Abstract:A method for the synthesis of propargylic amines has been developed via an efficient copper(I)-catalyzed three-component coupling reaction of alkynes, benzal halides and amines through C–H and C-halogen activation. This reaction is conducted under mild conditions and provides an alternative method for the synthesis of propargylic amines.

Abstract:The direct C-H and C-halogen activation for C-C bond formation is one of the most interesting reactions in organic chemistry. C-C bond formation using alkynes as a carbon nucleophilic source is a very useful method in synthesis. Herein, a copper(I) chloride catalyzed three-component coupling reaction of alkynes, dihalomethanes and amines through C-H and C-halogen activation to form propargylic amines under mild conditions has been established. The reaction can be conducted in water, in neat or in common organic solvents and it was applicable to both aromatic and aliphatic alkynes with good functional groups tolerance. It represents an excellent example of multi-component reactions (MCRs), provides an elegant method for the synthesis of propargylic amines which are frequent skeletal components and synthetically versatile key intermediates for the preparation of many nitrogen-containing biologically active compounds. From the mechanism point of view, this chemistry offers not only a new approach to propargylic amines with new C-C and C-N bonds formation through C-H and C-halogen activation, but also provides valuable mechanistic insight into the novel multi-component reactions.

Abstract:An iron(III)-catalyzed three-component coupling reaction of alkynes, CH2Cl2 and amines was developed for facile synthesis of propargylamines. Preliminary mechanism investigation using in situ FT-IR reveals that the crucial Fe-acetylide intermediate could be formed through C-H bond activation of alkynes thanks to cooperative effect of FeCl3 and 1,1,3,3-tetramethylguanidine.

Abstract:A copper nanoparticles-catalyzed three-component coupling reaction of alkyne, dihalomethane, and amine (AHA coupling) was reported for the synthesis of propargylic amine. The catalytic AHA coupling reaction was carried out under mild conditions with dihalomethane as substrate and solvent, and good to excellent yields were also achieved. Furthermore, the nanoparticie catalyst could be recovered and reused effectively, and no obvious reduction on catalytic activity was observed for five recycles.

Abstract:Rhodium-catalyzed three-component C-H bond activation of aromatics with amides and aldehydes to synthesize amines was established. The addition of copper was found to be essential to ensure the high reactivity. The mechanistic studies indicated that key intermediates formed by the transmetallization between rhodium and copper could further promote the addition between 2-(pyridin-2-yl)-phenyl-metal species and imines. A series of densely substituted amines could be conveniently prepared by this one-step, three-component procedure from commercially available substrates via C-H bond activation with water as the only by-product.

Abstract:A novel, efficient method has been developed for the synthesis of propargylamines through a three-component coupling of aryl azides, aldehydes, and alkynes in the presence of iron–iodine–copper(I) bromide as catalyst. This method is the first example for directly using aryl azides as an amino component in a three-component coupling reaction. Some of the major advantages of this protocol are: good yields, the involvement of a less-expensive and non-toxic catalyst, mild reaction conditions, and a wide range of substrates.

Abstract:An efficient microwave-assisted synthesis of tertiary propargylamines has been developed by copper(I)-catalyzed decarboxylative A(3)-coupling reaction of an alkynylcarboxylic acid with a secondary amine and an aldehyde. A wide range of diversely substituted propargylamines could be synthesized in high yields. The optimal reaction conditions are also effective for 3-alkylpropiolic acids and for terminal propiolic acid.
