Abstract:A practical, Pd(0)/Pd(II)-catalyzed reaction was developed for phenol-directed C-H activation/C-O cyclization using air as an oxidant. The turnover-limiting step of the process was found to be C-O reductive elimination instead of C-H activation. This reaction can tolerate a variety of functional groups and is complementary to the previous methods for the synthesis of substituted dibenzofurans.

Abstract:Although various methods for the preparation of dibenzofurans are available, an efficient, general synthesis of dibenzofurans from benzofurans remains an unsolved problem. Herein, we provide the first benzannulation of readily prepared 2‐nitrobenzofurans with alkylidene malononitriles. This methodology enables facile assembly of a wide variety of highly functionalized dibenzofurans in moderate to excellent yields under metal‐free conditions. Gram‐scale synthesis and further elaborations of the product were succesfully performed, demonstrating synthetic utility of this method.

Abstract:An efficient synthesis of novel fused heterocycles has been established via microwave-assisted palladium-catalyzed intramolecular direct arylation. The acyclic aryl bromides are readily available from microwave-assisted one-pot annulation of N-(2-bromobenzyl)-2-aminophenois and ethyl 2-bromoalkanoates. The intramolecular direct arylation is then performed in the presence of palladium(II) acetate and dppf (10 mol% each) in toluene using potassium carbonate (2 equiv) as the base under microwave heating (150 degrees C, 1 h) to afford the products in 43-99% yields. Steric effect is observed for Ar-Ar bond formation, giving a substantial amount of the debromination byproduct.

Abstract:We report a metal-, acid-, and base-free 2(bromomethyl)naphthalene (2-BMN)-promoted organocatalytic Cloke-Wilson rearrangement of chain doubly activated cyclopropanes for the construction of 2,3-dihydrofurans via a carbocation-initiated tandem intramolecular ring-opening/recyclization process. The strategy is especially suitable for the construction of furan units in complex molecules, providing a solution to the problem of heavy-metal residues in dihydrofuran-containing drugs synthesized by traditional metal-based protocols. Thus, it is of potential interest in synthetic and medicinal chemistry.

Abstract:Efficient synthetic routes to construct multisubstituted benzo[1,2-b:5,4-b']difurans and benzo[1,2-b:4,5-b']difurans from bis(allyloxy)bis(alkynyl)benzenes or bis(alkynyl)dihydroxybenzenes and allylic halides utilizing palladium-catalyzed double annulations is reported. Abstract:A new cascade process has been accomplished for the synthesis of tetrahydro-1H-cyclopenta[c]furans through palladium-catalyzed [2 + 2 + 1] cyclization of 1,6-enynes with vinyl bromides. Notably, the key feature of this transformation is the use of vinyl bromides as the C1 building block. Various functionalized tetrahydro-1H-cyclopenta[c] furans bearing two quaternary carbon centers could be obtained in good yields with excellent diastereoselectivities.

Abstract:A novel palladium-catalyzed cascade cyclization reaction of alkenyl ethers with alkynyl oxime ethers for the construction of poly-heterocyclic scaffolds has been developed, in which the electron-rich alkene moiety functions as a three-atom unit, simultaneously dealing well with the coordination and regioselectivity of electron-rich olefins under metal catalysis. The strategy features excellent regio- and chemoselectivities as well as good functional group tolerance. Moreover, the newly formed 2-isoxazolyl-2,3-dihydrobenzofuran products can be further transformed to diverse complex heterocycles, demonstrating their potential applications in organic synthesis and medicinal chemistry.

Abstract:A novel palladium-catalyzed tandem cyclization of 1-(allyloxy)-2-ethynylbenzene derivatives with isocyanides in the presence of water has been developed. The key intermediates, benzofuran-3-α-carbonyl aldehydes, were obtained through a simple acid hydrolysis process and could serve as precursors for structurally diverse 2,3-difunctionalized benzofuran derivatives such as important 2-benzofurylquinoxalines, benzofuran-3-α-ketoesters and benzofuryl ynediones. This transformation features convenient operation, simple and commercially available starting materials, broad functional-group compatibility, and moderate to good reaction yields.

Abstract:A series of phenanthridin-6-(5H)-ones were synthesized through a palladium-catalyzed C-H activation/cyclization strategy using diazonium salts in good yields. The best conditions included Pd(OAc)(2) as the catalyst, PPh3 as the ligand, toluene as the solvent, K2CO3 as the base and 60 degrees C as the optimal temperature.

Abstract:Herein, we report on the utilization of a heterogeneous catalyst, consisting of Pd nanoparticles supported on a siliceous mesocellular foam (Pd-0-AmP-MCF), for the synthesis of heterocycles. Reaction of o-iodophenols and protected o-iodoanilines with acetylenes in the presence of a Pd nanocatalyst produced 2-substituted benzofurans and indoles, respectively. In general, the catalytic protocol afforded the desired products in good to excellent yields under mild reaction conditions without the addition of ligands. Moreover, the structure of the reported Pd nanocatalyst was further elucidated with extended X-ray absorption fine-structure spectroscopy, and it was proven that the catalyst could be recycled multiple times without significant loss of activity.

Abstract:Stereoselective construction of highly functionalized heterocyclic molecules is an ongoing concern for the chemical community. Among the various strategies developed with this goal, catalytic asymmetric dearomatization, an attractive method for constucting cyclic molecules with multiple stereocenters from readily available aromatic compounds, has received extensive attention in recent years. Here, we report a highly stereoselective construction of tetrahydrofurobenzofurans and tetrahydrofurobenzothiophenes via palladium-catalyzed dearomative [3 + 2] cycloaddition of nitrobenzofurans and nitrobenzothiophenes, respectively. Good to excellent yields (63%–92%), diastereoselectivity (13/1 → >20/1 dr), and enantioselectivity (75%–95% ee) were obtained, leading to products with vicinal stereogenic carbon centers. The reaction features wide substrate scope and diverse transformations of the products.
