Abstract:Coupling nanomaterials with biomolecular recognition events represents a new direction in nanotechnology toward the development of novel molecular diagnostic tools. Here a graphene oxide (GO)‐based multicolor fluorescent DNA nanoprobe that allows rapid, sensitive, and selective detection of DNA targets in homogeneous solution by exploiting interactions between GO and DNA molecules is reported. Because of the extraordinarily high quenching efficiency of GO, the fluorescent ssDNA probe exhibits minimal background fluorescence, while strong emission is observed when it forms a double helix with the specific targets, leading to a high signal‐to‐background ratio. Importantly, the large planar surface of GO allows simultaneous quenching of multiple DNA probes labeled with different dyes, leading to a multicolor sensor for the detection of multiple DNA targets in the same solution. It is also demonstrated that this GO‐based sensing platform is suitable for the detection of a range of analytes when complemented with the use of functional DNA structures. A graphene oxide (GO)‐based multicolor fluorescent DNA nanoprobe is reported; it allows rapid, sensitive, and selective detection of DNA targets in homogeneous solution based on interactions between GO and DNA molecules. The large planar surface of GO allows simultaneous quenching of multiple DNA probes labeled with different dyes, leading to a multicolor DNA sensor.

Abstract:We explore the interactions between a fluorescein (FAM)-labeled single-stranded DNA (P), graphene oxide (GO), and a cationic conjugated polymer, poly [(9,9-bis(6'-N,N,N-trimethylammonium)hexyl)-fluorenylene phenylene dibromide] (PFP). It is found that the fluorescence change of P-GO-PFP system is dependent on the addition order of P and PFP. When adding PFP into P/GO complex, the fluorescence resonance energy transfer (FRET) from PFP to P is inefficient If P is added to PFP/GO complex, efficient FRET is obtained. This may be attributed to the equal binding ability for P and PFP to GO. The results of time resolved fluorescence and fluorescence anisotropy support the different fluorescent response under different addition order of P and PFP to GO. Based on the above phenomenon, we demonstrate a method to reduce the high background signal of a traditional PFP-based DNA sensor by introducing GO. In comparison to the use of single PFP, the combination of PFP with GO-based method shows enhanced sensitivity with a detection limit as low as 40 pM for target DNA detection.

Abstract:A fluorescent biosensor for sequence-specific recognition of double-stranded DNA (dsDNA) was developed based upon the DNA hybridization between dye-labeled single-stranded DNA (ssDNA) and double-stranded DNA. The fluorescence of FAM-labeled single-stranded DNA was quenched when it adsorbed on the surface of graphene oxide (GO). Upon addition of the target dsDNA, a homopyrimidine.homopurine part of dsDNA on the Simian virus 40 (SV40) (4424-4440, gp6), hybridization occurred between the dye-labeled DNA and the target dsDNA, which induced the dyelabeled DNA desorbed from the surface of GO, and turned on the fluorescence of the dye. Under the optimum conditions, the enhanced fluorescence intensity was proportional to the concentration of target dsDNA in the range 40.0-260 nM, and the detection limit was found to be 14.3 nM alongside the good sequence selectivity.

Abstract:Graphene is an increasingly important nanomaterial that has shown great promise in the area of nanotechnology. In this study, fluorescein-functionalized graphene oxide (GO) is synthesized via a polyethylene glycol (PEG) bridge and us application in intracellular imaging is explored GO is an oxide form of graphene that provides an ideal platform to prepare graphene-based functional nanomaterials via chemical modification. The PEG bridge was Introduced to prevent GO-induced quenching of conjugated fluorescein. The fluorescein-PEG-GO conjugate thus prepared exhibits excellent pH-tunable fluorescent properties and, more significantly, can be efficiently taken up by cells and serve as a fluorescent nanoprobe for intracellular imaging.

Abstract:Aggregation‐induced emission (AIE) probes have shown great potential applications in fluorescent sensing of biomolecules, and the integration of AIE probe and graphene oxide (GO) attracts intense interest in developing new tools for label‐free and “turn‐on” fluorescent biomolecular analysis. Herein, an ultrasensitive label‐free and “turn‐on” DNA sensing is realized by tuning the supramolecular interactions of AIE probe and GO with DNA. The investigation of supramolecular interactions of AIE probes and GO with DNA demonstrate that AIE probe with short alkyl chains substitute shows highest binding affinity with DNA strand, and GO with low oxidation degree possesses strong binding interactions to ssDNA and the highest fluorescence quenching efficiency. As a result, the optimized AIE probes and GO‐based fluorescent sensor can selectively detect the target DNA sequence and exhibits the detection limitation as low as 0.17 × 10–9m. It is believed that the research efforts will provide an efficient approach to improve the performance of DNA sensing assay and an indepth understanding of the supramolecular interactions of AIE probes and GO with DNA, and thus facilitate their extended applications in biosensors and biomedicine.
A Turn-on Fluorescence-Sensing Technique for Glucose Determination Based on Graphene Oxide–dna Interaction

Abstract:Graphene is a two-dimensional carbon nanomaterial one atom thick. Interactions between graphene oxide (GO) and ssDNA containing different numbers of bases have been proved to be remarkably different. In this paper we propose a novel approach for turn-on fluorescence sensing determination of glucose. Hydrogen peroxide (H2O2) is produced by glucose oxidase-catalysed oxidation of glucose. In the presence of ferrous iron (Fe2+) the hydroxyl radical (•OH) is generated from H2O2 by the Fenton reaction. This attacks FAM-labelled long ssDNA causing irreversible cleavage, as a result of the oxidative effect of •OH, producing an FAM-linked DNA fragment. Because of the weak interaction between GO and short FAM-linked DNA fragments, restoration of DNA fluorescence can be achieved by addition of glucose. Due to the excellent fluorescence quenching efficiency of GO and the specific catalysis of glucose oxidase, the sensitivity and selectivity of this method for GO-DNA sensing are extremely high. The linear range is from 0.5 to 10 μmol L−1 and the detection limit for glucose is 0.1 μmol L−1. The method has been successfully used for analysis of glucose in human serum.

Abstract:In the past few years, graphene oxide (GO) has emerged as a unique platform for developing DNA-based biosensors, given the DNA adsorption and fluorescence-quenching properties of GO. Adsorbed DNA probes can be desorbed from the GO surface in the presence of target analytes, producing a fluorescence signal. In addition to this initial design, many other strategies have been reported, including the use of aptamers, molecular beacons, and DNAzymes as probes, label-free detection, utilization of the intrinsic fluorescence of GO, and the application of covalently linked DNA probes. The potential applications of DNA-functionalized GO range from environmental monitoring and cell imaging to biomedical diagnosis. In this review, we first summarize the fundamental surface interactions between DNA and GO and the related fluorescence-quenching mechanism. Following that, the various sensor design strategies are critically compared. Problems that must be overcome before this technology can reach its full potential are described, and a few future directions are also discussed.
Partially Reduced Graphene Oxide As Highly Efficient DNA Nanoprobe.

Abstract:This work investigates the effect of reduction degree on graphene oxide (GO)-DNA interaction and the fluorescence quenching mechanism. Partial reduced graphene oxide (pRGO), which maintains well water-dispersibility, is synthesized using a mild reduction method by incubating GO suspension under alkaline condition at room temperature. The fluorescence quenching enhances with the restoration degree of sp(2) carbon bonds and follows the static quenching mechanism. The binding constant values imply that pRGO has much stronger affinity with ssDNA than GO. Utilizing this highly efficient nanoprobe, a universal sensing strategy is proposed for homogeneous detection of DNA. Compared with the reported GO-based DNA, this present strategy has obvious advantages such as requirement of low nanoprobe dosage, significantly reduced background, fast fluorescence quenching, and improved sensitivity. Even without any amplification process, the limit of detection can reach as low as 50 pM.

Abstract:In this work, we report the design of a novel graphene-based molecular beacon (MB) that could sensitively and selectively detect specific DNA sequences. The ability of water-soluble graphene oxide (GO) to differentiated hairpin and dsDNA offered a new approach to detect DNA. We found that the background fluorescence of MB was significantly suppressed in the presence of GO, which increased the signal-to-background ratio, hence the sensitivity. Moreover, the single-mismatch differentiation ability of hairpin DNA was maintained, leading to high selectivity of this new method.

Abstract:Reduced glutathione (GSH) is a crucial endogenous antioxidant that plays a central role in maintaining cell activity by involving in cellular defense against reactive oxygen species (ROS). Therefore, the detection of intracellular GSH is essential to monitor the cellular state. Herein, graphene oxide–MnO2–fluorescein (GO–MnO2–FL) nanocomposite was synthesized and used for GSH detection. GSH could reduce MnO2 to Mn2+ and block the energy transfer between FL and GO–MnO2, leading to the significant fluorescence enhancement of FL. The fluorescence-based GSH sensing has a limit of detection of 1.53 μM and a wide detection range of 10 μM–2 mM. Moreover, the GO–MnO2–FL nanocomposite was found to selectively image the cancer cells since the GSH content of cancer cells is higher than that of normal cells. In vivo experiments also confirmed the normal/cancer cell differentiation capability and excellent tumor-targeting ability of the GO–MnO2–FL nanocomposite. These properties make the GO–MnO2–FL nanocomposite an excellent fluorescent probe for GSH detection both in vitro and in vivo, and we believe it may hold great potential for GSH-mediated cancer diagnosis.

Abstract:Adsorption of a fluorophore-labeled DNA probe by graphene oxide (GO) produces a sensor that gives fluorescence enhancement in the presence of its complementary DNA (cDNA). While many important analytical applications have been demonstrated, it remains unclear how DNA hybridization takes place in the presence of GO, hindering further rational improvement of sensor design. For the first time, we report a set of experimental evidence to reveal a new mechanism involving nonspecific probe displacement followed by hybridization in the solution phase. In addition, we show quantitatively that only a small portion of the added cDNA molecules undergo hybridization while most are adsorbed by GO to play the displacement role. Therefore, it is possible to improve signaling by raising the hybridization efficiency. A key innovation herein is using probes and cDNA with a significant difference in their adsorption energy by GO. This study offers important mechanistic insights into the GO/DNA system. At the same time, it provides simple experimental methods to study the biomolecular reaction dynamics and mechanism on a surface, which may be applied for many other biosensor systems.

Abstract:The unique properties of graphene oxide (GO), such as good water solubility, non-toxicity, biocompatibility and cell permeability, make GO well suited for a number of biological applications. In this study, we explore the application of GO as an efficient fluorescent quencher for a highly fluorescent organic dye, resorufin (RF). The quenching effect of GO on resorufin is studied by noncovalent and covalent bonding of resorufin, respectively. The fluorescence is completely quenched when resorufin is covalently linked to GO; while resorufin's fluorescence could be still observed through noncovalent bonding to GO sheet. Interestingly, addition of hydrazine into RF-GO complex causes the fluorescence recovered, providing a potential ''OFF-ON'' fluorescence responsive probe for detecting hydrazine in vitro and in living cells.

Abstract:In this study, we develop an efficient method for multiple DNA detection by exploring silver nanoclusters (AgNCs)-graphene oxide (GO) nanohybrid materials. Because of the extraordinarily high quenching efficiency of GO, the ssDNA-AgNCs probe exhibits minimal background fluorescence, while strong emission is observed when it forms a double helix with the specific target DNA, leading to a high signal-to-background ratio. Therefore the AgNCs-GO nanohybrid materials can be successfully applied for DNA detection. The system described here exhibits not only high sensitivity with a detection limit of 1 nM, but also an excellent differentiation ability for single-base mismatched sequences. In addition, by exploring AgNCs as signal reporters and GO as the nanoquencher, this approach avoids labeling the probe DNA or target DNA, which offers the advantages of simplicity and cost efficiency. Moreover, the large planar surface of GO allows adsorption of different DNA-AgNCs probes, each with a distinct emission, leading to a multicolor sensor for the detection of multiple DNA targets in the same solution.

Abstract:A fluorescence biosensing strategy based on graphene oxide (GO) was reported for simple, rapid, sensitive, and selective DNA detection by T7 exonuclease assisted target recycling amplification. Due to the super fluorescence quenching efficiency of GO, the fluorescein amiditelabeled signal probe was firstly adsorbed onto the surface of GO and the fluorescence was quenched. Owing to its excellent selectivity for double-stranded DNA, T7 exonuclease was chosen as a signal-amplifying biocatalyst to improve the detection sensitivity. In the presence of target DNA, the signal probe could bind with target DNA and form a DNA duplex structure to trigger the digestion of the signal probe by T7 exonuclease, leading to the recycling of target DNA and the increasing of fluorescence intensity. Upon the recycling use of target DNA, this method achieved a high sensitivity towards target DNA with a detection limit of 0.3 pmol/L, which was lower than previously reported for GO-based DNA biosensors. Moreover, it does not require complex modifications of the molecular beacon and time-consuming thermal cycling procedures. Thus, the simple strategy provides a universal biosensing platform for DNA detection and it could find wide applications in DNA damage analysis and diagnostics.
