Abstract:The first example of a nickel‐catalyzed C–H trifluoromethylation of electron‐rich heteroarenes, including imidazopyridines, indoles and thiophenes, has been developed with the commercially available and relatively inexpensive industrial raw material iodotrifluoromethane (CF3I) as the trifluoromethylating reagent. The synthetic potential of this method is demonstrated by its successful application to the direct trifluoromethylation of the biologically active molecules melatonin and zolmitriptan.

Abstract:The incorporation of fluorine-containing moieties into organic compounds is of great importance in pharmaceutical, agricultural, and materials science. Within these organofluorides, the trifluoromethyl group is one of the most important motifs. In recent years, the trifluoromethyl group has attracted more and more attention, and many trifluoromethylated compounds have been found to possess special activities. However, until now, only a few methods have been developed to achieve this efficiently using Umemoto's reagents. This review highlights recent developments in the direct introduction of a trifluoromethyl group into organic compounds with Umemoto's reagents. Seven approaches to the trifluoromethylation of organic compounds are summarized: (i) trifluoromethylation of arenes, (ii) trifluoromethylation of alkenes, (iii) trifluoromethylation of terminal alkynes, (iv) deoxygenative trifluoromethylation of benzylic xanthates, (v) trifluoromethylation of ketoesters, (vi) trifluoromethylation of aryl boronic acids and aromatic amines (synthesis of ArCF3) and (vii) trifluoromethylation of biphenyl isocyanide derivatives.

Abstract:A highly efficient and regioselective direct C–H trifluoromethylation of pyridine based on an <i>N</i>-methylpyridine quaternary ammonium activation strategy has been developed. A variety of trifluoromethylpyridines can be obtained in good yield and excellent regioselectivity by treating the pyridinium iodide salts with trifluoroacetic acid in the presence of silver carbonate in <i>N,N</i>-dimethylformamide. The protocol features good functional group compatibility, easily available starting materials, and operational simplicity. Abstract:A highly selective nickel-catalyzed C-H trifluoroethylation of heteroarenes was developed with the assistance of a monodentate directing group. This protocol provides efficient access to various trifluoroethyl-substituted heteroarenes, including indoles, pyrroles, furans, and thiophenes, with commercially available CF3CH2I as an alkylation reagent. This robust catalytic procedure is scalable and tolerates a broad range of functional groups. Moreover, multifluoroalkylation of indoles is also viable.

Abstract:Abstract The structural motifs of trifluoromethylated aromatics (Ar‐CF3) have been widely employed in various industrial fields. The high lipophilicity, strong electron‐withdrawing ability, and characteristic size of the trifluoromethyl group is a key influence in the design of pharmaceuticals, agrochemicals, dyes, liquid crystals, and polymers. Therefore, a great deal of attention has been paid to convenient protocols of introducing trifluoromethyl groups into aromatic rings. With respect to the high regiochemical fidelity in aromatic substitution, the cross‐coupling of aryl halides with CF3Cu is one of the most powerful and versatile methods to construct trifluoromethylated aromatics.

Abstract:Despite the importance of difluoromethylene fragments in medicine, agrochemistry, and materials science, the precise and sustainable C−H difluoromethylation of aryl rings remains a challenge. Studies of metal-free difluoromethylation would be advanced if methods were as reliable and modular as metal catalysis. Herein, we present a modular platform for the sustainable synthesis of para-difluoromethylation of readily available aromatic amines via organophotoredox catalysis. The reaction exhibits excellent functional group tolerance and broad substrate scope, delivering a variety of valuable difluoromethylated products. Furthermore, the derivatization of various complex molecules further demonstrates the synthetic utility and practicality of this protocol.

Abstract:An umpolung strategy for the C(sp3)‐H aminomethylation of 2‐methylindole has been developed using trifluoroacetic acid to induce the transformation, with aminals serving as precursors of iminium ion. Mechanistic studies have clearly shown that trifluoroacetic acid may play a pivotal role in this strategy. The utilization of readily available starting materials, mild reaction conditions, tolerance to complex functional groups, combine with yields of up to 98% and potential biological applications render this protocol applicable and useful for organic synthesis and medicinal chemistry.

Abstract:In this personal account, several examples of the synthesis of fluoroalkylated aromatic compounds by cross‐coupling reactions is described. Our fundamental cross‐coupling reactions of haloarenes with nucleophilic fluoroalkylating reagents are effective to introduce CF3, C2F5, CF2CO2Et, CF2H, and CF2PO(OEt)2 groups into aromatic ring. Trifluoromethylated aromatic compounds (Ar−CF3) are the substances of considerable interest in various industrial fields. The high lipophilicity, strong electron‐withdrawing ability, and characteristic size of trifluoromethyl group are key influences in biologically active molecules. Due to these attractive properties, the benzotrifluoride structural motifs have been widely employed in the design of pharmaceuticals, agrochemicals, dyes, liquid crystals, and polymers. Therefore, the development of highly efficient methodologies for aromatic trifluoromethylation is of significant importance for wide fields of science and technology. Recently, a great deal of attention has been paid to catalytic protocols to introduce fluoroalkyl groups into aromatic rings selectively. This personal account highlights the copper‐mediated aromatic trifluoromethylation and the related transformations developed by our research group.

Abstract:The introduction of trifluoromethyl groups into organic molecules is of paramount importance in modern synthetic chemistry and medicinal chemistry. While methods for constructing C(sp2)-CF3 bonds have been well established, the advancement of practical and comprehensive approaches for forming C(sp3)-CF3 bonds remains considerably restricted. In this work, we describe an efficient and site-specific deaminative trifluoromethylation reaction of aliphatic primary amines to afford the corresponding alkyl trifluoromethyl compounds. The reaction proceeds at room temperature with readily accessible N-anomeric amide (Levin's reagent) and bench-stable bpyCu(CF3)3 (Grushin's reagent) under blue light. The protocol features mild reaction conditions, good functional group tolerance, and moderate to good yields. Remarkably, the method can be applied to the direct, late-stage trifluoromethylation of natural products and bioactive molecules. Experimental mechanistic studies were conducted, and a radical mechanism is proposed, wherein the dual roles of Grushin's reagent have been elucidated.

Abstract:Heterocyclic trifluoromethylation is efficiently initiated through a photochemical reaction utilizing an electron donor–acceptor (EDA) complex, proceeding smoothly without the use of photocatalysts, transition‐metal catalysts, or additional oxidants. This method has been optimized through extensive experimentation, demonstrating its versatility and efficacy across various substrates, including quinoxalinones, coumarins, and indolones. Notably, this approach enables the practical synthesis of trifluoromethylated quinoxalinones on a gram scale. Mechanistic investigations that incorporate radical trapping and ultraviolet/visible spectroscopy, confirmed the formation of the an EDA complex and elucidated the reaction pathways. This study highlights the crucial role of EDA photoactivation in trifluoromethylation, significantly expanding the application scope of EDA complexes in chemical synthesis.
