Abstract:The paper describes a new online quantitative electrophoretically mediated microanalysis (EMMA) for use in immunoassays based on the unique drop-by-drop introduction of a sample by means of an inkjet for capillary electrophoresis (CE). Plugs of a fluorescein-labeled antibody (Anti-humanIgG-DyL550) and human IgG were alternately injected into a capillary using the inkjet, followed by the merging of the plugs and the subsequent immune reaction. The antigen-antibody complex that was formed in the merged zone was then separated by CE. As a proof-of-concept, the method was used to determine human IgG. As a result, both the consumption of the reaction solution and the analysis time were significantly reduced. The method showed a wide linear range (10-2000 ng mL(-1), R-2 = 0.9912) of calibration and the detection limit (5 ng mL(-1)) was substantially lower than that by for conventional methods. Inkjet Nanoinjection for High-Thoughput Chemiluminescence Immunoassay on Multicapillary Glass Plate

Abstract:We report a novel chemiluminescence diagnosis system for high-throughput human IgA detection by inkjet nanoinjection on a multicapillary glass plate. As proof-of-concept, microhole-based polydimethylsiloxane (PDMS) sheets were aligned on a multicapillary glass plate to form a microwell array as microreactors for enzyme-linked immunosorbent assay (ELISA). The multicapillary glass plate was utilized as a switch that controlled the holding/passing of the solution. Further, anti-IgA-labeled polystyrene (PS) microbeads was assembled into the microwell array, and an inkjet nanoinjection was specially used to distribute the sample and reagent solution for chemiluminescence ELISA, enabling high-throughput detection of human IgA. As a result, the performance of human IgA tests revealed a wider range for the calibration curve and a lower limit of detection (LOD) of 0.1 ng mL(-1) than the ELISA by a standard 96-well plate. The analysis time and reagent consumption were significantly decreased. The IgA concentrations in saliva samples were determined after 10000-fold dilution by the developed ELISA system showing comparable results by conventional immune assay with 96-wells. Thus, we believe that the inkjet nanoinjection for high-throughput chemiluminescence immunoassay on a multicapillary glass plate will be promising in disease diagnosis.

Abstract:A portable heterogeneous immunoassay system is presented in this paper. It consists of a poly(dimethylsiloxane) (PDMS) based microfluidic chip as the immunoreactor, a miniaturized programmable high voltage sequencer as the power supply and the flow controller, and a laser-optical fiber fluorescence detection module as the signal reader. The operation of this immunoassay system is automatic. The sequential reagent dispensing and washing processes are controlled by the programmable sequencer. The reagent consumption was only 12 microL and the assay time was only 26 min. The detection limit for Escherichia coli O157:H7 bacterial antigen with this miniaturized system was 0.3 ng/muL, lower than that obtained using fluorescence microscope in previous studies.

Abstract:The first version of nano-injection device for capillary gas chromatography (cGC) based on inkjet microchip was developed. The nano-injector could accurately control the injection volume in nano-liter, even pico-liter range. Its configuration and mechanism were discussed in detail. Adopting photolithography and plasma etching technology, we firstly fabricated the inkjet microchip and stuck to a piezoelectric device to eject droplets. Then, a special feedback tube was added to make it function as a nano-injector for cGC, which was an important design to compensate pressure difference between the evaporation chamber of cGC and the sample extrusion chamber of inkjet microchip. The injected volume can be precisely controlled by the number of injected droplets. Excellent precision (RSDs were below 10.0%, n=5) was observed for the injection of ethanol at elevated pressure. Minimum injection volume was about 1.25nL at present. Additionally, good repeatability of the calibration curves for the hydrocarbons ethanolic solution (the RSDs of all components were below 5.30%, n=5) confirmed its feasibility in quantitative analysis regardless of concentration. These results suggested that it can be an accurate nano-injector for cGC.
Monolithic Integration of Nanorod Arrays on Microfluidic Chips for Fast and Sensitive One-Step Immunoassays

Abstract:Here, we present integrated nanorod arrays on microfluidic chips for fast and sensitive flow-through immunoassays of physiologically relevant macromolecules. Dense arrays of Au nanorods are easily fabricated through one-step oblique angle deposition, which eliminates the requirement of advanced lithography methods. We report the utility of this plasmonic structure to improve the detection limit of the cardiac troponin I (cTnI) assay by over 6 × 10 5 -fold, reaching down to 33.9 fg mL −1 (~1.4 fM), compared with an identical assay on glass substrates. Through monolithic integration with microfluidic elements, the device enables a flow-through assay for quantitative detection of cTnI in the serum with a detection sensitivity of 6.9 pg mL −1 (~0.3 pM) in <6 min, which was 4000 times lower than conventional glass devices. This ultrasensitive detection arises from the large surface area for antibody conjugation and metal-enhanced fluorescent signals through plasmonic nanostructures. Moreover, due to the parallel arrangement of flow paths, simultaneous detection of multiple cancer biomarkers, including prostate-specific antigen and carcinoembryonic antigen, has been fulfilled with increased signal-to-background ratios. Given the high performance of this assay, together with its simple fabrication process that is compatible with standard mass manufacturing techniques, we expect that the prepared integrated nanorod device can bring on-site point-of-care diagnosis closer to reality.

Abstract:An immunoassay readout method based on fluorescent imaging analysis with laser confocal scanning is described. The ZnS-coated CdSe quantum dots (ZnS/CdSe QDs) were linked to a detection antibody. Immunoassay was carried out on a glass chip using a sandwich assay approach, where antibody covalently bound to a glass chip was allowed to capture antigen specially. Afterwards, the detection antibody labeled with QD was allowed to bind selectively to the captured antigen. The fluorescent signals of the sandwich conjugate were detected by a laser confocal scanner. A diode laser was used to excite efficiently the fluorescent signals while bovine serum albumin was used to eliminate nonspecific binding sites. The detection limit of this approach was up to 10−9 M under current experimental conditions. The specificity of the QDs-labeled immunoglobulin (IgG) was tested by an experiment using goat IgG and human IgG samples. The result was consistent with the binding specificity in a sandwich-type assay. The potential of this method to function as a simple and efficient readout strategy for immunoassay in biochip is discussed.

Abstract:In recent years, inkjet printing, as a new method to fabricate microdroplet microarrays, has been increasingly applied in the field of biochemical diagnostics. To further improve the general applicability of the inkjet printing technology in fabricating biochemical chips, in this work, we introduce a model to describe the multiple injection procedure implemented by the inkjet printing approach, with experimental verification. The multiple injection model demonstrates a new sequential inkjet printing method that generates picoliter-scale multicomponent droplet-in-oil arrays via multistep printing on uniform planar substrates. Based on our previous work on double-inkjet printing, this technique adapts the piezoelectric inkjet printing technology to fabricate an oil droplet array, into which multiple precise injections of secondary droplets with different compositions and volumes can be automatically printed in the required sequence, simultaneously addressing the evaporation issues associated with printing picoliter droplets without external assistance. In this paper, we first describe the theory and characterize the model, which account for the basic principles of sequential inkjet printing, as well as validate the design in terms of multiple injections, droplet fusion, and rapid mixing. The feasibility and effectiveness of the method are also demonstrated in a dual fluorescence assay and a beta-galactosidase enzyme inhibition assay. We believe that applying the sequential inkjet printing methodology in existing inkjet printing devices will enhance their use as universal diagnostic tools as well as accelerate the adoption of inkjet printing in multistep screening experiments.
A Low-Cost, Disposable and Portable Inkjet-Printed Biochip for the Developing World

Abstract:Electrowetting on dielectric-based digital microfluidic platforms (EWOD-DMF) have a potential to impact point-of-care diagnostics. Conventionally, EWOD-DMF platforms are manufactured in cleanrooms by expert technicians using costly and time consuming micro-nanofabrication processes such as optical lithography, depositions and etching. However, such high-end microfabrication facilities are extremely challenging to establish in resource-poor and low-income countries, due to their high capital investment and operating costs. This makes the fabrication of EWOD-DMF platforms extremely challenging in low-income countries, where such platforms are most needed for many applications such as point-of-care testing applications. To address this challenge, we present a low-cost and simple fabrication procedure for EWOD-DMF electrode arrays, which can be performed anywhere with a commercial office inkjet printer without the need of expensive cleanroom facilities. We demonstrate the utility of our platform to move and mix droplets of different reagents and physiologically conductive buffers, thereby showing its capability to potentially perform a variety of biochemical assays. By combining our low-cost, inkjet-printed EWOD-DMF platform with smartphone imaging technology and a compact control system for droplet manipulation, we also demonstrate a portable and hand-held device which can be programmed to potentially perform a variety of biochemical assays.

Abstract:A lab-on-a-CD immunoassay system was designed, fabricated, and tested. The actuator for the centrifugal platform consists of a flyball governor and a group of spring plungers. The microfluidic structures were fabricated using replica processes with poly-dimethylsiloxane (PDMS) as structural material. For better control, the valves in the microfluidic system were designed to be mechanical-pinch ones and actuated. The burst frequencies of the micro-valves can be tuned independently by adjusting the heights and prestresses of corresponding spring plungers. For the research work reported in this paper, four sequential valves and one inward pump were integrated on a centrifugal microfluidic system to develop an automated immunoassay system. Mouse IgG was used as a model analyte to test the feasibility of using the proposed device for immunoassay applications. Sample solution of goat anti-mouse IgG conjugated with magnetic nanoparticles was used for magnetic trapping mechanism to eliminate the blocking process. Rabbit anti-mouse IgG labeled with fluorescent molecules was used as the signal reporter. The entire microfluidic assay process, including beads trapping, immunoassay reaction, washing and pumping was completed in a centrifugal platform. The detection sensitivity may also be improved by using higher quality monoclonal antibodies. This new immunoassay platform offers some significant advantages over those reported in the field: simple fabrication, robust control, easy integration, and lower cost. The system may also be modified for detection of multiple targets. The technology has great potential for applications in complex fluid handling needed in biological, chemical and medical application in Lab-on-a-CD systems. (C) 2017 Elsevier B.V. Abstract:Microreactors are of great importance for chemical reaction screening, nanoparticle synthesis, protein crystallization, DNA detection, organic synthesis, etc. Here, we reported an effective, flexible, and low-cost method for fabricating microreactor arrays by inkjet printing technology. This strategy utilizes the controllable sliding behavior of the three-phase contact line to form hydrophilic-hydrophobic micropatterns for microreactors with sizes low to several hundreds of nanometers. Reactions in the order of 1 × 10-21 mol molecules can be realized in these microreactors, and crystallization processes can also be conducted to synthesize single crystals.

Abstract:On-chip valves can simplify a microfluidic chip and make it easy to operate. However, most on-chip valves already reported still need complicated manufacture and sophisticated supporting devices. In this work, we present a straightforward on-chip valve, which can be serially connected, to form an on-chip pump. The liquid can horizontally flow one way by the regular deformations of flexure strips in the two valves at both sides of the chamber under pressure changes in microchannels generated by repeated vertical movements of linear actuators. The volume of this system including the chip and the supporting device is 0.65 cubic decimeters, which is much smaller than that of reported systems with a volume of at least 12 cubic decimeters, and the weight of this system is only 0.56 kg, making it possible for point-of-care testing. We carry out an immunoassay of folic acid on chip, and the results show satisfactory reproducibility with acceptable coefficients of variation. We determine 163 clinical human serum samples for folic acid. Furthermore, we detect transferrin, cobalamin and folic acid simultaneously on one chip with both sandwich and competitive binding immunoassay methods. We anticipate that this on-chip valve and pump can be applied in immunoassays and other biosensing applications.

Abstract:A novel, highly accurate sample injection system for capillary electrophoresis (CE) was developed based on an inkjet microchip capable of reproducing exact introduction volumes at the picoliter level. The difficulty in analyte discrimination using electrokinetic injection was also overcome using this injection method. The injection system consisted of an XY stage, an inkjet droplet ejection microchip, and a reservoir with a plug-in septum. To evaluate the precision of the system, a mixture of NBD-labeled amino acids consisting of Gly, L-Phe, L-Asp, and L-Ser was separated, and the performance was compared with that of traditional hydrodynamic and electrokinetic injection methods. The results demonstrated that the introduced volume highly relied on the number of droplets with low relative standard derivation (RSD) and good linear correction coefficient in the proposed injection method. In addition, a urine sample was analyzed via CE coupled with the inkjet injection system for the detection of the amino acid taurine. The concentration of urinary taurine was determined to be 2.42 +/- 0.08 mu M (confidence level, 95%; RSD, 1.05%; n = 4) with a recovery of 98.92-109.54% (n = 3). These results demonstrate the inkjet injection system we developed has the potential to revolutionize capillary electrophoretic separation in practical and commercial applications that require an automated accurate injection system.

Abstract:Highly sensitive detection of proteins offers the possibility of early and rapid diagnosis of various diseases. Microchip-based immunoassay integrates the benefits from both immunoassays (high specificity of target sample) and microfluidics (fast analysis and low sample consumption). However, direct capture of proteins on bare microchannel surface suffers from low sensitivity due to the low capacity of microsystem. In this study, we demonstrated a microchip-based heterogeneous immunoassay using functionalized SiO2 nanoparticles which were covalently assembled on the surface of microchannels via a liquid-phase deposition technique. The formation of covalent bonds between SiO2 nanoparticles and polydimethylsiloxane substrate offered sufficient stability of the microfluidic surface, and furthermore, substantially enhanced the protein capturing capability, mainly due to the increased surface-area-to-volume ratio. IgG antigen and FITC-labeled anti-IgG antibody conjugates were adopted to compare protein-enrichment effect, and the fluorescence signals were increased by ∼75-fold after introduction of functionalized SiO2 nanoparticles film. Finally, a proof-of-concept experiment was performed by highly efficient capture and detection of inactivated H1N1 influenza virus using a microfluidic chip comprising highly ordered SiO2 nanoparticles coated micropillars array. The detection limit of H1N1 virus antigen was 0.5 ng mL−1, with a linear range from 20 to 1,000 ng mL−1 and mean coefficient of variance of 4.71 %.
Development of a capillary flow microfluidic biosensor with on-chip reagent delivery using water-soluble nanofibers

Abstract:Although the potential role of microfluidics in point of care diagnostics is widely acknowledged, the practical limitations to their use still limit deployment. Here, we developed a capillary flow microfluidic with on-chip reagent delivery which combines a lateral flow assay with microfluidic technology. The horseradish peroxidase tagged antibody was electrospun in a water-soluble polyvinylpyrrolidone nanofibers and stored in a microfluidic poly(methyl methacrylate) chip. During the assay, the sample containing on immunomagnetic beads came in contact with the nanofibers causing them to dissolve and release the reagents for binding. Following hybridization, the solution moved by capillary flow toward a detection zone where the analyte was quantified using chemiluminescence. The limit of detection was found to be approximately 10 CFU/mL of . More importantly, the ability to store sensitive reagents within a microfluidic as nanofibers was demonstrated. The fibers showed almost instant hydration and dissemination within the sample solution.
Inkjet-printed paperfluidic immuno-chemical sensing device

Abstract:This paper reports on an inkjet printing method for the fabrication of lateral flow immunochromatographic devices made from a single piece of filter paper by patterning microfluidic channels and dispensing immunosensing inks, requiring only a single printing apparatus. This "paperfluidic" immunosensing device allows for a less time-consuming and more low-cost fabrication compared with the conventional immunochromatographic strips requiring multiple pads, plastic or nylon backing, and a plastic case. A sandwich immunoreaction was performed on the patterned immunosensing paper device, and the sensitivity of the device was optimized with an IgG model analyte. Inkjet-printed antibodies on the test line and the control line were immobilized by physical adsorption, resulting in a very simple fabrication method applicable for pure cellulose surfaces. The color intensity in the test line and the control line was determined both by naked eye and by means of a color scanner in combination with a simple computer program. With the resulting paperfluidic immunosensing device, human IgG concentrations at least down to 10 μg/l could be detected within 20 min. Additionally, in order to demonstrate the feasibility of a total multianalyte sensing system, a combined immuno-chemical sensing device was also fabricated by patterning an additional microfluidic channel for a chemical assay onto the same paper substrate. This low-cost multianalyte paperfluidic sensing device thus demonstrates the feasibility of simple, portable, and disposable tools for pathogen detection in the field of medical, environmental, and food analyses, possibly resulting in useful devices in remote settings and less-industrialized countries.
Integrated and Diffusion-Based Micro-Injectors for Open Access Cell Assays.

Abstract:Currently, most microfluidic devices are fabricated with embedded micro-channels and other elements in a close form with outward connections. Although much functionality has been demonstrated and a large number of applications have been developed, they are not easy for routine operation in biology laboratories where most in vitro cell processing still relies on the use of culture dishes, glass slides, multi-well plates, tubes, pipettes, etc. We report here an open access device which consists of an array of isolated micro-channels plated on a large culture surface, each of them having tiny nozzles for localized drug delivery. In a diffusion dominant regime, steady gradients of molecule concentration could be obtained and varied by changing the flow rate inside the micro-channels. As assay examples, cell staining and drug-induced cell apoptosis were demonstrated, showing fast cell responses in close proximity of the nozzles.

Abstract:This study describes a method to investigate the separation of cells by capillary electrophoresis (CE) coupled with inkjet printing system. The results validated the feasibility of inkjet printing for mammalian cells to achieve the drop-on-demand and convenient sampling into capillary then zone electrophoresis was applied to separate different cells according to their electrophoretic mobility, finally the peak signal were measured by UV detector. Linear relationship between the peak area and the droplet number was obtained within the range of 25-400 drops (R2 = 0.996) at a fixed cell concentration 106/mL, indicating that this system could be used for rapid and accurate quantification of cells.

Abstract:Microfluidics has emerged as a versatile technology that is applied to enhance the performance of analytical techniques, among others. Pursuing this, we present a capillary-driven microfluidic device that improves the sensitivity of lateral flow immunoassay rapid tests thanks to offering an automated washing step. A novel multilevel microfluidic chip was 3D-printed with a photocurable black resin, sealed by an optically clear pressure-sensitive adhesive, and linked to the lateral flow strip. To depict the efficacy of microfluidics and the washing step, cortisol was measured quantitatively within the proposed device. Measuring cortisol levels is a way to capture physiological stress responses. Among biofluids, saliva is less infectious and easier to sample than others. However, higher sensitivity is demanded because the salivary cortisol concentrations are much lower than in blood. We carried out a competitive lateral flow immunoassay protocol with the difference that the microfluidic device applies an automated washing step after the sample is drained downstream. It washes the trapped quantum-dot-labeled antibodies out from nitrocellulose, diminishing background noise as these are bonded to cortisols and not to the immobilized receptors. Fluorescence spectroscopy, as a high-precision analysis, was successfully applied to determine clinically relevant salivary cortisol concentrations within a buffer quantitatively. The microfluidic design relied on a 3D valve that avoids reagent cross-contamination. This cross-contamination could make the washing buffer impure and undesirably dilute the sample. The proposed device is cost-effective, self-powered, robust, and ideal for non-expert users.

A nanoinjector for microanalysis

Abstract:We describe a simple miniature injection device that can be used for introduction of nanoliter sample volumes in microfluidic systems. The hybrid microstructure consists of two hydraulically connected parts, a pulse micropump, and a multilevel cross-flow injector. Sample injection is accomplished by creating a transient pressure pulse in the sample line by means of the solenoid-based micropump. The sample line is aligned at right angles to the main carrier flow line. The two flow channels are located in two different parallel planes. The cross section of the two channels is defined by a self-sealing aperture in an elastomer. During the pressure pulse, the sample is introduced through this aperture directly into the main flow stream. Fast impulse-based injection causes rapid mixing of the injected sample with the main flow stream. This permits simple single-line manifold micro flow injection (MFI) systems. The deformation/relaxation of the elastomer is fast and repeatable; as such, rapid serial actuations essentially result in a larger injected sample volume without significantly affecting the peak shape. In the present form, 2-40-nL samples are easily injected by single injection, and the injected volume can be chosen by system parameters. The injection repeatability as observed by a photometric detector is better than 1.2% (n = 100).

Abstract:Developing rapid and inexpensive diagnostic tools for molecular detection has been pushed forward by the advancements of technical aspects. However, attention has rarely been paid to the molecular detection methodology using inkjet printing technique. Herein, we developed an approach that employed a self-assembled inkjet printer as the enabling technology to realize droplet digital loop-mediated isothermal amplification in a low-cost and practical format. An inkjet printer is a self-assembled tool for the generation of discrete droplets in controllable volumes from a picoliter to a nanoliter. A microfluidic chip serves as a droplets reservoir to perform droplet digital LAMP assays. The inkjet printer approach successfully quantified the HPV16 from CaSki cells. This self-assembled and practical inkjet printer device may therefore become a promising tool for rapid molecular detection and can be extended to on-site analysis.
