Abstract:In this work, we prepared gold nanoparticles (AuNPs) by employing gluconic acid (GlcA) as reducing-cum-stabilizing agent. The proposed GlcA-AuNPs successfully worked as a colorimetric sensor for visual chiral recognition of aromatic amino acid enantiomers, namely tyrosine (d/l-Tyr), phenylalanine (d/l-Phe), and tryptophan (d/l-Trp). After adding L-types to GlcA-AuNPs solution, the color of the mixture changed from red to purple (or gray), while no obvious color change occurred on the addition of D-types. The effect can be detected by naked eyes. The particles have been characterized by transmission electron microscopy, Fourier-transform infrared spectroscopy, zeta potential, the dynamic light scattering analysis as well as UV–Vis spectroscopy. This assay can be used to determine the enantiomeric excess of l-Trp in the range from 0 to + 100%. The method has advantages in simplicity, sensitivity, fast response, and low cost.

Abstract:AbstractChirality plays a key role in modern science and technology. Here, we report a simple and effective sensing platform for visual chiral recognition of enantiomers. In this sensing platform, gold nanorods (AuNRs) prepared through a common synthesis route are used as colorimetric probes for visual recognition of glutamine (Gln) enantiomers. D-Gln could rapidly induce the aggregation of AuNRs, thereby resulting in appreciable blue-to-gray color change of AuNRs solution; however, L-Gln could not induce color change of AuNRs. This distinct color change can be easily distinguished by the naked eyes; as a result, a visual method of chiral recognition was suggested. The method was applied to determine the enantiometric excess of D-Gln through the whole range of −100% ~ 100%. The chiral assay can be performed with a simple UV-vis spectrometer or the naked eyes. Notably, the AuNRs do not need any chiral labeling or modification, and the chiral recognition is based on the inherent chirality of AuNRs. This chiral assay method is simple, sensitive, cheap and easy to operate. This study is the first example using AuNRs for direct visual recognition of enantiomers, and will open new opportunity to construct more chiral recognition methods for some important compounds.

Abstract:Chiral recognition of enantiomers is fundamentally important. In this work, an effective and simple chiral sensing strategy for discrimination of leucine (Leu) enantiomers was constructed. In this chiral sensing, cube-shaped gold nanoparticles (AuNCs) as colorimetric probes are used to recognize Leu enantiomers. L-Leu can induce rapidly the aggregation of AuNCs, leading to change of AuNCs solution from red to colorless, while D-Leu cannot induce the color change. This distinct color changes allow naked-eye to distinguish chiral isomers of Leu. The chiral sensing was applied to measure the enantiometric excess of L-Leu in the whole range (from −100% to 100%). This chiral analysis can be performed by naked eye or simple ultraviolet-visible spectrometer. In addition, the mechanism of chiral recognition has been studied with circular dichroism (CD) spectra, UV–vis absorption spectra and zeta potential. In this method, the chiral recognition is due to the intrinsic chirality of AuNCs, and the AuNCs don't need any chiral labeling or modification. This chiral sensing method is simple, cheap, rapid and easy to operate. Furthermore, this work provides one experimental evidence for intrinsic chirality of nanoparticles, and helps people understand the chirality of nanostructures.
A Universal Strategy for Visual Chiral Recognition of Α-Amino Acids with L-Tartaric Acid-Capped Gold Nanoparticles As Colorimetric Probes

Abstract:The ability to recognize and quantify the chirality of alpha-amino acids constitutes the basis of many critical areas for specific targeting in drug development and metabolite probing. It is still challenging to conveniently distinguish the enantiomer of amino acids largely due to the lack of a universal and simple strategy. In this work, we report a strategy for the visual recognition of α-amino acids. It is based on the chirality of L-tartaric acid-capped gold nanoparticles (L-TA-capped AuNPs, ca. 13 nm in diameter). All of 19 right-handed α-amino acids can induce a red-to-blue color change of L-TA-capped AuNP solution, whereas all of the left-handed amino acids (except cysteine) cannot. The chiral recognition can be achieved by the naked eye and a simple spectrophotometer. This method does not require complicated chiral modification, and excels through its low-cost, good availability of materials and its simplicity. Another notable feature of this method is its high generality, and this method can discriminate almost all native α-amino acid enantiomers. This versatile method could be potentially used for high-throughput chiral recognition of amino acids.

Abstract:We report on a method for visual differentiation between the D- and L-forms of tryptophan (Trp). It is based on the inherent chirality of citrate-capped silver nanoparticles (AgNPs; ca. 15 nm in diameter) that can be used as chiral selector for D- and L-forms of Trp. On addition of D-Trp to a solution of the AgNPs, a color change from yellow to red can be seen, while no color change is found on addition of L-Trp. The chiral assay can be used to determine the enantiometric excess of D-Trp in the range from −50 % to 100 %. The effect can be detected with bare eyes and quantified by spectrophotometry. Notably, this method does not require any labeling or chiral modification. The method excels by its sensitivity, low cost, good availability of materials, and its simplicity.

Abstract:The nanohybrids which based on beta-cyclodextrin, platinum nanoparticles and graphene (beta-CD-PtNPs/GNs) were successfully synthesized and characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), fourier transform infrared spectroscopy (FT-IR) and electrochemical impedance spectroscopy (EIS). Then they were used to construct a simple and reliable chiral sensing platform to interact with tryptophan (Trp) enantiomers. Differential pulse voltammetry (DPV) was used to investigate the stereo selectivity of beta-CD-PtNPs/GNs to Trp enantiomers. After interaction, the obvious difference of peak currents of L-Trp and D-Trp was obtained, indicating this strategy could be employed to chiral recognition of Trp enantiomers. Under the optimum conditions, the chiral sensor exhibited a good linear response to Trp enantiomers in a linear range of 5.0x10(-5) to 5.0x10(-3) M with a low limit of detection of 1.7x10(-5) M (S/N=3). This approach provided a new available platform to recognize and determine Trp enantiomers.

Abstract:"The development and deployment of a practical and portable technology for on-site chiral identification of enantiomers hold immense significance in the fields of medical and biological sciences. Among the essential amino acids, Tryptophan (Trp) plays a crucial role in human metabolism and serves as a diagnostic marker for various metabolic disorders. In this study, we introduce an innovative approach that combines an enantio-selective ZIF-8-His MOF-MIPs packed-bed centrifugal microfluidic system with an enantioselective colorimetric sensor probe. This system is further integrated with smartphone-based on-site data recording. The basis of this colorimetric sensor's operation lies in the controlled morphology and surface passivation of gold nano-ovals (Au-NOs) through DL-Alanine. To confirm the successful synthesis of the chiral recognition elements, we employed various characterization techniques, including FE-SEM, TEM, FTIR, CD, UV-Vis, zeta potential, DLS, and XRD. Our focus was on optimizing operational parameters for the effective separation and determination of L-chiral tryptophan on-site. The sensor exhibited two linear ranges for L-Trp detection: 0-5.42 and 5.42-80.47 mM, with a detection limit of 0.5 mM. The integrated system possesses advantages such as ease of availability, preparation, high stability, desirable selectivity even in the presence of similar biomolecules, and rapid detection capabilities. Furthermore, our method demonstrated successful enantioselective sensing of L-Trp in various biological samples, including human blood plasma, urine, milk, and bovine serum albumin (BSA), yielding promising results. The integrated microfluidic platform follows a "sample-in and answer-out" approach, making it highly applicable in healthcare, environmental monitoring, food safety analysis, and point-of-care testing. The chiral recognition pretreatment assay and self-contained, automated colorimetric detection on the microfluidic disc represent a promising avenue for cutting-edge research in these domains".

Abstract:Herein, we introduce a novel integrated system that merges an enantio-discriminative bio-MOF-packed centrifugal microfluidic chip made from PDMS with a user-friendly on-site colorimetric sensor. This innovative approach enables the precise enantioselective recognition of L-tryptophane (L-Trp). This chiral recognition probe was successfully synthesized through meticulous control of nano-ovals-shaped gold nanoparticles morphology and surface passivation. The operational factor of this methodology was optimized to ensure simplicity, practicality, and efficiency. This optimization led to reduced reagent consumption and instantaneous analytical feedback. The integrated system was effectively applied for enantioselective separation and quantification of L-Trp across an extensive linear range of 50 μM-1.5 mM, impressive limit of detection as low as 15 μM. It is noteworthy that this integrated system demonstrated desirable selectivity even in the presence of similar biomolecules, showcasing its robust performance and rapid detection capability. Further extended the application of this strategy to exceptional performance across enantioselective sensing of L-Trp in various sample matrices, comprising bovine serum albumin, bovine milk, blood plasma and urine samples. This integrated microfluidic sample pretreatment, chiroptical sensing, and on-site signal recording with a smartphone hold tremendous potential for widespread implementation, practical applications engaging healthcare and environmental, food safety, and point-of-needs analysis, facilitating successive solution mixing and colorimetric detection.

Abstract:Chiral recognition is among the important and special modes of molecular recognition. It is highly desirable to develop a simple, rapid, sensitive, and high-throughput routine assay for chiral recognition. In this study, we demonstrate that nucleotide-capped Ag nanoparticles (AgNPs) can be used as an ultrahigh efficiency enantioseparation and detection platform for D-and L-cysteine. The aggregation of AgNPs is selectively induced by an enantiomer of cysteine, which allowed the rapid colorimetric enantiodiscrimination of cysteine without any prior derivatization and specific instruments and left an excess of the other enantiomer in the solution, thus resulting in enantioseparation. This is the first application of a nucleotide-capped AgNP-based biosensing platform for chiral recognition and opens new opportunities for design of more novel enantiosensing strategies and enantiospecific adsorbents and expansion of its application in different fields.

Abstract:Efficient enantiomer discrimination by a convenient method remains a challenge in biochemical and pharmaceutical fields. Herein we have modified silver nanoparticles (AgNPs) with beta-cyclodextrin (beta-CD) to obtain a probe for the chiral recognition of aromatic alpha-amino acids. This assay relies on the distance-dependent optical properties of AgNPs and the difference in inclusion binding strength between the aromatic amino acid enantiomers and the beta-CD host. The response of beta-CD functionalized-AgNPs to certain chiral aromatic amino acid enantiomers namely D- and L-phenylalanine (Phe), D- and L-tryptophan (Trp), and D- and L-tyrosine (Tyr) in phosphate buffer solutions was studied in detail. In the presence of D- Phe (or L-Trp or L-Tyr), an appreciable yellow-to-red color change of the AgNP solution can be observed, whereas no color change is found in the presence of L-Phe (or D-Trp or D-Tyr). This method can be used to quantitatively determine the percentage of D- Phe in a mixture of D- and L-Phe. The chiral assay described in this work is easily readout with the naked eye or using a cheap UV-vis spectrometer. This method excels by its simplicity, low cost, and good availability of materials.
Common materials, extraordinary behavior: An ultrasensitive and enantioselective strategy for D-Tryptophan recognition based on electrochemical Au@p-L-cysteine chiral interface

Abstract:The poly-L-cysteine modified Au nanoparticles (Au@p-L-Cys) were constructed on electrode surface as a highly efficient chiral interface for tryptophan (Trp) enantiomers recognition via one step electropolymerization. With the aid of Cu2+, L-Cys residues and D-Trp target formed a sandwich complex D-Trp-Cu2+-L-Cys, while L-Trp was unable to form such complex due to the steric hindrance provided by the chiral interface, which was confirmed by the electrochemical and SEM results. With the introduction of ferricyanide probe, D-Trp produced significant current decrease while L-Trp produced a slight current increase, which implied the successful enantioselective recognition of Trp enantiomers (specifically D-Trp) in the true sense. This novel sensor showed a surprisingly wide linear range toward D-Trp of 6 x 10- 7 M to 1 x 10-2 M, with a detection limit as low as 75 nM (S/N = 3). Moreover, the exclusive enantioselectivity toward D-Trp was discovered since other amino acids showed negligible interference to detection of D-Trp. The recovery of D-Trp in human serum was between 91.30 and 109.3%, which further verified the satisfying specificity and practicality of the proposed strategy. The coordination thermodynamics by UV-Vis spectroscopy and DFT simulation were also used to investigate the enantioselective mechanism. These results highlight the great potential of using Au@p-L-Cys to construct chiral interface for enantiomers recognition and hold the promise of practical application of electrochemical chiral sensors in fields like pharmaceutics and bioanalysis.
Study on Au nanoflowers-human serum albumin for enantioselective of tryptophan enantiomers

Abstract:A simple chiral sensing platform for enantioselective recognition of the tryptophan enantiomers was fabricated via the self-assembled of human serum albumin (HSA) on the gold nanoflowers (AuNFs) modified glassy carbon electrode. And the interaction between the modified electrodes and the tryptophan enantiomers (D-Trp and L-Trp) was studied via square wave voltammetry (SWV) . The difference of peak currents in SWV display that the transmission of the electron on the electrode surface can be hindered by the interaction between D-or L-Trp and the chiral interface. The peak currents of L-Trp were significantly lower than that of D-Trp and the difference of peak currents reached 123.7μA. The peak currents of L-Trp or D-Trp were linearly related to the concentration in the range of 1.0 × 10-6 mol/L to 5.0 × 10-3 mol/L.

Abstract:A new colorimetric analysis approach for chiral recognition of d- and l-forms of phenylalanine (phe) was developed based on triangular silver nanoplates (TAg-NPs). The TAg-NPs could be used as chiral colorimetric probes for d- and l-forms of phe. Upon addition of d-phe to TAg-NPs solution, a color change from blue to purple to pink could be observed, while no obvious color change was found on addition of l-phe. l-phe could prevent the TAg-NPs from being etched to small size particles while the protective effect of d-phe was weak. Moreover, the enantiomeric excess of d-phe could be determined using the proposed chiral assay in the percentage of l-phe from 0 to 100% with a correlation coefficient of 0.9855. The phenomenon could be monitored by bare eyes and quantified analysis by UV–Vis spectrophotometry. The developed approach had several advantages, such as simplicity, visualization, short analysis time and low cost. This study presented a fast visualization analysis method of chiral d/l-phenylalanine and may lay the foundation for the development of visualization chiral recognition of other target analytes.

Abstract:Chirality is an important property of nature and it regulates fundamental phenomena in nature and organisms. Here, we develop a chiral electrochemical sensor based on copper–amino acid mercaptide nanorods (L-CuCys NRs) to discriminate tryptophan (Trp) isomers. The chiral L-CuCys NRs are prepared in alkaline solution based on the facile coordination reaction between the sulfhydryl groups of L-Cys and copper ions. Since the stability constant ( K ) of L-CuCys NRs with L-Trp (752) are much higher than that of L-CuCys NRs with D-Trp (242), the cross-linking bonds between L-CuCys NRs and L-Trp are more stable than those between L-CuCys NRs and D-Trp. Consequently, this electrochemical sensor can selectively recognize the Trp isomers with an enantiomeric electrochemical difference ratio ( I L-Trp / I D-Trp ) of 3.22, and it exhibits a detection limit of 0.26 μM for L-Trp. Moreover, this electrochemical sensor can quantitatively measure Trp isomers in complex samples. Importantly, this electrochemical sensor has the characteristics of high stability, good repeatability, easy fabrication, low cost, and efficient discrimination of tryptophan (Trp) isomers.

Abstract:Since chiral carboxylic acids (CCAs) are a class of important biological molecules and common functional moieties found in pharmaceutical molecules, the chiral analysis of CCAs has received much attention. Herein, we developed a simple, rapid and cost-effective method for visual and colorimetric high-throughput analysis of CCAs using chiral di-imine structures (CDIS) modified gold nanoparticles (C-AuNPs) as the probe. The C-AuNPs are positively charged in the presence of zinc ion, and they can be enantioselectively shielded by the negative charged CCA enantiomers. Therefore, upon the addition of different concentration and enantiomeric excess (ee) of CCAs, the C-AuNPs-based sensor shows the different levels of aggregation along with the visual changes in solution color, which can achieve simultaneous analysis of the concentration and ee of CCAs. The chiral recognition mechanism based on C-AuNPs was investigated by the determination of binding constants (K) and molecular simulation methods. Our approach is expected to have the wide-ranging applications in the developing region for enantio-sensing of various chiral drugs and biomolecules.

Abstract:The novel nanocomposites were obtained by covalently linking arginine enantiomers with the sidewall of multi-walled carbon nanotubes, and utilized as sensing materials for biomolecular recognition in electrochemical immunoassay. They were characterized by X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM) and circular dichroism spectroscopy (CD). Nano-gold and Prussian Blue were employed to amplify the analytical signal responses. Prostate specific antibody/antigen and carcinoembryonic antibody/antigen were used as model systems. The results showed that the different configurations of nanocomposites could readily facilitate chiral recognition of antibodies, and D-nanocomposites displayed a larger signal.

Abstract:Due to the crucial role of amino acids in life sciences and pharmaceutics, identification of optical amino acid molecules is of great significance. In this study, the two materials (CNT and PANI) were combined together to obtain the magnification of electrochemical signal by substrate material (CNT/PANI). Then a self-assembled multiwalled carbon nanotubes/polyaniline/sodium alginate (CNT/PANI/SA) nanocomposite with chiral sites and conductive material was synthesized as the electrochemical sensing interface. Next, a novel electrochemical sensing interface was fabricated via modifying the as-prepared chiral material on a polished glassy carbon electrode (CNT/PANI/SA/GCE) for precisely, efficiently, and rapidly differentiation of tryptophan (Trp) enantiomers. It was observed that CNT/PANI/SA/GCE showed desirable stereoselective recognition effect in the variety of signal strength to peak current (Ip) to the different optical activity of Trp enantiomers. In the case of optimal conditions, the peak current ratio in the solution of l-Trp and d-Trp (ID /IL ) was observed to be 2.1 at CNT/PANI/SA/GCE by differential pulse voltammogram (DPV). UV-visible spectroscopy further showed that CNT/PANI/SA had a greater binding energy to l-Trp. Also different factors affecting the enantioselectivity of CNT/PANI/SA/GCE, such as the incubation time, pH, and dropcoating volume of CNT/PANI/SA were optimized. Moreover, the proposed CNT/PANI/SA/GCE showed excellent specific stereoselectivity and anti-interference ability. Besides, the proposed chiral sensing platform can be effectively applied in real samples to detect Trp enantiomers sensitively. This work inspires us a new path for the preparation of substrate material with excellent electrical conductivity, as well as extend its application potential in chiral recognition.
