Abstract:Various multifunctional chromone-spiroindolinone-cyclopentene derivatives were synthesized through a phosphine-catalyzed (3+2) annulation of Morita-Baylis-Hillman (MBH) carbonates with oxindole-chromones in generally high yields with good to excellent diastereoselectivities. The synthetic method has several advantages such as mild reaction conditions, wide substrate scope, high yields and good stereoselectivity. The gram-scale reaction indicated the synthetic method could be a practical tool for synthesis of spirocyclic compounds and will find a nice application in the synthesis of diverse biologically important compounds.

Abstract:The first tertiary amine catalyzed enantioselective Morita-Baylis-Hillman (MBH) reaction of isatins with acrylates has been demonstrated, allowing asymmetric synthesis of biologically significant 3-substituted-3-hydroxy-2-oxindoles in good yields and with excellent enantioselectivities. The C6'-OH group of β-isocupreidine (β-ICD) is believed to facilitate the key proton transfer step in the MBH reaction, via an intramolecular proton relay process.

Abstract:Phosphine-squaramide derivatives were developed to catalyze the enantioselective Morita-Baylis-Hillman reaction of acrylates with isatins to construct 3-hydroxy-2-oxindoles with quaternary stereocenters. In the presence of 2 mol% H-bonding catalyst 3e, the desired products were achieved in high yields and good-to-excellent enantioselectivities (up to 95% ee).

Abstract:A highly regio- and stereoselective [3 + 2] cycloaddition reaction for constructing novel 3,3'-cyclopentenyldispirooxindoles incorporating two adjacent quaternary spirostereocenters is reported. Under the mild conditions, the asymmetric annulation of isatin-derived MBH carbonates with 3-methyleneoxindoles involving a chiral tertiary amine catalyst provides the corresponding dispirooxindole frameworks with an extraordinary level of enantioselective control. Further synthetic utility of this method was demonstrated by the gram-scale experiment and simple transformation of the obtained product. Moreover, a plausible mechanism for this annulation reaction was also proposed on the basis of the control experiments.

Abstract:Oxazolones are structural subunits in numerous natural products and designed molecules with substantial pharmacological properties. Here, a palladium-catalyzed chemodivergent regio-and enantioselective allylic alkylation of 4-or 5 -substituted oxazol-2(3H)-ones with Morita-Baylis-Hillman (MBH) adducts has been developed using a spiroketal-based diphosphine as the ligand (50 examples). Interestingly, 4-substituted oxazol-2(3H)-ones acted as a C-nucleophiles in the reaction to afford a range of chiral 4,5-substituted oxazol-2(3H)-ones in high yields (72-99%) with good to excellent chemo-, regio-, and enantioselectivities (C/N 95:5->99:1, b/l 91:9->99:1, 85-98% ee). When a N-nucleophile was used under otherwise identical conditions, 5 -substituted oxazol-2(3H)-ones delivered a range of chiral 3,5-substituted oxazol-2(3H)-ones in high yields (68- 98%) with good regio-and enantioselectivities (b/l 71:29-91:9, 66-94% ee). The synthesis can be readily performed on gram scale under fairly low catalyst loadings, and the utility of the protocol was showcased in the facile transformation of the products into more elaborate chiral molecules.

Abstract:Exploration of the diastereodivergent synthesis of spirocyclic oxindoles has been challenging. Herein we report asymmetric [3 + 2] annulations of isatin-derived Morita–Baylis–Hillman (MBH) carbonates and 5-alkenylthiazol-4(5H)-ones. Interestingly, two different chiral catalysts, amide-phosphine and 4-dimethylaminopyridine (DMAP)-thiourea, could lead to two kinds of diastereomeric dispiro oxindoles with three contiguous stereogenic centers. The hexafluoroisopropanol (HFIP) additive plays a vital role in accelerating the reaction and tuning the diastereoselectivity. Abstract:A novel phosphine-catalyzed asymmetric aza-Morita–Baylis–Hillman reaction of endocyclic ketimines with vinyl ketone or acrolein has been presented. This approach resulted in densely functionalized adducts with moderate to high yields and enantioselectivities (up to 97% yield, up to 97% ee), with the feature of a C2 tetra -substituted chiral center on a 3-oxindole scaffold. A wide variety of substrates were compatible with this methodology, and diverse enantiomeric products could be obtained through a catalyst switching strategy. A series of transformations further demonstrated the utility of this methodology.
