Abstract:Since the 21st century, nanocomposites based on layered double hydroxide (LDH) and carbon nanotubes (CNTs) are extensively employed as heat stabilizers, supercapacitors and electrocatalysts, due to their high conductivity, outstanding stability and environmental friendliness. Herein, CNTs/NiAl-LDH composite were obtained by onestep hydrothermal method and used as a platform for Ru(phen)3Cl2 (tris(1,10-phenanthroline)ruthenium (II) dichloride) sensor to selectively detect CT DNA (Calf thymus DNA). It was found that CNTs/NiAl-LDH composite could effectively quench the emission of the Ru(phen)3Cl2 sensor in aqueous solution. After adding a certain amount of CT DNA to the above system, the Ru(phen)3Cl2 interacted with CT DNA immediately and released from the CNTs/NiAl-LDH, resulting in the fluorescence recovery. In the concentration range of 4.4-26.3 mu g/mL, the increase of luminescence was linearly proportional to the amount of CT DNA added, and the correlation coefficient was 0.9967. The detection limit was 6.09 x 10-8 g/mL.

Abstract:In this work, a novel fluorescent DNA sensor system based on polydopamine (PDA) modified layered double hydroxides (Mg/Al-LDH) was designed and PDA-co-LDH composites were examined for DNA sensing owing to their fluorescence quenching capability. We demonstrated that the fluorescence of ssNDA could be effectively quenched after mixing with PDA-co-LDH composites, while the fluorescence could be recovered after the target DNA was added to form dsDNA. Based on the fluorescence changes, the target DNA can be facilely and quantitatively determined by the DNA sensor system. Based on 3σ method, the detection limit is calculated to be as low as 1.0 pM. Moreover, PDA-co-LDH composites have an excellent sensitivity and selectivity that can discriminate the complementary and mismatched target sequences even for the single-base mismatched target sequences. More importantly, this DNA sensor system could be expected for detection of many other molecules through design DNA sequences.
Electrochemical detection of DNA hybridization based on three-dimensional ZnO nanowires/graphite hybrid microfiber structure.

Abstract:In the present work, zinc oxide (ZnO) nanowire (NWs)/graphite microfiber hybrid electrodes were used for the detection of DNA hybridization. ZnO NWs were in-situ synthesized onto graphite fibers using the hydrothermal technique, and the result was characterized by X-ray diffraction (XRD), scanning electron microscope (SEM) as well as transmission electron microscope (TEM). An electrochemical sensor for the analysis of DNA hybridization was developed based on immobilizing DNA probes onto the ZnO NWs/graphite microfiber electrodes surface via electrostatic interaction. Cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and X-ray photoelectron spectroscopy (XPS) were used to corroborate the fabrication of the sensor. Based on the results, ZnO NWs/graphite microfiber hybrid electrodes showed a great hybridization capacity for the determination of target DNA. A wide dynamic detection range from 1.0 × 10-14 M to 1.0 × 10-7 M with an ultralow detection limit of 3.3 × 10-15 M was achieved for detecting the target DNA. Moreover, it showed good selectivity and stability during the detection process.

Abstract:We demonstrated an easy and efficient approach to produce amino-group-functionalized graphene (GNH(2)) for highly sensitive DNA biosensing through intermolecular interaction. The nucleotide molecules prefer to anchor onto the as-prepared G-NH2 surface rather than be immobilized on pristine graphene. The electrostatic interaction between the positive charges of the ionized amino on G-NH2 and the negative charges of phosphate groups on nucleotide chains enabled label-free probe DNA to be immobilized on the G-NH2 surface. The electrochemical performance and quality variations during the preparation of G-NH2 and nucleotide immobilization processes were determined by electrochemical measurements and quartz crystal microbalance in situ, respectively. Results showed that complementary target DNA could be hybridized with probe DNA within the concentration range of 0.1 nM to 200 nM, and the detection limit was 0.8 nM. Therefore, this kind of G-NH2 could be an alternative for DNA biosensing.

Abstract:A novel 2-dimensional graphene analog molybdenum disulfide/multi-walled carbon nanotube (MoS2/MWCNT) was synthesized by a simple hydrothermal method to achieve excellent electrochemical properties. An ultrasensitive electrochemical DNA biosensor was subsequently constructed by assembling a thiol-tagged DNA probe on a MoS2/MWCNT and gold nanoparticle (AuNP)-modified electrode that has already been coupled with glucose oxidase (GOD). In this work, GOD was used as a redox marker. The heteronanostructure formed on the biosensor surface appeared relatively good conductor for accelerating the electron transfer, while the modification of GOD and AuNPs provided multiple signal amplification for electrochemical biosensing. The multiple signal amplification strategy produced an ultrasensitive electrochemical detection of DNA down to 0.79fM with a linear range from 10fM to 107fM, and appeared high selectivity to differentiate three-base mismatched DNA and one-base mismatched DNA. The developed approach provided a simple and reliable method for DNA detection with high sensitivity and specificity, and would open new opportunities for sensitive detection of other biorecognition events.

Abstract:Graphene combines with single-stranded DNA by a self-assembly process under strong ultrasonication and in the resulting water-dispersible graphene-DNA hybrids, monolayers of globular ss-DNA molecules are adsorbed on both sides of the graphene sheets by a non-covalent pi-pi stacking. The cyclic voltammetry results of the graphene-DNA hybrids coated electrodes demonstrate a well-defined and nearly symmetrical redox characteristic which means an enhanced electron transfer on the electrode surface as compared to the uncoated glassy carbon electrodes. Accordingly, the coated ones show apparently better sensing performance towards hydrogen peroxide which is characterized by large detection range, rapid response and high sensitivity.

Abstract:Layered double hydroxides (LDHs), also known as hydrotalcite-like anionic clays, have been investigated widely as promising electrochemical active materials. Due to the inherently weak conductivity, the electrochemical properties of LDHs were improved typically by utilization of either functional molecules intercalated between LDH interlayer galleries, or proteins confined between exfoliated LDH nanosheets. Here, we report a facile protocol to prepare NiAl-LDH/graphene (NiAl-LDH/G) nanocomposites using a conventional coprecipitation process under low-temperature conditions and subsequent reduction of the supporting graphene oxide. Electrochemical tests showed that the NiAl-LDH/G modified electrode exhibited highly enhanced electrochemical performance of dopamine electrooxidation in comparison with the pristine NiAl-LDH modified electrode. Results of high-resolution transmission electron microscopy and Raman spectra provide convincing information on the nanostructure and composition underlying the enhancement. Our results of the NiAl-LDH/G modified electrodes with the enhanced electrochemical performance may allow designing a variety of promising hybrid sensors via a simple and feasible approach.

Abstract:This work reports the synthesis of novel CdTe quantum dot (QD)-functionalized porous ZnO nanosheets via a covalent binding method with (3-aminopropyl)triethoxysilane as a linker. The functional nanosheets showed an excellent visible-light absorbency and much higher photoelectrochemical activity than both CdTe QDs and ZnO nanosheets due to the porous structure and appropriate band alignment between the CdTe QDs and ZnO nanosheets. Using hydrogen peroxide as an electron acceptor the nanosheet-modified electrode showed a sensitive photocurrent response. This speciality led to a novel methodology for the design of hydrogen peroxide-related biosensors by the formation or consumption of hydrogen peroxide. Using biotin-labeled DNA as capture probe, a model biosensor was proposed by immobilizing the probe on a nanosheet-modified electrode to recognize target DNA in the presence of an assistant DNA, which produced a "Y" junction structure to trigger a restriction endonuclease-aided target recycling. The target recycling resulted in the release of biotin labeled to the immobilized DNA from the nanosheet-modified electrode, thus decreased the consumption of hydrogen peroxide by horseradish peroxidase-mediated electrochemical reduction after binding the left biotin with horseradish peroxidase-labeled streptavidin, which produced an increasing photoelectrochemical response. The 'signal on' strategy for photoelectrochemical detection of DNA showed a low detection limit down to the subfemtomole level and good specificity to single-base mismatched oligonucleotides. The sensitized porous ZnO nanosheets are promising for applications in both photovoltaic devices and photoelectrochemical biosensing.

Abstract:The detection of specific DNA sequences provides a powerful tool for early diagnosis and prevention of disease. Herein, an enzyme-free electrochemiluminescence (ECL) DNA biosensor was developed by coupling inorganic-organic nanocomposite as the luminophore and ZnCo2O4 as the efficient accelerator. ZnCo2O4 nanoparticles (ZCO NPs), derived from Co and Zn based bimetallic zeolite imidazole framework, was used as the ECL substrate due to their large surface area and excellent catalytic effect. AgInS2-ZnS quantum dots (ZAIS) were covalently linked with meso-tetra(4-carboxyphenyl)-porphine (TCPP) to form ZAIS/TCPP inorganic-organic nanocomposites, which were used for the first time as the cathodic ECL luminophore with the coreactant of K2S2O8. Detection of target DNA (tDNA) about chronic myeloid leukemia (CML) was achieved based on dual-signal amplification via cascade toehold-mediated strand displacement reaction (TMSDR). On the basis of experimental results, a limit of detection (LOD) of 200 aM was achieved in the range of 0.5 fM to 1 nM, which showed the excellent performance of this biosensor. This work not only indicates that inorganic-organic nanocomposites as the ECL luminophore is a feasible strategy for efficient emission, but also expands the application of nanomaterials derived from metal organic frameworks.
Self-assembled, Functionalized Graphene and DNA As a Universal Platform for Colorimetric Assays

Abstract:We have demonstrated a robust sensing strategy by employing single-stranded probe DNA and the hemin-graphene hybrid (GH) to detect a broad range of targets including metal ions, DNA and small molecules. This nearly “universal” biosensor approach is based on the DNA-mediated assembly of the hemin-graphene composite upon addition of the targets. Afterwards, GH aggregate resulting from DNA hybridization will occur. The DNA-GH hybrids will settle on the bottom of the vial after centrifugation, leaving behind a transparent supernatant. After incubation with TMB and H2O2, the colorimetric signal of the centrifugal supernatant will be significantly lower compared to that in the absence of targets. Therefore, mediation of the assembly of DNA on GH by targets can yield a facile means with tunable optical properties in response to concentration changes of the targets. This colorimetric “readout” offers great advantages such as the simple operation process, low-cost portable instrument and easy-to-use applications. Therefore, we believe that this method promises a great potential of becoming a routine tool for quantitative detection of a wide spectrum of analytes for specific applications in biodiagnostics, nanoelectronics, and bionanotechnology.

Abstract:Graphene/cadmium sulphide (GR-CdS) nanocomposite was synthesized via a low temperature process in aqueous solution. The as-prepared nanocomposite was characterized by scanning electron microscopy, UV-visible spectroscopy, Fourier transform infrared spectroscopy, and X-ray diffraction. The impedance analysis indicated that GR-CdS nanocomposite possessed outstanding electrochemical performance for facile electron transfer. When DNA was immobilized on GR-CdS (DNA/GR-CdS) modified electrode, the electrochemical oxidation of guanine and adenine in DNA residue bases was significantly promoted. Due to the interaction of DNA with phenformin, the voltammetric current of guanine or adenine on the DNA/GR-CdS electrode was decreased when phenformin was present in the electrolytic solution. Under optimized conditions, the signal of guanine on DNA/GR-CdS electrode decreased linearly with increasing the concentration of phenformin in the range of 1.0 x 10(-6) mol L-1 to 1.0 x 10(-3) mol L-1. Abstract:Graphene quantum dots (GQDs), an alternative of conventional luminescent reagents, has been widely used in electrochemiluminescent (ECL) analysis because of its low cost, non-toxicity and ease of preparation. To further improve the ECL signal of GQDs, we herein succeeded in synthesizing hydrazide modified graphene quantum dots (HM-SGQDs). In comparison with GQDs, HM-SGQDs possessed abundant luminol like units, thus greatly enhancing the ECL intensity. On the other hand, luminol involved ECL system typically needed the participation of H2O2 as a coreactant, while hemin/G-quadruplex DNAzyme (hGQ) DNAzyme could efficiently catalyze the decomposition of H2O2. Accordingly, taking p53 gene as a model, a novel ECL DNA biosensor was developed based on HM-SGQDs and hGQ DNAzyme. The experimental results indicated target DNA can be quantified in a linear range from 100fM to 100nM with a detection limit of 66fM (S/N=3). Meanwhile, the discrimination of single-base mismatch was also achieved with the proposed analytical approach, suggesting broad potential applications of nucleic acid-related clinical diagnosis.

Abstract:In this paper, we constructed a new electrochemical biosensor for DNA detection based on a molecule recognition technique. In this sensing protocol, a novel dual-labeled DNA probe (DLP) in a stem–loop structure was employed, which was designed with dabcyl labeled at the 3′ end as a guest molecule, and with a Pb nanoparticle labeled at the 5′ end as electrochemical tag to indicate hybridization. One α-cyclodextrin-modified electrode (α-CD/MCNT/GCE) was used for capturing the DNA hybridization. Initially, the DLP was in the “closed” state in the absence of the target, which shielded dabcyl from the bulky α-CD/MCNT/GCE conjugate due to a steric effect. After hybridization, the loop sequence (16 bases) formed a rigid duplex with the target, breaking the relatively shorter stem duplex (6 bases). Consequently, dabcyl was forced away from the Pb nanoparticle and became accessible by the electrode. Therefore, the target hybridization event can be sensitively transduced via detecting the electrochemical reduction current signal of Pb. Using this method, as low as 7.1×10−10M DNA target had been detected with excellent differentiation ability for even a single mismatch.
A photoelectrochemical biosensor for determination of DNA based on flower rod-like zinc oxide heterostructures

Abstract:The work describes a photoelectrochemical (PEC) biosensor for the detection of DNA. It employs oriented hierarchical ZnO flower-rod architectures (ZnO FRs) and DNA dendrimers. ZnO FRs act as photoactive material that yields a photocurrent of up to 23 μA. The photogenerated electron transfer is inhibited once the probe DNA and the target DNA oligonucleotides hybridize, and this results in a reduced photocurrent. The use of DNA dendrimers with scores of DNA branches further amplifies the signal of the PEC biosensor. The PEC sensor displays a response that is linear in the DNA concentration range from 10 fM to 0.1 μM with a detection limit of 3.7 fM (at S/N = 3). The sensor was applied to the determination of DNA in human serum samples and was found to work with acceptable accuracy. Abstract:A layer-by-layer (LBL) assembly was employed for preparing multilayer thin films with a controlled architecture and composition. In this study, we report the one-step coelectrodeposition-assisted LBL assembly of both gold nanoparticles (AuNPs) and reduced graphene oxide (rGO) on the surface of a glassy carbon electrode (GCE) for the ultrasensitive electrochemical impedance sensing of DNA hybridization. A self-healable nanohybrid thin film with a three-dimensional (3D) alternate-layered nanoarchitecture was obtained by the one-step simultaneous electro-reduction of both graphene oxide and gold chloride in a high acidic medium of H2SO4 using cyclic voltammetry and was confirmed by different characterization techniques. The DNA bioelectrode was prepared by immobilizing the capture DNA onto the surface of the as-obtained self-healable AuNP/rGO/AuNP/GCE with a 3D LBL nanoarchitecture via gold-thiol interactions, which then served as an impedance sensing platform for the label-free ultrasensitive electrochemical detection of DNA hybridization over a wide range from 1.0 x 10(-9) to 1.0 x 10(-13) g ml(-1), a low limit of detection of 3.9 x 10(-14) g ml(-1) (S/N = 3), ultrahigh sensitivity, and excellent selectivity. This study presents a promising electrochemical sensing platform for the label-free ultrasensitive detection of DNA hybridization with potential application in cancer diagnostics and the preparation of a self-healable nanohybrid thin film with a 3D alternate-layered nanoarchitecture via a one-step coelectrodeposition-assisted LBL assembly.

Abstract:A new type of PtCo(x) @graphene nanocomposite is prepared by a simple chemical solution method, which can dramatically enhance the chemiluminescence (CL) intensity of luminol-H(2)O(2) system, making it possible for the detection of glucose through measuring the H(2)O(2) produced from its catalytic oxidation.
