Abstract:Gray mold, caused by Botrytis cinerea, is one of the most serious and widespread diseases that affect tomato plants and fruit and reduce yield. In the present study, Bacillus altitudinis B1-15 isolated from saline-alkali soil showed strong antifungal activity against B. cinerea. In pot experiments, a significant biocontrol efficacy of strain B1-15 was observed against tomato gray mold at 3-leaf stage (87.63%) and 6-leaf stage (76.43%), and the biocontrol efficacy was found to be 39.83% even after inoculation for 35 d at 6-leaf stage. A significant improvement of stem diameter, plant height, dry weight and fresh weight was also observed after tomato plants inoculated with strain B1-15. Strain B1-15, applied to tomato fruit by wounded or unwounded inoculation, could significantly inhibit gray mold and prolong the storage period of postharvest tomato fruit. The antifungal substances of strain B1-15 were found both in intraspore and extraspore, and the strain was also able to produce volatile organic compounds (VOCs), which could cause the mycelia of B. cinerea swelling and dehydrated malformation, the shortening of mycelia septum and the decomposition of cell wall and vacuole. Besides that, strain B1-15 could produce protease at a greater level, and chitinase and amylase at a lower level, which contributed to its antifungal activity. Additionally, 15 and 9 chemical compounds (similarity ≥ 80%) were individually identified from the crude extracts of the supernatant and precipitate of strain B1-15 via gas chromatography-mass spectrometry (GC–MS), including two VOCs 3-methylbutanoic acid and 2-methylbutanoic acid detected in both the supernatant and precipitate. Therefore, strain B1-15 could be used as a biocontrol agent and has great potential to control tomato gray mold and improve tomato yield and storage quality in practical production.

Abstract:Gray mold is a destructive plant disease caused by a fungal pathogen Botrytis cinerea . The use of plant growth promoting rhizobacteria (PGPR) has proven to be a promising method to control this disease. Bacillus velezensis K01 was isolated from the rhizosphere of planting tomatoes. Strain K01 has a range of roles, including the ability to solubilize phytate phosphorus, stimulate resistant response, and produce indoleacetic acid (IAA), protease, cellulase, and antimicrobial substances. Strain K01 was found to inhibit 12 phytopathogenic fungi and 5 phytopathogenic bacteria. Specially, strain K01 demonstrated a biocontrol efficiency of over 78% against gray mold caused by B. cinerea on the leaves and fruits of tomato and pepper. Additionally, K01 was found to promote the growth of maize seedlings. Further genomic analysis revealed that K01 belongs to B. velezensis , which is consistent with phylogenetic analysis, average nucleotide polymorphism (ANI), and digital DNA–DNA hybridization (dDDH). The genome of strain K01 had a size of 3,927,799 bp and deduced 3866 predicted genes, with an average guanine-cytosine (GC) content of 46.5%. Based on the analyses of genomic secondary metabolites, over 18.4% of the genome was annotated to 12 gene clusters related to antimicrobial metabolite synthesis. Additionally, genome annotation and comparative genomics identified several genes associated with plant growth promotion and environmental adaption. These findings suggest that B. velezensis K01 has the potential to serve as a new biocontrol agent for management of gray mold on tomato and pepper.

Biocontrol of Postharvest Gray Mold of Cherry Tomatoes with the Marine Yeast Rhodosporidium Paludigenum

Abstract:The marine yeast Rhodosporidium paludigenum was tested for its biocontrol efficacy in reducing postharvest gray mold of cherry tomatoes caused by Botrytis cinerea in vitro and in vivo tests. A previously unreported gray mold antagonist, R. paludigenum, significantly reduced disease incidence on cherry tomatoes. The results showed that suspensions containing 1×108CFU/ml washed cells of R. paludigenum provided the highest levels of inhibition of gray mold, while the unwashed cell suspension provided less protection against the disease after 5days at 25°C. Increasing concentrations of R. paludigenum influenced decay incidence in cherry tomato wounds: the disease incidence decreased as the concentration of R. paludigenum was increased. Moreover, in the small-scale experiment, the incidence of gray mold on cherry tomatoes treated with R. paludigenum was 14.7% and 46.3% of the control at 15°C after 10days, respectively. In cherry tomato wounds, the population of R. paludigenum reached peak levels 72h after inoculation, and then began to decline slowly. In conclusion, our results showed that competition for nutrients may have an important role in biological control of gray mold of cherry tomatoes with R. paludigenum.
Identification of a Novel Antifungal Strain and Its Biocontrol Effect on Plant Pathogenic Fungi

Abstract:With the increase in the incidence of plant pathogenic fungi, the discovery and development of efficient, low-toxic and security antifungal compounds became even more important. A strain KZ001, antagonising plant pathogenic fungi, was isolated from a soil sample of Zhejiang Province, China. The strain was identified as Bacillus subtilis by sequencing 16S rRNA gene and BLAST searching GenBank. One of its metabolites from the microorganism was purified and identified as a macrolide compound with molecular weight of 402 Da by ESI/MS,(1)H-NMR and (13)C-NMR. The macrolide compound from Bacillus subtilis KZ001 showed strong inhibition against the fungal pathogens Aspergillus niger, Fusarium graminearu, Afternaria alternata, Exserohilum turcium, etc. As a result, the compound has the potential application value in biocontrol of phytopathogens in agriculture.

Abstract:Gray mold (Botrytis cinerea) is a serious disease causing considerable losses to harvested tomato during storage and transportation. Extensive and unjudicial use of fungicides have certain deleterious effects such as evolved resistance in pathogen and environmental pollution. The ice cold chain transportation is the most commonly used method, but the results are not satisfactory. Currently, the trend is shifting towards prevention and control measures which ensures environmental protection, therefore, the biological control of postharvest diseases has become a hot spot. In this study, Bacillus subtilis L1-21 was used to control gray mold on tomato fruits, by adopting plate confrontation and fruit acupuncture method. To evaluate the preservative effect of biocontrol agent L1-21 on tomato, the gfp-tagged L1-21 was used to study colonization potential in harvested tomatoes assay. Colonization of L1-21-gfp in postharvest tomatoes was stable at 103 cfu/g by soaking and fluorescence method. Whereas Inhibition rate of B. cinerea was 39.63±4.73% in plate confrontation. Culture suspension of L1-21 at a concentration of 108 cfu/ml displayed best control effect: 86.57%±2.55% against tomato gray mold. In addition, different components exhibited different results such as cell pellet+saline (72.94%±8.49%), supernatant (41.18%±16.22%), sterilized culture (28.56%±6.54%). Further, suspension of B. cinerea and Bacillus L1-21 injected in tested tomatoes led to maximum control effect (81.40%±2.74%) after 4 h. Injection of L1-21 culture into tomato followed by pathogen B. cinerea spore solution resulted in 100%±0% control effect after 48 h. There was no significant difference in fruit quality between treated and untreated tomatoes. Taken together, B. subtilis L1-21 may lay theoretical foundation to be used in prevention of gray mold and other postharvest diseases during storage time.

Abstract:About 200 Bacillus isolates were isolated from tomato and potato rhizosphere and examined for their antagonistic activities against Ralstonia solanacearum T-91, the causal agent of tomato bacterial wilt (TBW), in vitro and in vivo . Four strains, AM1, D16, D29 and H8, have shown high potential of antagonistic activity against the pathogen in laboratory and greenhouse experiments. In greenhouse, 81.1 to 89.0% reduction of disease incidence of TBW was recorded in treated tomato plants with 4 isolates, which also significantly (p > 0.05) increased plant height by 22.7 to 43.7% and dry weight by 47.93 to 91.55% compared with non-treated control. 16SrRNA gene sequence, the biochemical and physiological tests and fatty acid methyl esters analysis assigned strains AM1 and D29 as Bacillus amyloliquefaciens , while strains D16 and H8 as Bacillus subtilis and B. methylotrophicus , respectively. In addition, the 4 strains showed ability to inhibit growth of the three soil-borne fungi, produce indole-3- acetic acid, siderophores and also with exception of strain D16, the other 3 strains were capable of solubilizing phosphate. Therefore, these results suggest that out of 200 isolates, Bacillus stains AM1, D16, D29 and H8 support good antagonistic activity and could be applied as biocontrol agents against TBW under greenhouse conditions beside their potential to promote tomato plants growth. Abstract:Botrytis cinerea, a major phytopathogenic fungus, has been reported to infect more than 200 crop species worldwide, and it causes massive losses in yield. The aim of this study was to evaluate the inhibitory abilities and effects of Bacillus amyloliquefaciens RS-25, Bacillus licheniformis MG-4, Bacillus subtilis Z-14, and Bacillus subtilis Pnf-4 and their culture filtrates and extracts against the gray mold caused by B. cinerea on postharvest tomato, strawberry, and grapefruit. The results revealed that the cells of Z-14, culture filtrate of RS25, and cells of Z-14 showed the strongest biocontrol activity against the gray mold on the strawberry, grape, and tomato fruit, respectively. All the strains produced volatile organic compounds (VOCs), and the VOCs of Pnf-4 displayed the highest inhibition values. Based on headspace solid-phase microextraction in combination with gas chromatography-mass spectrometry, esters accounted for the largest percentage of the VOCs produced by RS-25, MG-4, Z-14, and Pnf-4 (36.80%, 29.58%, 30.78%, and 36.26%, respectively). All the strains showed potent cellulase and protease activities, but no chitinase activity. RS-25, Z-14, and MG-4, but not Pnf-4, grew on chrome azurol S agar, and an orange halo was formed around the colonies. All the strains showed biofilm formation, fruit colonization, and lipopeptide production, which may be the main modes of action of the antagonists against B. cinerea on the fruit. This study provides the basis for developing natural biocontrol agents against the gray mold caused by B. cinerea on postharvest fruit.
Unlocking antagonistic potential of Bacillus amyloliquefaciens KRS005 to control gray mold

Abstract:To establish a safe, efficient, and simple biocontrol measure for gray mold disease caused by Botrytis cinerea , the basic characteristics and antifungal activity of KRS005 were studied from multiple aspects including morphological observation, multilocus sequence analysis and typing (MLSA–MLST), physical-biochemical assays, broad-spectrum inhibitory activities, control efficiency of gray mold, and determination of plant immunity. The strain KRS005, identified as Bacillus amyloliquefaciens , demonstrated broad-spectrum inhibitory activities against various pathogenic fungi by dual confrontation culture assays, of which the inhibition rate of B. cinerea was up to 90.3%. Notably, through the evaluation of control efficiency, it was found that KRS005 fermentation broth could effectively control the occurrence of tobacco leaves gray mold by determining the lesion diameter and biomass of B. cinerea on tobacco leaves still had a high control effect after dilution of 100 folds. Meanwhile, KRS005 fermentation broth had no impact on the mesophyll tissue of tobacco leaves. Further studies showed that plant defense-related genes involved in reactive oxygen species (ROS), salicylic acid (SA), and jasmonic acid (JA)-related signal pathways were significantly upregulated when tobacco leaves were sprayed with KRS005 cell-free supernatant. In addition, KRS005 could inhibit cell membrane damage and increase the permeability of B. cinerea . Overall, KRS005, as a promising biocontrol agent, would likely serve as an alternative to chemical fungicides to control gray mold.

Abstract:[Aims] This paper aims to study the compound microbial disinfectant against cucumber gray mold.[Methods] The affinity test was carried out by plate confrontation method;The optimum ratio was determined by measuring the effect of strain fermentation on the spores and hyphae of gray mold pathogens.The pot test was used to determine the prophylactic effect of preparation.[Results] The strains D1,BSD-2,ch2-22 have synergistic effect.When the ratio of the three fermentation filtrates was 5∶4∶1 and the number of live spore was 1.5x107 cfu/mL,the compound fungicide for control of gray mold of cucumber was 86.25％.[Conclusions] Compound biocides have a strong anti-disease and promoting effect on botrytis cinerea,which has a potential to develop as a new type of biological pesticide.
Gender abuse, depressive symptoms, and substance use among transgender women: a 3-year prospective study.

Abstract:OBJECTIVES We examined the effects of gender abuse (enacted stigma), depressive symptoms, and demographic, economic, and lifestyle factors on substance use among transgender women. METHODS We conducted a 3-year prospective study (December 2004 to September 2007) of 230 transgender women aged 19 to 59 years from the New York Metropolitan Area. Statistical techniques included generalized estimating equations with logistic and linear regression links. RESULTS Six-month prevalence of any substance use at baseline was 76.2%. Across assessment points, gender abuse was associated with alcohol, cannabis, cocaine, or any substance use during the previous 6 months, the number of days these substances were used during the previous month, and the number of substances used. Additional modeling associated changes in gender abuse with changes in substance use across time. Associations of gender abuse and substance use were mediated 55% by depressive symptoms. Positive associations of employment income, sex work, transgender identity, and hormone therapy with substance use were mediated 19% to 42% by gender abuse. CONCLUSIONS Gender abuse, in conjunction with depressive symptoms, is a pervasive and moderately strong risk factor for substance use among transgender women. Improved substance abuse treatment is sorely needed for this population.
Actinomycete as biocontrol agents against tomato gray mold disease caused by Botrytis cinerea

Abstract:The present work aims to isolate actinomycete bacteria with antagonistic abilities towards Botrytis cinerea, the causal agent of gray mold, from a soil sample collected from the rhizosphere of a healthy tomato grove. In vitro confrontation led to the isolation of 104 actinomycete isolates; fifteen isolates have shown the most significant mortality rate of the mycelial growth of B. cinerea (>50%). Based on the results of this screening, representative strains were selected to verify their in vivo antagonistic activity on tomato fruits; the reduction of B. cinerea has a percentage ranging from 52.38% to 96.19%. Furthermore, the actinomycete isolates were evaluated for their plant growth-promoting (PGP) properties and their ability to produce biocontrol-related extracellular enzymes viz., amylase, protease, cellulase, chitinase, esterases, and lecithinase. Indeed, Ac70 showed high β-1,3-glucanase activity and siderophore production (17U/ml and 43% respectively), and the highest chitinase activity (39μmol/ml) was observed for Ac24. These results indicated that these actinomycetes might potentially control gray mold caused by B. cinerea on tomato fruits. Investigations on enhancing the efficacy and survival of the biocontrol agent in planta and finding out the best formulation are recommended for future research.

Bio-perfume guns: Antifungal volatile activity of Bacillus sp. LNXM12 against postharvest pathogen Botrytis cinerea in tomato and strawberry

Abstract:Gray mold disease, caused by Botrytis cinerea is a major postharvest disease impacting fruits such as strawberries and tomatoes. This study explores the use of volatile organic compounds (VOCs) produced by Bacillus spp. as eco-friendly biocontrol agents against B. cinerea. In vitro experiments demonstrated that VOCs from Bacillus sp. LNXM12, B. thuringiensis GBAC46, and B. zhanghouensis LLTC93-VOCs inhibited fungal growth by 61.2%, 40.5%, and 21.6%, respectively, compared to the control. LNXM12 was selected for further experiments due to its highest control efficacy of 58.3% and 76.6% on tomato and strawberry fruits, respectively. The LNXM12 VOCs were identified through gas chromatography-mass spectrometry (GC-MS) analysis, and 22 VOCs were identified. Synthetic VOCs with the highest probability percentage, namely ethyloctynol, 3-methyl-2-pentanone (3M2P), 1,3-butadiene-N, N-dimethylformamide (DMF), and squalene were used in experiments. The results showed that the synthetic VOCs ethyloctynol and 3M2P were highly effective, with an inhibition rate of 56.8 and 57.1% against fungal mycelium radial growth at 120 μg/mL on agar plates. Trypan blue staining revealed strongly disrupted, deeper blue, and lysed mycelium in VOC-treated B. cinerea. The scanning and transmission electron microscope (SEM and TEM) results showed that fungal mycelium was smaller, irregular, and shrunken after synthetic VOC treatments. Furthermore, the synthetic VOCs Ethyloctynol and 3M2P revealed high control efficacy on tomatoes and strawberries infected by B. cinerea. The control efficacy on leaves was 67.2%, 66.1% and 64.5%, 78.4% respectively. Similarly, the control efficiency on fruits was 45.5%, 67.3% and 46.3% 65.1%. The expression of virulence genes in B. cinerea was analyzed, and the results revealed that selected genes BcSpl1, BcXyn11A, BcPG2, BcNoxB, BcNoxR, and BcPG1 were downregulated after VOCs treatment. The overall result revealed novel mechanisms by which Bacillus sp. volatiles control postharvest gray mold disease.
Biological Control of Verticillium Wilt and Growth Promotion in Tomato by Rhizospheric Soil-Derived Bacillus amyloliquefaciens Oj-2.16

Abstract:Verticillium wilt disease caused by Verticillium dahliae seriously affects tomato quality and yield. In this work, strain Oj-2.16 was isolated from rhizosphere soil of the medicinal plant Ophiopogon japonicas and identified as Bacillus amyloliquefaciens on the basis of morphological, physiological, and biochemical characteristics and 16S rDNA sequencing. Strain Oj-2.16 exhibited a high inhibition rate against V. dahliae, and the hyphae inhibited by Oj-2.16 were found to be destroyed on scanning electron microscopy. Lipopeptide and dipeptide genes were detected in the Oj-2.16 genome by PCR amplification involved in surfactin, iturin, fengycin, and bacilysin biosynthesis. In pot experiments, the biocontrol efficacy of strain Oj-2.16 against Verticillium wilt in tomato was 89.26%, which was slightly higher than the efficacy of the chemical fungicide carbendazim. Strain Oj-2.16 can produce indole acetic acid, siderophores, assimilate various carbon sources, and significantly promoted the growth of tomato seedlings by increasing plant height, root length, stem width, fresh weight, and dry weight by 44.44%, 122.22%, 80.19%, 57.65%, 64.00%, respectively. Furthermore, defense-related antioxidant CAT, SOD, POD, and PAL enzyme activities significantly increased and MDA contents significantly decreased in tomato seedlings treated with strain Oj-2.16 upon inoculation of V. dahliae compared with the pathogen-inoculated control. In summary, we concluded that B. amyloliquefaciens Oj-2.16 could be used as a promising candidate for the biocontrol of Verticillium wilt and as plant growth stimulator of tomato.

Abstract:The overuse of chemical fungicides against fungal pathogens adversely affects soil and plant health, resulting in environmental problems and food safety. Therefore, biocontrol is considered as an environmentally friendly and cost-effective green technique in environmental protection and agricultural production. We obtained a bacterial strain N23 from a contaminated plate which showed significant inhibition to anthracnose. The strain N23 was identified as Bacillus velezensis based on 16S rRNA gene, gyrA gene, and whole-genome sequence. The bacterium N23 was able to suppress the mycelial growth of numerous plant pathogenic fungi on solid media. Tomato seeds treated with strain N23 showed significantly higher germination levels than untreated ones. Moreover, strain N23 effectively reduced the lesion area of pepper anthracnose disease in planta. The gene clusters responsible for antifungal metabolites (fengycin, surfactin, and iturin) were identified in the genome sequence of N23 based on genome mining and PCR. Furthermore, methanol extracts of the bacterial culture caused significant inhibition in growth of the fungal Colletotrichum sp. and Botrytis cinerea. These findings suggested that B. velezensis N23 could be a potential biocontrol agent in agricultural production and a source of antimicrobial compounds for further exploitation.

Abstract:To evaluate the potential of a bacterial strain as a fungal disease control agent and plant growth promoter, its inhibitory effects on phytopathogens such as Bipolaris sorokiniana, Botrytis cinerea, Colletotrichum capsici, Fusarium graminearum, F. oxysporum, Neocosmospora rubicola, Rhizoctonia solani, and Verticillium dahliae were investigated. The results showed that the inhibitory rates in dual-culture and sterile filtrate assays against these eight phytopathogens ranged from 57% to 83% and from 36% to 92%. The strain was identified as Bacillus velezensis based on morphological and physiological characterization as well as phylogenetic analyses of 16S rRNA and the gyrase subunit A protein (gyrA) regions. The results demonstrated that B. velezensis was able to produce fungal cell-wall-degrading enzymes, namely, protease, cellulase, and β-1,3-glucanase, and the growth-promotion substances indole-3-acetic acid (IAA) and siderophore. Furthermore, B. velezensis BV01 had significant control effects on wheat root rot and pepper Fusarium wilt in a greenhouse. Potted growth-promotion experiments displayed that BV01 significantly increased the height, stem diameter, and aboveground fresh and dry weights of wheat and pepper. The results imply that B. velezensis BV01, a broad-spectrum biocontrol bacterium, is worth further investigation regarding its practical applications in agriculture.

Abstract:Trichoderma harzianum (TM) is a soil-borne beneficial fungus that positively affects plant growth and defense. TM shows antagonism against multiple plant pathogens; however, the TM as a biocontrol agent against Botrytis cinerea (BC) and underlying mechanisms remain unclear. Herein we studied the effects of TM on BC by dual-culture assay and greenhouse bioassay in tomato plants. In vitro experiments showed that TM rapidly occupied most of the nutrient space and eventually covered BC on potato dextrose agar (PDA) medium. The TM inhibition rate against BC was 62.05% on the sixth day of culture. The inhibitory effect of TM spore suspension on gray mold inoculated with spore suspension and solid inoculants was 33.66% and 69.44%, respectively in tomato plants. Notably, TM decreased the leaf concentrations of BC-induced hydrogen peroxide and malondialdehyde but improved the redox state. TM inoculation caused biphasic peaks in the expression of secondary metabolism and defense-related genes and increased the concentrations of flavonoids, phenols and lignin in tomato leaves. Moreover, TM alleviated stomatal closure and enhanced the net photosynthesis rate and the activity of RuBisCO and FBPase compared with only BC inoculation. Our results suggest that TM spore suspension improves tomato resistance to gray mold disease by stimulating cellular redox, secondary metabolites and stomatal movement.

Abstract:Gray mold, caused by Botrytis cinerea is a major cause of post-harvest rot of fresh fruits and vegetables. The utilization of selected microorganisms as biocontrol agents is a promising alternative to effectively control gray mold on tomatoes. The current study was conducted to explore potential biocontrol mechanisms of the Pseudomonas strain to control infections on post-harvest tomatoes. Among the 8 tested bacterial isolates, Pseudomonas protegens ML15 demonstrated antagonistic activity to Botrytis cinerea . Moreover, P. protegens ML15 exhibited the production of siderophores, hydrogen cyanide, ammonia, exopolysaccharides, lipase, biosurfactant, 2,4-diacetylphloroglucinol, and several other antifungal compounds, such as 1-tetradecanol, cyclododecane, 2,4-di-tert-butylphenol, and 2-methyl-1-hexadecanol. A comprehensive genomic analysis of P. protegens ML15 unravels 18 distinct genetic regions with the potential for biosynthesizing secondary metabolites, known for their pivotal role in biocontrol responses against plant pathogens. In vivo , experiments showed that both culture suspension and cell-free supernatant of P. protegens ML15 significantly reduced fungal growth (53.0 ± 0.63%) and mitigated disease development (52.8 ± 1.5%) in cherry tomatoes at four days post- B . cinerea inoculation. During the infection, the application of P. protegens ML15 resulted in the augmentation of total antioxidant, phenolic content, and ascorbic acids content. Thus, our results suggested that P. protegens ML15’s role as a biocontrol agent against B. cinerea -induced postharvest tomato decay achieved through the secretion of antifungal substances, induction of tomato defense responses, and inhibition of mycelial growth of B. cinerea . These findings provide a significant contribution to the ongoing search for alternative, eco-friendly methods of controlling gray mold in fresh products. The utilization of P. protegens ML15 as a biocontrol agent could help to reduce the reliance on chemical fungicides and promote sustainable agriculture practices.

Abstract:Soil-borne plant diseases seriously threaten the tomato industry worldwide. Currently, eco-friendly biocontrol strategies have been increasingly considered as effective approaches to control the incidence of disease. In this study, we identified bacteria that could be used as biocontrol agents to mitigate the growth and spread of the pathogens causing economically significant diseases of tomato plants, such as tomato bacterial wilt and tomato Fusarium wilt. Specifically, we isolated a strain of Bacillus velezensis (RC116) from tomato rhizosphere soil in Guangdong Province, China, with high biocontrol potential and confirmed its identity using both morphological and molecular approaches. RC116 not only produced protease, amylase, lipase, and siderophores but also secreted indoleacetic acid, and dissolved organophosphorus in vivo. Moreover, 12 Bacillus biocontrol maker genes associated with antibiotics biosynthesis could be amplified in the RC116 genome. Extracellular secreted proteins of RC116 also exhibited strong lytic activity against Ralstonia solanacearum and Fusarium oxysporum f. sp. Lycopersici. Pot experiments showed that the biocontrol efficacy of RC116 against tomato bacteria wilt was 81%, and consequently, RC116 significantly promoted the growth of tomato plantlets. Based on these multiple biocontrol traits, RC116 is expected to be developed into a broad-spectrum biocontrol agent. Although several previous studies have examined the utility of B. velezensis for the control of fungal diseases, few studies to date have evaluated the utility of B. velezensis for the control of bacterial diseases. Our study fills this research gap. Collectively, our findings provide new insights that will aid the control of soil-borne diseases, as well as future studies of B. velezensis strains.

Screening and identification of antagonistic actinomycete LA-5 against Botrytis cinerea

Abstract:In this study, actinomycetes were isolated from rhizosphere soil of tomato by spread plate method, with the pathogen Botrytis cinerea as the target fungus. An actinomycete isolate LA-5 with strong antifungal activity was obtained by confrontation culture and Oxford cup double screening method. According to cultural characteristics, physiological and biochemical properties and 16S rDNA analysis, the strain LA-5 was primarily identified as Streptomyces chungwhensis. The second screening results showed that the fermentation filtrate of LA-5 could substantially inhibit the spore germination and mycelium growth of B. cinerea. Both conidium germination and mycelia growth could be inhibited by 100 times fermentation broth of LA-5, with the inhibiting rate being higher than 50%. Furthermore, the inhibited colony showed white mycelia, with sparse and thin aerial hypha, and the obviously decreased branches. Results from in vitro biocontrol experiment showed that control efficiency of filtrate from LA-5 fermentation liquid on B. cinerea could be up to 83.4%, indicating that the strain was a potential antagonist against tomato gray mold.
