Abstract:An array of chiral [1,2,4]triazino[5,4-<i>a</i>]isoquinoline derivatives were obtained in excellent yields (up to 98%) and with excellent enantioselectivities (up to 99% ee) via a new highly asymmetric (3 + 3) cycloaddition reaction of diazo compounds and isoquinolinium methylides, with a bifunctional chiral phase-transfer catalyst (PTC). Density functional theory calculations show that PTC has a bridge role in the deprotonation/protonation process. The obtained products were transformed into densely functionalized polycyclic heterocompounds with multiple stereocenters.

Abstract:An efficient, stereoselective, and base-controlled procedure for the preparation of polyhydroxy bicyclic diazenes and polyhydroxy bicyclic hydrazones is described, starting from sugar-derived methyl 6-deoxy-6-iodo-hexosides. The one-pot synthesis involves a three-step reaction, including ring-opening, condensation with tosylhydrazine, and intramolecular cycloaddition with terminal alkene, which affords important synthetic precursors of bicyclic diazenes and hydrazones.

Abstract:A Pd(II)-catalyzed aerobic intermolecular 1,2-diamination of conjugated dienes was developed for the regio- and chemoselective preparation of a variety of functionalized tetrahydroquinoxalines, using simple sulfonyl protected o-phenylendiamines as a nitrogen source. This methodology provides a direct and efficient synthesis of tetrahydroquinoxalines. O2 was used as the stoichiometric oxidant, and reaction conditions were applied to a series of o-phenylendiamines and conjugated dienes. 35 examples are described, and good yields and selectivities are obtained for the majority of the products.

Abstract:A diastereoselective three-component reaction of diazo compounds with alcohols and pyrazolinone ketimines by utilizing rhodium(ii) catalysis via interception of transient oxonium ylides is reported. The reaction provides an efficient approach for the facile construction of polyfunctionalized pyrazolone derivatives bearing two contiguous quaternary stereocenters in good yields with high regioselectivities and excellent diastereoselectivities.

Abstract:We have reported the 1,4-diazabicyclo[2.2.2] octane (DABCO)-catalyzed [3 + 3] cycloaddition reaction of a-arylidene pyrazolinones and 2-benzylidenemalononitriles under mild reaction conditions, which afforded the spirocyclohexadiene-pyrazolones in good yields with moderate to good diastereoselectivities. By the use of a cinchona alkaloid derived bifunctional squaramide-tertiary amine catalyst, an asymmetric variant of this [3 + 3] cycloaddition reaction has also been developed to provide optically active spirocyclohexadiene-pyrazolones bearing an all-carbon quaternary chiral center in good yields with excellent enantioselectivities and moderate diastereoselectivities.
