Abstract:A novel palladium catalyzed homodimerization of ortho-hydroxyphenyl substituted p-QMs has been developed via [4 + 2] cycloaddition/oxidative dehydrogenation coupling domino reactions. Abstract:A facile access to thiazolidin-4-ones from alpha-enolic dithioesters and alpha-halohydroxamates in situ derived active 1,3-dipolar aza-oxyallyl cations was achieved under mild conditions. Abstract:Herein, we have developed a method for the construction of spiro[benzofuran-2,2'-bicyclo[2.1.1]hexanes] via BF3·Et2O-catalyzed formal [2π + 2σ] cycloaddition of bicyclo[1.1.0]butanes with benzofuran-derived oxa(aza)dienes. This transformation allowed for facile access to a variety of functionalized spiro-bicyclo[2.1.1]hexanes in good yields (up to 99% yield) with excellent chemoselectivities and a broad substrate scope (34 examples) under mild reaction conditions. Moreover, the synthetic utility of the cycloadducts was further emphasized through a scale-up experiment and subsequent synthetic transformations.

Abstract:A mechanistically interesting and practical method for the synthesis of functionalized spiro[4.5]decanes is developed, featuring oxidative dearomatization-induced ring expansion of cyclobutanes as the key element. The new method enables facile access to a variety of spiro[4.5]cyclohexadienones with good efficiency and generality. Further elaboration of the resulting products into other valuable scaffolds is also explored, leading to the discovery of an interesting compound that displays a promising biological profile. Moreover, we have also conducted a comprehensive computational study that provides a deep insight into the mechanism of the reaction.

Abstract:An anodic oxidation/cyclization of 2-arylbenzoic acids for the synthesis of dibenzopyranones has been developed. The reaction proceeds at room temperature with no oxidant or electrolyte required and exhibits a high atom economy with H2 being the only byproduct. A series of dibenzopyranones was obtained in good to excellent yields. Urolithins A, B, and C are formally synthesized by adopting this method as a key step to demonstrate its synthetic utility.

Abstract:Bicyclo[3.2.1] type 1,2,4-trioxanes are readily synthesized from precursors that may form intramolecular hemiketals using UHP (H2O2–urea complex) as the source of the peroxy bond and p-TsOH or CSA as the catalyst. The ring closure through an intramolecular Michael addition occurred in a highly stereoselective way, giving only one diasteromer as shown by the NMR spectra.

Abstract:A squaramide-catalyzed asymmetric N,O-acetalization/aza Michael addition domino reaction between N-Boc ketimines derived from pyrazolin-5-ones and γ-hydroxyenones has been developed for the construction of pyrazolinone embedded spirooxazolidines. A hydroquinine derived bifunctional squaramide catalyst was found the most effective for this cascade spiroannulation. This new protocol allows the generation of two stereocenters and the desired products are obtained in good yields, moderate to good diastereoselectivity (up to 3.3:1 dr) and high enantioselectivity (up to >99% ee) from a range of substituted N-Boc pyrazolinone ketimines and γ-hydroxyenones. The developed protocol is amenable for a scale-up reaction.

Abstract:An enantioselective [5 + 1] annulation of α-hydroxyl aryl ketones with cyclohexadienone-tethered enones via chiral dinuclear zinc catalysts is reported. A Brønsted base and Lewis acid cooperative activation model is the critical factor for the chirality transfer process, giving access to a wide array of enantioenriched bicyclic pyrans scaffolds bearing two oxa-quaternary carbon centers with good yields and high optical purities. Notably, this reaction offers a new reaction mode of α-hydroxyl ketones as C,C-bisnucleophiles in catalytic asymmetric transformation.
