Abstract:The combination of supramolecular chemistry and nanotechnology has potentially applied in the construction of biosensors, and thus improves the analytical performance and robustness of electron devices. Herein, a new sandwich-type DNA sensor was constructed for ultrasensitive determination of hepatitis B virus (HBV) DNA, a recognized marker for chronic hepatitis B. The water-soluble pillar[5]arene stabilized Pd NPs combined with reduced graphene oxide nanosheet (WP5-Pd/RGO) was synthesized and employed as supporting material for the modification of electrode surface. The probe DNA was immobilized onto the electrode surface through a new strategy based on the host-guest interaction between WP5 and methylene blue labeled DNA (MB-DNA). Moreover, MOF-derived cobalt sulfide nanobox was prepared to anchor the hydroxylatopillar[5]arene stabilized Au NPs (HP5-Au/CoS), which had superior electrocatalytic performance towards H2O2 reduction to achieve signal amplification. Under the optimized conditions, the proposed sensor displayed a linear relationship between amperometric currents and the logarithm of tDNA solution from 1 × 10-15 mol/L to 1 × 10-9 mol/L, and a low detection limit of 0.32 fmol/L. What's more, the DNA sensor had remarkable behaviors of stability, reproducibility, specificity, and accuracy, which provided a potential and promising prospect for clinical diagnosis and analysis.

Abstract:Based on the strong interaction between single-stranded DNA and graphene material, we have designed a simple but smart electrochemical platform to detect HBV-DNA by using a graphene quantum dot (GQD) modified glassy carbon electrode coupled with specific sequence DNA molecules as probes. The probe DNA is designed to be complementary to the HBV-DNA, when the probe DNA is strongly bound to the surface of the GQD modified electrode the transfer of an electron from the electrode to the electrochemically active species K-3[Fe(CN)(6)] will become difficult. Nevertheless, if the target HBV-DNA is found in the test solution, the probe DNA will bind with the target HBV-DNA instead of GQDs. As a result, the obtained peak currents of K-3[Fe(CN)(6)] will have a different degree of increase with the different concentrations of the target HBV-DNA. In particular, the proposed sensor exhibits high sensitivity with a detection limit of 1 nM, and the linear detection range is from 10 nM to 500 nM. Additionally, the sensor could be used in detecting other probe DNA, which may have potential applications in the future.

Abstract:Persistent infection with hepatitis B virus (HBV) is a major health problem worldwide that may lead to chronic hepatitis, cirrhosis and primary liver cancer. In this paper, we report a QDs-DNA nanosensor based on fluorescence resonance energy transfer (FRET) for sensitive and specific detection of HBV DNA hybridization where the separation of un-ligated DNA strands from hybridized system are unnecessary. Experimental results showed that the proposed approach was suitable for label-free detection of specific DNA targets at low DNA concentration. The practical utility of the proposed method was demonstrated in the detection of synthetic 30-mer oliginucleotide targets derived from the hepatitis B virus with a sensitivity of 35 nM by a multilabel plate assay, which might make the application of the proposed method possible due to the simplicity and of the multilabel counter.

Abstract:As one of the most prominent global public health problems, which seriously threats public health security and human health, hepatitis B virus (HBV) infection is the main cause of liver cirrhosis and cancer. Herein, an innovative and sensitive biosensor for HBV-DNA detection was developed based on the surface etching of the helical gold nanorods (HGNRs) at single-particle level under a dark-field microscope (DFM). In this work, the unique HGNRs with high surface activity were functionalized as the optical probe, which were etched to the smooth gold nanorods and even to gold nanospheres, resulting in a distinct color and light scattering change of single particle with a more sensitive result of HBV-DNA detection than the common gold nanorods. Furthermore, the hybridization chain reaction amplification and DFM technique enabled the proposed strategy with a high sensitivity, offering the limit of detection (LOD) as low as 30.15 fM (3σ/k) and realizing sensitive detection of HBV-DNA from 0.05 to 10 pM. Moreover, the proposed strategy supplied the good specificity, which was capable of distinguishing mismatch sequence and other viral nucleic acids. In addition, this strategy was universal and provided a new idea for early diagnosis of other viral nucleic acids, such as hepatitis C virus (HCV) and human immunodeficiency virus (HIV).

Abstract:Hepatitis B virus (HBV) infection is associated with the progression of liver disease. Occult HBV infection (OBI) is defined as the existence of detectable HBV DNA in HBV surface antigen negative individuals. However, HBV DNA is negative in serum while HBV surface antigen remains positive in incomplete cured chronic HBV infection. Hence, combined detection of HBV surface antigen and HBV DNA is essential for the accurate detection and rehabilitation of HBV infection. Therefore, a multiplex detection strategy based on branched DNA nanostructures was developed. The single-stranded segment of branched DNA nanostructure (segment of S4 and S2) assembled by four single-stranded DNA was hybridized with aptamer of HBV surface antigen and DNA hairpin to construct the DNA nanosensor, which can achieve high specificity identification and highly sensitive fluorescence responding to the targets. The detection limits of the developed nanosensor for HBV and HBV DNA are 50 pM and 5 nM, respectively. The combined detection of HBV surface antigen and HBV DNA provides a new insight for more thorough diagnostic evaluation of HBV infection and rehabilitation.

Abstract:The prevalence of hepatitis B virus (HBV) is a global healthcare threat, particularly chronic hepatitis B (CHB) that might lead to hepatocellular carcinoma (HCC) should not be neglected. Although many types of HBV diagnosis detection methods are available, some technical challenges, such as the high cost or lack of practical feasibility, need to be overcome. In this study, the polycrystalline silicon nanowire field-effect transistors (pSiNWFETs) were fabricated through commercial process technology and then chemically functionalized for sensing hepatitis B virus surface antigen (HBsAg) and hepatitis B virus X protein (HBx) at the femto-molar level. These two proteins have been suggested to be related to the HCC development, while the former is also the hallmark for HBV diagnosis, and the latter is an RNA-binding protein. Interestingly, these two proteins carried opposite net charges, which could serve as complementary candidates for evaluating the charge-based sensing mechanism in the pSiNWFET. The measurements on the threshold voltage shifts of pSiNWFETs showed a consistent correspondence to the polarity of the charges on the proteins studied. We believe that this report can pave the way towards developing an approachable tool for biomedical applications.

Abstract:Sequence-known short-stranded hepatitis B virus (HBV) DNA fragment (181 bps) was obtained by PCR method. The strategy for its electrochemical detection was designed by covalently immobilizing single-stranded HBV DNA on gold electrode surface via carboxylate ester as a linkage between 3′-hydroxy end of DNA and carboxyl group of thioglycolic acid (TGA) self-assembled monolayer. The hybridization reaction on surface was evidenced by electrochemical methods using ferrocenium hexafluorophosphate (FcPF6) as an electroactive indicator. The interactions of Fc+ with single-stranded (ss) and double-stranded (ds) HBV DNA immobilized on TGA monolayer were studied. The difference between the responses of Fc+ at ss- and ds-DNA/Au electrodes suggested that this hybridization biosensor could be conveniently used to monitor DNA hybridization with a high sensitivity. AC impedance and XPS techniques have been employed to characterize the immobilization of ss-DNA on the gold surface.

Abstract:A novel fluorescent resonant energy transfer (FRET) biosensor was fabricated for the detection of hepatitis B virus (HBV) DNA using poly(ethylenimine) (PEI) modified upconversion nanoparticles (NH2-UCNPs) as energy donor and gold nanoparticles (Au NPs) as acceptor. The PEI modified upconversion nanoparticles were prepared directly with a simple one-pot hydrothermal method, which provides high quality amino-group functionalized UCNPs with uniform morphology and strong upconversion luminescence. Two single-stranded DNA strands, which were partially complementary to each other, were then conjugated with NH2-UCNPs and Au NPs. When DNA conjugated NH2-UCNPs and Au NPs are mixed together, the hybridization between complementary DNA sequences on UCNPs and Au NPs will lead to the quenching of the upconversion luminescence due to the FRET process. Meanwhile, upon the addition of target DNA, Au NPs will leave the surface of the UCNPs and the upconversion luminescence can be restored because of the formation of the more stable double-stranded DNA on the UCNPs. The sensor we fabricated here for target DNA detection shows good sensitivity and high selectivity, which has the potential for clinical applications in the analysis of HBV and other DNA sequences.

Abstract:A novel hepatitis B virus (HBV) DNA biosensor was developed by immobilizing covalently single-stranded HBV DNA fragments to a gold electrode surface via carboxylate ester to link the 3′-hydroxy end of the DNA with the carboxyl of the thioglycolic acid (TGA) monolayer. A short-stranded HBV DNA fragment (181bp) of known sequence was obtained and amplified by PCR. The surface hybridization of the immobilized single-stranded HBV DNA fragment with its complementary DNA fragment was evidenced by electrochemical methods using [Os(bpy)2Cl2]+ as a novel electroactive indicator. The formation of double-stranded HBV DNA on the gold electrode resulted in a great increase in the peak currents of [Os(bpy)2Cl2]+ in comparison with those obtained at a bare or single-stranded HBV DNA-modified electrode. The mismatching experiment indicated that the surface hybridization was specific. The difference between the responses of [Os(bpy)2Cl2]+ at single-stranded and double-stranded DNA/TGA gold electrodes suggested that the label-free hybridization biosensor could be conveniently used to monitor DNA hybridization with a high sensitivity. X-ray photoelectron spectrometry technique has been employed to characterize the immobilization of single-stranded HBV DNA on a gold surface.

Abstract:The direct electron transfer of guanine and adenine on HBV cccDNA strand through the protein membrane and/or host cell membranes of HBV was captured at PGCE by microwave irradiation which can melt cccDNA into ssDNA. Meanwhile, the difference of oxidation peaks in the whole blood after microwave irradiation can be used to screen HBV carriers and infection. The direct electrochemistry of hepatitis B virus containing the covalently closed circular DNA (HBV cccDNA) is realized at pretreated glassy carbon electrode (PGCE). It is essential that the direct electron transfer of guanine (G) and adenine (A) on HBV cccDNA strand is captured by microwave irradiation to melt cccDNA into ssDNA. The irreversible adsorption and diffusion controlled processes of G and A are revealed by cyclic voltammetry with participating 2 protons and 2 electrons respectively. In addition, we found that there are two oxidation peaks of G and A in whole blood containing HBV. Nevertheless, there is only one oxidation peak of G in whole blood of normal people. According to this difference of oxidation peaks in whole blood sample, it can be used screen quickly HBV infection or carriers in clinic and large area of blood donors.

Abstract:Hepatitis B Surface Antigen (HBsAg) is usually recognized as an earliest diagnostic markers for Hepatitis B Virus (HBV) infections. The precise and sensitive determination of HBsAg is crucial for the diagnostic of HBV infection. In this paper, a colorimetric immunosensor has been successfully designed and fabricated based on conjugating magnetic separation and enzymatic catalysis, for point-of-care detection of HBsAg. The HBsAg was simultaneously captured by the biotinylated HBsAg primary antibody (biotin-Ab1) labeled immunomagnetic nanoparticles and detection antibody (Ab2). In conjugation with Ab2, horseradish peroxidase (HRP) labeled rabbit anti-goat secondary antibody was linked with chromogenic agents to induce the colorimetric signal. As a result, a good linear relation was achieved between the concentration of HBsAg and OD450 nm (absorbance values at 450 nm) in the 0.1–20 ng mL−1 ranges, demonstrating a great specificity to HBsAg with a limit of detection of 0.012 ng mL−1. Furthermore, considering its succeed in real human serum sample test, this immunosensor could make contribute to potential applications in rapid diagnosis, effective prevention of epidemic and infectious diseases.

Abstract:Poly(acrylic acid) (PAA) brushes coated onto silica nanoparticles have been widely utilized in bioassays due to their abilities of providing favorable microenvironments and ensuring good biological activities for biomolecules. However, traditional PAA brushes are synthesized by reversible addition–fragmentation chain transfer polymerization. Hence, it is generally difficult to control and characterize the molecular weight of the PAA brushes, which may depress the reproducibility and bring more uncertain results. Herein, atom transfer radical polymerization method is employed to synthesize β-cyclodextrin-cored PAA with uniform and controllable molecular weight. After loading on the surfaces of adamantane-functionalized silica nanoparticles via host-guest interactions, glucose oxidase and probe single strand DNA (ssDNA) are further immobilized on the as-prepared nanoparticles. Meanwhile, capture ssDNA is functionalized on amino modified magnetic beads. In the presence of ssDNA sequence of Hepatitis B Virus (HBV) containing completely matched sequence of both probe and capture ssDNA, a bioconjugate is formed and can be separated by an external magnet. The isolated glucose oxidase can further catalyze glucose into gluconic acid and H2O2, and then reduce HAuCl4 on Au seeds. By monitoring the absorption intensity change of the Au NPs at 530nm, the proposed biosensor with novel signal amplification probes can be used to detect DNA sequence of HBV with high sensitivity and selectivity in both buffer and serum samples. This developed strategy has presented a new way to construct silica nanoparticles coated by PAA brushes for the fields of clinical diagnosis and other life sciences.

Abstract:Genotyping of hepatitis B virus (HBV) can be used for clinical effective therapeutic drug-selection. A novel microfluidic biochip for HBV genotyping has been fabricated, for the first time, integrating loop-mediated isothermal amplification (LAMP), line probes assay (LiPA) and giant magnetoresistive (GMR) sensors. Coupling LAMP with LiPA in microfluidic chip shortened reaction time substantially, and combining LAMP with GMR sensor enabled limit of detection to attain 10copiesmL−1 target HBV DNA molecules in 1h. Furthermore, the independent designed GMR sensors and microfluidic chip can decrease manufacturing cost and patient's test-cost, and facilitate GMR detector repeating use for signal detection. In addition, the detection system has a lower background signal owing to application of superparamagnetic nanoclusters. And it can be expected to use for multiple target molecules synchronous detection in microfluidic chip based on a characteristic of stationary reaction temperature of LAMP. In conclusion, the neoteric detecting system is well suitable for quick genotyping diagnosis of clinical HBV and other homothetic biomolecule detection in biological and medical fields.

Abstract:Hepatitis B virus (HBV) acts as a severe public health threaten through causing the chronic liver diseases. Although the quantified evaluation of HBV infection can be directly characterized by estimating the capacity of HBV DNA genome, it still lacks of effective and robust detection method without using enzyme or chemical labeling. Herein, we have designed a binary split fluorescent DNA aptasensor (bsFDA) through rationally splitting the Lettuce aptamer into two functional DNA short chains with utilizing the HBV DNA segment (HDs) complementary sequences. In this strategy, bsFDA has been investigated to be specifically recognized by HDs and then forming a triplex DNA contained the structure of Lettuce aptamer. Meanwhile, the turn-on fluorescence of bsFDA can be obtained when forming the fluorescent complex between DFHO and triplex DNA structure, and allowed the enzyme-free, label-free, fast-responsive, and reliable fluorescence readout for detecting HDs and the potential HDs mutants. Moreover, bsFDA has been applied for the spiked HDs analysis in different real matrixes including human serum and cell lysates. The satisfied recovery rates and reproducibility of bsFDA revealed the potentially detecting efficacies of HDs analysis in biological samples. Overall, bsFDA will hold the great potential in developing the functionalized aptasensor and realizing the viral genome analysis in biological research.

Noble metal-molybdenum disulfide nanohybrids as dual fluorometric and colorimetric sensor for hepatitis B virus DNA detection

Abstract:Hepatitis B virus (HBV) infection is one of the global healthcare burdens, and its early diagnosis is crucial for the prevention of HBV-induced chronic hepatitis, liver fibrosis, cirrhosis, and hepatocellular carcinoma. Although different detection approaches have been reported, most of these methods either rely on sophisticated machines or procedures, which limit their use particularly in the high endemic, developing countries. In this work, we report a dual-sensing nanoplatform built on noble metal-molybdenum disulfide (MoS2) nanohybrids, and this platform can detect the HBV DNA target through either fluorometric or colorimetric readouts. The design with the silver nanocluster (AgNC)–MoS2 nanohybrid enables multiplex fluorescent detection, while the HBV DNA-regulated growth of platinum nanoparticles (PtNPs) on the MoS2 nanosheets offers signal-on colorimetric detection. Both AgNC–MoS2 and PtNP–MoS2 nanohybrids show high sensitivity with pico-molar detection limit and single nucleotide specificity, even with the spiked human serum. Collectively, the proposed nanohybrids possess their potential in the use of early HBV diagnosis, particularly suitable for the high endemic areas with limited medical and instrumental supports.

Flexible Terahertz Metamaterial Biosensor for Ultra-Sensitive Detection of Hepatitis B Viral DNA Based on the Metal-Enhanced Sandwich Assay

Abstract:The high sensitivity and specificity of terahertz (THz) biosensing are both promising and challenging in DNA sample detection. This study produced and refined a flexible THz MM biosensor for ultrasensitive detection of HBV in clinical serum samples based on a gold magnetic nanoparticle-mediated rolling circle amplification (GMNPs@RCA) sandwich assay under isothermal conditions. Typically, solid-phase RCA reactions mediated by circular padlock probes (PLPs) are triggered under isothermal conditions in the presence of HBV DNA, resulting in long single-stranded DNA (ssDNA) with high fidelity and specificity. Then, the resultant ssDNA was conjugated with detection probes (DPs) immobilized on gold nanoparticles (DP@AuNPs) to form GMNPs-RCA-AuNPs sandwich complexes. The HBV DNA concentrations were quantified by introducing GMNPs-RCA-AuNPs complexes into the metasurface of a flexible THz metamaterial-based biosensor chip and resulting in a red shift of the resonance peak of the THz metamaterials. This biosensor can lead to highly specific and sensitive detection with one-base mismatch discrimination and a limit of detection (LOD) down to 1.27E + 02 IU/ml of HBV DNA from clinical serum samples. The HBV DNA concentration was linearly correlated with the frequency shift of the THz metamaterials within the range of 1.27E + 02∼1.27E + 07 IU/ml, illustrating the applicability and accuracy of our assay in real clinical samples. This strategy constitutes a promising THz sensing method to identify virus DNA. In the future, it is hoped it can assist with pathogen identification and clinical diagnosis.
