Abstract:The synthesis of dihydrofurans via a dynamic kinetic asymmetric domino oxa-Michael-carbocyclization procedure is reported. The optimization of the reaction parameters (solvent, temperature, metal source, additives, organocatalyst) is described. The requirements for both a metal catalyst and a proline-derived organocatalyst for the transformation were established. Although aryl substitution in the aldehyde component was well tolerated, aliphatic aldehydes were found to give lower yields. The use of secondary and tertiary propargylic alcohols was also shown to afford good enantioselectivity, but poor diastereoselectivity.

Abstract:The bifunctional organocatalyst C3 N,N'-dioxide has been successfully applied to the asymmetric cascade Michael/hemiacetalization reaction of α-substituted cyano ketones and β,γ-unsaturated α-ketoesters for the synthesis of multifunctionalized chiral dihydropyrans. The corresponding products were obtained in excellent yields (up to 99%) with high to excellent enantioselectivities (up to 99% ee).

Abstract:A highly regio- and stereoselctive palladium-catalyzed domino reaction of functionalized aryl allyl ethers has been developed. Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:A Pd-catalyzed asymmetric allylic substitution cascade of allylic meso-dicarbonates with 3-oxo-nitriles has been developed for the synthesis of chiral bicyclic dihydrofurans bearing two vicinal carbon stereocenters. The reaction proceeds via an asymmetric desymmetrization process with the desired products being obtained in high yields and with up to 97% ee. The reaction was performed on a gram-scale and the corresponding bicyclic dihydrofurans could undergo several transformations. The methodology provides an efficient synthetic route to biologically active chiral bicyclic dihydrofurans derivatives.

Abstract:An asymmetric domino double Michael addition reaction was developed using accessible substrates to construct biologically important and synthetically challenging cyclopentanes with four contiguous stereocenters. The proline-like molecules, trans-perhydroindolic acids, proved to be efficient organocatalysts in this reaction. Under the optimal reaction conditions, the asymmetric domino double Michael addition provided good yields (up to 98%), and excellent diastereoselectivities (up to 100% dr) and enantioselectivities (up to 99% ee). The obtained polysubstituted aliphatic cyclopentanes not only exist in biologically active natural products and medicines, but can also be converted into many other useful scaffolds via a simple transformation, such as cis-fused bicyclic lactams. Our current methodology is suitable for the synthesis of polysubstituted aliphatic cyclopentanes with contiguous multiple stereocenters.

Abstract:The enantioselective synthesis of chiral compounds containing multiple stereogenic elements via a single catalytic step is a challenging process. In the presence of α‐chloronitrostyrenes and a chiral squaramide catalyst, C–C or C–N pro‐axially chiral 2‐naphthol substrates, featuring low barriers to enantiomerization, underwent a remote diastereo‐ and enantioselective domino Michael/O‐alkylation. It provided the desired benzodihydrofurans bearing two stereogenic carbon atoms and a configurationally stable C–C or a C–N bond, thanks to a high increase of the barrier to rotation upon dihydrofurannulation.

Abstract:Organocatalysis: a modified Feist-Bénary reaction employing acyclic β-ketoesters as the dinucleophile and (E)-β,β-bromo nitrostyrenes as the dielectrophile was developed for the asymmetric synthesis of 2,3-dihydrofurans. L-Threonine-derived tertiary amine/thiourea catalysts were found to be the most efficient catalysts, inducing the reaction in a highly enantioselective manner (TBDPS=tert-butyldiphenylsilyl).

Abstract:An unprecedented multicomponent cascade asymmetric inverse-electron-demand Diels-Alder/nucleophilic addition/ring-opening reaction involving 2-methoxyfurans as efficient dienophiles was successfully developed with Cu(I)/(t)Bu-Box complex as the catalyst. Abstract:A direct method to construct 3,4-dihydropyridin-2-ones by enzymatic condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds was developed. One ring and four new bonds (two C–C, one C–N, one CC) were formed in one pot. And reaction conditions involving hydrolases, solvents, substrate molar ratios, and hydrolase loading were optimized. A series of new compounds based on the 3,4-dihydropyridin-2-one core were synthesized by the unprecedented three-component domino reaction.

Abstract:Organocatalyzed reactions of α,α-difluoro-β-keto esters with acetone were demonstrated. In the presence of L-proline, the Aldol reaction occurred under mild conditions to give the corresponding tertiary α,α-difluoroalcohols in good yields with high enantioselectivities. Using pyrrolidine as catalyst, the domino Michael-aldol reaction took place readily to give the corresponding addition products in high yields with excellent diastereoselectivities.

Abstract:: A straightforward and atom-economical approach to the synthesis of dihydrofurans employing electro-oxidative [3+2] annulation of 1,3-dicarbonyl compound and alkenes has been developed. Neither metal catalysts nor chemical oxidants are required to promote the dehydrogenation process. Moreover, the use of base to decrease the oxidative potential and 1,1,1,3,3,3-hexafluoro-2-propanol to stabilize the reactive intermediates are key to the success of the cyclization.

Abstract:Described herein is the development of a palladium(II)-catalyzed two- or three-component reaction of 2-(1-alkynyl)-2-alken-1-ones with nucleophiles and allylic chlorides. Various types of nucleophiles such as O-, N-, C-based nucleophiles and olefin-tethered O-, N-, C-based nucleophiles were investigated. The scope, mechanism and application of this Pd(II) -catalyzed domino reaction were studied. In these transformations, the palladium catalyst exhibits a dual role, serving simultaneously as a Lewis acid and a transition metal. Two possible reaction pathwavs (cross-coupling reaction vs. Heck reaction) from the same intermediate furanylpalladium species were observed. The reaction pathway is dependent on the property of the nucleophile and the length of the tethered chain as well. When olefin-tethered O- based nucleophiles were used., only the cross-coupling reaction pathway was observed, in contrast, both reaction pathways were observed when olefintethered C-based nucleophiles were employed. The product ratio is dependent on the length of the tethered chain. Furthermore, ring-closing metathesis (RCM) of corresponding furans with C=C bonds provides an easy method for the preparation of functionalized oxygen-heterocycles - 3,4-fused bicyclic furans. It is also noteworthy that allylic chloride can be as an oxidant besides its well known function as an alkylating reagent.
