing cysteine (Cys) in physiological pH. The probe can detect Cys by fluorescent spectrometry within 40 min with a detection limit of 5.08 mu M. The mechanism of DPCA sensing Cys involved conjugate addition/cyclization which was confirmed by HRMS and fluorescence spectra analysis. Moreover, DPCA was able to be applied to complicated conditions with negligible interference from these species and successfully applied in cell imaging. (C) 2014 Elsevier B.V. All rights reserved.

Abstract:Ratiometric near-infrared (NIR) fluorescent probes are attractive tools for bioimaging applications. Although many fluorescent probes have been developed for detection of the biologically important cysteine (Cys), ratiometric NIR fluorescent probes for Cys are very rare, particularly those with significant fluorescence enhancement in the NIR region. In this work, a new ratiometric NIR fluorescent probe with significant NIR fluorescent turn-on changes was reported for rapid, colorimetric and ratiometric fluorescent detection of Cys. This probe is able to detect Cys in aqueous solution under mild conditions, with high selectivity and sensitivity for Cys over various analytes including the similar structured homocysteine (Hcy) and glutathione (GSH). The detection limit of this probe for Cys was found to be as low as 0.2 μM, which is far below the normal content of Cys (30–200 μM) in cells. Besides the excellent sensing properties, this probe shows low cytotoxicity and can be conveniently applied to image intracellular Cys in living cells, indicating it is a promising tool for biological applications.

Abstract:A merocyanine-based highly selective colorimetric and ratiometric fluorescent probe is described for Cys detection in water and diluted deproteinized human serum. Upon reaction with Cys in aqueous buffer solution, the probe showed a dramatic color change from faint yellow to pink and remarkable ratiometric fluorescence enhancement signals were also observed, which are ascribed to an intramolecular charge transfer (ICT) process. This strategy was based on modulating the merocyanine π-electron system by conjugation and removal of the acrylate group to release the chromophore group, resulting in a specific colorimetry and fluorescence response. The probe has low cytotoxicity and good cell permeability. It is readily employed for assessing the change of the intracellular Cys level.

Abstract:A benzothiazole-based near-infrared (NIR) ratiometric fluorescent probe (HBT-Cys) was developed for discriminating cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH). The probe was designed by masking phenol group in the conjugated benzothiazole derivative with methacrylate group that could be selectively removed by Cys, and therefore an intramolecular charge transfer (ICT) fluorescence was switched on in the NIR region. In the absence of Cys, the probe exhibited a strong blue fluorescence emission at 431 nm, whereas a NIR fluorescence emission at 710 nm was significantly enhanced accompanied by a decrease of emission at 431 nm in the presence of Cys, allowing a ratiometric fluorescence detection of Cys. The fluorescence intensity ratio (I710nm/I431nm) showed a good linear relationship with Cys concentration of 1-40 μM with the detection limit of 0.5 μM. The sensing mechanism was explored based on MS experimental analysis and DFT theoretical calculation. Moreover, the fluorescent probe was successfully used for fluorescence bioimaging of Cys in living cells.
