Abstract:Intracellular pH (pHi) is an important parameter associated with cellular behaviors and pathological conditions. Sensing pHi and monitoring its changes in live cells are essential but challenging due to the lack of effective probes. We herein report a pH-sensitive fluorogen for pHi sensing and tracking. The dye is a tetraphenylethene-cyanine adduct (TPE-Cy). It is biocompatible and cell-permeable. Upon diffusing into cells, it responds sensitively to pHi in the entire physiological range, visualizing the acidic and basic compartments with intense red and blue emissions, respectively. The ratiometric signal of the red and blue channels can thus serve as an indicator for local proton concentration. The utility of TPE-Cy in pHi imaging and monitoring is demonstrated with the use of confocal microscopy, ratiometric analysis, and flow cytometry.

Abstract:Acid-base balance in cells is vital to maintain the normal biological processes, and its disorders could lead to various diseases. It is significant to develop a reliable and portable method for tracing intracellular pH changes. Herein, a novel colorimetric and fluormetric dual-modal probe 2-(((6,6-dimethylbicyclo [3.1.1]hept-2-en-2-yl)methylene)hydrazono)-3-ethyl-5-(2-hydroxybenzylidene)thiazolidine-4-one (MTHB) was constructed for sensitive and rapid detection of alkaline pH changes by employing myrtenal as a starting material. MTHB showed a significantly enhanced yellow fluorescence upon increasing pH from 7.45 to 12.89, and the pKa was determined as 9.84. The probe MTHB was featured with large Stokes shift, high selectivity, good reversibility, and excellent photostability. The sensing mechanism of MTHB for alkaline pH was verified by HRMS, 1H NMR titration, and theoretical calculations. Additionally, in view of its low toxicity and good biocompatibility, MTHB was successfully utilized to visualize the pH variations in living HeLa cells and zebrafish.

Abstract:In this work, a red-emissive zwitterionic hemicyanine dye, named TPE–Cy, containing tetraphenylethene (TPE) and N-alkylated indolium is designed and synthesized. TPE–Cy inherits the aggregation-induced emission (AIE) feature of TPE and displays a large Stokes shift (>185 nm), overcoming the limitations of the concentration-quenching effect and small Stokes shift (from a few to 20 nm) encountered by conventional cyanine dyes. By taking advantage of the photophysical AIE property and chemical reactivity towards OH−/H+, TPE–Cy is able to sense pH in a broad range (the broadest to date) by showing different emission colors and intensities: strong to moderate red emission at pH 5–7, weak to no emission at pH 7–10, and no emission to strong blue emission at pH 10–14. The acid/base-switched red/blue emission transition is reversible and can be repeated for many cycles. By means of NMR and HRMS analyses, we have drawn a mechanistic picture at molecular level to illustrate how this dye works as a pH-sensitive fluorescent probe.

Abstract:pH plays a crucial role in multiple biological processes, therefore, the development of pH imaging and measurement using the fluorescent probes in biological systems is highly active and has an urgent demand. In this work, a new ratiometric near-infrared (NIR) fluorescent probe DCIQ based on conjugated dicyanoisophorone and quinoline with double vinylene linkers was obtained for pH sensing. The remote conjugated quinoline and middle phenol used as an acceptor for protons may induce dual-modal colorimetric and fluorescence changes as a function of tiny variations in pH in the range of 6–8. Compared to previously reported probes, the proposed DCIQ probe was advantageous in terms of near-infrared emission signal (734 nm), large Stockes shift (173 nm), high selectivity, and good sensitivity. In addition, DCIQ could be as potential probe for viscosity by the fluorescence change. Moreover, the DCIQ -immobilized test papers exhibited reversible colorimetric changes as a function of variations in the environmental acid/base (>4 cycles) through naked-eye colorimetric detection. The desirable pKa (7.24) indicated that DCIQ probe could be used as a molecular tool for readily entering living cells with low cytotoxicity. Importantly, the DCIQ probe was successfully demonstrated for real-time detection of pH fluctuation in HeLa cells through ratiometric fluorescence images.

Abstract:Intracellular pH (pHi) plays an essential role in organelles. Fluorescent probe combined with fluorescence imaging analytical approach has been used for detection pH fluctuation due to high sensitivity and good photostability. Herein, a benzoindole-based colorimetric and naked-eye hemicyanine fluorescent probe 2,3-trimethy-3-[2,4-(dihyoxyl-4-yl)vinyl]-3H-benzo[e]indole (BiDD) was developed in one step. Upon the decreasing of pH, BiDD exhibited strong a pH-dependent characteristic with pKa 4.98 and responded linearly within the pH range of 4.4-6.2. BiDD also showed high sensitivity and selectivity, colorimetric and fluorometric dual-modal response, high photostability, low cytotoxicity as well as good cell membrane permeability. More importantly, the probe was applied to sense and visualize the pH fluctuations in HeLa cells successfully by the fluorescence confocal microscope.

Abstract:It is very important to measure the pH in the cells because abnormal pH in the body can cause many neurological diseases. In this work, a simple emission fluorescent pH probe 2-(benzothiazol-2-yl)-1-((pyren-3-yl) methylene) hydrazine (BMH) was synthesized by connecting pyrene-1-carboxaldehyde and 2-hydrazinobenzothiazole. BMH displayed fluorescence emission characteristics in which the pKa was 3.49 and responded linearly to measure pH range from 4.70 to 3.16. Meanwhile, it possessed high fluorescence quantum yield (64.60%). Moreover, BMH exhibited good stability and highly selectivity which promotes its ability in analytical application and the detection limit of approach was 0.41 μΜ. Simultaneously, it was fully able to image pH fluctuations due to its cell membrane permeability, biocompatibility and no auto-fluorescence effects.

Abstract:A simple pH fluorescent probe based on new fluorophorepyrido[1,2-a]benzimidazole derivative has been synthesized and characterized. This probe responds to acidic pH (pKa =5.18) with high quantum yield (phi)=0.96), fast response (within 1 min), high selectivity and sensitivity. Abstract:Intracellular pH plays a crucial role in many cellular processes, and abnormal intracellular pH has been linked to common diseases such as cancer and Alzheimer's. To address this issue, a water-soluble fluorescent pH probe was designed based on the protonation/deprotonation of the 4-methylpiperazin-1-yl group, using dicyanoisophorone as the fluorophore. In the neutral form of the probe, fluorescence is quenched due to charge transfer from the 4-methylpiperazin-1-yl group to the fluorophore upon excitation. Under acidic conditions, protonation of the 4-methylpiperazin-1-yl group inhibits the photoinduced electron transfer process, leading to an increase in fluorescence intensity. Density-functional theory calculations also verified the fluorescence OFF-ON mechanism. The probe exhibits high selectivity, photostability, fast response to pH changes, and low cytotoxicity to cells. Additionally, the probe selectively accumulates in lysosomes, with a high Pearson coefficient (0.95) using LysoTracker Green DND-26 as a reference. Notably, the probe can monitor lysosomal pH changes in living cells and track pH changes stimulated by chloroquine. We anticipate that the probe has potential for diagnosing pH-related diseases.

Abstract:In this study a new ratiometric pH probe, PHE-C, was designed and synthesized with the aim of enlarging the wavelength difference (Δλfl) between the two peaks of the typical hemicyanine fluorophore without significantly affecting its pKa value. The two emission maxima at 682 and 730 nm, associated with the respective protonation states of the hydroxyl group in PHE-C, have a larger Δλfl (48 nm) than that of unsubstituted hemicyanine. Importantly, the pKa of PHE-C (6.55) is the same as that of the unsubstituted counterpart. Consequently, the ratio of the intensities of the 682 and 730 nm emission bands is linearly related to pH in the range of 5.5–7.5, which is appropriate for monitoring small changes in pH of weakly acidic physiological environments. Moreover, PHE-C possesses good photostability, sensitivity and reversibility, and it displays the same response as pH meter when determining the pH in bovine serum solutions, indicating that it can be utilized in complex biological environments. Finally, the results of studies aimed at assessing the applicability to monitoring pH changes in living cells and mice demonstrate that the new probe is a highly sensitive ratiometric probe for in vivo pH determinations.

Abstract:Acidic pH is an important physiological parameter in environment and biological systems. Thus, designing a probe (Hcy-F) with good characters to monitor acidic pH is really important. Herein, a small molecular water-soluble probe is designed based on indole with high selectivity and sensitivity for detection the acidic pH in aqueous solution and living cells. When pH variation of aqueous is detected by probe, an obvious ratiometric and good linear response behavior are found in UV–vis absorption spectrum (R2 = 0.9973, A434 nm/A371 nm = 6.9929–4.2231 pH) and fluorescence emission spectrum (R2 = 0.9987, I571 nm/I514 nm = 7.2935–2.981 pH), which further proved the feasibility of Hcy-F to monitor the change of acidic pH in aqueous, respectively. Furthermore, Hcy-F is successfully applied for monitoring pH variation in living cells in dual channels (green and red) with higher cell survival rate, indicating the probe has great potential for pH imaging in biological applications.

Abstract:pH regulates the function of many organelles and plays a pivotal role in requiring multitud cellular behaviors. Compared with single fluorescent probes, ratio fluorescent probes have higher sensitivity and immunity to interference. Herein, a novel Janus ratio nanoprobe was developed for intracellular pH detection. Modified rhodamine B probe and fluorescein isothiocyanate (FITC) were individually encapsulated in the independent hemispheres of Janus microparticles fabricated via Pickering emulsion. Moreover, it exhibits a satasified ratiometric detection of pH compared to the previous core-shell structure and organic small molecule probe. Accordingly, the Janus nanoprobe possesses many important features as an attractive sensor, including high antijamming capability, excellent stability, good reversibility and low cytotoxicity. Variations of the two fluorescence intensities (Fgreen/Fred) resulted in a ratiometric pH fluorescent sensor, which can respond to wide range of pH values from 3 to 8. To be more specific, with a single excitation wavelength of 488 nm, there are dual emission bands centered at 538 nm and 590 nm. Also the Janus nanoprobe displays a excellent linear relationship in the physiologically relevant pH range of 4.0-6.0. Consequently, detecting of pH and imaging was successfully achieved in living cells, which provides a simple and reliable method for detecting intracelluar pH and other similar substances.

Abstract:Abnormal pH values in the organelles are closely associated with inappropriate cellular functions and many diseases. Monitoring subcellular pH values and their variations is significant in biological processes occurring in living cells and tissues. Herein, we develop a series of ratiometric fluorescence nanoprobes for quantification and imaging of pH values with a single-wavelength excitation in cytoplasm, lysosomes, and mitochondria. The nanoprobes consist of mesoporous silica nanoparticles assembled with aminofluorescein as the recognition unit for pH measurement and ethidium bromide as reference fluorophore. Further conjugation of subcellular targeting moiety enables the nanoprobes to specifically target lysosome and mitochondria. Confocal fluorescence imaging demonstrated that the nanoprobes could effectively monitor the pH fluctuations from 5.0 to 8.3 in living cells by ratio imaging with 488 nm excitation. Subcellular pH determination and imaging in lysosome and mitochondria could also be achieved in different conditions. The current method can offer a general strategy to determine subcellular analytes and investigate the interactions in biological samples.

Abstract:A fluorescent pH probe, N,N'-bi( l-phenylalanine amine)-perylene-3,4;9,10-dicarboxylic diimide (PDCDA) was synthesized and used for pH sensing in living cells. A significant fluorescence intensity change was observed over a pH range from 7.0 to 4.0. Electrostatic potential maps (MEP) suggested that the electronic repulsion between PDCDAs was increased by the high negative electrostatic potential which resulted in a high water solubility of PDCDA. PDCDA was successfully applied as a high-performance fluorochrome for living HeLa cell imaging. The results demonstrate that the probe PDCDA is a good candidate for monitoring pH fluctuations in living cells with good water solubility, low cytotoxicity, high fluorescence quantum yield and photostability.

Abstract:Lysosome as the typical acidic organelle involved in many biological processes, the dysfunction of its pH would alter many diseases. Small-molecule fluorescent probe has been considered as vital tool for monitoring pH fluctuation in living cells. Herein, a hemicyanine based ratiometric fluorescent probe was synthesized, namely, 2,3-trimethy 3-[2-(dimethyl-amino-4-yl) vinyl]-3H-benzo[e]inclole (BiDL), for rapidly detection pH under acidic conditions. BiDL exhibited ratio fluorescence emission (F-534nm/F-622nm) Characteristic with pK(a) 4.25. BiDL showed good linearly response in pH 3.4-4.32, indicating that the probe can be used for quantitative detection pH. The probe also displayed large Stokes shift (112 nm/201 nm) under neutral and acidic conditions, respectively, which could effectively reduce the excitation interference. BiDL had excellent cell membrane permeability, good photo-stability and low toxicity in living cells. The dual-channel confocal fluorescent microscopic ratiometric imaging application of pH in HeLa cells and lysosome were achieved successfully, indicating that BiDL has good potential for investigating lysosome-relevant pathological and physiological processes. Abstract:A new ratiometric fluorescent probe based on cell-penetrating peptides (CPPs) was constructed for whole-cell pH mapping and simultaneous measurement of pH changes in the cytoplasm and lysosomes. The arginine-rich CPP, R12K worked as linker, carrier and part of the fluorophore. Benefiting from R12K, the fluorescent probe is completely water soluble, membrane permeable and well biocompatible. It shows high selectivity, sensitivity and reversibility to pH fluctuations. The ratio of fluorescence intensities F519/F582 increased from 0.2 to 9.2 over the pH range from 3.3 to 8.1. Intracellular pH mapping was successfully realized owing to the wide distribution of the probe in live cells (even in nucleus). Moreover, cytosolic and lysosomal pH change caused by the stimuli can be simultaneously detected. Compared to other ratiometric pH probes, RhB-R12K-FITC can provide more precise information about H+ redistribution between different cellular compartments.

Abstract:pH plays an essential role in virtually all biological processes, which thus needs to be tightly regulated. Especially small changes in pH value in biological systems will affect the normal metabolic functions of animals and plants. Therefore, it is very important to accurately track changes in pH in biological systems. Herein, we rationally fabricated a new molecular pH probe (NBD-pbz) based on conjugation of the 2-Piperazin-1-yl-1,3-benzothiazole (pbz) and 7-nitro-1,2,3-benzoxadiazole (NBD) building blocks. The following test on NBD-pbz revealed that it can offer a sensitive and selective tracking of pH changes from 3.2 to 7.6 with pKa 5.51, and not only in eukaryotic cells (especially the imaging of ocular tumor cell OCM-1) and zebrafish but also in mung bean sprouts via fluorometric turn on response and an intramolecular charge transfer (ICT) based working mechanism strategy. Therefore, this NBD-pbz probe not only replenishes the current repertory of molecular pH probes but also extends the application of pH probes to monitor pH variation in the plant kingdom, which will undoubtedly arouse greater research interest in the field.

Abstract:A novel FRET-ICT-based ratiometric fluorescent and colorimetric pH probe (BDMNP) was developed. BDMNP exhibited 96% energy transfer efficiency and good linear response in the pH ranges of 5.4–6.8 (pKa = 6.10) with high selectivity and sensitivity. BDMNP has been successfully applied to monitor lysosomal pH changes in living cells. Meanwhile, BDMNP can also be used to detect the subtle pH changes of a variety of environmental water samples.

Abstract:A novel pH-sensitive fluorescent probe has been designed and synthesized for sensing intracellular pH. This probe showed excellent water solubility, it was sensitive toward the pH range from 4 to 12, and it was especially sensitive in alkaline environments. During the pH changes from acidic to alkaline environments, the color of the solution turned from yellow to purple, thus the difference in color can be used to distinguish between acidic and alkaline systems. The other major features of probe pH-DCN including high selectivity, low toxicity, good reversibility and stability allowed pH-DCN to visualize fluctuations of the pH in live cells. Moreover, probe pH-DCN has successfully discriminated cancer cells from normal cells by monitoring their different intracellular pH levels.

This sensing behavior is reversible and not interfered by normal intracellular metal cations such as Na+, K+, Ca2+, Mg2+ and other transition metal cations. Comparison between the pH sensing behaviors of NBD-Py and its analogue without the N-(4-pyridyl) methyl group disclosed that the pyridylmethyl group is essential its pH sensing ability for weak acidic and near neutral pH, which is important for cell biology. Confocal imaging in HeLa cells confirmed the pH, imaging ability and the lysosome-targetability of NBD-Py, which is valuable for lysosome cell biology.
