Abstract:Microencapsulation is a promising and feasible method to help probiotics against adverse situation and overcome storage instability. In this study, we prepared Bacillus smithii XY1 microcapsules by spray drying. Firstly, wall materials of the microcapsules were screened and the ratio between wall materials and protective agent was optimized. The results showed that 9% gum arabic and 5% soluble starch along with 4% skim milk powder were the best condition for spray drying through response surface method. Through scanning electron microscope (SEM), bacteria were successfully encapsuled in the wall materials and microcapsules presented complete structure without cracks. Compared to non-encapsulated bacteria, encapsulation increased tolerance of B. smithii XY1 in gastrointestinal environment, improved shelf time and showed a better effect on alleviating intestine inflammation of zebrafish induced by dextran dulfate sodium (DSS). Thus, this encapsulation approach may have the potential application for improving efficacy of B. smithii XY1 and other probiotics.

Abstract:Abstract The aim of these studies was to develop an effective formulation for microbial strains active in plant protection. Emulsification technique in oil was used to produce alginate microcapsules supplemented with organic additives to improve production process and stability of the preparations during storage. The release of microorganisms from the capsules in soil and their effectiveness in biocontrol of Fusarium wilt were evaluated. Three bacterial strains Burkholderia cepacia strain CAT5, Bacillus spp. strains PZ9 and SZ61, and fungus Trichoderma virens TRS106 were immobilized separately in calcium alginate supplemented with chitosan, peat powder, or skim milk. The productivity of microcapsules was enhanced by 60% when peat was added to the alginate matrix. Peat reduced also contamination of the capsules during storage, significantly enhancing their quality. By contrast, the addition of skim milk reduced quality of the microcapsules. The additives did not influence the viability of entrapped microorganisms and their release in soil. The survival of the microbial cells was mainly related to the kind of microorganism used, and the highest viability showed Bacillus sp. PZ9 and Trichoderma TRS106. Lyophilization of the microcapsules appeared to be unfavorable by reducing microbial viability in the capsules and in the soil after application. The best properties: good storage ability and sufficient microbial release in the soil, exhibited wet microcapsules amended with peat. These capsules were used to control Fusarium wilt in tomato plants. The protective effect was obtained when the microcapsule-entrapped bacteria PZ9 were used. The effectiveness of this bacterium was comparable with fungicide. Peat-amended microcapsules entrapping Bacillus PZ9 showed the best quality and may have potential for commercial use.
Preparation of Trichoderma asperellum Microcapsules and Biocontrol of Cucumber Powdery Mildew

Abstract:Microencapsulation is an important technique for protecting the viability and activity of microorganisms under adverse environmental conditions. To improve biological control, controlled-release microcapsules of Trichoderma asperellum were prepared and embedded in combinations of the biodegradable wall materials sodium alginate (SA). The microcapsules were evaluated for their ability to control cucumber powdery mildew in the greenhouse. The results showed that the highest encapsulation efficiency of 95% was obtained by applying 1% SA and 4% calcium chloride. The microcapsules provided good, controlled release and UV resistance, and could be stored for a long time. The greenhouse experiment revealed that the T. asperellum microcapsules had a maximal biocontrol efficiency of 76% against cucumber powdery mildew. In summary, embedding T. asperellum in microcapsules is a promising technique to improve the survivability of T. asperellum conidia. The T. asperellum microcapsules exerted significant biocontrol efficiency against cucumber powdery mildew. IMPORTANCE Trichoderma asperellum is widely found in plant roots and soil and has been used for the biocontrol of various plant pathogens; however, the control efficiency of T. asperellum is usually unstable in field trials. To improve the control efficiency of T. asperellum, in the present study, T. asperellum microcapsules were prepared using sodium alginate as wall material to reduce the effects of temperature, UV irradiation, and other environmental factors on its activity, and to significantly improve its biocontrol efficiency on cucumber powdery mildew. Microcapsules can prolong the shelf life of microbial pesticides. This study provides a new way to prepare a biocontrol agent against cucumber powdery mildew with high efficiency.
Characterization and evaluation of Bacillus isolates for their potential plant growth and biocontrol activities against tomato bacterial wilt

Abstract:About 200 Bacillus isolates were isolated from tomato and potato rhizosphere and examined for their antagonistic activities against Ralstonia solanacearum T-91, the causal agent of tomato bacterial wilt (TBW), in vitro and in vivo . Four strains, AM1, D16, D29 and H8, have shown high potential of antagonistic activity against the pathogen in laboratory and greenhouse experiments. In greenhouse, 81.1 to 89.0% reduction of disease incidence of TBW was recorded in treated tomato plants with 4 isolates, which also significantly (p > 0.05) increased plant height by 22.7 to 43.7% and dry weight by 47.93 to 91.55% compared with non-treated control. 16SrRNA gene sequence, the biochemical and physiological tests and fatty acid methyl esters analysis assigned strains AM1 and D29 as Bacillus amyloliquefaciens , while strains D16 and H8 as Bacillus subtilis and B. methylotrophicus , respectively. In addition, the 4 strains showed ability to inhibit growth of the three soil-borne fungi, produce indole-3- acetic acid, siderophores and also with exception of strain D16, the other 3 strains were capable of solubilizing phosphate. Therefore, these results suggest that out of 200 isolates, Bacillus stains AM1, D16, D29 and H8 support good antagonistic activity and could be applied as biocontrol agents against TBW under greenhouse conditions beside their potential to promote tomato plants growth. Abstract:This study aimed to develop microencapsulation technology using alginate to improve the viability and performance of Trichoderma harzianum. The method of ionic gelation was used to prepare the microparticles, and the efficiency of encapsulation was estimated to be 99%. The average size of the prepared microspheres was 2600 μm (wet) and 1000 μm (dry). Scanning electron microscopy revealed that the microspheres were approximately spherical. Fourier transform infrared spectrophotometer analysis indicated an interaction between T. harzianum and the microspheres. The results of temperature resistance and light stability against ultraviolet radiation emphasized the positive impact of microencapsulation in improving the viability and resistance of T. harzianum compared to the non-microencapsulated state. The disease percentage of Rhizoctonia solani and Sclerotinia sclerotiorum in plants treated with microencapsulated T. harzianum microcapsules was 8.88 % and 20 % respectively, but in the control group was 73.33 % (p ≤ 0.05).

Abstract:Gray mold (Botrytis cinerea) is a serious disease causing considerable losses to harvested tomato during storage and transportation. Extensive and unjudicial use of fungicides have certain deleterious effects such as evolved resistance in pathogen and environmental pollution. The ice cold chain transportation is the most commonly used method, but the results are not satisfactory. Currently, the trend is shifting towards prevention and control measures which ensures environmental protection, therefore, the biological control of postharvest diseases has become a hot spot. In this study, Bacillus subtilis L1-21 was used to control gray mold on tomato fruits, by adopting plate confrontation and fruit acupuncture method. To evaluate the preservative effect of biocontrol agent L1-21 on tomato, the gfp-tagged L1-21 was used to study colonization potential in harvested tomatoes assay. Colonization of L1-21-gfp in postharvest tomatoes was stable at 103 cfu/g by soaking and fluorescence method. Whereas Inhibition rate of B. cinerea was 39.63±4.73% in plate confrontation. Culture suspension of L1-21 at a concentration of 108 cfu/ml displayed best control effect: 86.57%±2.55% against tomato gray mold. In addition, different components exhibited different results such as cell pellet+saline (72.94%±8.49%), supernatant (41.18%±16.22%), sterilized culture (28.56%±6.54%). Further, suspension of B. cinerea and Bacillus L1-21 injected in tested tomatoes led to maximum control effect (81.40%±2.74%) after 4 h. Injection of L1-21 culture into tomato followed by pathogen B. cinerea spore solution resulted in 100%±0% control effect after 48 h. There was no significant difference in fruit quality between treated and untreated tomatoes. Taken together, B. subtilis L1-21 may lay theoretical foundation to be used in prevention of gray mold and other postharvest diseases during storage time.

Biological Control of Bacterial Wilt of Tomato by Bacillus spp. under Greenhouse Environment

Abstract:Eighty-one isolates of Bacillus were evaluated for their ability to inhibit the growth of Ralstonia solanacearum in vitro. Four isolates (B2,B5, B7 and B8) showed inhibitory effect on bacterial growth, were identified as Brevibacillus brevis B2, Bacillus subtilis B5, Ba. subtilis B7, and Ba. subtilis B8, respectively, by bacteriological assays, Biolog test and 16S rDNA sequence analysis. Four isolates promoted plant growth and reduced the infection of tomato wilt in sterile and nonsterile soil under greenhouse conditions to a varying degree as compared with non-treated control. Strain B2 showed significantly reduction of the disease infection by 80.0% and 87.4% in sterile and nonsterile soil,respectively.

Abstract:The biocontrol mechanisms and the fungistatic activities of the Bacillus marinus B-9987 were preliminarily studied under various treatments.The results showed that the fermentation broth of B-9987 had the highest activity in both fungitoxicity and controlling diseases.The minimum inhibitory concentration(MIC) to both Alternaria solani and Botrytis cinerea was 4μL/mL and the control effect was 85.1% and 88.4%,respectively.The activities of five defense enzymes in each treatment were significantly improved.The fermentation broth had the highest induction activities to phenylalanine ammonialyase(PAL),polyphenol oxidase(PPO) and superoxide dismutase(SOD),with 4.13,2.54 and 1.85 times higher activities than the control,respectively.These data indicated that the biocontrol effect of B-9987 might be resulted from the synergetic effect of its antifungal activity and the induced resistance of the plant.

Methods: The wall material and spray drying conditions were optimised. Morphological characteristics of microcapsule were measured, and soil remediation effects were tested under field conditions. Results: A relatively higher survival rate of B. megaterium microcapsule was obtained with 1:1 of chitosan/maltodextrin (w/w) when spray drying was performed at 150.0 degrees C, with the feed flow rates of 800 mL h(-1) and 1000 mL h(-1), respectively. The span value of 0.93 +/- 0.01 was obtained under above conditions. Microcapsule survival rate was 64.09 +/- 0.12% after 6 months of storage. Moreover, microcapsule successfully decreased NO3- and EC value in strongly saline soil by 46.5 +/- 1.48% and 45.2 +/- 1.51%, respectively. Conclusion: Bacillus megaterium NCT-2 microcapsules have application potential in the remediation of secondary salinisation soil.
Biofilm Formation Ability of Paenibacillus Polymyxa and Paenibacillus Macerans and Their Inhibitory Effect Against Tomato Bacterial Wilt

Abstract:The potential of biofilm formation of 16 Paenibacillus strains and their inhibitory effect against bacterial wilt of tomato seedlings were examined in this study. The crystal violet assay indicated that all strains of Paenibacillus except strain MB02-1202 formed biofilm after 96 and 144 h of incubation while there was not significant difference in biofilm formation between strains of Paenibacillus polymyxa and strains of Paenibacillus macerans. However, the increase level of biofilm formation was associated with the increase of the incubation time and the initial inoculum density. In addition, all Paenibacillus strains except strain MB02-428 reduced wilt incidence in tomato seedlings inoculated with Ralstonia solanacearum while the cell numbers of R. solanacearum in rhizosphere soil was reduced by all Paenibacillus strains compared to the pathogen control. In general, most strains of Paenibacillus were able to both form biofilm and protect tomato seedlings from bacterial wilt, indicating that biofilm formation may play an important role in the biocontrol of Paenibacillus. This is first study regarding the relationship between in vitro biofilm formation ability of Paenibacillus strains and their inhibitory activity against R. solanacearum.
Synergistic Effect Of Acetyl Salicylic Acid And Dl-Beta-Aminobutyric Acid On Biocontrol Efficacy Of Bacillus Strains Against Tomato Bacterial Wilt

Abstract:This study was conducted to assess the biocontrol efficacy of Bacillus subtilis (strain 4812) and Bacillus methylotrophicus (strain H8) individually or in combination with two plant defense inducers including Acetyl salicylic acid (ASA) and DL-Beta-aminobutyric acid (BABA) against tomato wilt caused by Ralstonia solanacearum. The pathogen was significantly inhibited by all treatments in the greenhouse and in vitro tests. The treatments H8+ASA and 4812+H8 were found to be the most effective treatments in in vitro tests. Applied on tomato seeds and as soil drenching, the disease was most inhibited by H8 whereas 4812+H8+ASA was the least effective treatment. High activity of phenylalanine ammonia-lyase was observed in the plants treated with 4812+H8, H8+ASA and 4812+H8+ASA. The highest expression of peroxidase and polyphenoloxidase enzymes was found in the plants treated with H8, 4812+H8 and H8+ASA. The result of real time PCR showed that concentration of the pathogen in stem tissues was significantly reduced in all treated plants and H8+ASA was the most effective treatment. This study revealed that combined application of Bacillus strains can be considered as a more effective biocontrol agent against tomato bacterial wilt. Furthermore, the synergistic effect of plant defense inducers was variable to different Bacillus strains.
Biocontrol of Tomato Wilt Disease by Bacillus Subtilis Isolates from Natural Environments Depends on Conserved Genes Mediating Biofilm Formation.

Abstract:Bacillus subtilis and other Bacilli have long been used as biological control agents against plant bacterial diseases but the mechanisms by which the bacteria confer protection are not well understood. Our goal in this study was to isolate strains of B. subtilis that exhibit high levels of biocontrol efficacy from natural environments and to investigate the mechanisms by which these strains confer plant protection. We screened a total of 60 isolates collected from various locations across China and obtained six strains that exhibited above 50% biocontrol efficacy on tomato plants against the plant pathogen Ralstonia solanacearum under greenhouse conditions. These wild strains were able to form robust biofilms both in defined medium and on tomato plant roots and exhibited strong antagonistic activities against various plant pathogens in plate assays. We show that plant protection by those strains depended on widely conserved genes required for biofilm formation, including regulatory genes and genes for matrix production. We provide evidence suggesting that matrix production is critical for bacterial colonization on plant root surfaces. Finally, we have established a model system for studies of B. subtilis-tomato plant interactions in protection against a plant pathogen.

Abstract:The use of lactic acid bacteria (LAB) for the biocontrol of fungi that cause fruit and vegetable deterioration is a highly promising strategy. However, one of the main challenges lies in maintaining both viability and antifungal activity even in conditions that are unfavorable for LAB. The microencapsulation of LAB can minimize the impact of environmental conditions, helping to maintain viability, but there is still little information on what occurs with respect to antifungal activity. Therefore, the objective of this study was to evaluate the effect of microencapsulation with several polymer blends on the viability and antifungal activity of Lactiplantibacillus plantarum TEP15 and L. pentosus TEJ4. Sodium alginate (2%), sodium alginate–gum arabic (2%:1%), sodium alginate–k-carrageenan (2%:0.05%), and sodium alginate–whey protein (0.75%:1.5%) were used as encapsulating polymers. After processing (day 0), as well as after 14 and 28 days of storage, the encapsulation efficiency, moisture content, bead size, and survival were evaluated. Likewise, the encapsulated bacteria were subjected to in vitro tests against Colletotrichum gloeosporioides, Penicillium AT21H10, and Fusarium sp. Capsules with sizes ranging from 1.47 mm to 1.88 mm were obtained, and all the wall materials tested had more than 85% encapsulation efficiency and allowed survival rates greater than 90% and 80% to be maintained after 14 and 28 days of storage, respectively. The encapsulated LAB inhibited the growth of mycelia by up to 100%, but, against spores, the greatest inhibition was 22.08% for all the fungi tested.

Abstract:The seed germination and tomato seedling tests showed that Bacillus subtilis SL-13 could promote the sprouting and seedling growth of tomato. The fresh and dry weight of tomato seedlings increased 42.86% and 18.75%, respectively. The control efficacies of the SL-13 to tomato Rhizoctonia rot were 20.65% and 35.23% in the greenhouse and field, respectively. The growth of the plant-pathogenic fungus Rhizoctonia solani was considerably inhibited in the presence of the strain SL-13 culture supernatant. The main antifungal protein was detected to be chitinase through vitro assay. The chitinase was purified with DEAE-Sepharose fast flow ion exchange column chromatography and Sephadex G-75 gel filtration for further characterization. The optimal pH and temperature for the chitinase activity were 7.0 and 50 (C, respectively. It was demonstrated that the enzyme was stable at pH 5-9 and 40-60 (C. 70% of the enzyme activity was retained when incubated at 121 (C and 0.11 MPa for 20 min, and the enzyme was not sensitive to protease K and ultraviolet radiation. Thus it is suitable for effective biological control in relatively unstable environment.

Abstract:Ralstonia solanacearum (Smith) has been a severe pathogen on tomato worldwide and no effective control measure is available yet. To develop a new bio-product for controlling this disease, antagonistic Bacillus amyloliquefaciens strains QL-5 and QL-18 were isolated from rhizosphere soil of tomato and used separately or together to fortify organic fertilisers to control bacterial wilt of tomato (BWT), both in the greenhouse and in the field. The bio-organic fertiliser (BOF) fortified with a mixed inoculation of QL-5 and QL-18 effectively decreased R. solanacearum disease incidence in both greenhouse and field conditions. However, the suppressive efficacy in the field depended on the season. Disease incidence in the spring crop season in 2009 and 2010 was significantly reduced, while a low suppressive effect was observed in the autumn crop season in 2008 and 2009. The initial R. solanacearum population in the soil was nearly three times lower in the spring before transplanting seedlings than in the autumn crop season. The pathogen population in the field was positively and significantly correlated with air temperature but was not correlated with relative humidity. The interaction between BOF and other soil and climatic factors that affect pathogen density in the field should be clarified.

Abstract:Biocontrol microbes are environment-friendly and safe for humans and animals. To seek biocontrol microbes effective in suppressing tomato gray mold is important for tomato production. Therefore, serial experiments were conducted to characterize the antagonism of Bacillus velezensis HY19, a novel self-isolated biocontrol bacterium, against Botrytis cinerea in vitro and the control on tomato gray mold in greenhouse. This bacterium produced extracellular phosphatase, protease, cellulose and siderophores, and considerably inhibited the growth of B. cinerea. A liquid chromatography-mass spectrometry (LC-MS) detected salicylic acid and numerous antifungal substances present in B. velezensis HY19 fermentation liquid (BVFL). When B. cinerea was grown on potato glucose agar, BVFL crude extract remarkably suppressed the fungal growth and reduced protein content and the activities of catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD). Transcriptome studies showed that BVFL crude extract significantly induced different expression of numerous genes in B. cinerea, most of which were down-regulated. Theses differently expressed genes were involved in the biological process, cell compartment, molecular functions, and metabolisms of glycine, serine, threonine, and sulfur in pathogen hyphae. Thus, this biocontrol bacterium antagonized B. cinerea in multiple ways due to the production of numerous antifungal substances that acted on multiple targets in the cells. BVFL significantly increased antioxidant enzyme activities in tomato leaves and decreased the incidence of tomato gray mold, with the control efficacies of 73.12-76.51%. Taken together, B. velezensis HY19 showed a promising use potential as a powerful bioagent against tomato gray mold.
Preparation of microcapsules and evaluation of their biocontrol efficacy

Abstract:In this study, a combination of Serratia nematophila L2 and Bacillus velezensis W24 was used to biocontrol Sclerotinia sclerotiorum. When the mixed ratio of L2 to W24 was 1:1, the inhibition rate on the growth of S. sclerotiorum was 88.1 %. To gain a large number of bacteria, the culture medium and conditions were optimized. When the medium formula involved molasses (8.890 g/L), soy peptone (6.826 g/L), and NaCl (6.865 g/L), and the culture conditions were 32 °C, inoculum 4%, rotation speed 200 rpm, and pH 7, the maximum amounts of bacterial cells obtained. In order to prepare microcapsules, spray drying conditions were optimized. These conditions included the soluble starch concentration of 30 g/100 mL, the inlet air temperature of 160 °C, and the feed flow rate of 450 mL/h. Under these optimized conditions to prepare microcapsules, the mixed strain (L2 and W24) exhibited a survival rate of 93.9 ± 0.9% and a viable bacterial count of 6.4 × 1012 cfu/g. In addition, microcapsules (GW24Ms) which contained strains L2 and W24 had good storage stability. In the pot experiment, GW24Ms could effectively reduce the disease of soybean plants and the control effect was 88.4%. Thus, the microbial agent represents a promising biocontrol solution for managing Sclerotinia in soybean.

Abstract:Present study was conducted to improve microencapsulation process of Clostridium tyrobutyricum (Ct) by optimizing process parameters to improve its in-vitro characteristics over that of free cells. All process parameters including wall material (w/v concentration: modified starch 3-7%; gelatin 2-6%; maltodextrin 3-7%), sample flow rate (250-450 mLh-1) and inlet air temperature (105-145°C) were analysed through single factor analysis. Response surface design test was used to develop multiple quadratic regression equations to fit the functional relationship between factors and response values and to choose the optimal conditions. The optimal conditions for maximum survival rate (82.030%) of encapsulated Ct were: 4% gelatin, 5% modified starch and 5% maltodextrin concentration with sample flow rate of 350 mLh-1 at inlet air temperature of 105°C. Encapsulation reduced the survival loss of Ct from 1.990 to 1.080 lgCFUg-1 under strong acidic condition (pH1) than free Ct. Survival loss of free Ct was 31.914% more than encapsulated Ct under high temperature treatment (90°C). Similarly, protected Ct showed higher survival rate under simulated gastric condition with long storage life. Encapsulation of Ct through optimized spray drying method efficiently improved its survival rate under strong acidic or high temperature environment with safe transit through gastrointestinal tract and also eradicates the technological limitations which preventing the use of many probiotic strains

Abstract:Bacillus subtilis is a potential biocontrol agent for management of fungal diseases on greenhouse crops. Colonization of plant roots by B. subtilis enhances growth and reduces disease incidence and severity to minimize yield losses and improve quality. The mechanisms through which B. subtilis may contribute to control of fungal diseases include competition with other microbes, production of inhibitory chemicals, and induction of plant resistance. Specific strains of B. subtilis exhibit strong antagonistic activity against mycelial growth of different fungal pathogens through antibiotic production. The effectiveness of B. subtilis strain QST 713 against several important diseases of cucumber and tomato grown under greenhouse conditions was assessed. The major diseases on cucumber are Pythium crown and root rot (Pythium aphanidermatum), Fusarium root and stem rot (Fusarium oxysporum f. sp. radicis-cucumerinum), gummy stem blight (Didymella bryoniae, anamorph Phoma cucurbitacearum), and powdery mildew (Podosphaera xanthii). On greenhouse tomatoes, the most important diseases are gray mold (Botrytis cinerea), Pythium damping-off (Pythium aphanidermatum), Fusarium wilt (Fusarium oxysporum), and powdery mildew (Oidium neolycopersici). Applications of B. subtilis made on cucumber and tomato plants before or after the onset of pathogen inoculation demonstrated efficacy against various diseases. The potential for broad-spectrum activity against different pathogens on these crops is demonstrated.
