Abstract:An iridium(III)-catalyzed ortho-C(sp(2))-H amidation of O-methyl ketoximes with sulfonamides has been accomplished under mild conditions. This protocol can be successfully applied to a broad scope of oxime substrates and works well with a range of aryl- and alkylsulfonamides.

Abstract:The functionalization of remote unactivated C(sp3)-H and the reaction selectivity are among the core pursuits for transition-metal catalytic system development. Herein, we report Pd-catalyzed γ-C(sp3)-H-selective alkylation and alkenylation with removable 7-azaindole as a directing group. Acid and base were found to be the decisive regulators for the selective alkylation and alkenylation, respectively, on the same single substrate under otherwise the same reaction conditions. Various acrylates were compatible for the formation of C(sp3)-C(sp3) and C(sp3)-C(sp2) bonds. The alkenylation protocol could be further extended to acrylates with natural product units and α,β-unsaturated ketones. The preliminary synthetic manipulation of the alkylation and alkenylation products demonstrates the potential of this strategy for structurally diverse aliphatic chain extension and functionalization. Mechanistic experimental studies showed that the acidic and basic catalytic transformations shared the same six-membered dimer palladacycle.

Abstract:A palladium-catalyzed stereospecific alkylation of allylamines with primary and secondary alkyl iodides is described. Isoquino-line-1-carboxamide (IQA) acts as directing group to generate multi-substituted olefin products in cis configuration in moder-ate to good yields. Mechanistic studies suggest that alkenyl C–H bond activation is the rate-determining step.

Abstract:A palladium(II)-catalyzed β-methylene C(sp(3))-H bond alkenylation of acyclic aliphatic amides with alkenyl halides has been developed. Both (E)-olefins and (Z)-olefins can be readily accessed using this method and a possible (Z)/(E)-olefin isomerization pathway is proposed. A solvent effect-promoted sequential C(sp(3))-H bond alkenylation and C(sp(2))-H bond alkenylation was also studied, and can provide a convenient route to novel diene compounds.

Abstract:A palladium-catalyzed monoarylation of β-methyl C(sp(3))-H of an alanine derivative with aryl iodides using an 8-aminoquinoline auxiliary is described. The reaction is highly efficient, scalable and compatible with a variety of functional groups with complete retention of chirality, providing a general and practical access to various β-aryl-α-amino acids. The synthetic potential of this protocol is further demonstrated in the sequential synthesis of diverse β-branched α-amino acids.

Abstract:A direct ethynylation reaction between N-vinylacetamides and (bromoethynyl)triisopropylsilane catalyzed by palladium is reported. A broad scope of cyclic N-vinylacetamide derivatives was tolerated and the corresponding products could be obtained in acceptable yields. This work provides a general tool for the synthesis of conjugated enyne products using palladium(II) acetate as catalyst. The mild reaction conditions and reasonable results should make this method a useful synthetic protocol. Unfortunately, other bromo-substituted alkynes are found to be unreactive and could not afford the desired products under the current reaction conditions.
