Abstract:The ability to recognize and quantify the chirality of alpha-amino acids constitutes the basis of many critical areas for specific targeting in drug development and metabolite probing. It is still challenging to conveniently distinguish the enantiomer of amino acids largely due to the lack of a universal and simple strategy. In this work, we report a strategy for the visual recognition of α-amino acids. It is based on the chirality of L-tartaric acid-capped gold nanoparticles (L-TA-capped AuNPs, ca. 13 nm in diameter). All of 19 right-handed α-amino acids can induce a red-to-blue color change of L-TA-capped AuNP solution, whereas all of the left-handed amino acids (except cysteine) cannot. The chiral recognition can be achieved by the naked eye and a simple spectrophotometer. This method does not require complicated chiral modification, and excels through its low-cost, good availability of materials and its simplicity. Another notable feature of this method is its high generality, and this method can discriminate almost all native α-amino acid enantiomers. This versatile method could be potentially used for high-throughput chiral recognition of amino acids.

Abstract:In this work, we prepared gold nanoparticles (AuNPs) by employing gluconic acid (GlcA) as reducing-cum-stabilizing agent. The proposed GlcA-AuNPs successfully worked as a colorimetric sensor for visual chiral recognition of aromatic amino acid enantiomers, namely tyrosine (d/l-Tyr), phenylalanine (d/l-Phe), and tryptophan (d/l-Trp). After adding L-types to GlcA-AuNPs solution, the color of the mixture changed from red to purple (or gray), while no obvious color change occurred on the addition of D-types. The effect can be detected by naked eyes. The particles have been characterized by transmission electron microscopy, Fourier-transform infrared spectroscopy, zeta potential, the dynamic light scattering analysis as well as UV–Vis spectroscopy. This assay can be used to determine the enantiomeric excess of l-Trp in the range from 0 to + 100%. The method has advantages in simplicity, sensitivity, fast response, and low cost.

Abstract:Chiral recognition of enantiomers is fundamentally important. In this work, an effective and simple chiral sensing strategy for discrimination of leucine (Leu) enantiomers was constructed. In this chiral sensing, cube-shaped gold nanoparticles (AuNCs) as colorimetric probes are used to recognize Leu enantiomers. L-Leu can induce rapidly the aggregation of AuNCs, leading to change of AuNCs solution from red to colorless, while D-Leu cannot induce the color change. This distinct color changes allow naked-eye to distinguish chiral isomers of Leu. The chiral sensing was applied to measure the enantiometric excess of L-Leu in the whole range (from −100% to 100%). This chiral analysis can be performed by naked eye or simple ultraviolet-visible spectrometer. In addition, the mechanism of chiral recognition has been studied with circular dichroism (CD) spectra, UV–vis absorption spectra and zeta potential. In this method, the chiral recognition is due to the intrinsic chirality of AuNCs, and the AuNCs don't need any chiral labeling or modification. This chiral sensing method is simple, cheap, rapid and easy to operate. Furthermore, this work provides one experimental evidence for intrinsic chirality of nanoparticles, and helps people understand the chirality of nanostructures.

Abstract:Since chiral carboxylic acids (CCAs) are a class of important biological molecules and common functional moieties found in pharmaceutical molecules, the chiral analysis of CCAs has received much attention. Herein, we developed a simple, rapid and cost-effective method for visual and colorimetric high-throughput analysis of CCAs using chiral di-imine structures (CDIS) modified gold nanoparticles (C-AuNPs) as the probe. The C-AuNPs are positively charged in the presence of zinc ion, and they can be enantioselectively shielded by the negative charged CCA enantiomers. Therefore, upon the addition of different concentration and enantiomeric excess (ee) of CCAs, the C-AuNPs-based sensor shows the different levels of aggregation along with the visual changes in solution color, which can achieve simultaneous analysis of the concentration and ee of CCAs. The chiral recognition mechanism based on C-AuNPs was investigated by the determination of binding constants (K) and molecular simulation methods. Our approach is expected to have the wide-ranging applications in the developing region for enantio-sensing of various chiral drugs and biomolecules.

Abstract:AbstractChirality plays a key role in modern science and technology. Here, we report a simple and effective sensing platform for visual chiral recognition of enantiomers. In this sensing platform, gold nanorods (AuNRs) prepared through a common synthesis route are used as colorimetric probes for visual recognition of glutamine (Gln) enantiomers. D-Gln could rapidly induce the aggregation of AuNRs, thereby resulting in appreciable blue-to-gray color change of AuNRs solution; however, L-Gln could not induce color change of AuNRs. This distinct color change can be easily distinguished by the naked eyes; as a result, a visual method of chiral recognition was suggested. The method was applied to determine the enantiometric excess of D-Gln through the whole range of −100% ~ 100%. The chiral assay can be performed with a simple UV-vis spectrometer or the naked eyes. Notably, the AuNRs do not need any chiral labeling or modification, and the chiral recognition is based on the inherent chirality of AuNRs. This chiral assay method is simple, sensitive, cheap and easy to operate. This study is the first example using AuNRs for direct visual recognition of enantiomers, and will open new opportunity to construct more chiral recognition methods for some important compounds.

Abstract:A new colorimetric analysis approach for chiral recognition of d- and l-forms of phenylalanine (phe) was developed based on triangular silver nanoplates (TAg-NPs). The TAg-NPs could be used as chiral colorimetric probes for d- and l-forms of phe. Upon addition of d-phe to TAg-NPs solution, a color change from blue to purple to pink could be observed, while no obvious color change was found on addition of l-phe. l-phe could prevent the TAg-NPs from being etched to small size particles while the protective effect of d-phe was weak. Moreover, the enantiomeric excess of d-phe could be determined using the proposed chiral assay in the percentage of l-phe from 0 to 100% with a correlation coefficient of 0.9855. The phenomenon could be monitored by bare eyes and quantified analysis by UV–Vis spectrophotometry. The developed approach had several advantages, such as simplicity, visualization, short analysis time and low cost. This study presented a fast visualization analysis method of chiral d/l-phenylalanine and may lay the foundation for the development of visualization chiral recognition of other target analytes.

Abstract:Recently, chiral ionic liquids have attracted increasing attention in analytical chemistry. However, only a few papers focus on the application of them in visual chiral recognition. Herein, two functionalized chiral ionic liquids derived from (S)-mandelic acid (1-butyl-3-methylimidazolium mandelate, CIL1 and N-butyl-N-methyl-pyrrolidinium mandelate, CIL2) were prepared for visual chiral recognition of aromatic amino acids for the first time. In the presence of Cu(II) and appropriate solvents, visual enantiomeric responses of phenylalanine, tryptophane, tyrosine and phenylglycine were observed. Relying on solubility or color differences, all chiral recognition could be finished within 5 min. The potential mechanism was investigated by means of infrared spectroscopy, ultraviolet spectroscopy, thermal gravity analysis, elemental analysis and scanning electron microscope. Results revealed that CuSO 4 interacted with CIL1 and D-tryptophane in the ratio of 1:1.96:0.43 in relevant precipitate, and the different stability of complex was responsible for the chiral recognition. In addition, resolution of racemic tryptophane was performed, which offered excellent enantiomeric excess values (94.2% for CIL1 and 95.1% for CIL2 in solid phase). The proposed ionic liquids had strong enantioselectivity for aromatic amino acids and great potential in visual chiral recognition.

Abstract:We report on a method for visual differentiation between the D- and L-forms of tryptophan (Trp). It is based on the inherent chirality of citrate-capped silver nanoparticles (AgNPs; ca. 15 nm in diameter) that can be used as chiral selector for D- and L-forms of Trp. On addition of D-Trp to a solution of the AgNPs, a color change from yellow to red can be seen, while no color change is found on addition of L-Trp. The chiral assay can be used to determine the enantiometric excess of D-Trp in the range from −50 % to 100 %. The effect can be detected with bare eyes and quantified by spectrophotometry. Notably, this method does not require any labeling or chiral modification. The method excels by its sensitivity, low cost, good availability of materials, and its simplicity.

Abstract:Recognizing and quantifying enantiomers of chiral molecule is of great importance in chemical, biological and pharmaceutical fields. Herein, we presented one simple-yet-efficient method of sensing tyrosine (Tyr) enantiomers. In this sensing, silver triangular nanoplates (AgTNPs) were used as colorimetric probes. L -Tyr quickly induced the color of AgTNPs solution to change from dark blue to light gray, whereas D -Tyr induced no change of the AgTNPs solution color. The obvious color change enables the naked eye to recognize Tyr enantiomer. The visual method was used to detect the enantiometric excess value of L -Tyr in the whole range (-100 % similar to 100 %). This chiral sensing can be finished within 5 min using one simple ultraviolet-visible spectrometer or naked eye. Furthermore, the mechanism of this chiral sensing was explored. It was confirmed that this chiral sensing was based on AgTNPs' intrinsic chirality. This chiral sensing is rapid, simple, and low-cost, and has great potential for chiral determination of Tyr.

Abstract:In this work, we proposed one simple and effective chiral-sensing method for the discrimination of carnitine enantiomers. The gold nanorods (AuNRs) were used as colorimetric probes to recognize carnitine enantiomers in this method. l-Carnitine can induce rapidly the blue-to-gray change of AuNR solution color, whereas d-carnitine cannot induce the obvious change of the color. Based on this distinct difference of color, a visual method for recognition of carnitine enantiomers was suggested. This method has been applied to detect the enantiometric excess of l-carnitine in the range of − 100~100%. The presented chiral measurement can be performed with a simple ultraviolet-visible spectrometer and even the naked eyes. In this method, AuNRs need no modification with chiral molecule, and the chiral recognition is based on the inherent chirality of AuNRs. This new method opens up possibility for fast and easy assay of carnitine enantiomers.

Abstract:Chiral recognition is among the important and special modes of molecular recognition. It is highly desirable to develop a simple, rapid, sensitive, and high-throughput routine assay for chiral recognition. In this study, we demonstrate that nucleotide-capped Ag nanoparticles (AgNPs) can be used as an ultrahigh efficiency enantioseparation and detection platform for D-and L-cysteine. The aggregation of AgNPs is selectively induced by an enantiomer of cysteine, which allowed the rapid colorimetric enantiodiscrimination of cysteine without any prior derivatization and specific instruments and left an excess of the other enantiomer in the solution, thus resulting in enantioseparation. This is the first application of a nucleotide-capped AgNP-based biosensing platform for chiral recognition and opens new opportunities for design of more novel enantiosensing strategies and enantiospecific adsorbents and expansion of its application in different fields.

Abstract:Enantioselective identification of chiral molecules holds immense significance in the fields of biological processes, chemical reactions, and drug development due to the optical rotation properties of enantiomers. Moreover, the chiral nanocomposite plays an important role in capable of selectively interacting with specific chiral molecules. By utilizing the chiral surfaces of metal polyphenol framework, an enantioselective identification chiral nanocomposite is fabricated on polyphenol colloidal spheres (PCS). The multivariate metal-based nanocomposite use (L)-tartaric acid (TA) and copper as a chiral recognition selector and signal corrector, aminothiophenol (PATP) anchoring Ag NPs as artificial traction skeleton. Applying (L)-/ (D)-cysteine as patterns enantiomers, the chiral recognition selector Cu- (L)-TA framework perform stereoselective recognition of target molecules through strong affinity and hydrogen interaction. The discriminative SERS signal and CT-related totally symmetric (a1) value of Cu- PCS@Ag NPs and PATP molecules is regarded as discriminating part and internal standard to correct the signal drifting. The put forward method has a good linearity in the concentration range of 1 nM similar to 10 mM and detection limits as low as 0.37 pM and 0.30 pM for L/D-Cys. This work provides specific method for the chiral molecule recognition and determination in complex matrix sample.

Abstract:Chirality sensing is a very challenging task. Here, we report a method for ultrasensitive detection of chiral molecule L/D-carnitine based on changes in the recognition tunneling current across self-assembled core satellite gold nanoparticle (GNP) networks. The recognition tunneling technique has been demonstrated to work at the single molecule level where the binding between the reader molecules and the analytes in a nanojunction. This process was observed to generate a unique and sensitive change in tunneling current, which can be used to identify the analytes of interest. The molecular recognition mechanism between amino acid L-cysteine and L/D-carnitine has been studied with the aid of SERS. The different binding strength between homo- or heterochiral pairs can be effectively probed by the copper ion replacement fracture. The device resistance was measured before and after the sequential exposures to L/D-carnitine and copper ions. The normalized resistance change was found to be extremely sensitive to the chirality of carnitine molecule. The results suggested that a GNP networks device optimized for recognition tunneling was successfully built and that such a device can be used for ultrasensitive detection of chiral molecules.

Abstract:Efficient enantiomer discrimination by a convenient method remains a challenge in biochemical and pharmaceutical fields. Herein we have modified silver nanoparticles (AgNPs) with beta-cyclodextrin (beta-CD) to obtain a probe for the chiral recognition of aromatic alpha-amino acids. This assay relies on the distance-dependent optical properties of AgNPs and the difference in inclusion binding strength between the aromatic amino acid enantiomers and the beta-CD host. The response of beta-CD functionalized-AgNPs to certain chiral aromatic amino acid enantiomers namely D- and L-phenylalanine (Phe), D- and L-tryptophan (Trp), and D- and L-tyrosine (Tyr) in phosphate buffer solutions was studied in detail. In the presence of D- Phe (or L-Trp or L-Tyr), an appreciable yellow-to-red color change of the AgNP solution can be observed, whereas no color change is found in the presence of L-Phe (or D-Trp or D-Tyr). This method can be used to quantitatively determine the percentage of D- Phe in a mixture of D- and L-Phe. The chiral assay described in this work is easily readout with the naked eye or using a cheap UV-vis spectrometer. This method excels by its simplicity, low cost, and good availability of materials.

Abstract:A facile one-pot synthetic method for preparing gold nanoparticles by employing sugammadex (SUG), a carboxylic acid functionalized gamma-cyclodextrin derivative, as reducing-cum-stabilizing agent herein was reported for the first time. The SUG protected gold nanoparticles (SUG-AuNPs) can work as a colorimetric sensor for visual chiral recognition of a-amino acids enantiomers, especially for lysine (Lys) and asparagine (Asn) enantiomers. The chiral recognition assay was successfully applied to determining the enantiometric excess of L-Lys and L-Asn ranging from -100 % to 100 % respectively. Moreover, the prepared SUG-AuNPs was found to exhibit efficient catalytic activity towards the reduction of toxic 4-nitrophenol by NaBH4 and the efficiency of the system was further demonstrated through the reduction of other typical nitroaromatics under mild condition. The as-synthesized SUG-AuNPs shows good performance for both chiral sensing and reduction activity and thus may facilitate the practical application in the area of both chiral discrimination and catalysis.
