Abstract:Intracellular pH (pHi) is an important parameter associated with cellular behaviors and pathological conditions. Sensing pHi and monitoring its changes in live cells are essential but challenging due to the lack of effective probes. We herein report a pH-sensitive fluorogen for pHi sensing and tracking. The dye is a tetraphenylethene-cyanine adduct (TPE-Cy). It is biocompatible and cell-permeable. Upon diffusing into cells, it responds sensitively to pHi in the entire physiological range, visualizing the acidic and basic compartments with intense red and blue emissions, respectively. The ratiometric signal of the red and blue channels can thus serve as an indicator for local proton concentration. The utility of TPE-Cy in pHi imaging and monitoring is demonstrated with the use of confocal microscopy, ratiometric analysis, and flow cytometry.

Abstract:Intracellular pH plays a pivotal role in various biological processes. In eukaryotic cells, lysosomes contain numerous enzymes and proteins exhibiting a variety of activities and functions at acidic pH (4.5-5.5), and abnormal variation in the lysosomal pH causes defects in lysosomal function. Thus, it is important to investigate lysosomal pH in living cells to understand its physiological and pathological processes. In this work, we designed a one-step synthesized rhodamine derivative (RM) with morpholine as a lysosomes tracker, to detect lysosomal pH changes with high sensitivity, high selectivity, high photostability and low cytotoxicity. The probe RM shows a 140-fold fluorescence enhancement over a pH range from 7.4 to 4.5 with a pKa value of 5.23. Importantly, RM can detect the chloroquine-induced lysosomal pH increase and monitor the dexamethasone-induced lysosomal pH changes during apoptosis in live cells. All these features demonstrate its value of practical application in biological systems.

Abstract:A new near-infrared fluorescent probe Qcy-OH with a turn-on mechanism based on a acceptor-donor-acceptor (ADA) π-electron system that can undergo an internal charge transfer to form new fluorochromes was developed for monitoring pH fluctuations in biological systems. With the pH value increases, Qcy-OH exhibited a strong pH-sensitive response from 1.1 to 9.0 (pKa = 4.41) and a good linear response from pH 3.0 to 5.5 in aqueous solution and in living cells. In addition, based on the phenolic and ketonic structure interconversions of the fluorescent backbone, Qcy-OH showed rapidly and reversibly response to pH with high selectivity. Because the introduce of two benzothiazolium units in the backbone of the probe as the electron-withdrawing group to enhance the selectivity for intracellular lysosomes, the probe had been applied successfully for tracking lysosomal pH changes and the fluorescence changes showed a good linear enhancement from pH 3.0 to 7.4 in living cells. We believe that this sulfur-driving lysosomes-targeting ability of the probe affords a guarantee for achieving long-term monitoring of lysosomal pH biology by the elimination of harmful protonating effects of the probe.

Abstract:A dual-emission pH-sensitive fluorescent probe was developed, which displays green fluorescence in alkaline after deprotonation, while orange emission in acid at aggregated state. This novel probe allows the specific light-on in acidic lysosomes of cancer cells and tumors in nude mice, which indicates its potential application in cancer diagnosis.

Abstract:Two rhodamine-based pH probes (RhP and RhPA) were synthesized via the click reaction. The probes exhibited high pH sensitivity and selectivity with significant fluorescence intensity enhancement. Cell imaging experiments demonstrated RhPA was a good lysosome targeting probe in living cells with low cytotoxicity and excellent photostability.

Abstract:pH regulates the function of many organelles and plays a pivotal role in requiring multitud cellular behaviors. Compared with single fluorescent probes, ratio fluorescent probes have higher sensitivity and immunity to interference. Herein, a novel Janus ratio nanoprobe was developed for intracellular pH detection. Modified rhodamine B probe and fluorescein isothiocyanate (FITC) were individually encapsulated in the independent hemispheres of Janus microparticles fabricated via Pickering emulsion. Moreover, it exhibits a satasified ratiometric detection of pH compared to the previous core-shell structure and organic small molecule probe. Accordingly, the Janus nanoprobe possesses many important features as an attractive sensor, including high antijamming capability, excellent stability, good reversibility and low cytotoxicity. Variations of the two fluorescence intensities (Fgreen/Fred) resulted in a ratiometric pH fluorescent sensor, which can respond to wide range of pH values from 3 to 8. To be more specific, with a single excitation wavelength of 488 nm, there are dual emission bands centered at 538 nm and 590 nm. Also the Janus nanoprobe displays a excellent linear relationship in the physiologically relevant pH range of 4.0-6.0. Consequently, detecting of pH and imaging was successfully achieved in living cells, which provides a simple and reliable method for detecting intracelluar pH and other similar substances.

Abstract:Abnormal pH values in the organelles are closely associated with inappropriate cellular functions and many diseases. Monitoring subcellular pH values and their variations is significant in biological processes occurring in living cells and tissues. Herein, we develop a series of ratiometric fluorescence nanoprobes for quantification and imaging of pH values with a single-wavelength excitation in cytoplasm, lysosomes, and mitochondria. The nanoprobes consist of mesoporous silica nanoparticles assembled with aminofluorescein as the recognition unit for pH measurement and ethidium bromide as reference fluorophore. Further conjugation of subcellular targeting moiety enables the nanoprobes to specifically target lysosome and mitochondria. Confocal fluorescence imaging demonstrated that the nanoprobes could effectively monitor the pH fluctuations from 5.0 to 8.3 in living cells by ratio imaging with 488 nm excitation. Subcellular pH determination and imaging in lysosome and mitochondria could also be achieved in different conditions. The current method can offer a general strategy to determine subcellular analytes and investigate the interactions in biological samples.

Abstract:Cell, enzyme, and tissue activity in living organisms are closely related to intracellular pH. Detecting the changes of intracellular pH is important to understanding the physiological and pathological changes in the process of crucial cell metabolism. A pH probe (HTBI) based on hemicyanine was synthesized. The probe solution displayed a marked colour change from yellow to amaranth with the pH increase from neutral to basic; simultaneously, the emission spectra showed a significant red shift. The probe exhibited a ratiometric fluorescence emission (F586nm /F542nm ) characteristic of pKa 8.82. As expected, HTBI exhibited high sensitivity and selectivity for pH, fine photostability, reversibility, and low cytotoxicity. Therefore, it would be a very useful tool for measuring the intracellular pH changes.

Abstract:Acid-alkaline balance plays a crucial role in all biological processes. Accordingly, monitoring pH changes will help us to understand the functional status of these physiological and pathological processes. Though fluorescent probes may be a useful tool for detecting pH changes, and there are many limitations to currently available probes, such as background interference, potential cytotoxicity, and poor cell permeability, which call for a solution urgently. In this work, a rhodamine-derived colorimetric and ratiometric sensor (Rh-HN) was fabricated for monitoring pH change via the mechanism of fluorescence resonance energy transfer (FRET). Rh-HN has been shown to possess several advantages over other probes, such as high sensitivity, outstanding permeability, and low toxicity. Besides, the fluorescence intensity ratio (F-526/F-592) of Rh-HN displays a pH-sensitive response from 2.0 to 7.5 (pKa = 5.05) and linear response from pH 3.8 to 6.4, which was desirable for mapping pH change in the biological systems. Besides, the results indicated that Rh-HN generated a pH-dependent response regulated by switchable forms between closed and opened spirolactam ring. Overall, Rh-HN has accomplished sensing and mapping of pH in living cells, bacteria, and zebrafish. Those results demonstrated that the great potential of Rh-HN in sensing and visualizing pH in the living biosystem. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:A lysosome-targeting ratiometric fluorescent nanoprobe based on cell-penetrating peptides (CPPs) and quantum dots (QDs) has been developed for monitoring pH fluctuation in living cells. The as-prepared nanoprobe is constructed by Rhodamine B labeled R9RGD CPPs as H+ response unit and the red fluorescent QDs as reference unit to achieve ratiometric pH measurement. With the help of RhB-R9RGD CPPs, the nanoprobe efficiently stains lysosomes and enables discernment of lysosomal pH fluctuation in cells treated with different pH buffers and drug stimulation. The method of using dye labeled CPPs to realize functionalization of nanoparticle in one-step reported herein is expected to obtain wider applications in the detection of subcellular active substances by combining different small molecular probes and functional peptides.

Abstract:In this work, we reported a novel near-infrared (NIR) fluorescent probe RQNN with large Stokes shift (98 nm) for monitoring pH under extremely acidic conditions. For the preparation of this probe, a 1,4-diethylpiperazine moiety was introduced in rhodamine scaffold to tune the electron-donating character, and an o-phenylenediamine was introduced in spironolactone to provide larger steric hindrance. The deprotonated-protonated equilibrium between RQNN, RQNN-H+ and RQNN-H++ were evaluated in different pH by absorption and emission spectra. As expected, RQNN exhibited lower pk(a) values (pk(a)1 = 4.83, pk(a)2 = 2.99), indicating that the probe can be used in extremely acidic pH. Moreover, RQNN possessed highly selective response to H+ over essential metal ions and biologically related redox molecules, high photo-stability, rapid response time, and excellent reversibility. Importantly, the probe had excellent cell membrane permeability and was further applied successfully to monitor pH fluctuations in live cells. (c) 2020 Published by Elsevier B.V.

Abstract:A novel lysosome-targeting ratiometric fluorescent probe (CQ-Lyso) based on the chromenoquinoline chromorphore has been developed for the selective and sensitive detection of intracellular pH in living cells. In acidic media, the protonation of the quinoline ring of CQ-Lyso induces an enhanced intramolecular charge transfer (ICT) process, which results in large red-shifts in both the absorption (104 nm) and emission (53 nm) spectra which forms the basis of a new ratiometric fluorescence pH sensor. This probe efficiently stains lysosomes with high Pearson's colocalization coefficients using LysoTrackerDeep Red (0.97) and LysoTrackerBlue DND-22 (0.95) as references. Importantly, we show that CQ-Lyso quantitatively measures and images lysosomal pH values in a ratiometric manner using single-wavelength excitation.
Fluorescent Probes Based on Π-Conjugation Modulation Between Hemicyanine and Coumarin Moieties for Ratiometric Detection of Ph Changes in Live Cells with Visible and Near-infrared Channels.

Abstract:We report two ratiometric fluorescent probes based on pi-conjugation modulation between coumarin and hemicyanine moieties for sensitive ratiometric detection of pH alterations in live cells by monitoring visible and near-infrared fluorescence changes. In a pi-conjugation modulation strategy, a coumarin dye was conjugated to a near-infrared hemicyanine dye via a vinyl connection while lysosome-targeting morpholine ligand and o-phenylenediamine residue were introduced to the hemicyanine moieties to form closed spirolactam ring structures in probes A and B, respectively. The probes show only visible fluorescence of the coumarin moiety under physiological and basic conditions because the hemicyanine moieties retain their closed spirolactam ring structures. However, decrease of pH to acidic condition causes spirolactam ring opening, and significantly enhances pi-conjugation within the probes, thus generating new near-infrared fluorescence peaks of the hemicyanine at 755 nm and 740 nm for probes A and B, respectively. Moreover, the probes display ratiometric fluorescence response to pH with decreases of the coumarin fluorescence and increases of the hemicyanine fluorescence when pH changes from 7.4 to 2.5. The probes are fully capable of imaging pH changes in live cells with good ratiometric responses in visible and near-infrared channels, and effectively avoid fluorescence blind spots under neutral and basic pH conditions - an issue that typical intensity-based pH fluorescent probes run into. The probe design platform reported herein can be easily applied to prepare a variety of ratiometric fluorescent probes for detection of biological thiols, metal ions, reactive oxygen and nitrogen species by introducing appropriate functional groups to hemicyanine moiety. Abstract:In this work, we presented a naphthalimide-rhodamine based fluorescence resonance energy transfer system (FRET) NR1 as a ratiometric and intracellular pH probe, in which 1,2,3-triazole was identified as an ideal bridge and biocompatibility. It could selectively monitor pH variations in methanol/HEPES solution at room temperature. When the pH changed from 6.20 to 2.00, both the fluorescence intensities at 580 nm and the intensity ratios, R (I-580 nm/I-538 nm) increased, which allowed the detection of pH changes by both normal fluorescence and ratiometric fluorescence methods. The observation is consistent with the increased FRET from the 1,8-naphthalimide (donor) to the ring-opened, colored form of rhodamine (acceptor). Moreover, as NR1 is lysosomal with low cytotoxicity, it will be helpful for investigating the pivotal role of H+ in a biological context, especially in lysosomes through direct intracellular imaging. Abstract:Intracellular pH is a key physiological factor for controlling the activities and functions of cells and lysosome is a vital subcellular organelle. Thus, developing a novel lysosome-targeting fluorescence probe for selective and sensitive detection of lysosomal pH in living cells is very important. In this work, we synthesized a series of fluorescence probes based on imidazole-fused benzothiadiazole. The optical properties of these probes were easily adjusted by modifying the substituents with different electron-withdrawing/donating ability in imidazole moiety. All of them showed acid-response and decreased fluorescence intensity during pH values changing from 4.0 to 8.5. The introduction of morpholine group allowed them to specifically respond to the changes of lysosomal pH. Among them, probe MIBTAA possessed a suitable pK(a) value (5.3) and showed good linear response to pH (R-2 = 0.9918) with red emission when pH changed from 4.4 to 5.6. The probe was successfully applied for monitoring pH variation in living cells induced by proton-pump inhibitor Baf-A1 and chloroquine, indicating its great potential for pH imaging in biological applications.

Abstract:The pH values of lysosomes in cancer cells is slightly lower than that in normal cells, which can be used to distinguish cancer cells from normal cells. According to this, a naphthalimide-rhodamine based fluorescent probe (hereafter referred to as RBN) with a pKa of 4.20 was designed and synthesized for ratiometric sensing of cellular pH via fluorescence resonance energy transfer (FRET), which can respond to different pH precisely through ratiometric fluorescence intensity (I577/I540). RBN can be employed to distinguish cancer cells from normal cells on the basis of different fluorescent response, in particular, RBN showed excellent water solubility and low cell toxicity, all these are quite significant for potential application in cancer diagnose and therapy.

Abstract:Since the intracellular pH plays an important role in the physiological and pathological processes, however, the probes that can be used for monitoring pH fluctuation under extreme acidic conditions are currently rare, so it is necessary to construct fluorescent probes for sensing pH less than 4. In this work, we developed a new near-infrared (NIR) fluorescent probe Cy-SNN for sensing pH fluctuation under extremely acidic conditions. For the preparation of this probe, benzothiozolium moiety was chosen as lysosomal targeting unit and NIR fluorophore, and barbituric acid moiety was fused in the polymethine chain of probe to introduce protonation center. Surprisingly, on the basis of the balance of quaternary ammonium salts and free amines, the pk(a) value of Cy-SNN was calculated as low as 2.96, implying that Cy-SNN can be used in acidic conditions with pH < 4. Moreover, Cy-SNN exhibited highly selective response to H+ over diverse analytes in real-time with dependable reversibility. Importantly, Cy-SNN can be used to specifically target lysosome, providing potential tools for monitoring the function of lysosome in autophagy process.
