Abstract:A novel highly enantioselective one-pot dithiolation through sulfa-Michael addition/thioesterification of thiols with α,β-unsaturated N-acylated succinimides catalysed by squaramide has been developed. This organocatalysed reaction proceeded well in high to excellent yields (up to >99%) to afford useful bioactive β-sulfated thioester derivatives with high enantioselectivities (up to 96% ee).

Abstract:A highly enantioselective sulfa-Michael addition (SMA) of various thiols to alpha,beta-unsaturated N-acylated oxazolidinones has been achieved with a chiral squaramide catalyst under very mild reaction conditions. In addition, the conversion of the beta-thio-substituted adduct to the corresponding methyl beta-tosylbutanoate was demonstrated in high yield through a methanolysis/oxidation sequence.

Abstract:A highly enantio- and diastereoselective sulfa-Michael addition of thioacetic acid to α,β-disubstituted nitroalkenes catalysed by a chiral squaramide organocatalyst has been described. This organocatalytic reaction at an extremely low catalyst loading (0.2 mol%) furnished synthetically useful β-nitro sulfides in excellent yields with good diastereoselectivities and high enantioselectivities (up to 94 : 6 dr, 95% ee). In addition, the catalytic reaction can be performed on a 10 gram scale, and facile transformation into taurine derivative is also presented.

Abstract:A highly efficient asymmetric Michael addition of thiols to exocyclic enones was achieved by using chiral spiro phosphoramide catalysts. The precisely chiral control in the protonation of the enol intermediate ensured high enantioselectivity. The reaction features high activity (yields up to 99%, turnover numbers up to 8400) and high enantioselectivity (up to 97% ee) with a broad substrate scope, and it has the potential for wide application in the synthesis of chiral sulfides.

Abstract:An efficient, asymmetric sulfa-Michael/aldol cascade reaction catalyzed by a chiral squaramide catalyst has been developed. This organocatalytic cascade reaction provides easy access to highly functionalized spirothiochromanones with three contiguous stereocenters, including one quaternary center, in excellent yields of up to 99% with excellent diastereoselectivity up to >99:1 d.r. and enantioselectivity up to 98% ee. In addition, the key to the present method is introducing indenones that are readily accessible as hydrogen-bond acceptors with very low catalyst loading of 0.5 mol%.
