Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:A Lewis-acid-promoted approach to the synthesis of highly functionalized dihydrobenzofuran derivatives was developed. A diverse range of functional groups are tolerated in this type of reaction. The reaction mechanism investigation indicates that the highly reactive phenyl cation intermediate is probably involved in this process. The chirality of substrate is retained under the reaction conditions.

Abstract:CuI-catalyzed O-arylation of phenols with electron-rich 4-bromoanisole and electron-deficient 3-bromoanisole was described. Electron-deficient 3-bromoanisole showed high reactivity, and electron-rich 4-bromoanisole had low reactivity. O-Arylation of 4-bromoanisole occurred to afford the product by increasing reaction temperature and time. This reaction was applied to the synthesis of 1-chloro-4-(4-methoxyphenoxy)benzene, which was further synthesized to herbicide Clofop.

Abstract:The first aryl iodide catalyzed intramolecular C-H amination of phenylurea has been disclosed for high-efficiency synthesis of benzimidazolone derivatives in excellent yields (up to 97%) by an operationally simple one-step organocatalytic oxidative process. Fluorinated protic alcohols can efficiently accelerate the conversion of this transformation. The straightforward method has good functional group tolerance and can be performed with an inexpensive and readily accessible catalyst with high proficiency.

In the presence of triethylamine, a tandem Michael addition and nucleophilic substitution progress was generated. The current method features readily available raw materials, mild reaction conditions, high atom economy, and wide tolerance for the coupling partners.

Abstract:An unprecedented I2/FeCl3-promoted cascade reaction of aryl methyl ketones with 8-aminoquinolines for the convenient synthesis of (E)-3-(2-acyl-1H-benzo[d]imidazol-4-yl)acrylaldehydes was developed by merging annulation with ring deconstruction. This novel strategy unlocked the new reactivity of 8-aminoquinolines and provided an attractive platform for the ring opening of unactivated N-heteroaromatic compounds. Preliminary mechanistic investigation suggested that dual C(sp3)–H amination/C–N bond cleavage were key reaction steps. Abstract:The reaction of 1-benzyl- and 1-(2-phenylethyl)- 3-methyl-5-pyrazolone with POCl3 and DMF affords N,N- dimethylaminothylidene derivatives that in acid or basic medium can be transformed into the corresponding aldehydes at the carbon 4 of the heterocycle. The dimethylenamine can also be more easily obtained by reacting pyrazolones with the dimethyl acetal of N,N-dimethylformamide. Starting from the aldehydes or the N,N-dimethylenamines is possible to obtain new enamines. The advantages of using dimethylenamines are the absence of the enol as an undesired byproduct and that there is no need to prepare the aldehyde to obtain Schiff bases. Overall, the yields can be classified as acceptable. The aldehydes can be described as 5-hydroxypyrazole-4-carbaldehydes in chloroform solutions, but in solid phase they are dicarbonyl compounds. The Schiff bases exhibit an enamine structure in chloroform solution and in solid phase.

Abstract:A simple, environmentally benign and catalyst‐free method for the synthesis of chirally enriched imidazo[1,2‐a]pyrimidinone glycohybrids has been successfully developed. The protocol is based on a base‐induced annulation of α‐iodo‐pyranone with Michael addition of 2‐aminopyrimidinones followed by intramolecular nucleophilic substitution reaction. The privilege of this method includes mild reaction conditions, eco‐friendly solvent and a transition metal‐free approach. Moreover, using straightforward simple methods, this reaction method exhibits a broad range of substrate scope and remarkable tolerance toward various functional groups.
