Abstract:The asymmetric intermolecular Heck reaction has been limited to aryl and vinyl triflates. Herein, we extend the reaction to aryl and vinyl bromides. Various cyclic olefins coupled with high enantioselectivity. Only bisphosphine oxides on a Spiro backbone formed highly stereoselective Pd catalysts. The use of alcoholic solvents and alkylammonium salts were essential to promote halide dissociation from neutral arylpalladium complexes.

Abstract:Asymmetric Wittig-type olefination of 4-substituted cyclohexanones with chiral ligand-modified stable arsonium ylides has been examined. The 8-phenylmenthol-derived chiral arsonium ylide of 4 reacted with prochiral ketones 9a-d at -15 degrees C to give the 4-substituted cyclohexylideneacetates 11a-d in 58-69% yield and in up to 80% diastereomeric excess (de). Abstract:Acyclic internal alkenes are a class of challenging substrates in asymmetric Heck-type reactions due to difficulties related to both reactivity and selectivity control. Employing acyclic alkenyl alcohols, an asymmetric Heck–Matsuda reaction is developed through the strategy of chiral anion phase transfer. Various chiral ketones could be obtained in high levels of enantioselectivity. Abstract:We describe herein a new method that allows selective production of Catellani-Heck isomers from various aryl halides, including ones without ortho-groups. Under previous conditions, unhindered aryl halides were plagued with the formation of simple Heck isomers and multiple arylation and norbornene insertion. The use of a bulky PtBu3 ligand accelerates C-C reductive elimination from the key palladacycle.

Abstract:A chiral anion-mediated asymmetric Heck/Tsuji–Trost reaction of aryl iodides and 1,3-dienes is presented. Chiral indoline derivatives could be afforded with remarkably higher yields and enantioselectivities than our previous chiral ligand-based method. Silver carbonate is employed as both base and halide scavenger to ensure fast and recyclable exchange of the catalytic amount of chiral anions. Abstract:Recent advances in Heck reaction are highlighted according to the types of carbon and heteroatom electrophiles. The last two decades have witnessed significant progress in asymmetric Heck reaction of aryl/alkenyl halides, sulfonates and aryldiazonium salts as in Heck Matsuda reaction. Heck-type reactions that used new types of carbon electrophiles also flourished, examples including unactivated alkyl halides possessing eliminable β-hydrogens, activated alkyl halides (e.g., α-haloesters and α-halomalonates), benzylic, allylic and propargylic electrophiles. Significant progress was also made in Heck reactions using heteroatom electrophiles, such as the innovation of enantioselective Narasaka-Heck cyclization of oxime derivatives, aza-Heck cyclization of hydroxamate derivatives and the emergence of Heck-type reactions of silyl and boryl halides.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

The key to the success is the discovery and fine-tuning of the different N-substituents of Xu-Phos, which ensure the enantioselectivity and reactivity. Abstract:A highly enantioselective palladium‐catalyzed intramolecular Heck reaction of ortho‐iodophenol‐derived allyl ether, which provides an efficient method to synthesize various chiral 3,3‐disubstituted 2,3‐dihydrobenzofuran compounds bearing an all‐carbon quaternary stereocenter has been developed. Comprehensive Summary A highly enantioselective palladium‐catalyzed intramolecular Heck reaction of unactivated alkenes is developed, which provides a powerful route to access a broad range of chiral 3,3‐disubstituted‐2,3‐dihydrobenzofurans bearing all‐carbon quaternary stereocenter of interest in pharmaceutical research. Its salient features include high yield, excellent chemo‐ and enantioselectivity, mild conditions, a broad substrate scope as well as versatile transformations of the product.

Abstract:A palladium-catalyzed aryl-to-alkyl radical relay Heck reaction of amides at α-C(sp3)-H sites with vinyl arenes is described. This process displays a broad substrate scope with respect to both amide and alkene components and provides access to a diverse class of more complex molecules. The reaction is proposed to proceed via a hybrid palladium-radical mechanism. The core of the strategy is the fast oxidative addition of aryl iodide and fast 1,5-HAT overcome the slow oxidative addition of alkyl halides, and photoexcitation effect suppress the undesired β-H elimination. It is anticipated that this approach would inspire the discovery of new palladium-catalyzed alkyl-Heck methods.
