Abstract:Herein, novel photoactive materials, MOF-derived porous hollow carbon nanobubbles@ZnCdS multi-shelled dodecahedral cages (C@ZnCdS MSDCs)， were synthesized via continuous chemical etching, sulfurization, cation-exchange and calcination strategies. Due to the synergistic effect between the porous shells and the carbon-layer coating， C@ZnCdS MSDCs displayed superior photoelectrochemical (PEC) performance. The synthesized C@ZnCdS MSDCs were assembled onto TiO2 modified ITO electrodes to form a type-II heterostructures. Then, Au nanoparticles (NPs) were deposited on the surface of ITO/TiO2/C@ZnCdS MSDCs. Benefiting from the unique structure and performance merits of photoactive materials, a label-free PEC sensing platform based on ITO/TiO2/C@ZnCdS MSDCs/Au was successfully constructed for CEA detection. Under optimal conditions, the PEC biosensor exhibited a wide linear range (0.00005-500 ng mL-1) and low detection limit (2.28 fg mL-1). The proposed PEC biosensor also showed good stability, specificity, reproducibility and acceptability in human serum. The prepared C@ZnCdS MSDCs would be a promising photoactive material for PEC biosensors. Most importantly, this work opens up new horizons for the application of MOFs-derived hollow carbon materials in sensing.

Abstract:Liposomal photoelectrochemical (PEC) bioanalysis has recently emerged and exhibited great potential in sensitive biomolecular detection. Exploration of the facile and efficient route for advanced liposomal PEC bioanalysis is highly appealing. In this work, we report the split-type liposomal PEC immunoassay system consisting of sandwich immunorecognition, CdS quantum dots (QDs)-loaded liposomes (QDLL), and the release and subsequent capture of the QDs by a separated TiO2 nanotubes (NTs) electrode. The system elegantly operated upon the protein binding and lysis treatment of CdS QDLL labels within the 96-well plate, and then the CdS QDs-enabled sensitization of TiO2 NTs electrode. Exemplified by cardiac markers troponin I (cTnI) as target, the proposed system achieved efficient activation of TiO2 NTs electrode and thus the signal generation toward the split-type PEC immunoassay. This work features the first use of QDs for liposomal PEC bioanalysis and is expected to inspire more interests in the design and implementation of numerous QDs-involved liposomal PEC bioanalysis.
A Label-Free Photoelectrochemical Immunosensor Based on Water-Soluble Cds Quantum Dots

Abstract:We demonstrate herein a newly developed lable-free photoelectrochemical immunosensor using a CdS quantum dots (QDs) multilayer film coupled with a biospecific interaction. The CdS QDs multilayer film was prepared by layer-by-layer assembling positively charged poly(dimethyldiallylammonium chloride) (PDDA) and thioglycolic acid (TGA)-capped water-soluble US QDs with negative charges on the surface of an indium-tin oxide (ITO) electrode. Ascorbic acid (AA) was exploited as an efficient and nontoxic electron donor for scavenging photogenerated holes under mild solution medium. The photoexcitation of CdS QDs modified electrode potentiostated at 0 V (vs. Ag/AgCl) in the presence of 0.1 M AA led to a stable anodic photocurrent. To perform the immunoassay, goat antimouse IgG was conjugated onto CdS QDs modified electrode by using the classic EDC coupling reactions between COOH groups on the surfaces of the TGA capped CdS QDs and NH2 groups of the antibody. The concentrations of mouse IgG were measured through the decrease in photocurrent intensity resulting from the increase in steric hindrances due to the formation of the immunocomplex. The synthetic conditions (different Cd/S ratio and different pH) of CdS QDs and the number of PDDA/CdS bilayers could influence the photoelectrochemical properties of US QDs modified electrodes used for immunosensor construction. Under the optimal conditions, a linear relationship between photocurrent decrease and mouse IgG concentration was obtained in the range of 10 pg/mL to 100 ng/mL with a detection limit of 8.0 pg/mL. This strategy opens a new perspective for the application of QDs, which might be of great significance for QDs in photoelectrochemical bioanalysis in the future.

Abstract:Recently developed localized surface plasmon resonance (LSPR) enhanced photoelectrochemical (PEC) biosensing has emerged as a unique label-free technology owing to the high sensitivity and robust detection. Herein, a feasible PEC biosensing platform based on receptor-functionalized plasmonic Au-MoS2 nanosheets was designed for the capture of target proteins. The biomolecular interaction substantially enhances the hot electron transfer efficiency between Au and MoS2, leading to a direct change in electrocatalytic current on MoS2 as electricity readout signal. The biosensor exhibits excellent analytical performance for protein detection in a range from 41.7 nM to 4.17 mu M, with a detection limit of 4.17 nM. Additionally, this system also affords real-time analysis of binding kinetics between antigen and antibody, allowing for obtaining their rate constants of association/disassociation (k(on)/k(off)) and equilibrium constant K-d of 3.95 mu M. LSPR-enhanced label-free PEC monitoring system, without the participation of ultraviolet excitation and sacrifice agents, bears no harm to biomolecules and shows high-sensitivity detection, which holds great potential applications in clinical and biological analyses. Abstract:A highly efficient photocathode, based on graphite-like carbon nitride nanosheets (CNNS) sensitized CdTe quantum dots (QDs), was constructed for photoelectrochemical (PEC) biosensing. Using dissolved oxygen as an electron acceptor, the hybrid photocathode showed a sensitive photocurrent response at −0.2V bias potential under 405nm illumination. The CdTe/CNNS hybrid photocathode demonstrates about 100% increase of photocurrent compared to CdTe QDs modified electrode owing to the formation of heterojunction through contact of two semiconductor materials. The improved charge separation efficiency was identified by the extension of electron transit time (τd) and electron lifetime (τn) in this PEC system. The introduction of Cu2+ on the surface of hybrid photocathode could decrease photocurrent via the exciton trapping quenching effect. A sensitive PEC sensor for Cu2+ was thus developed with a good linear range from 20nM to 100μM and a detection limit of 3.3nM, and was successfully applied in the detection of Cu2+ in human hair samples. The CNNS-sensitized photocathode provides a good alternative for enhancement of the PEC signal transduction and could be widely used in biosensing and clinical diagnosis.

Abstract:In this study, semiconducting organic polymer dots (Pdots) and inorganic quantum dots (Qdots) were first utilized to construct the organic-inorganic nanodots heterojunction for the photoelectrochemical (PEC) bioanalysis application. Specifically, n-type CdS Qdots, p-type CdTe Qdots, and tetraphenylporphyrin (TPP)-doped poly[(9,9-dioctylfluoreny1-2,7-diyl)-co(1,4-benzo-{2,1',3}-thiadazole)] (PFBT) Pdots were fabricated, and their energy levels, that is, their valence band (VB)/conduction band (CB) or lowest unoccupied molecular orbital (LUMO)/highest occupied molecular orbital (HOMO) values, were also determined. Then, these nanodots were integrated to construct four types of p-n and p-p organic-inorganic nanodots heterojunctions, that is, CdS Qdots/TPP-doped PFBT Pdots, TPP-doped PFBT Pdots/CdS Qdots, CdTe Qdots/TPP-doped PFBT Pdots, and TPP-doped PFBT Pdots/CdTe Qdots, on the transparent glass electrode. Upon light irradiation, four heterojunctions exhibited different PEC behaviors with some having prominent photocurrent enhancement. With the model molecule L-cysteine (L-cys) as target, the proposed PEC sensor exhibited good performances. In brief, this work presents the first semiconducting organic-inorganic nanodots heterojunction for PEC bioanalysis application, which could be easily used as a general platform for future PEC bioanalysis building. Besides, it is expected to inspire more interest in the design, development, and implementation of various organic-inorganic heterojunctions for advanced PEC bioanalysis in the future.
Long-lived Charge Carriers in Mn-doped CdS Quantum Dots for Photoelectrochemical Cytosensing.

Abstract:Photoelectrochemical (PEC) biosensing with semiconductor quantum dots (QDs) has received great attention because it integrates the advantages of both photo-excitation and electrochemical detection. During the photon-to-electricity conversion in PEC processes, electron-hole (charge) separation competes with electron-hole recombination, and the net effect essentially determines the performance of PEC biosensors. Herein, we propose a new approach for slowing down electron-hole recombination to increase charge separation efficiency for PEC biosensor development. Through doping with Mn(2+) , a pair of d bands ((4) T1 and (6) A1 ) is inserted between the conduction and valence bands of CdS QDs, which alters the electron-hole separation and recombination dynamics, allowing the generation of long-lived charge carriers with ms-scale lifetime that decay about 10(4) -10(5) -fold more slowly than in the case of undoped QDs. Photocurrent tests indicated that Mn(2+) doping resulted in an approximately 80 % increase in photocurrent generation compared with undoped CdS QDs. For application, the Mn-doped CdS QDs were coated on the surface of a glassy carbon electrode and functionalized with a cell surface carbohydrate-specific ligand (3-aminophenylboronic acid). In this way, a sensitive cytosensor for K562 leukemia cells was constructed. Moreover, the sugar-specific binding property of 3-aminophenylboronic acid allowed the electrode to serve as a switch for the capture and release of cells. This has been further explored with a view to developing a reusable PEC cytosensing platform.

Abstract:A sensitive photoelectrochemical (PEC) biosensor for silver ions (Ag+) was developed based on Zn-Co doped C and CdS quantum dots (CdS QDs) nanomaterials. Hydrophobic modified sodium alginate (HMA), which could stabilize and improve the PEC performance of CdS QDs, was also used for the construction of PEC sensors. Specially, Zn-Co doped C, CdS QDs and HMA were sequentially modified onto the electrode surface via the drop-coated method, and the C base rich DNA strand was then immobilized onto the modified electrode. As the C base in DNA specifically recognized Ag+, it formed the C-Ag+-C complex in the presence of Ag+, which created a spatial steric hindrance, resulting in a reduced PEC response. The sensing platform is sensitive to Ag+ in the range of 10.0 fM to 0.10 μM, with a limit of detection of 3.99 fM. This work offers an ideal platform to determine trace heavy metal ions in environmental monitoring and bioanalysis.

Abstract:Benefiting from the maximum atom-utilization efficiency and distinct structural features, single-atom catalysts open a new avenue for the design of more functional catalysts, whereas their bioapplications are still in their infancy. Due to the advantages, platinum single atoms supported by cadmium sulfide nanorods (Pt SAs-CdS) are synthesized to build an ultrasensitive photoelectrochemical (PEC) biosensing platform. With the decoration of Pt SAs, the PEC signal of CdS is significantly boosted. Furthermore, theory calculations indicate the positively charged Pt SAs could change the charge distribution and increase the excited carrier density of CdS. Meanwhile, it also suggests that Cu2+ can severely hinder the photoexcitation and electron–hole separation of CdS. As a proof of concept, prostate-specific antigen is chosen as the target analyte to demonstrate the superiority of the Pt SAs-CdS-based PEC sensing system. Abstract:Nanocatalysts are a promising alternative to natural enzymes as the signal labels of electrochemical biosensors. However, the surface modification of nanocatalysts and sensor electrodes with recognition elements and blockers may form a barrier to direct electron transfer, thus limiting the application of nanocatalysts in electrochemical immunoassays. Electron mediators can accelerate the electron transfer between nanocatalysts and electrodes. Nevertheless, it is hard to simultaneously achieve fast electron exchange between nanocatalysts and redox mediators as well as substrates. This work presents a scheme for the design of electrochemical immunosensors with nanocatalysts as signal labels, in which pyrroloquinoline quinone (PQQ) is the redox-active center of the nanocatalyst. PQQ was decorated on the surface of carbon nanotubes to catalyze the electrochemical oxidation of tris(2-carboxyethyl)phosphine (TCEP) with ferrocenylmethanol (FcM) as the electron mediator. With prostate-specific antigen (PSA) as the model analyte, the detection limit of the sandwich-type immunosensor was found to be 5 pg/mL. The keys to success for this scheme are the slow chemical reaction between TCEP and ferricinum ions, and the high turnover frequency between ferricinum ions, PQQ. and TCEP. This work should be valuable for designing of novel nanolabels and nanocatalytic schemes for electrochemical biosensors.

Abstract:This work designs a competitive consumption strategy of surface electron donor and light absorption for quenching the photocurrent of ZnSnO3 nanocubes/BiOI nanoarrays/polydopamine (ZnSnO3 NCs/BiOI NAs/PDA) as a photoactive material. This material can be formed on electrode surface by successive coating and deposition to provide a substrate for immobilization of capture antibody, and producing strong photocurrent in the presence of ascorbate acid as a surface electron donor due to the well matching structure of band gaps between ZnSnO3 NCs and BiOI NAs, the excellent light absorption ability and high photo-electron conversion efficiency of BiOI NAs and PDA, and the accelerated electron transfer. Using ascorbate oxidase loaded dopamine-melanin nanosphere (DAM-AAO) as a label of the signal (secondary) antibody, the sandwich-type immunoreaction leads to dual photocurrent quenching of the label through the competitive consumption of ascorbate acid with enzymatic oxidation and the light absorption by DAM nanosphere. Thus, a sensitive "On-Off" photoelectrochemical (PEC) immunosensing method is constructed for the analysis of neuron specific enolase (NSE). The proposed method shows a detection range of 0.1 pg/mL -50 ng/mL and a detection limit of 0.03 pg/mL. The excellent performance and the recovery text demonstrated the practicability of the designed strategy and label in immunoassay of different protein biomarkers.
CdS Quantum-Dots-Decorated V2O5 Nanosheets as Chemically Etchable Active Materials for Sensitive Photoelectrochemical Immunoassay of Carcinoembryonic Antigen

Abstract:We report here CdS quantum-dots (QDs)-decorated V2O5 nanosheets as high-performance and chemically etchable photoelectric active materials for constructing a photoelectrochemical (PEC) immunoassay platform. CdS QDs-decorated V2O5 nanosheets as new photoelectric materials can show superior photocurrent to V2O5 nanosheets and CdS QDs under visible-light irradiation because of the promoted photogenerated electron-hole separation and the increased visible-light absorption. V2O5 nanosheets can be etched by ascorbic acid (AA) because of the reduction of V2O5 to V4+, and the photocurrent of CdS/V2O5-nanocomposite-modified indium tin oxide electrode decreases significantly after being etched by AA. Inspired by this phenomenon, a PEC immunoassay platform is constructed for carcinoembryonic antigen (CEA) detection by using CdS/V2O5 nanocomposite as the photoelectric material and AA-encapsulated liposome immunonanocapsules as labels. The linear detection range for detecting CEA is from 0.5 pg mL-1 to 1 ng mL-1, with a limit of detection of 0.1 pg mL-1. The proposed method also shows good selectivity, excellent reproducibility, and satisfactory recovery in detection of CEA in human serum samples. We believe that this work will lay the foundation for the future development of V2O5-based materials for PEC analysis, and also provide a reasonable design and implementation for the development of PEC immunoassay.

Abstract:In this paper, Bi2S3/AgBiS2 composite nanomaterials and PDA@Ag@N-CQDs were synthesized, and used as substrates and second antibody label respectively to construct a sandwich photoelectrochemical (PEC) sensor. The upconversion luminescence effect of N-CQDs can convert long wavelength light into short wavelength light that can be utilized by the substrate material, which can provide additional excitation light energy for the substrate material and further enhance the photoelectric response. Besides, Ag has SPR effect and can also promote electron transfer. The proposed sandwich immunosensor achieves detection of NSE in the concentration range of 0.001 ng mL-1 to 100 ng mL-1, with a detection limit of 0.28 pg mL-1 (S/N = 3). What's more, the proposed sensor also exhibits good stability, selectivity, as well as reproducibility, indicating its promising application prospects.

Abstract:Integrating the advantages of excellent anti-interference and high stability superior to photoanode, cathodic photoelectrochemical (PEC) bioanalysis is promising and competitive in precise monitoring targets in complex matrices. However, serious consideration of the photocathode is far behind the anodic one. Herein, we report a high quality MoS2 QDs-BiOI p-n heterojunctioned material and exemplify the feasibility of constructing cathodic PEC aptasensing platform. Intense visible light-harvesting, high photoelectric conversion efficiency and magnified photocurrent is observed, which origin is detailed explored by various spectroscopic and electrochemical study. Taking the tumor necrosis factor-alpha (TNF-α) as a model analyte and by coupling the simplest target-induced conformational change mechanism, high selectivity and ultrasensitive self-power cathodic PEC detection is exemplified. Stable and reproducible PEC responses are achieved in a wide linear range of 10 fg/mL to 0.1 ug/mL with an ultralow detection limit (5.2 fg/mL) surpassing most sensors reported so far. Monitoring TNF-α in biological system is realized with desired accuracy and satisfactory recovery.

Abstract:This work reports the first preparation and characterization of a hybrid PbS quantum dot (QD)/nanoporous NiO film nanostructure as well as its application for novel self-powered cathodic photoelectrochemical (PEC) sensing. Specifically, we synthesized the thioglycolic acid-capped PbS QDs and then assembled them onto the hydrothermally fabricated three-dimensional (3D) NiO nanostructured films on the transparent indium tin oxide-coated glass substrates, followed by the subsequent conjugation with the glucose oxidase as a model biocatalyst. Favorable alignment existed between the NiO and PbS QDs, and the as-obtained p-type heterostructure was characterized by various techniques and found to have good PEC activities. In the self-powered PEC biosensing of glucose, the system exhibited high sensitivity toward the presence of dissolved oxygen in the electrolyte, and thereby, a novel PEC enzymatic sensor was developed. With a PbS QD/3D NiO nanofilm, this work manifested the great promise of a heterostructure photocathode for a self-powered PEC biosensor that to the best of our knowledge has not been reported. We believe that it could inspire more interest in the design and development of numerous other p-type heterostructures for advanced self-powered PEC biosensors.
Photoelectrochemical DNA Biosensor Based on G-C3n4/mos2 2D/2D Heterojunction Electrode Matrix and Co-Sensitization Amplification with CdSe QDs for the Sensitive Detection of Ssdna

Abstract:A novel enhanced photoelectrochemical (PEC) DNA biosensor, based on a compact heterojunction g-C3N4/MoS2 and co-sensitization effect with CdSe quantum dots (QDs), was first proposed for simple and accurate analysis of a short ssDNA. In this work, the g-C3N4/MoS2 was successfully synthesized and used as the electrode matrix material to construct PEC biosensor. 2D/2D heterojunction was formed between g-C3N4 and MoS2, which could promote the separation of photogenerated electron-hole pairs resulting in an enhanced photocurrent. In the presence of target DNA, CdSe QDs labeled reporter DNA was complementary pairing with target DNA which was specific recognized by capture DNA loading on self-assembled CdS QDs film, leading to close contact between CdSe QDs and g-C3N4/MoS2 modified electrode surface, thereby resulting in the enhanced photocurrent intensity due to the co-sensitization effect. Under the optimal operating conditions, the photoelectrochemical biosensor demonstrated favorable accuracy and could respond to 0.32 pM (S/N = 3) with a linear concentration range from 1.0 pM to 2.0 mu M. Moreover, the proposed PEC DNA biosensor exhibits high sensitivity, excellent specificity, acceptable reproducibility and accuracy, showing a promising potential in DNA bioanalysis and other relative fields.
