Abstract:An asymmetric Sulfa‐Michael/Michael domino reaction of (E)‐4‐mercapto‐2‐butenoates with difluoromethyl‐eneoxindoles catalyzed by a quinidine‐derived ammonium salts as phase transfer catalyst has been disclosed. Under mild conditions, a broad range of CF2H‐containing spiro[indoline‐3,3′‐thiophene]s, bearing three adjacent chiral centers including two vicinal spiro quaternary stereocenters, catalysed by only 5 mol% PTC catalyst were obtained in yields of 75–98%, excellent diastereoselectivities (>20:1, in almost all cases) and enantioselectivities of 19–98% in short reaction times (almost less than 20 minutes).

Abstract:Reported here is the development of a class of chiral spirosilabiindane scaffolds by Rh-catalyzed asymmetric double hydrosilation, for the first time. Enantiopure SPSiOL (spirosilabiindane diol), a new type of chiral building block for the preparation of various chiral ligands and catalysts, was readily prepared on greater than 10 gram scale using this protocol. The potential of this new spirosilabiindane scaffold in asymmetric catalysis was preliminarily demonstrated by development of the corresponding monodentate phosphoramidite ligands (SPSiPhos), which were used in both a Rh-catalyzed hydrogenation and a Pd-catalyzed intramolecular carboamination.

Abstract:Spiro[azetidine-indolines] are important scaffolds in diverse bioactive compounds. Current efforts to synthesize spiro[azetidine-indolines] are limited to chiral spiro[azetidine-2,3'-indolines]. Asymmetric synthesis of structurally similar chiral spiro[azetidine-3,3'-indolines] remains unexplored. In this work, the first copper(I)-catalyzed asymmetric Kinugasa/aryl C-C coupling cascade reaction is described. This provides a straightforward access to densely functionalized chiral spiro[azetidine-3,3'-indoline]-2,2'-diones in good yields and with high enantioselectivity.

Abstract:Aurones and their analogues are convenient two‐atom units for the rapid construction of valuable spiroheterocycles. Here, we demonstrate that the Morita−Baylis−Hillman carbonates of isatins can be efficiently assembled with thioaurones, aurones, or even methyleneindolinones in a highly asymmetric [3+2] annulation manner, affording a broad spectrum of densely functionalized spiro‐benzothiophene, ‐benzofuran and bisspirooxindole frameworks in high yields with good to excellent stereoselectivity.

Abstract:Asymmetric synthesis of spiro[indoline‐3,4‐pyrrolo [3,4‐b]pyridines] derivatives was first developed through organocatalytic cascade Knoevenagel/Michael/cyclization reaction using a quinidine ‐derived squaramide. Under the optimized conditions, the three‐component reaction of isatins, cyanoacetates, and 3‐aminomaleimides yield the desired heterocycle‐fused spirooxindoles in good yields (78–91%) with 53‐99% enantiomer excess (ee). Notably, this reaction enabled a broad substrate scope under mild conditions, and provided a convenient method for enantioselective construction of diverse spirooxindoles combined with dihydropyridine and maleimide skeleton, which brought great potential to build new bioactive chemical entities.

Abstract:An asymmetric sulfa-Michael/aldol cascade reaction of 2-ylideneoxindoles with 1,4-dithiane-2,5-diol catalyzed by squaramide derived from cinchona was first developed to realize the chiral synthesis of unprecedented C2-spirooxindoles with a 3-amine-tetrahydrothiophene moiety incorporated a spiro hetero-quaternary carbon center and three consecutive chiral centers (30 examples; up to 95% yields, >20:1 dr and 99% ee). For some examples of poor enantioselectivity, the optical purity of spiro[indoline-2,3'-thiophene]-3-ones can be further improved by cocrystallization without the addition of other pure cocrystallization additives. In addition, DFT calculations offered a mechanistic explanation for this high stereoselectivity of the sulfa-Michael/aldol cascade reaction.

The synthesis features a semipinacol rearrangement reaction to stereoselectively construct the two-vicinal quaternary chiral centers at C8 and C10, a tungsten-mediated cyclopropene-based Pauson-Khand reaction to install the C13 quaternary chiral center, and a furan-based oxidative cyclization to stereoselectively form the spiroketal motif.

Abstract:A mild and efficient organocatalyzed [3+2] cycloaddition of isatin-derived ketimines and 2-ylideneindane-1,3-diones has been realized. Abstract:An interesting asymmetric organocatalytic cascade reaction of 3-hydroxyoxindoles with coumarins was accomplished by chiral bisguanidinium hemisalt. A series of enantioenriched spirobenzofuran indolinones were afforded in high yields with high diastereo- and enantioselectivities. The reaction was disclosed to involve in several steps: intermolecular Michael reaction, transesterification, retro-Michael reaction, substitution and decarboxylation, then an intramolecular Michael reaction to yield the product, in which the final step is disclosed to be stereo-determining step. In addition, it provided a facile route for the late-stage modification of several drug molecules via installation of spirobenzofuran indolinone scaffold.

Abstract:A concise and diastereoselective construction of the ABCD ring system of spirochensilide A is described. The key steps of this synthesis are a semipinacol rearrangement reaction to stereoselectively construct the AB ring system bearing two vicinal quaternary chiral centers and a Co-mediated Pauson-Khand reaction to form the spiro-based bicyclic CD ring system. This chemistry leads to the stereoselective synthesis of 13(R)-demethyl spirochensilide A, paving the way for the first asymmetric total synthesis of (-)-spirochensilide A.
