Abstract:We report the development of an ultrasensitive amperometric biosensor based on Ag nanoparticles-decorated graphene oxide nanosheets (GO) (Ag-GO) for the rapid detection of clenbuterol (CLB). The morphology and structure of the Ag-GO labeled CLB (Ag-GO-CLB) were characterized by transmission electron microscope (TEM), atomic force microscope (AFM), and ultraviolet-visible spectroscope (UV-vis). The immunosensor was prepared by covalently immobilizing capture antibodies on a multi-walled carbon nanotubes-modified glassy carbon electrode. Through competitive immunoreactions, the Ag-GO-CLB nanocomposites were captured on the immunosensor and the silver was measured by positive differential pulse voltammetry (DPV) in KCl solution for the detection of antigen. The experimental results show a linear response over the range from 0.01 to 10.0 ng mL(-1) with a lower detection limit of 6.8 pg mL(-1) (signal-to-noise ratio of 3). The Ag-GO based immunosensor offers a simple and convenient route for metal-immunoassay labels, which can avoid the complicated and time-consuming dissolving of metal component for ultrasensitive determination. Moreover, the electrochemical immunoassay shows acceptable specificity and stability and is suitable for the determination of CLB in real samples.
Flow Injection Chemiluminescent Competitive Immunoassay for the Β-Adrenergic Agonist Salbutamol Using Carboxylic Resin Beads and Enzymatic Amplification

Abstract:A simple, rapid and sensitive competitive immunoassay for the p-adrenergic agonist salbutamol (SAL) was developed with flow injection chemiluminescence (FI-CL) detection. The method was based on immobilizing coating-antigen on carboxylic resin beads and introducing the second antibody tagged with horseradish peroxidase (HRP) into the CL reaction of luminol-PIP-H2O2. The carboxylic resin beads were creatively applied for amplifying signals by carrying more coating-antigen through the chemical bonds between functional groups. With a competitive format, the salbutamol in solution competed with the coating-antigen for the limited binding sites of the antibody. Then the second antibody labeled with HRP was simply immobilized and an enzyme-enhanced CL emission was obtained due to the catalysis of HRP for the CL reaction. Under the optimal conditions, the CL intensity was linearly related to the concentration of salbutamol in the range of 0.5-100 ng mL(-1) with a detection limit (3 sigma) of 0.15 ngmL(-1). This immunosensor exhibited favorable stability, specificity and reproducibility, and was applied to practical samples with satisfactory results. Molecularly Imprinted Polymers As the Extracted Sorbents of Clenbuterol Ahead of Liquid Chromatographic Determination.

Abstract:A pre-treatment methodology for clenbuterol hydrochloride (CLEN) isolation and enrichment in a complex matrix environment was developed through exploiting molecularly imprinted polymers (MIPs). CLEN-imprinted polymers were synthesized by the combined use of ally-β-cyclodextrin (ally-β-CD) and methacrylic acid (MAA), allyl-β-CD and acrylonitrile (AN), and allyl-β-CD and methyl methacrylate (MMA) as the binary functional monomers. MAA-linked allyl-β-CD MIPs (M-MAA) were characterized by Fourier transform-infrared (FT-IR) spectroscopy and a scanning electron microscope (SEM). Based upon the results, M-MAA polymers generally proved to be an excellent selective extraction compared to its references: AN-linked allyl-β-CD MIPs (M-AN) and MMA-linked allyl-β-CD MIPs (M-MMA). M-MAA polymers were eventually chosen to run through a molecularly imprinted solid-phase extraction (MISPE) micro-column to enrich CLEN residues spiked in pig livers. A high recovery was achieved, ranging from 91.03% to 96.76% with relative standard deviation (RSD) ≤4.45%.

Abstract:This study described the use of a rapid and sensitive flow injection chemiluminescence immunoassay based on the specific binding of antigens and antibodies for the detection of brombuterol residues in the swine meat and feed samples. Carboxylic resin beads were used as a solid-phase carrier for supporting more coating antigens through acylamide bonds. Under the mechanism of competitive immunity, the brombuterol in solution competed with the coating antigen immobilized on the resin beads for the limited binding sites of antibody. Then, horseradish peroxidase-labeled secondary antibodies were introduced into the immunosensor to combine with polyclonal antibodies. The chemiluminescence intensity of the luminol-p-iodophenol-H2O2 system was catalyzed by horseradish peroxidase, and thus, signal amplification was achieved. Under the optimal conditions of the concentration of coating antigen, antibody and enzymatic secondary antibody, brombuterol could be detected quantitatively. The chemiluminescence intensity decreased linearly with the logarithm of the brombuterol concentration in the range of 0.001 to 300 ng mL−1. The limit of detection (3σ) and the limit of quantitation were 0.33 pg mL−1 and 1.1 pg mL−1, respectively. This method had undergone a series of test conditions and exhibited good specificity, stability, and reproducibility. The immunosensor can also detect real samples and provide prospects for the detection of other small molecule compounds, such as isoproterenol, enrofloxacin, and ofloxacin.
An Automated Immunoassay Based on Magnetic Microspheres for the Determination of Clenbuterol in Food

Abstract:An automated magnetic immunoassay for the determination of clenbuterol in food is reported. Magnetic microspheres were employed as the solid phase for binding anti-clenbuterol antibodies. Fe3O4 microspheres (506.9nm) were synthesized from FeCl3 and ethylene glycol. The magnetic saturation value was 72.4emu g(-1) for the microspheres. The magnetic microspheres were subsequently surface modified using tetraethyl orthosilicate. Carboxylate groups were produced on the surface of the microspheres and the anti-clenbuterol antibodies were covalently attached using 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide and N-hydroxysuccinimide. The incubation, washing, and detection procedures were automated. The limits of detection and quantification were 0.02ng mL(-1) and 0.1ng mL(-1), respectively. Recoveries of clenbuterol in sausage, pork, and beef were 93.5% to 110.0%, 98.9- 109%, and 94.0-101.9%, respectively. This approach allowed analysis in 25min compared to 50min for a commercial method while reducing the reagents required.

Abstract:A novel flow injection chemiluminescence immunoassay for simple, sensitive and low-cost detection of diclofenac was established based on specific binding of antigen and antibody. Carboxylic resin beads used as solid phase carrier materials provided good biocompatibility and large surface-to-volume ratio for modifying more coating antigen. There was a competitive process between the diclofenac in solution and the immobilized coating antigen to react with the limited binding sites of the polyclonal antibody to form the immunocomplex. The second antibody labelled with horseradish peroxidase was introduced into the immunosensor and trapped by captured polyclonal antibody against diclofenac, which could effectively amplify chemiluminescence signals of luminol-PIP-H2O2. Under optimal conditions, the diclofenac could be detected quantitatively. The chemiluminescence intensity decreased linearly with the logarithm of the diclofenac concentration in the range of 0.1-100ngmL-1 with a detection limit of 0.05ngmL-1 at a signal-to-noise ratio of 3. The immunosensor exhibited high sensitivity, specificity and acceptable stability. This easy-operated and cost-effective analytical method could be valuable for the diclofenac determination in real water samples.

Abstract:The determination of clenbuterol is crucial for food safety and sport science. In this work, we use tetraethyl orthosilicate and (3-aminopropyl)triethoxysilane as cross-linking agents and functional monomers for the electrodeposition of SiO2 films with the addition of clenbuterol. After the elution process, the prepared electrode can be used for the sensitive determination of clenbuterol due to the molecular imprinting effect. Under optimum conditions, results show that the current of clenbuterol decreases linearly in the range of 2 μM to 0.1 mM, and the detection limit is 31 nM. The proposed electrochemical sensor also exhibits excellent stability and reproducibility, showing its high potential for use in sensing clenbuterol in food samples.

Abstract:A rapid immunochromatographic lateral flow test strip was developed in a competitive format with the gold-conjugated monoclonal antibody to specifically determine the residues of Clenbuterol (CL), a β-adrenergic agonist. The test strip is made up of a sample pad, a conjugate reagent pad, a test membrane containing a control line and a test line, and an absorbent pad. CL standard samples of 0, 0.1, 0.3, 0.9, 2.7, 8.1 ng/ml in swine urine were determined by the test strip. It was shown that detection limit of the test strip was as low as 0.1 ng/ml of CL and that the half of maximal inhibition concentration (IC50) in relative optical density was calculated to be 1.78±0.17 ng/ml under an optical density scanner. The sensitivity by eye measurement was 1.0 ng/ml. It takes 10 min to accomplish a test. Parallel analysis of urine samples from pigs fed with CL showed comparable results obtained from the test strip and GC-MS. Therefore, the test strip is very useful as a screening method for quantitative, semi-quantitative or qualitative detection of CL residues in swine urine.

Abstract:Objective A novel technology of microcantilever sensor platform for label-free detection of clenbuterol antigen-antibody specificity binding is reported. Method Thiolated clenbuterol antibody was immobilized on the gold-coated side of the microcantilever by self-assembled monolayer technique.Consequently the deflections of the microcantilever corresponding to the process of injecting different concentration of clenbuterol antigen were real-time monitored by optical lever technique.The activity of thiolated clenbuterol antibody and the clenbuterol antigen-antibody binding on the gold-coated side of the microcantilever were confirmed by an Enzyme-Linked Immunosorbent Assay(ELISA). Result The sensitivity of detecting clenbuterol is better than 1 μg/L.Microcantilevers functionalized with thiolated clenbuterol antibodies were exposed to 1 mg/L of Chloramphenicol(CAP),and no significant response was detected,which confirmed the specificity of the immobilization method.In addition,clenbuterol antigen-antibody binding on the microcantilever generates a compressive surface stress and the surface stress is linear to logarithm of clenbuterol concentration over a range of concentration from 1ng/mL to 1 μg/mL. Conclusions It is possible for detection of clenbuterol by microcantilever sensor platform.

Abstract:A dual signal amplification immunosensor based on glucose oxidase（GOD）-functionalized nanocomposites was developed for the detection of clenbuterol.The characteristics of the nanocomposites were confirmed by scanning electron microscopy（SEM） and spectrophotometry.The results indicate that GOD in the nanocomposites keeps good enzyme dynamic response and electro-catalysis performance.The detection mechanism of the immunosensor was investigated by electrochemical methods of differential pulse voltammetry（DPV）.A set of experimental conditions were also optimized for the detection of clenbuterol.The developed immunosensor exhibited high sensitivity,selectivity and reproducibility,which had a wide linear range of 0.01～100 ng/mL and a low detection limit of 4.50 pg/mL for clenbuterol.Accurate detection of clenbuterol in pig liver samples was demonstrated by standard addition method.

Abstract:A simple and ultrasensitive flow injection chemiluminescence competitive immunoassay based on gold nanoparticle-loaded enzyme for the detection of chloramphenicol (CAP) residues in shrimp and honey has been developed. Due to their good biocompatibility and large specific surface area, carboxylic resin beads can be used as solid phase carriers to immobilize more coating antigens (Ag). In addition, gold nanoparticles could provide an effective matrix for loading more CAP antibody and horseradish peroxidase, which would effectively catalyze the system of luminol-p-iodophenol (PIP)-H2O2. A competitive immunoassay strategy was used for detection of CAP, in which CAP in the sample would compete with the coating Ag for the limited antibodies, leading to a chemiluminescence (CL) signal decrease with increase in CAP concentration. A wide linear range 0.001-10 ng ml(-1)(R-2= 0.9961) was obtained under optimized conditions, and the detection limit (3 sigma) was calculated to be 0.33 pg ml(-1). This method was also been successfully applied to determine CAP in shrimp and honey samples. The immunosensor proposed in this study not only has the advantages of high sensitivity, wider linear range, and satisfactory stability, but also expands the application of flow injection CL immunoassay in antibiotic detection.

Abstract:A novel immunoaffinity column used as an immunosensor for flow-injection chemiluminescent (CL) immunoassay was prepared by immobilizing antibody on carboxylic resin beads. The immunosensor could fast recognize and trap the immunocomplex of horseradish peroxidase (HRP)-labeled antibody and antigen, which was firstly formed with a micro-bubble accelerated pre-incubation process, to produce a sandwich immunocomplex. The HRP introduced in the immunoaffinity column could catalyze the CL reaction to produce enzyme-enhanced emission. With α-fetoprotein (AFP) as a mode, a flow-injection CL immunoassay was proposed. The whole assay for one sample, including the pre-incubation and the regeneration of immunoaffinity column, could be performed within 16min. The linear range was 1.0–80ng/ml with a correlation coefficient of 0.998 and a detection limit of 0.1ng/ml at a signal/noise ratio of 3. The intra- and inter-assay coefficients of variation at 20ng/ml AFP were 1.2% and 8.5%, respectively. The storage stability of the immunoaffinity column and the accuracy for sample detection were acceptable. This flexible, sensitive, low-cost, and rapid method is valuable for clinical immunoassay.

Abstract:We report a highly sensitive competitive-type assay for multiplexing electrochemical detection of clenbuterol in pig urine using a 16-sensor array. In this design, the clenbuterol was first conjugated with bovine serum albumin (BSA), and then the conjugates were immobilized on the electrode surface to compete with the free clenbuterol in the sample for specific antibody. Under the optimal conditions, the reproducibility of the clenbuterol electrochemical immunosensor was evaluated to be 3.4% (coefficient of variation, CV) and the limit of detection was estimated to be 1.3pg/mL. The very low detection limit was probably derived from the higher efficiency of the competitive immunoreaction caused by appropriate quantities of the clenbuterol immobilized on the 16-sensor array and the suitable amount of anti-clenbuterol antibody in the assay system.

Abstract:A label-free fluorescent biosensor based on clenbuterol binding aptamer and the fluorescent intercalator SYBR Green I (SGI) was established for the sensitive and selective detection of clenbuterol. In the absence of clenbuterol, a weak fluorescence signal of SGI was observed. In the presence of clenbuterol, the specific recognition and high-affinity binding of aptamer with clenbuterol induced a conformational change of the aptamer, longer double-stranded DNA was formed. As a result, an enhancement fluorescence of SGI could be found. Under optimal conditions, the fluorescence change ratio increased linearly with clenbuterol concentration from 50 nM to 450 nM, with a detection limit of 18.8 nM. In addition, this proposed aptasensor showed good selectivity and was successfully applied for clenbuterol quantification in pork samples with recoveries of 90.37-104.98%.

Abstract:A multiple amplified electrochemiluminescence immunosensor is described for the determination of the illicit β-adrenergic agonist brombuterol. Firstly, cystein-coated silver nanowires (SCNW) were modified with polyamidoamine dendrimers (PAMAM) and then immobilized on a glassy carbon electrode (GCE). Then, Ag@Au core/shell nanoparticles (NPs) were deposited on the surface of the modified GCE via gold-nitrogen bonds. The use of Cys-coated silver nanowires accelerates the electron transfer process and also prevents the aggregation of the Ag@Au NPs. The nanocomposites on the GCE have a large surface and outstanding electrical conductivity. It can carry a large amount of coating antigen to amplify the ECL signal. Moreover, PAMAM-Au-CdSe quantum dot bioprobes were prepared, and the unique dendrimer-encapsulated gold nanoparticles with numerous functional amino groups were employed to load abundant activated CdSe quantum dots for further enhancement of the ECL signal. On the basis of signal amplification of the SCNW-PAMAM-Ag@Au-based immunosensor and the PAMAM-Au-CdSe quantum dot bioprobes. The assay has a wide linear range that extends over the 0.001–1000 ng·mL−1 brombuterol concentration range and a lower detection limit of 37 fg·mL−1. The method is well reproducible, stable and specific. It was applied to the determination of brombuterol in spiked samples of pork and feed where it gave satisfactory recoveries.

Abstract:A portable fluorescence biosensor with rapid and ultrasensitive response for Clenbuterol (CL) has been built up with fluorescent nanosilica and a lateral flow test strip. Quantitative detection of CL was realized by recording the fluorescence intensity of fluorescent nanosilica captured on the test line. The sensing results indicated that the sensitivity of the fluorescent nanosilica-based strip was better than that of conventional colloidal gold-based strips. The visual limit of detection of the strip for qualitative detection was 0.1ng/mL while the LOD for quantitative detection could down to 0.037ng/mL by using fluorescence biosensor. The recoveries of test samples were from 89.3% to 97.7%. The assay time for CL detection was less than 8min, suitable for rapid testing on-site.

Abstract:In this work, an ultra-sensitive and simple chemiluminescence (CL) immunosensor has been constructed to detect Hg2+ in real water samples. It is based on the excellent properties of oxidized graphene and chitosan modified a novel flow cell to achieve satisfactory results. Oxidized graphene has good biocompatibility, can effectively immobilize antigens and construct an efficient biosensor interface. Chitosan has excellent biodegradability, excellent film-forming and biocompatibility. Hereby, oxidized graphene-chitosan composites provide a favorable microenvironrnent for immobilizing coating-antigen and introducing HRP-labeled secondary antibodies into the CL reaction of luminol-PIP-H2O2. With a competitive-type assay mode, the Hg2+ in solution competed with the immobilized coating antigen for the limited binding sites of monoclonal antibody, which resulted in the CL of this system decreases because of less HRP. At the optimal conditions, the CL signal displayed a good linear relation toward Hg2+ in the range of 0.001-300 ng mL(-1) with a detection limit (3 sigma) of 0.33 pg mL(-1). The immunosensor possessed acceptable accuracy, high specificity and reproducibility, and was applied to examine Hg2+ in real water samples and cream with favorable results. The successful development of this method also provides reliable prospects and technical support for the follow-up detection of similar environmental pollutants.
An Electrochemical Biosensor for Clenbuterol Detection and Pharmacokinetics Investigation.

Abstract:Clenbuterol is a member of β2 adrenergic agonists, which is widely used not only as a food additive for livestocks, but also a kind of stimulant for athletes; however, the abuse of clenbuterol may pose a significant negative impact on human health. Since it is highly required to develop fast, sensitive and cost-effective method to determine clenbuterol level in the suspected urine or blood, we herein have fabricated an electrochemical biosensor for the determination of clenbuterol. Measurement of the species with the proposed biosensor can also have the advantages of simplicity, high sensitivity and selectivity. Moreover, the sensor can be directly used for clenbuterol determination in rat urine. We have further studied the pharmacokinetics of clenbuterol by using this proposed electrochemical biosensor, so a new tool to investigate pharmacokinetic is developed in this work.
