Abstract:A palladium-catalyzed C-H functionalization/intramolecular asymmetric allylation cascade of N-sulfonyl benzamides with 1,3-dienes has been developed. In the presence of a chiral pyridine-oxazoline ligand, this protocol enables the synthesis of chiral 3,4-dihydroisoquinolones in yields of up to 83% with enantioselectivities of up to 96%, using environmentally friendly air as the terminal oxidant.

Abstract:Diverse 1-imidazolylisoquinolines are generated through a silver triflate-catalyzed cascade reaction of 2-alkynylbenzaldoxime with imidazole or benzoimidazole using parallel diversity-oriented synthesis. A small library of 1-imidazolylisoquinolines can be constructed efficiently under mild conditions. A plausible mechanism is proposed. We discovered that the presence of silver triflate and bromotrispyrrolidinophosphonium hexafluorophosphate (PyBroP) is essential for the reaction transformation. In the reaction process, treatment of 2-alkynylbenzaldoxime with silver triflate affords isoquinoline-N-oxide. Then, isoquinoline-N-oxide acts as a nucleophile to attack PyBroP to replace the bromide. Subsequently, an intermolecular nucleophilic addition of imidazole and further deprotonation take place to produce the desired 1-imidazolylisoquinolines. A range of great diversity could be easily introduced under the standard conditions during the reaction process.

Abstract:An efficient synthesis of 1,3-/1,1-dialkoxy 1,2-dihydroisoquinolines from o-ethynylbenzacetals and sulfonyl azides via a cascade process combining copper-catalyzed alkyne-azide cycloaddition (CuAAC), Dimroth rearrangement, 1,5-OR shift/1,5-H shift, and 6π-electrocyclic ring closure (6π-ERC) is described. Extension of the produced 1,3-dialkoxy-1,2-dihydroisoquinolines to isoquinolium salts is also disclosed.

Abstract:A novel annulation reaction of amidines with sulfur ylides and nitrobenzenes has been developed, affording various novel 4-hydroxy-5-phenylaminoimidazoles in moderate to excellent yields. The 4-hydroxy-5-phenylaminoimidazoles have been further converted into α-ketoamide and imidazol-4-imine derivatives. Moreover, a plausible mechanism for this multicomponent reaction is proposed.

Abstract:An approach to access 1-substituted isoquinolones has been developed through the addition-cyclization of imines with Grignard reagents in the presence of 2,2'-dipyridyl. A number of substituted aromatic magnesium reagents were amenable to this process, and the desired products were obtained with excellent yields and outstanding diastereoselectivities ( dr > 99:1). The utility of this convenient approach is demonstrated by the formal synthesis of ( S)-cryptostyline II. Moreover, N-methylmorpholine (NMM) was found to be an effective additive for the formation of 3-substituted isoindolin-1-ones using one-pot addition-cyclization-deprotection of imine with Grignard reagents.

Abstract:Rh(II)-catalyzed reactions of 3-diazoindolin-2-imines with 3-(2-bromoethyl)indoles, 3-(3-bromopropyl)indoles, and 3-(4-bromobutyl)indoles, followed by treatment with 1,8-diazabicyclo [5.4.0]undec-7-ene (DBU) in a one-pot operation furnished azepino [2,3-b:4,5-b']diindoles, azocino [2,3-b:4,5-b']diindoles, and azonino[2,3-b:4,5-b']diindoles, respectively. Structural uniqueness of the products, broad substrate scope, mild reaction conditions, and readily available starting materials are the merits of this approach.

Abstract:An acid‐controlled regional diverse synthesized process of CF3‐containing ketoimines by the condensation of benzo[b]thiophene‐2,3‐dione and 2,2,2‐trifluoroethlamine has been described. The imines obtained at either the 2‐position or 3‐position can further be used as substrates in asymmetric organocatalyzed [3+2] cycloaddition reactions, leading to a library of regiodiverse bispiro‐compounds with 85‐96% yield and 65‐99% ee.
