Abstract:Several new 6-(1-aryl-5-methyl-1, 2, 3-triazol-4-yl)-3-(4-pyridyl)-s-triazolo[3,4-b]-1, 3, 4-thiadiazoles 3a-j have been synthesized by the condensation of 4-amino-5-mercapto-3-(4-pyridyl)-1, 2, 4-triazole 2 with 1-aryl-4-carboxy-5-methyl-1, 2, 3-triazoles 1a-j in the presence of phosphorus oxychloride. The structures of the products obtained are characterized by elemental analyses and spectral data and the fragmentation pattern of the mass spectra of 3a-j is discussed. 3a-j have been screened for their antibacterial activity against B. subtilis and E. coli.

Abstract:5-acetylamino-1,3,4-thiadiazole-2-sulfonyl chloride compound has been synthesized using 5-amino-2-mercapto-1,3,4-thiadiazole, and the compound were been acyltion and oxidation by gaseous chlorine. The target produces is 5-acetylamino-1,3,4-thiadiazole-(2-N- methyl piperazine)sulfonylanilin and 5-acetylamino-1,3,4-thiadiazole-(2-sulfonyl)- aniline. The structures of the target compound were confirmed by melting pointing, IR, 1H-NMR、and LC-MS elementary analysis.

Abstract:A series of acylhydrazone compounds 3a-i and acylthioscmicarbazides 4a-h containing thiadiazole rings have been synthesized conveniently in high yields. Some heterocyclic derivatives 5 are prepared by the method of dehydration of compounds 4. Structure of all these compounds have been confirmed by elemental analysis, 1R, 1 H and 13 C NMR. The bioassay result indicates that some compounds have relatively low antibacterial activity and other compounds have bioactivity for improving plant growth.

Abstract:In view of various biological activities of benzoxazolinone and thiadiazole derivatives, the preparation of 1,3,4-thiadiazole derivatives containing benzoxazolinone moiety is of great significance. In this paper, ten novel 2- arylformylamino-5-(2-benzoxazolinone-3-yl) methyl-1,3,4-thiadiazoles (Ta -7j) were synthesized by 2-aminophenol and urea as the starting materials via a series of reactions. The structures of the target compounds were characterized by IR, 1H NMR and elemental analysis.

Abstract:In this paper, we reported the development of an efficient and highly productive synthetic protocol for preparing new 2,5-diamino-1,3,4-thiadiazole derivatives functionalized with N -(2,2,2-trichloroethyl)carboxamide group. The method of their preparation was based on the reaction of oxidative dehydrosulfurization of N -(2,2,2-trichloro-1-(2-(phenylcarbamothioyl)hydrazine-1-carbothioamido)ethyl)carboxamides and N -(1-(2-carbamothioylhydrazine-1-carbothioamido)-2,2,2-trichloroethyl)benzamide under the action of a mixture of iodine and triethylamine in DMF. This reaction took place at room temperature for two hours and allowed obtaining the target products with a 69-75% yield. Using the developed protocol, we obtained ten new derivatives of 1,3,4-thiadiazole. Their structure was proven by IR, 1 H NMR, 13 C NMR, 1 H- 1 H COSY, 1 H- 13 C HSQC, 1 H- 13 C HMBC spectroscopy, and LC/HRMS spectrometry.

Abstract:A series of novel N-1-(5-substituted-1,3,4-thiadiazole-2-yl)-N-3-substituted phenyl-urea derivatives have been synthesized and evaluated for their anti-tumor activity. The effect of the title compounds on tumor metastasis was analyzed by Lewis-lung-carcinoma model. The bioassay showed that some title compounds dramatically reduced the numbers of lung metastasis and depressed the primary tumor growth. The structures of compounds were identified from the elemental analysis, IR and H-1 NMR.

Abstract:A series of new N′-[N-(2,3,4-tri-O-acetyl-β-d-xylopyranosyl)thiocarbamoyl]-2-[(1-aryl-1H-tetrazol-5-yl)sulfanyl]acetohydrazides 5a–5e were synthesized rapidly in high yields from 2-(1-aryl-1H-tetrazol-5-ylsulfanyl)acetohydrazides 3a–3e and 2,3,4-tri-O-acetyl-β-d-xylopyranosyl isothiocyanate 4, then 5a–5e were converted to a series of new 5-(1-aryl-1H-tetrazol-5-ylsulfanylmethyl)-N-(2,3,4-tri-O-acetyl-β-d-xylopyranosyl)-1,3,4-oxadiazole-2-amines 6a–6e and 5-(1-aryl-1H-tetrazol-5-ylsulfanylmethyl)-N-(2,3,4-tri-O-acetyl-β-d-xylopyranosyl)-1,3,4-thiadiazole-2-amines 7a–7e, respectively under mercuric acetate/alcohol system or acetic anhydride/phosphoric acid system, then deacetylated in the solution of CH3ONa/CH3OH. All of the novel compounds were characterized by IR, 1H NMR, 13C NMR, MS and elemental analysis. The structures of compounds 2e, 3e, 5a and 5c have been determined by X-ray diffraction analysis. Some of the synthesized compounds displayed PTP1B inhibition and microorganism inhibition.

Abstract:Six new 2-alkylthio-5-(3,4,5-trimethoxyphenyl)-1,3,4-thiadiazole derivatives were synthesized from gallic acid by six steps: etherification, esterification, hydrazidation, salt formation, cyclization, and thioetherification. The synthesis of the title compounds catalyzed by indium in aqueous media was easy in process with high yield and environmental-friendliness. The products were characterized by elemental analysis, IR, H-1 NMR and C-13 NMR spectra. The crystal structure of compound 8a was determined by X-ray diffraction analysis. 8a belongs to the monoclinic system with space group C2/c and cell dimensions of a = 1.5730(19) nm, b=0.5477 nm, c=4.2244 nm, beta=92.015(6)degrees, V=3.6370(2) nm(3), Z=8, D-c=1.497 g/cm(3), mu=0.463 mm(-1), F(000)=1696, R-1=0.0783, wR(2)=0.1832. It was found that the title compounds 8 possess good antitumor activity to PC3 and BGC-823 cells in vitro by MTT method. For example, the antiproliferation activity of compound 8e to PC3 cells at the concentration of 5 mu mol center dot L-1 at 48 h was 55.71% and the antiproliferation activity of compound 8b to BGC-823 cells at the concentration of 5 mu mol center dot L-1 at 48 h was 66.21%.

Abstract:Thirteen title compounds (2) were obtained by nucleophilic addition of benzyl carbanion of 2-aroylamino-5-benzyl-1,3,4-thiadiazoles(1) with anisaldehyde in EtONa/EtOH solution. The structures of the compounds 2 were confirmed by elemental analysis, IR,1H NMR,and MS. The compounds were found to have some fungicidal activity.

Abstract:In order to investigate the biological activity of novel thiourea compounds, some novel 1,2,3-thiadiazole derivatives containing 1,3,4-thiadiazole were synthesized under phase transfer catalyzed condition (PEG-600) by multi-step reactions. The chemical structures of all compounds were established by H-1 NMR, FTIR, MS, and elemental analysis, and some of these compounds were investigated for fungicidal activity and plant growth regulatory activity. The bioassay results indicated that some of these compound exhibited moderate activities.

Abstract:The annelation of 1,2,3-thiadiazole rings was accomplished by the reaction of N-acylhydrazone 2a bearing an adjacent α-methyl with thionyl chloride to give α-chloro-N-methyl-1,2,3-thiadiazole-4-acetamide 4 and was demonstrated by the X-ray crystal structure of its derivative 5a. A novel series of α-substituted phenoxy-N-methyl-1,2,3-thiadiazole-4-acetamide 5 were synthesized through the reaction of the compound 4 and phenols. The results of bioassays show that the title compounds exhibit good anti-HBV activities. The crystal of compound 5a, N-methyl-α-2-bromophenyl-1,2,3-thiadiazole-4-acetamide, has been prepared and determined by X-ray diffraction.

Abstract:A facile one-pot synthesis of 2,5-disubstituted-1,3,4-thiadiazoles was achieved by ultrasonic irradiation and phase transfer catalysis for the first time and eight novel 2,5-disubstituted-1,3,4-thiadiazoles were synthesized. This method has the advantages of mild conditions, short time, good yields, convenience, economy and environmental friendliness. The structures of the target molecules were characterized by IR, H-1 NMR,C-13 NMR, 1D NOE, MS spectra and elemental analysis. The experiments of the biological activity for the target molecules were accomplished. The experimental results of the inhibiting activity against COX-2 indicated that the compound 5f had the highest inhibition rate, up to 95.59%, and yet the compound 5g exhibited weakly inhibiting activity. The target compounds did not possess anticonvulasional activity.

Abstract:Some new sulfoxide compounds containing 1,2,3-thiadiazole moiety were synthesized from diethyl carbonate and hydrazine hydrate by multi-step reactions. Their structures were confirmed by NMR, MS, and elemental analysis. One of the title compounds, 5-(4-cyclopropyl-5-((3-fluorobenzyl)sulfinyl)-4H-1,2,4-triazol-3-yl)-4-methyl-1,2,3-thiadiazole (C15H14FN5OS2), was structurally determined by a single crystal X-ray diffraction study. The biological activity of the title compound was determined, and the results showed that it displays moderate biological activities.

Abstract:Fifteen novel title compounds were synthesized from 3-(4-methyl-1,2,3-thiadiazolyl)-4-amino-1,2,4-triazolo-5-thione with substituted benzaldehyde in acetic acid under refluxing.The structures of the compounds were confirmed by elemental analysis,IR and 1H NMR.The single crystal of compound 5c was cultured and determined by X-ray single-crystal diffraction as of a monoclinic system,space group P2(1)/c with cell parameters a=1.403 7(3) nm,b=1.570 5(3) nm,c=0.686 4(4) nm,β= 102.06(3)°,V=1.479 8(9) nm3,Z=4,F(000)=656.Bioassaywas conducted by fungi growth inhibition method using 10 commonly used fungi which represented most disease occurring in China,the results showed that,all the title compounds had satisfactory fungicidal activity against Botrytis cinerea,the growth inhibition rate of compound 5l against Botrytis cinerea was up to 87%,for compound 5c,5d and 5f were more than 78%,compound 5a exhibited fungicidal activity against Gibberella zeae with growth inhibition of 78.7%,the growth inhibition rate of compound 5m against Colletotrichum lagenarium was 65.6%.
