Abstract:A convenient synthesis of polyoxygenated tetrahydrothiopyrans and thiepanes from various alditol derivatives with xylo, ribo, manno, gluco, galacto, and fuco configurations is described. The preparation started from the corresponding partially protected alditols and proceeded in a ‘one-pot’ manner to afford the final products in 80%–95% yield. Furthermore, thiosugar compounds 2b, 2d, and 2g were converted to the optically pure polyoxygenated cycloalkenes through Ramberg–Backlund reaction in moderate to good overall yield. The procedures can be used for the preparation of polyoxygenated thiosugars and cycloalkenes on a relatively large scale.

Abstract:Aim To develop a concise method for the synthesis of bicyclic azasugar and thiosugar with novel scaffold. Methods The two primary hydroxyl groups of compound 1 were selectively protected with tosyl cloloride in pyridine, followed by ring-closure with sodium sulfide or primary amine to form the oxy-bridged bicyclic molecules in good yields. Results Two bicyclic azasugars and a thiosugar were produced from L-sorbose in several steps. Conclusion The described procedures provide an efficient method to synthesize bicyclic azasugar and thiosugar with novel scaffold as potential glycosidase inhibitors.
Synthesis of C-Oligosaccharides Through Versatile C(sp3)-H Glycosylation of Glycosides

Abstract:C-oligosaccharides are pharmacologically relevant because they are more hydrolysis-resistant than O-oligosaccharides. Despite indisputable advances, C-oligosaccharides continue to be underdeveloped, likely due to a lack of efficient and selective strategies for the assembly of the interglycosidic C-C linkages. In contrast, we, herein, report a versatile and robust strategy for the synthesis of structurally complex C-oligosaccharides via catalyzed C(sp3 )-H activations. Thus, a wealth of complex interglycosidic (2→1)- and (1→1)-C-oligosaccharides becomes readily available by palladium-catalyzed C(sp3 )-H glycoside glycosylation. The isolation of key palladacycle intermediates and experiments with isotopically-labeled compounds identified a trans-stereoselectivity for the C(sp3 )-H glycosylation. The glycoside C(sp3 )-H activation manifold was likewise exploited for the diversification of furanoses, pyranoses and disaccharides.

Abstract:A collective synthesis of 3,6-dideoxysugars, including seven naturally known congeners, has been reported using commercially available methyl lactates in five steps. The essential tandem process involving the olefin cross-metathesis and isomerization steps was enabled by the dual function of Grubbs-II catalyst, affording the products in good yields and providing concise and practical access to a class of biologically important deoxysugars.

Abstract:A series of semi‐saturated fused polycyclic iminosugars were synthesized by one‐pot stereoselective three‐component reactions of D‐ribose tosylate, aniline and cycloenones under heating conditions. The N‐aryl enamine derived from an iminium ion is the key intermediate for the reaction. In this way, various novel complex fused iminosugars were obtained through a normal Diels‐Alder mechanism at 80 °C. This strategy will enable the preparation of bioactive iminosugar analogues with structural diversity.

Abstract:An efficient, convenient protocol for the preparation of per-O-acetylated p-tolylthio glycosides is described. Treatment of various unprotected sugars, including 2-deoxy-2-amino sugars, sialic acid, lactose, and maltose, with acetic anhydride using SnCl4 as a catalyst, and subsequently with p-tolylthiol, furnished the corresponding thioglycosides in 71%-90% yield under solvent-free conditions.

Abstract:Benzenesulfinyl morpholine (BSM) has been applied as a new promoter for the activation of thioglycosides. This proceeds by the in situ reaction of BSM with trifluoromethanesulfonic anhydride, which subsequently activates thioglycosides for glycosyla- tions. Importantly, this reagent works well in our pre-activation one-pot oligosaccharide assembly strategy.

Abstract:A highly regio- and chemo-selective heteroannulation protocol for the synthesis of unreported polysubstituted thiaazatricyclododecanes has been developed by a sequential four-component reaction of isoquinoline, 2-bromoacetophenones, aryl isothiocyanates, and methyl 4-chloro-3-oxobutanoate under mild conditions in CH3CN. To the best of our knowledge there are no previous reports for the synthesis of these classes of thiaazatricyclododecanes. The method features formation of various CN, CS, and CC bonds, low-cost and readily available substrates, as well as simple one-pot operation, which makes this process highly profitable. This synthesis serves as a nice addition to chemistry in which purification via chromatography and recrystallization can be avoided, and the pure products were obtained simply by washing the crude products with hexane–ethyl acetate (10:1).

Abstract:An efficient route to prepare l-glucose and l-galactose is described. The l-sugars are achieved by using the strategy of switching the functional groups at C1 and C5 of d-glucose and d-mannose. The oxidation and reduction of the silyl enol ether at C1 and the lead(IV) tetraacetate mediated oxidative decarboxylation at C5 are the key steps. l-Glucose and l-galactose are prepared in a convenient and inexpensive way.

Abstract:Abstract: 1-Thiosugars and their glycosides play crucial roles in carbohydrate chemistry, primarily due to their stability and potential for mimicking O-glycosides. The synthetic methodologies for thioglycosides pose ongoing challenges. In recent years, researchers have shown a growing interest in exploring the applications of thiosugars in various fields, including the development of natural product derivatives, oligo- and polysaccharide mimics, metallodrugs, and dendrimers. Numerous approaches and protocols have been devised for the synthesis of thiosugars. This review aims to comprehensively cover the efforts towards preparing thiosugars and their application as synthetic precursors.
