Abstract:Inspired by the assembly of Lego toys, hydrogel building blocks with heterogeneous responsiveness are assembled utilizing macroscopic supramolecular recognition as the adhesion force. The Lego hydrogel provides 3D transformation upon pH variation. After disassembly of the building blocks by changing the oxidation state, they can be re-assembled into a completely new shape.

Abstract:DNA supramolecular hydrogels are hydrogels cross-linked through DNA hybridization. They have attracted wide attention due to their great molecular permeability, biocompatibility, degradability, thixotropy, and self-healing properties, which make them very useful in cell culture, tissue engineering, and 3D printing. The designability and responsiveness of DNA endows these hydrogels with specific stimuli responsiveness, thus enhancing their utility for more applications, e.g., detection devices and shape-memory materials. In this review, we outlined and discussed the development of responsive DNA supramolecular hydrogels, and their applications have also been summarized.

Abstract:Stimuli-responsive hydrogels are "smart" materials with diverse applications. We now report short peptide conjugates with merocyanine (MC) that are able to form stimuli-responsive hydrogels. Systematic investigation reveals that merocyanine is a highly effective promoter for the self-assembly of its oligopeptide conjugates. Hydrogels formed by MC-peptide conjugates showed responses towards light and heat, and their sol-gel phase transition could be manipulated by the reverse photochromism of the corresponding spiropyran moiety. Impressively, a MCI-RGD conjugate formed a supramolecular hydrogel with responses to multiple stimuli, including visible light irradiation, pH change and the presence of Ca2+ ions. An erasable photo-lithograph on the MCI-RGD hydrogel was demonstrated using visible light to write and heat-and-cool treatment to erase for multiple rounds without significant loss of sensitivity.

Abstract:A novel pH and oxidation dual-responsive and injectable supramolecular hydrogel was developed, which was formed from multi-block copolymer poly(ether urethane) (PEU) and α-cyclodextrin (α-CD) inclusion complexes (ICs). The PEU copolymer was synthesized through a simple one-pot condensation polymerization of poly(ethylene glycol), di(1-hydroxyethylene) diselenide, dimethylolpropionic acid and 3-isocyanatomethyl-3,5,5-trimethylcyclohexyl isocyanate. In aqueous solution, the amphiphilic PEU copolymers could self-assemble into nanoparticles with dual pH and oxidation sensitivities, which can efficiently load and controllably release a hydrophobic drug indomethacin (IND). Then a dual-drug loaded supramolecular hydrogel was obtained by addition of α-CD and hydrophilic model drug (rhodamine B, RB) into the resulting IND-loaded PEU nanoparticle solution. The rheology studies showed that the supramolecular hydrogels with good injectability underwent a pH-induced reversible sol-gel transition and an oxidation-triggered degradation behavior. The in vitro drug release results demonstrated that the hydrogels showed dual drug release behavior and the release rates could be significantly accelerated by addition of an oxidizing agent (H2O2) or increasing the environmental pH. Therefore, this injectable and dual stimuli-responsive supramolecular hydrogel based codelivery systems could potentially be a promising candidate for controlled drug delivery systems.

Abstract:In this study, we report the formation of a multi-stimuli-responsive supramolecular hydrogel through the self-assembly of an oxidized glutathione derivative. The hydrogels were obtained in aqueous DMF solutions after a heating–cooling process. These hydrogels exhibited fibrous structures, which formed via the self-assembly of the hydrogelator driven by the combination of hydrogen bonding, π–π stacking, and hydrophobic effect. Besides the thermo-sensitivity, the hydrogels showed redox- and light-responsiveness attributing to the presence of a disulfide bond and two azobenzene terminal groups, respectively, in the molecular structure of the hydrogelator. The multi-stimuli-responsiveness endows the hydrogels with the potential applications in the field of smart materials.

Abstract:Controlled drug release has sparked much attention in the field of drug delivery by prolonging the duration of drug effectiveness and reducing drug toxicity. However, the development of controlled-release drug delivery systems remains a significant challenge, primarily due to inadequate drug compatibility, low biocompatibility, and low response sensitivity of the drug carriers. Herein, an innovative strategy for fabricating multifunctional organohydrogels as drug carrier systems is designed. 1-Tetradecanol/water emulsions, stabilized by cellulose acetoacetate (CAA), are initially prepared. The organohydrogels are fabricated by the ternary enzyme-mediated polymerization of the monomer - containing emulsion. The amphipathy of CAA ensures the stability of emulsions during polymerization, and CAA participates in polymerization as an initiator. Through the reversible crystallization-melting transition of 1-tetradecanol, the drug can be encapsulated in/released from the organohydrogel. Moreover, 1-tetradecanol enhances the bioavailability of water-insoluble drugs without any additional cosolvent. Therefore, biofriendly organohydrogels can serve as ideal dressings with adjustable mechanical properties, effective temperature regulation, and temperature-triggered drug release ability.

Abstract:By using a disulfide-functionalized crosslinker, a pH- and thermo-responsive 2-(dimethylamino) ethyl methacrylate (DMAEMA) monomer and a zwitterionic sulfobetaine methacrylate (SBMA) monomer were conjugated to fabricate a multi-responsive P(DMAEMA-SS-SBMA) copolymeric hydrogel. Apparent UCST and LCST volume transitions were observed in the P(DMAEMA-SS-SBMA) hydrogels with equivalent weight fractions of monomers. Different pore size and response sensitivity of shrunken structures below UCST and above LCST were visualized by SEM images. The hydrogel exhibited a highly swollen state with a swelling ratio of 17.8 and a pore size of 106μm at 45°C, they deswelled unequally at 5°C with a compact surface with pore size of 30μm and a loose bulk with pore size of 83μm, while they deswelled uniformly at 65°C with dense shrunken structure with small pore size of 12μm. The dual-thermoresponsive hydrogel was promising in controlled drug release. The initial drug release was predominantly controlled by diffusion, and the long-term release was influenced by the swelling ratio. Below UCST, the relatively hydrophilic shrunken structure and slow diffusion had a synergistic effect on the sustained release. Above LCST, the fast diffusion and the rapid "off" effect of hydrophobic skin layer resulted in a burst release. Additionally, pH-tunable swelling and redox-sensitive degradation were also observed.

Abstract:Supramolecular peptide hydrogels have many important applications in biomedicine, including drug delivery applications for the sustained release of therapeutic molecules. Targeted and selective drug administration is often preferential to systemic drug delivery, as it can allow reduced doses and can avoid the toxicity and side-effects caused by off-target binding. New discoveries are continually being reported in this rapidly developing field. In this review, we report the latest developments in supramolecular peptide-based hydrogels for drug delivery, focusing primarily on discoveries that have been reported in the last four years (2018–present). We address clinical points, such as peptide self-assembly and drug release, mechanical properties in drug delivery, peptide functionalization, bioadhesive properties and drug delivery enhancement strategies, drug release profiles, and different hydrogel matrices for anticancer drug loading and release.

Abstract:A multi-footed composite hydrogel with double responsiveness was prepared by the combination of sodium alginate (SA), gelatin and ferroferric oxide nanomaterials. Under magnetic field control, the multi-footed composite hydrogel could walk and move to the designated locations and release some drugs under the specific environment (pH 7.4) due to the pH sensitivity of SA. The gelatin addition effectively improved the mechanical properties of composite hydrogel, adjusting the drug release behavior of composite hydrogel by the decreased porous structure. Our results demonstrated that the proposed dual stimulus-responsive composite hydrogel could offer a promising choice for medical fixed-point drug release and disease treatment.

Abstract:During the past decade, a series of thermo-responsive hydrogels based on poly(N-isopropylacrylamide) (PNIPAM) were prepared for drug delivery. However, the release performance of those hydrogels was limited because the LCST (lower critical solution temperature) of PNIPAM (about 32 °C) is slightly lower than human body temperature, and the drug loading is complicated. To develop a new drug delivery matrix with the suitable responsive interval, a novel, temperature-sensitive copolymer with two monomers, N-isopropylacrylamide and methylacrylic polyethylene glycol monomethyl ether ester (MPEGMA) [poly(NIPAM-co-MPEGMA)], was synthesized by free radical polymerization. Then, the supramolecular hydrogels with tunable response temperature, whose structures were confirmed by X-ray diffraction (XRD), were fabricated via inclusion complexation with α-cyclodextrin (α-CD) and the copolymers in aqueous solution. The rheology results indicated that response interval could be modulated by controlling the contents of PNIPAM in the copolymers. Compared to the normal PNIPAM ge1, the succinct method of gel preparation streamlined the process of loading and made the drug loading capacity controllable. The release time was proved to be prolonged by the release kinetics of 5-fluoroucrail (5-Fu), which showed feasibility as a drug delivery.

Abstract:In this study, we prepared a novel multifunctional two-component supramolecular hydrogel (T-G hydrogel) via two organic molecules in ethanol/water mixed solvents. In addition, we prepared gold nanoparticle/T-G (AuNPs/T-G) composite hydrogels using T-G hydrogel as a template for stabilizing AuNPs by adding HAuCl 4 and NaBH 4 during the heating and cooling process of T-G hydrogels. The morphology and microstructure of the as-prepared hydrogels were characterized using SEM, TEM, XRD, and FT-IR. The hydrogels prepared by solutions that contained different ethanol/water volume ratios exhibited different microstructures, such as sheets, strips, and rods. The obtained T-G hydrogels exhibited a sensitive response to pH changes in the process of sol–gel transformation and showed good adsorption properties for model organic dyes. In the presence of NaBH 4 , the obtained AuNP/T-G composite hydrogels exhibited the excellent catalytic performance for 4-nitrophenol (4-NP) degradation. Thus, the current research provides new clues in developing new multifunctional two-component supramolecular gel materials and exhibits potential applications for wastewater treatment.

Abstract:It has been attracted much attention to fabricate stimulus responsive supramolecular hydrogels as smart materials in recent years. However, the multi-stimuli responsiveness often requires the complicated chemical synthesis and rational molecular design. Herein, a quadruple-stimuli responsive supramolecular hydrogel has been designed through the host-guest interaction between a β-CD dimer and a methoxy-azobenzene (mAzo) and ferrocene (Fc) grafted poly(acrylic acid) derivative, as well as the electrostatic interaction of negatively charged carboxyl side groups. Owing to the dynamic properties of the host-guest and electrostatic interactions, the reversible sol-gel transition can be triggered by various stimuli, including temperature, light irradiations, pH changes and chemical redox reagents. As a result, the release of rhodamine B loaded in the hydrogel can be accelerated by green light irradiation, oxidizing agents and low pH, demonstrating the potential applications in biomedical materials.

Abstract:Poly(ethylene glycol) methyl ether-grafted-graphene oxide (GO) was synthesized by a coupling reaction and formed inclusion complexes (ICs) after selective threading of the mPEG segments of the GO-mPEG through the cavities of α-cyclodextrins (α-CDs) units. The polypseudorotaxane structures of the as-obtained hydrogels were confirmed by X-ray diffraction, TGA, DSC, and SEM. The complexation of the mPEG segments with α-CDs and the hydrogen-bond interaction between CDs resulted in the formation of supramolecular hybrid hydrogels with a strong network. The resultant hybrid hydrogels were found to be thermo-responsive, and could be applied as a promising drug delivery system.

Abstract:<p>It has been attracted broad interest to construct stimulus responsive supramolecular hydrogels as smart materials over the past few decades; however, the design of multi-stimuli responsive hydrogels remains a formidable challenge. Herein, a multi-stimuli responsive supramolecular hydrogel has been designed on the basis of methoxy-azobenzene-grafted poly(acrylic acid) (PAA-mAzo) and β-CD dimer, which involves the host-guest and electrostatic interactions. Besides the common thermo-responsive behavior, the hydrogel also exhibits reversible sol-gel transition under green and blue light irradiations because of the disassociation and association of host–guest interaction between mAzo and β-CD. In addition, the hydrogel turns into precipitate at low pH due to the decrease of solubility of PAA in aqueous solution. The electrostatic interaction between the negatively charged carboxylic acid groups and hydrated protons at high pH makes the recovery of hydrogel. At last, the release of rhodamine B loaded in the triple-stimuli responsive hydrogel can be accelerated by green light irradiation and low pH, which demonstrates the potential applications in biomedical materials.</p>

Supramolecular hydrogel for programmable delivery of therapeutics to cancer multidrug resistance

Abstract:Multidrug resistance (MDR) has been considered as a major adversary in oncologic chemotherapy. To simultaneously overcome drug resistance and inhibit tumor growth, it is essential to develop a drug delivery system that can carry and release multiple therapeutic agents with spatiotemporal control. In this study, we developed a hydrogel containing an enzyme-cleavable peptide motif, with a network structure formed by 4-armed polyethylene glycol (PEG) crosslinked by complementary nucleic acid sequences. Hydrogen bond formation between nucleobase pairing allows the hydrogel to be injectable, and the peptide motif grants deliberate control over hydrogel degradation and the responsive drug release. Moreover, MDR-targeted siRNAs are complexed with stearyl-octaarginine (STR-R8), while doxorubicin (Dox) is intercalated with DNA and nanoclay structures in this hydrogel to enhance therapeutic efficacy and overcome MDR. The results show a successful configuration of a hydrogel network with in situ gelation property, injectability, and degradability in the presence of tumor-associated enzyme, MMP-2. The synergistic effect by combining MDR-targeted siRNAs and Dox manifests with the enhanced anti-cancer effect on drug resistant breast cancer cells in both in vitro and in vivo tumor models. We suggest that with the tailor-designed hydrogel system, multidrug resistance in tumor cells can be significantly inhibited by the co-delivery of multiple therapeutics with spatial-temporal control release.

Abstract:Hydrogels are widely used as excellent drug carriers in the field of biomedicine. However, their application in medicine is limited by their poor mechanical properties and softness. To improve the mechanical properties of hydrogels, a novel triple-network amphiphilic hydrogel with three overlapping crosslinking methods using a one-pot free-radical polymerization was synthesized in this study. Temperature-sensitive and pH-sensitive monomers were incorporated into the hydrogel to confer stimulus responsiveness, making the hydrogel stimuli-responsive. The successful synthesis of the hydrogel was confirmed using techniques, such as proton nuclear magnetic resonance spectroscopy (1H NMR), Fourier-transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD). In order to compare and analyze the properties of physically crosslinked hydrogels, physically–chemically double-crosslinked hydrogels, and physically–chemically clicked triple-crosslinked hydrogels, various tests were conducted on the gels' morphology, swelling behavior, thermal stability, mechanical properties, and drug loading capacity. The results indicate that the triple-crosslinked hydrogel maintains low swelling, high mechanical strength, and good thermal stability while not significantly compromising its drug delivery capability.
