Abstract:From the fermentation extract of Xylarialean sp. A45, an endophytic fungus of Annona squamosa L., three new triterpenes, namely xylariacins A-C (1-3, resp.) were obtained. Their structures were determined by spectroscopic analyses, including 1D- and 2D-NMR experiments and HR-ESI-Q-TOF mass spectrometry. The in vitro cytotoxic activities of compounds 1-3 were tested against human tumor cell line HepG2, and these compounds showed modest cytotoxic activity.

Abstract:Three new triterpenoid derivatives, named (15 alpha)-15-hydroxysoyasapogenol B (1), (7 beta,15 alpha)-7,15-dihydroxysoyasapogenol B (2), and (7 beta)-7,29-dihydroxysoyasapogenol B (3), were isolated from cultures of the plant endophytic fungus Pestalotiopsis clavispora. Their structures and relative configurations were elucidated by extensive spectroscopic analysis and X-ray crystallography.

Abstract:A new mixture of epimeric isomers furanosesquiterpenes (1 and 1a), together with two known furanosesquiterpenes were isolated from Xanthopappus subacaulis, which was used as Tibetan medicine in China. The structures of these compounds were elucidated on the comprehensive analysis of spectroscopic data including UV, IR, MS and NMR. The biosynthetic pathway of furanosesquiterpenes is proposed and the chemotaxonomic significance of these compounds is also discussed.

Abstract:One new diterpenoid, xylarenolide (1), and three new sesquiterpenoids, xylaranol A (2), xylaranol B (3), and xylaranic acid (4), were obtained from the fungal strain Xylaria sp. 101, which was isolated from the fruiting body of Xylaria sp. collected in Gaoligong Mountain, Yunnan Province. Their structures were elucidated by spectroscopic analyses, including 1D- and 2D-NMR experiments, and by HR-Q-TOF mass spectrometry. Their antimicrobial activities were evaluated.

Abstract:Chemical investigation of the culture extract of an endophyte Xylaria curta YSJ-5 from Alpinia zerumbet (Pers.) Burtt. et Smith resulted in the isolation of eight previously undescribed compounds including five eremophilane sesquiterpenes xylarcurenes A-E, one norsesquiterpene xylarcurene F, and two α-pyrone derivatives xylarpyrones A-B together with eight known related derivatives. Their chemical structures were extensively established based on the 1D- and 2D-NMR spectroscopic analysis, modified Mosher's method, electronic circular dichroism calculations, single-crystal X-ray diffraction experiments, and the comparison with previous literature data. All these compounds were tested for in vitro cytotoxic, anti-inflammatory, α-glucosidase inhibitory, and antibacterial activities. As a result, 6-pentyl-4-methoxy-pyran-2-one was disclosed to display significant antibacterial activity against Staphylococcus aureus and methicillin-resistant S. aureus with minimal inhibitory concentration value of 6.3 μg/mL.

Abstract:A new xanthone, 2,8-dihydroxy-9-oxo-9H-xanthene-6-carboxylic acid (1), together with six known compounds, 2,8-dihydroxy-1-methoxycarbonyl-9-oxo-9H-xanthene-6-carboxylic acid (2), 8-hydroxy-3-methyl-9-oxo-9Hxanthene-1-carboxylic acid methyl ether (3), 1,3,6-trihydroxy-8-methylxanthone (4), 1,6-dihydroxy-3-methoxy-8-methylxanthone (5), decarboxycitrinone (6), and formoic acid A (7), were isolated from an endophytic fungus Arthrinium arundinis of Anoectochilus roxburghii. Compounds 2, 3, 5–7 were also isolated from this genus for the first time.

Abstract:Four new metabolites, named pyrenochaetamide A (1), pyrenochaetolide A (2), pyrenochaetoxy A (3) and pyrenochaetolide B (4), together with five known compounds (5-9), were isolated from strain Pyrenochaeta sp. B36, an endophytic fungus of Annona squamosa L. Their structures were elucidated by spectroscopic analyses including 1D- and 2D-NMR (HMQC, HMBC, 1H-1H COSY and NOESY) and HRFT-MS. The cytotoxic activities of these compounds were evaluated and some of them showed substantial effect.

Abstract:Three new sesquiterpene acids, xylaric acids A-C (1-3, resp.), and a new tetralone (=3,4-dihydronaphthalen-1(2H)-one) derivative, 4, along with nine known compounds, xylaric acid D (5), hydroheptelidic acid (6), gliocladic acid (7), chlorine heptelidic acid (8), trichoderonic acid A (9), 16-(alpha-D-mannopyranosyloxy)isopimar-7-en-19-oic acid (10), 16-(alpha-D-glucopyranosyloxy) isopimar-7-en-19-oic acid (11), 5-carboxymellein (12), and naphthalen-1,8-diol 1-O-alpha-D-glucopyranoside (13) have been isolated from the solid culture of the ascomycete fungus Xylaria sp. associated with termite nest. The structures of these compounds were elucidated primarily by NMR experiments. The absolute configurations of compounds 1-3 and 5-9 were determined by combination of X-ray data and CD spectral analysis. The absolute configuration of 4 was assigned by Snatzke's method. Compounds 8 and 11 showed slight cytotoxicities against two cell lines A549 and SGC7901.

Abstract:Three new eremophilane sesquiterpenes (1-3) were isolated from the mangrove endophytic fungus Xylaria sp. BL321 together with 07H239-A (4), a known analogue of the new compounds. The structures of these compounds were elucidated by analysis of their MS, 1D and 2D NMR spectroscopic data. Compound 4 showed activation activity on α-glucosidase at 0.15 μM (146%), and then, 4 gradually produced inhibitory activity on α-glucosidase with increasing concentration, and the IC₅₀ value is 6.54 μM.

Abstract:Six new xanthone dimers, diaporxanthones A-F (1-6), and an unusual xanthone monomer diaporxanthone G (7), in addition to seven known analogues (8-14), were isolated and identified from endophytic Diaporthe goulteri L17 harbored in the fruits of the salt-tolerant plant Vitex trifolia. The chemical structures of these metabolites were elucidated on the basis of nuclear magnetic resonance, high-resolution electrospray ionization mass spectrometry, and reported data in the literature. Their absolute configurations were established by single-crystal X-ray diffraction analysis together with time-dependent density functional theory electronic circular dichroism calculations. Among these compounds, compounds 1 and 6 exhibited moderate antifungal activities against Nectria sp. and Colletotrichum musae and compound 4 showed significant cytotoxicity against all selected five cancer cell lines.
