Abstract:An efficient copper‐catalyzed trifluoromethylation of substituted phthalic anhydrides using (trifluoromethyl) trimethylsilane (Me 3 SiCF 3 ) as a nucleophilic trifluoromethylating reagent in the presence of 1,10‐phenanthroline and KF, followed by quenching the reaction mixture with water has been developed. A possible mechanism was suggested.

Abstract:A new method for the generation of trifluoromethylcopper ("CuCF3") species from readily available phenyl trifluoromethyl sulfoxide has been developed. The "CuCF3" reagent can be applied in efficient trifluoromethylations of aryl iodides and activated aryl bromides in the absence of additional ligands. Furthermore, the "CuCF3" species can also undergo oxidative cross-coupling with terminal alkynes and arylboronic acids.

Abstract:Trifluoroethyl (CH2CF3) is an important functional group in many pharmaceutical and agrochemical compounds. Herein, we report an efficient method for the copper-catalyzed direct trifluoroethylation of heteroarenes. The reaction exhibited good compatibility to various substrates, and the desired products were obtained in good yields. Preliminary mechanistic investigations indicate the trifluoroethyl radical is involved in the catalytic circle. Moreover, the late-stage modification of bioactive molecules further confirmed the practical applications of this method.

Abstract:Abstract The structural motifs of trifluoromethylated aromatics (Ar‐CF3) have been widely employed in various industrial fields. The high lipophilicity, strong electron‐withdrawing ability, and characteristic size of the trifluoromethyl group is a key influence in the design of pharmaceuticals, agrochemicals, dyes, liquid crystals, and polymers. Therefore, a great deal of attention has been paid to convenient protocols of introducing trifluoromethyl groups into aromatic rings. With respect to the high regiochemical fidelity in aromatic substitution, the cross‐coupling of aryl halides with CF3Cu is one of the most powerful and versatile methods to construct trifluoromethylated aromatics.

Abstract:In this personal account, several examples of the synthesis of fluoroalkylated aromatic compounds by cross‐coupling reactions is described. Our fundamental cross‐coupling reactions of haloarenes with nucleophilic fluoroalkylating reagents are effective to introduce CF3, C2F5, CF2CO2Et, CF2H, and CF2PO(OEt)2 groups into aromatic ring. Trifluoromethylated aromatic compounds (Ar−CF3) are the substances of considerable interest in various industrial fields. The high lipophilicity, strong electron‐withdrawing ability, and characteristic size of trifluoromethyl group are key influences in biologically active molecules. Due to these attractive properties, the benzotrifluoride structural motifs have been widely employed in the design of pharmaceuticals, agrochemicals, dyes, liquid crystals, and polymers. Therefore, the development of highly efficient methodologies for aromatic trifluoromethylation is of significant importance for wide fields of science and technology. Recently, a great deal of attention has been paid to catalytic protocols to introduce fluoroalkyl groups into aromatic rings selectively. This personal account highlights the copper‐mediated aromatic trifluoromethylation and the related transformations developed by our research group.

Abstract:We report copper(II) and copper(III) trifluoromethyl complexes supported by pyridinedicarboxamide ligand (L) as a platform for investigating the role of electron transfer in C(sp2)-H trifluoromethylation. While the copper(II) trifluoromethyl complex is unreactive towards (hetero)arenes, the formal copper(III) trifluoromethyl complex performs C(sp2)-H trifluoromethylation of a wide range of (hetero)arenes. Mechanistic studies using the copper(III) trifluoromethyl complex suggest that the mechanism of arene trifluoromethylation is substrate-dependent. When the thermodynamic driving force for electron transfer is high, the reaction proceeds through a previously unidentified single electron transfer (SET) mechanism, where an initial electron transfer occurs between the substrate and oxidant prior to CF3 group transfer. Otherwise, a CF3 radical release/electrophilic aromatic substitution (SEAr) mechanism is followed. These studies provide valuable insights into the role of strong oxidants and potential mechanistic dichotomy in Cu-mediated C(sp2)-H trifluoromethylation.

Abstract:A copper-catalyzed oxidative cyclization of vinyl azides with benzylic C-sp3-H bonds via a tandem dual C-H functionalization process has been developed. This procedure allows access to substituted phenanthridines containing a variety of functional groups.
