Abstract:A novel signal-off photoelectrochemical (PEC) immunosensor was proposed for procalcitonin (PCT) sensitive detection based on the Au nanoparticles/BiOI nanosheets/Black TiO2 nanoparticles (Au NPs/BiOI NSs/B-TiO2 NPs) as photoactive matrix and PbS quantum dots/Co3O4 polyhedron (PbS/Co3O4) as a quenching signal label for signal amplification. Concretely, BiOI NSs was fabricated by successive ionic layer absorption and reaction (SILAR) and formed the interlaced flake-like structure. Then Au NPs dropped on the BiOI NSs/B-TiO2 NPs electrode forming Au NPs/BiOI NSs/B-TiO2 NPs sensitization structure, which increased the utilization of visible light and accelerated electron transfer rate, resulting in a desirable PEC signal. PbS/Co3O4, an effective dual suppression signal quencher, was proposed for signal-off PEC immunosensor to weaken the photocurrent response because of the steric impedance, competitive absorption of light and competing consumption of electron donor. In this proposal, PCT was chosen to measure the performance of the suggested PEC immunosensor. Consequently, the fabricated PEC immunosensor showed a widely detection ranged from 0.1 pg/mL to 50 ng/mL and a low detection limit of 0.02 pg/mL (S/N = 3). What is more, the proposed PEC immunosensor possessed satisfactory selectivity, stability and reproducibility.

Abstract:An original signal amplification photoelectrochemical (PEC) immunosensor for procalcitonin detection was fabricated based on ZnxBi2S3+x sensitized NiTiO3 nanorods. Using ethylene glycol as medium, the NiTiO3 nanorods were successfully manufactured after calcination in air atmosphere, the rod-like structure have rough surface that produced benefit to capture lots of functional ZnxBi2S3+x the sensitizer sensitized NiTiO3 on ITO substrate by means of a sequential ionic layer adsorption reaction, and the obtained ZnxBi2S3+x/NiTiO3 composites showed an excellent absorption of visible light owing to the matched band gap. By the way, ZnxBi2S3+x/ NiTiO3 composites as mimic enzyme of glucose oxidase, high sensitivity was achieved due to the generated H2O2 as holes scavenger when glucose added, that promote the photo electrons transfer quickly, further increased the PEC signal. The as prepared sensor showed a large linear range of 0.0001-50 ng.mL(-1) with 0.04 pg.mL(-1) detection limit. Simultaneously, the signal amplification strategy of this sensor provides applicable to other PEC immunosensors for detection of other targets.

Abstract:In this paper, we constructed a sandwich-type photoelectrochemical (PEC) immunosensor for the quantitative detection of procalcitonin (PCT) based on the sensitization of Mn2+ doped CdS (CdS:Mn) nanocomposites to Bi2WO6 and the signal amplification effect of an upconversion material NaYF4:Yb,Tm. Bi2WO6 was synthesized with a three-dimensional flowered structure. CdS:Mn reduced the recombination of photogenerated carriers, and significantly improved photocurrent response. Lanthanide-doped upconversion nanomaterials were used as the label of secondary antibody. NaYF4:Yb,Tm have two functions, not only connected with the secondary antibody, but also can further amplify the photocurrent response. The proposed immunosensor for detecting PCT provided a desired linear range of 0.5 pg mL-1-100 ng mL-1 and a detection limit of 0.13 pg mL-1 under optimal experimental conditions. Besides, the PEC immunosensor demonstrated good reproducibility, specificity and stability. The results of determination of PCT in real human serum samples were satisfactory. Thus, the immunosensor may be applied in the clinical diagnosis of PCT and other biomarkers.

Abstract:As an inflammatory marker, procalcitonin (PCT) is more representative than other traditional inflammatory markers. In this work, a highly efficient photoelectrochemical (PEC) immunosensor was constructed based on the photoactive material Bi2S3/Ag2S to realize the sensitive detection of PCT. Bi2S3 was prepared by a hydrothermal method, and Ag2S quantum dots were deposited on the ITO/Bi2S3 surface via in situ reduction. Bi2S3 is a kind of admirable photoelectric semiconductor nanomaterial on account of its moderate bandgap width and low binding rate of photogenerated electron holes, which can effectively convert light energy into electrical energy. Therefore, based on the energy level matching principle of Bi2S3 and Ag2S, a labeled Bi2S3/Ag2S PEC immunosensor was constructed, and the sensitive detection of PCT was successfully established. The linear detection range of the PEC immunosensor was 0.50 pg∙mL−1 to 50 ng∙mL−1, and the minimum detection limit was 0.18 pg∙mL−1. Compared with the traditional PEC strategy, the proposed PEC immunosensor is simple, convenient, and has good anti-interference, sensitivity, and specificity, which could provide a meaningful theoretical basis and reference value for the clinical detection of PCT.

Abstract:A label-free photoelectrochemical (PEC) immunoassay system based on the one-step synthesis of SnS2/CdS nanocomposites is successfully constructed for sensitively analyzing procalcitonin (PCT). The one-step synthesis of SnS2/CdS enables the rapid construction of an immunoassay system and avoids the waste of energy. In the constructed immunoassay system, CdS as a sensitizer for PCT analysis not only enhances the light absorption of SnS(2)as a photocurrent generator but also can accelerate the separation of photosensitive electrons and holes by forming a heterostructure. Under optimum conditions, the energy-saving assay platform based on the enhancement photocurrent response of SnS2/CdS nanocomposites exhibited an excellent performance in analyzing a series of PCTs with different concentrations in a wide linear range of 0.5 pg mL(-1)to 100 ng mL(-1)and the detection limit was 0.17 pg mL(-1). In view of the advantages of good selectivity, high sensitivity, and facile preparation procedure, the PEC immunoassay is expected to be a promising analytical platform for rapid analysis of PCT and other biomarkers in clinical medical samples.

Abstract:A split-type photoelectrochemical (PEC) sensor for procalcitonin (PCT) detection was successfully prepared. This split-type sensor adopted controlled-release strategy, which separated the two processes of antigen-antibody specific recognition and photoelectric conversion, so that the two processes can be performed independently. In addition, CdS sensitized Bi2Sn2O7 is combined for the first time to form a heterojunction as the matrix to provide the basic PEC response, which could effectively absorb visible-light, and reduced electron-hole pair recombination, ensured the stability of the sensor. Distinctly important, the skillfully process was performed in a 96-well plate where the specific reaction was completed proceeded between antigen and antibody. For improving sensitivity of the sensor, acetylcholinesterase (ACHE) connected to SiO2 nanospheres (SiO2 NSs) modified by second antibody (Ab2-ACHE@SiO2 NSs) was designed, the thiocholine (THCH) can be released when acetylthiocholine iodide (ATCH) exists, and the THCH as an electron donor could enhance PEC response. The proposed PEC immunoassay with high-performance photosensitive materials, and the smart electron donor released strategy have a wide detection liner range from 0.0005 to 100 ng/mL with a low detection limit of 0.17 pg/mL, also provide great clinical diagnostic potential for other target proteins.

Abstract:Sensitive detection of CA15-3 is important for postoperative monitoring and early diagnosis of breast cancer. In this work, a photoelectrochemical (PEC) biosensing platform was constructed of CA15-3 by using Bi2O3/CdLa2S4/Bi2S3 ternary type-II heterojunction as the photoactive transduction material for sensitive detection, and the raspberry-shaped Cu2O nanospheres were exploited as the signal sensitizing tags. On one hand, the use of Bi2S3 as a sensitizer can effectively enhance the visible light excitation of the substrate material. On the other hand, Bi2S3 and Bi2O3/CdLa2S4 have the ability to create dual type-II heterojunctions with matched energy levels, which significantly enhances the separation and migration efficiency of the photogenerated carriers and effectively suppresses the recombination of the e(-)/h(+) pairs. Raspberry-like semiconducting Cu2O nanospheres can mediate the PEC process of the heterojunctions. An immunosensor was developed combining the signal transduction capacity of ternary heterojunction and the signal sensitizing effect of Cu2O nanospheres. The proposed PEC immunosensor showed good selectivity and stability for detecting CA15-3 (0.001 similar to 100 U/mL) with a detection limit of 0.0003 U/mL (S/N = 3). The constructed PEC immunosensor had potential applications in the clinical detection of CA15-3, and was expected to play a great role in the early diagnosis of breast cancer in the future. Moreover, in the design engineering of photoactive materials, the improvement of visible excitation and the enhancement of carrier separation efficiency have been realized simultaneously using a single material, which provides new ideas for the design of photoactive materials.

Abstract:The outstanding photoactive materials are the imperative for the construction of a front-illuminated photoelectrochemical (PEC) biosensor, which is crucial step for improving the detection sensitivity. Yet, the weak and unstable initial PEC signals of the photoelectrodes have limited evidently the detection performance. Herein, a front-illuminated "on-off" PEC immunosensor was constructed based on Mo:BiVO4 as photoactive matrix and Au/CeO2 as signal quencher for sensitive detection of procalcitonin (PCT). Systematic studies reveal that the Mo doped BiVO4 can increase the charge carrier density of BiVO4, leading to much higher initial signal under front illumination than back illumination. Moreover, Mo:BiVO4 was directly grown on conducting substrates, which effectively overcomes the loose combination of sensing substrate ensuring good electrical contact and continuity. Upon coupling with Au/CeO2 as signal quencher, the initial photocurrent signal can be significantly quenched. As a result, the proposed PEC immunosensor presents a wide linear range from 10 fg mL-1 to 50 ng mL-1 with a detection limit of 2.45 fg mL-1. Impressively, this study will open a new avenue for the construction of highly efficient and stable photoelectrode, as well as extend the application of PEC biosensor for biomarkers detection in early disease diagnosis.
Self-Assembled Perylene Diimide (PDI) Nanowire Sensitized In2O3@MgIn2S4 S-Scheme Heterojunction as Photoelectrochemical Biosensing Platform for the Detection of CA15-3

Abstract:Inorganic/organic heterojunctions show promising applications as high-performance sensing platforms for photoelectrochemical (PEC) immunosensors. This work reports constructing a PEC biosensor for CA15-3 based on a self-assembled perylene diimide (PDI) nanowire sensitized In2O3@MgIn2S4 S-scheme heterojunction platform. P-type semiconductor Cu2O nanoparticles were designed as a signal burst source and were used as immunoassay labels. The carboxyl group on self-assembled PDI nanowires eliminates the step of additional surface functionalization for covalent immobilization of the capture elements. The π-π stacking of PDI enhances electron generation efficiency, while the carboxylic acid groups on PDI promote electron transfer. The performance of the constructed sensor was validated using CA15-3 as a model. The experimental results showed that the sensor based on In2O3@MgIn2S4/PDI has excellent selectivity, stability, and reproducibility, and can sensitively detect CA15-3 in the range of 0.001-100 U·mL-1 with the detection limit of 0.00056 U·mL-1. The sensor has a broad application prospect. It is hoped that this research work based on the unique advantages of the organic compound PDI will inspire other researchers to design light-responsive materials and promote the development of the field of photoelectrochemical sensing.
A Label-Free Photoelectrochemical Immunosensor Based on Water-Soluble Cds Quantum Dots

Abstract:We demonstrate herein a newly developed lable-free photoelectrochemical immunosensor using a CdS quantum dots (QDs) multilayer film coupled with a biospecific interaction. The CdS QDs multilayer film was prepared by layer-by-layer assembling positively charged poly(dimethyldiallylammonium chloride) (PDDA) and thioglycolic acid (TGA)-capped water-soluble US QDs with negative charges on the surface of an indium-tin oxide (ITO) electrode. Ascorbic acid (AA) was exploited as an efficient and nontoxic electron donor for scavenging photogenerated holes under mild solution medium. The photoexcitation of CdS QDs modified electrode potentiostated at 0 V (vs. Ag/AgCl) in the presence of 0.1 M AA led to a stable anodic photocurrent. To perform the immunoassay, goat antimouse IgG was conjugated onto CdS QDs modified electrode by using the classic EDC coupling reactions between COOH groups on the surfaces of the TGA capped CdS QDs and NH2 groups of the antibody. The concentrations of mouse IgG were measured through the decrease in photocurrent intensity resulting from the increase in steric hindrances due to the formation of the immunocomplex. The synthetic conditions (different Cd/S ratio and different pH) of CdS QDs and the number of PDDA/CdS bilayers could influence the photoelectrochemical properties of US QDs modified electrodes used for immunosensor construction. Under the optimal conditions, a linear relationship between photocurrent decrease and mouse IgG concentration was obtained in the range of 10 pg/mL to 100 ng/mL with a detection limit of 8.0 pg/mL. This strategy opens a new perspective for the application of QDs, which might be of great significance for QDs in photoelectrochemical bioanalysis in the future.

Abstract:This work designs a competitive consumption strategy of surface electron donor and light absorption for quenching the photocurrent of ZnSnO3 nanocubes/BiOI nanoarrays/polydopamine (ZnSnO3 NCs/BiOI NAs/PDA) as a photoactive material. This material can be formed on electrode surface by successive coating and deposition to provide a substrate for immobilization of capture antibody, and producing strong photocurrent in the presence of ascorbate acid as a surface electron donor due to the well matching structure of band gaps between ZnSnO3 NCs and BiOI NAs, the excellent light absorption ability and high photo-electron conversion efficiency of BiOI NAs and PDA, and the accelerated electron transfer. Using ascorbate oxidase loaded dopamine-melanin nanosphere (DAM-AAO) as a label of the signal (secondary) antibody, the sandwich-type immunoreaction leads to dual photocurrent quenching of the label through the competitive consumption of ascorbate acid with enzymatic oxidation and the light absorption by DAM nanosphere. Thus, a sensitive "On-Off" photoelectrochemical (PEC) immunosensing method is constructed for the analysis of neuron specific enolase (NSE). The proposed method shows a detection range of 0.1 pg/mL -50 ng/mL and a detection limit of 0.03 pg/mL. The excellent performance and the recovery text demonstrated the practicability of the designed strategy and label in immunoassay of different protein biomarkers.

Abstract:Benefiting from the maximum atom-utilization efficiency and distinct structural features, single-atom catalysts open a new avenue for the design of more functional catalysts, whereas their bioapplications are still in their infancy. Due to the advantages, platinum single atoms supported by cadmium sulfide nanorods (Pt SAs-CdS) are synthesized to build an ultrasensitive photoelectrochemical (PEC) biosensing platform. With the decoration of Pt SAs, the PEC signal of CdS is significantly boosted. Furthermore, theory calculations indicate the positively charged Pt SAs could change the charge distribution and increase the excited carrier density of CdS. Meanwhile, it also suggests that Cu2+ can severely hinder the photoexcitation and electron–hole separation of CdS. As a proof of concept, prostate-specific antigen is chosen as the target analyte to demonstrate the superiority of the Pt SAs-CdS-based PEC sensing system. Abstract:In this paper, Bi2S3/AgBiS2 composite nanomaterials and PDA@Ag@N-CQDs were synthesized, and used as substrates and second antibody label respectively to construct a sandwich photoelectrochemical (PEC) sensor. The upconversion luminescence effect of N-CQDs can convert long wavelength light into short wavelength light that can be utilized by the substrate material, which can provide additional excitation light energy for the substrate material and further enhance the photoelectric response. Besides, Ag has SPR effect and can also promote electron transfer. The proposed sandwich immunosensor achieves detection of NSE in the concentration range of 0.001 ng mL-1 to 100 ng mL-1, with a detection limit of 0.28 pg mL-1 (S/N = 3). What's more, the proposed sensor also exhibits good stability, selectivity, as well as reproducibility, indicating its promising application prospects.

Abstract:Herein a novel electmchemiluminescence resonance energy transfer (ECL-RET) pair of isoreticular metal organic framework-3 (IRMOF-3) enriched nitrogen-doped graphene quantum dots (N-GQDs) luminescent material (N-GQDs@IRM0E-3@N-GQDs, donor) and Au nanoparticles modified zinc oxide nanomd (ZnO@Au, acceptor) were firstly designed to fabricate a dual-quenching sandwich-type ECL immunosensor for ultrasensitive analysis of pmcalcitonin (PCT). IRMOF-3 not only acted as a carrier for abundantly enriching luminescent N-GQDs by internal encapsulation and external decoration, but also a coreaction accelerator which could boost the transformation of persulfate ion S2O82- - to generate more anion radical SO4 center dot-, so as to promote the ECL emission of N-GQDs. Additionally, Au nanoparticles were loaded onto the ZnO nanomds through in-situ modification to obtain ZnO@Au, which was firstly applied as a dual-quencher of ECL signal. In the best test environment, a wide detection range from 0.0001 to 100 ng mL(-1) was obtained and the limit of detection is 12.58 fg mL(-1) (S/N = 3). With excellent reproducibility and sensitivity, the practical test consequence of PCT in human serum samples was satisfactory. These results suggested that this sensing system could provide a reference model for effective detection of disease markers in clinical medicine.
Electrochemiluminescence Double Quenching System Based on Novel Emitter GdPO4:Eu with Low-Excited Positive Potential for Ultrasensitive Procalcitonin Detection

Abstract:Nowadays, the electrochemiluminescence (ECL) immunosensor with the unique superiority of tunable luminescence and ultrahigh sensitivity has become one of the most promising immunoassay techniques, especially for low-abundance biomarkers analysis. However, the use of signal probes with high excited potential and applied emitters which owned good intensity but biotoxicity limited its application. Herein, an ECL resonance energy transfer strategy was developed based on protein bioactivity protection utilizing europium-doped phosphoric acid gadolinium (GdPO4:Eu) as novel low-potential luminophor (donor) and Pd@Cu2O as the quenching probe (acceptor). Specifically, GdPO4:Eu was first prepared by using the hydrothermal synthesis method to apply in ECL, and when it coexisted with K2S2O8, cathode, a strong ECL signal would be generated at a low potential of -1.15 V (vs Ag/AgCl), where the immunocompetence of antigens and antibodies can be maintained well. Electrical pair Eu3+/Eu2+, as the coreactant promoter, produced by potential excitation could produce more SO4 center dot- to accelerate the oxidation process of GdPO4:Eu. Meanwhile, Cu2O coated onto Pd (Pd@Cu2O), as a dual-quencher, enhanced the quenching effect of Pd alone and controlled the ECL intensity of the "signal on" state within a reasonable range. As a result, the proposed biosensor for detection of trace procalcitonin, a biomarker of systemic inflammatory response syndrome, exhibited a far low detection limit, 0.402 fg/mL (S/N = 3). Importantly, this work not only utilized a promising ECL emitter for biosensing platform construction but also had momentous potential in biomarker detection of disease diagnosis and clinical analysis.

Abstract:A sandwich-type photoelectrochemical (PEC) immunosensor for detecting N-terminal pro-brain natriuretic peptide (NT-proBNP) was constructed on the basis of SnO2/SnS2/mpg-C3N4 nanocomposites and PbS/SiO2, with the former as a photoactive matrix and the latter as an efficient quencher. SnO2/SnS2/mpg-C3N4 was synthesized via in situ growth of SnO2 and SnS2 on mesoporous graphene like C3N4 nanocomposites (mpg-C3N4). Specifically, SnO2/SnS2/mpg-C3N4 exhibited intense PEC signal responses, which are tens of times stronger than its each single component. Because of its superior performance, SnO2/SnS2/mpg-C3N4 was applied as a photoactive matrix and signal indicator for fabricating PEC immunosensor. Interestingly, the excellent PEC signals from SnO2/SnS2/mpg-C3N4 could be reduced severely with the addition of PbS/SiO2. Hence, the secondary antibody bioconjugates (PbS/SiO2-Ab2) were prepared as an efficient quencher. The mechanism of the quench reaction was further discussed in detail. On the basis of the interaction between the matrix and the quencher, the NT-proBNP immunosensor was fabricated and a wide linear range of 0.1 pg·mL-1 to 50 ng·mL-1 was obtained with a low detection limit of 0.05 pg·mL-1. Additionally, the PEC immunosensor manifested good stability, reproducibility, and selectivity, which could underlie robust platforms for detecting multitudinous biomarkers or other targets of interest.
Cu Nanoclusters-Encapsulated Liposomes: Toward Sensitive Liposomal Photoelectrochemical Immunoassay.

Abstract:Herein we report the strategy of liposome-mediated Cu2+-induced exciton trapping upon CdS quantum dots (QDs) for amplified photoelectrochemical (PEC) bioanalysis application. Specifically, the Cu nanoclusters (NCs)-encapsulated liposomes were first fabricated and then processed with antibodies bound to their external surfaces. After the sandwich immunocomplexing, the confined liposomal labels were subjected to sequential lysis treatments for the release of Cu NCs and numerous Cu2+ ions, which were then directed to interact with the CdS QDs electrode. The interaction of Cu2+ ions with CdS QDs could generate CuxS and form the trapping sites to block the photocurrent generation. Since the photocurrent inhibition is closely related with the Cu NCs-loaded liposomal labels, a novel and general "signal-off" PEC immunoassay could thus be tailored with high sensitivity. Meanwhile, a complementary "signal-on" fluorescent detection could be accomplished by measuring the fluorescence intensity originated from the Cu NCs. This work features the first use of Cu NCs in PEC bioanalysis and also the first NCs-loaded liposomal PEC bioanalysis. More importantly, by using other specific ions/reagents-semiconductors interactions, this protocol could serve as a common basis for the general development of a new class of liposome-mediated PEC bioanalysis.
