Abstract:A novel tetralignan, tatarinan T (1) with the rare C8-C7′ linkage pattern, along with a known monolignan (2) were isolated from the roots of Acorus tatarinowii Schott. Their chemical structures were elucidated on the basis of NMR and X-ray diffraction analysis. We evaluated the protective effects of two rare lignans against β-amyloid toxicity by using CL4176 transgenic C. elegans model for the first time, and found that they significantly delayed paralysis of worms at the concentration of 100μM. Compound 2 exhibited the more potential protective effect against β-amyloid toxicity, its value of PT50 extended up to 62.3% at 100μM compared with control, especially, it still has 30.8% extension at 10μM.

Abstract:In our efforts to find potential agents for the treatment of Alzheimer's disease in naturally occurring compounds, a systematic investigation for the active constituents of Flemingia philippinensis was carried out. Four new prenylated isoflavones, philippinone A-D (1-4), together with six known analogues (5-10), were obtained from the roots of Flemingia philippinensis. Their structures were established through extensive physical and spectroscopic data analysis. All the isolates were evaluated for their inhibitory effect of self-induced Aβ aggregation among which compound 5 showed significant Aβ aggregation inhibitory ability with the inhibitory rate of 70.56%. The results of molecular docking experiment for compounds 1 and 6 corresponded to that of the thioflavin-T assay. Moreover, the results further clarified the effects of different substituent group of tested compounds on the Aβ aggregation inhibition. A preliminary structure-activity relationship is further discussed. Our results suggested that the isoflavonoids may mitigate the progression of AD and compounds 2 and 5 may be a candidate in the treatment of AD.

Abstract:Abstract One new flavonol, chlorflavonin A (1), four new diterpenoids, aspergiloids E–H (3, 5–7), together with eight known compounds (2, 4, 8–13) were isolated from solid fermentation of Aspergillus sp. (strain no. YXf3), an endophytic fungus from Ginkgo biloba. Their structures were determined through detailed spectroscopic analysis combined with comparison of NMR spectra data with reported ones. All of them were screened on cytotoxicity against KB, SGC-7901, SW1116, and A549 cell lines; compounds 4, 9–11 exhibited moderate activities with IC50 values ranging from 6.74 to 46.64 µM.

Abstract:Alzheimer's disease (AD) is a slow but progressive neurodegenerative disease. One of the pathological hallmarks of AD is the progressive accumulation of β-amyloid (Aβ) in the form of senile plaques, and Aβ insult to neuronal cells has been identified as one of the major causes of AD onset. In the present study, we investigated the anti-AD potential of four flavonoids, naringenin, didymin, prunin, and poncirin, by evaluating their ability to inhibit acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and β-site amyloid precursor protein cleaving enzyme 1 (BACE1). All four flavonoids displayed promising inhibitory activity against AChE, BChE, and BACE1. Structure–activity relationships suggested that glycosylation of naringenin at sugar moieties, and at different positions of the glycosidic linkage, might be closely associated with anti-AD potential. Kinetic and docking studies showed the lowest binding energy and highest affinity for the mixed, competitive, and non-competitive type inhibitors didymin, prunin, and poncirin. Hydrophobic interactions and the number of hydrogen bonds determined the strength of the protein–inhibitor interaction. We also examined the neuroprotective mechanisms by which flavonoids act against Aβ25–35-induced toxicity in PC12 cells. Exposure of PC12 cells to 10 μM Aβ25–35 for 24 h resulted in a significant decrease in cell viability. In addition, pretreatment of PC12 cells with different concentrations of flavonoids for 1 h significantly reversed the effects of Aβ. Furthermore, treatment with the most active flavonoid, didymin, significantly reduced BACE1, APPsβ, and C99 expression levels in a dose-dependent manner, without affecting amyloid precursor protein (APP) levels in the amyloidogenic pathway. Together, our results indicate that flavonoids, and in particular didymin, exhibit inhibitory activity in vitro, and may be useful in the development of therapeutic modalities for the treatment of AD.

Abstract:Many Chinese herbs are well known for their neuroprotective and anti-oxidant properties. Extracts of Salvia miltiorrhiza and Anemarrhenae asphodeloides, tanshinone IIA (tanIIA), salvianolic acid B (Sal B) and sarsasapogenin (ML-1), were selected to study their dissociation potential towards Aβ42 peptide fibrils and neuroprotective effect on cells. Moreover, derivatives of sarsasapogenin (ML-2, ML-3 and ML-4) have been prepared by the addition of modified carbamate moiety. TanIIA and Sal B have shown to possess a strong ability to dissociate Aβ42 fibrils. The dissociation potential of ML-1 increased upon the introduction of carbamate moiety with N-heterocycles. In silico data revealed that derivatives ML-4 and Sal B interact with Aβ42 regions responsible for fibril stabilization through hydrogen bonds. Contrary, tanIIA binds close to a central hydrophobic region, which may lead to destabilization of fibrils. Sarsasapogenin derivative ML-2 decreased nitride oxide production, and derivative ML-4 enhanced the growth of neurites. The reported data highlight the possibility of using active compounds to design novel treatment agents for Alzheimer's disease.

Common plant flavonoids prevent the assembly of amyloid curli fibres and can interfere with bacterial biofilm formation

Abstract:Like all macroorganisms, plants have to control bacterial biofilm formation on their surfaces. On the other hand, biofilms are highly tolerant against antimicrobial agents and other stresses. Consequently, biofilms are also involved in human chronic infectious diseases, which generates a strong demand for anti-biofilm agents. Therefore, we systematically explored major plant flavonoids as putative anti-biofilm agents using different types of biofilms produced by Gram-negative and Gram-positive bacteria. In Escherichia coli macrocolony biofilms, the flavone luteolin and the flavonols myricetin, morin and quercetin were found to strongly reduce the extracellular matrix. These agents directly inhibit the assembly of amyloid curli fibres by driving CsgA subunits into an off-pathway leading to SDS-insoluble oligomers. In addition, they can interfere with cellulose production by still unknown mechanisms. Submerged biofilm formation, however, is hardly affected. Moreover, the same flavonoids tend to stimulate macrocolony and submerged biofilm formation by Pseudomonas aeruginosa. For Bacillus subtilis, the flavonone naringenin and the chalcone phloretin were found to inhibit growth. Thus, plant flavonoids are not general anti-biofilm compounds but show species-specific effects. However, based on their strong and direct anti-amyloidogenic activities, distinct plant flavonoids may provide an attractive strategy to specifically combat amyloid-based biofilms of some relevant pathogens.

Abstract:Three undescribed isosteroidal alkaloids, przewalskines A − C (1–3), as well as seven known alkaloids (4–10) were obtained from Fritillaria przewalskii bulbs. Their structures were deduced by extensive HRESIMS, 1D NMR, and 2D NMR analyses and their bioactivities were evaluated involving the anti‐inflammatory and inhibitory potencies on AChE, BChE, and Aβ aggregation. Compound 4 revealed the potent effect on inhibiting Aβ aggregation activity with IC50 value of 33.1 μM, AChE activity with IC50 value of 6.9 μM, and also showed NO release inhibitory acitivity with IC50 value of 32.6 μM. These findings contribute new multi‐target anti‐AD agents and embody the chemical diversity of F. przewalskii.

Binding Research on Flavone Derivatives As Probes for Beta-Amyloid Plaques

Abstract:2315 Objectives 26 flavone analogues were afforded as probes for β-amyloid peptides (1~40) using fluorescence titration, demonstrating nano-molar ranges of Kd. Methods Brief Aβ (1~40) dissolved in PBS was under a magnetic stir to result in a visibly cloudy solution (a), confirmed by a transmission electron microscope. Additional test for fibrils formation used ThT (b). Flavone analogues were obtained in a final concentration range of 10~30 nM probes in 500 μL PBS, with 5 μL Aβ(1~40) fibrils. Fibrils were vortexed with each probe for 30 s followed by 1 min incubation before measuring the fluorescence intensity with suitable excitation wavelengthes. Results Flavone analogues showed high affinities in binding to fibrils, such as 9.1 nM of formononetin (c). Conclusions Formononetin is a candidate for Aβ aggregates imaging agent as a labeling precursor. Comparative Transcriptome Analysis to Identify Candidate Genes Related to Chlorogenic Acid and Flavonoids Biosynthesis in Iridaceae

Abstract:Iris (Iridaceae) is one of the most widely admired ornamental plants. It has been used mainly in medicine due to the high concentration of chlorogenic acid (CGA), flavonoids, isoflavones, lignans, and other compounds in its rhizomes. In iris, the gene functions related to CGA and flavonoids biosynthesis are still unclear. In this study, we compared the I. germanica rhizome with a high accumulation level of CGA but a low accumulation level of flavonoids, and the I. pallida rhizome with a low accumulation level of CGA but a high accumulation level of flavonoids at the transcriptome and metabolome levels. A total of 761 metabolites were detected, including 202 flavonoids and 106 phenolic acids based on metabolome profiling. In total, 135 flavonoids were highly accumulated in I. pallida, including three flavanols, 51 flavonoids, 12 flavonoid carbonosides, 31 flavonols, and 21 isoflavones. Based on single-molecule long-read sequencing technology, 94,461 transcripts were identified in iris. Expression analysis indicated that the high accumulation level of C4H and 4CL in I. germanica were essential for CGA accumulation, while CHS, DFR, ANS, ANR, LAR, and 3GT were essential for flavonoids biosynthesis in I. pallida. Many transcription factors such as transcript_83288 (MYB), transcript_57970 (WRKY), and transcript_77465 (WRKY) act as regulators, playing important roles in these biological processes. Our findings provide new insights into the molecular mechanisms associated with the biosynthesis and regulation of flavonoids and CGA in the iris rhizome, and highlight the usefulness of an integrated approach for understanding this process.

Abstract:Eight cyclopenta[ b ]benzofurans ( 1 , 2 , 4 , and 5 - 9 ) and eight cyclopenta[ bc ]benzopyrans ( 3 , 10 - 16 ), including a revised ( 4 ) and three undescribed compounds ( 1 - 3 ), were isolated from the twigs and leaves of Aglaia edulis (Roxb.) Wall. Their structures were determined by a combination of spectral analysis in conjunction with NMR and ECD calculations. Moreover, based on the findings from 13 C NMR calculations and DP4+ statistical analysis, an empirical guideline was established to differentiate the structures of cyclopenta[ bc ]benzopyrans and cyclopenta[ b ]benzofurans by aggregating chemical shift data from known compounds. This guideline facilitated the proposal of structural revisions for three previously reported analogs ( R-1 , R-2 , R-3 ). Biological assay indicated that cyclopenta[ b ]benzofuran flavalines ( 2 , and 4-8 ) could significantly inhibit NO production in LPS-induced BV-2 cells with IC 50 values from 0.002 to 0.05 μM.

Abstract:Corallodiscus flabellata B. L. Burtt (CF) is a Chinese folk herb with reported potential for the treatment of Alzheimer's disease (AD). 3,4-Dihydroxyphenylethanol-8-O-[4-O-trans-caffeoyl-β-D-apiofuranosyl-(1→3)-β-D-glucopyranosyl (1→6)][1]-β-D-glucopyranoside (SDC-1-8) and hydroxytyrosol (HT) are two polyphenolic compounds isolated from CF. The aim of this study was to investigate the protective effects of SDC-1-8 and HT on an Aβ 25-35 -induced AD model and to study the underlying mechanism. The AD mouse model was established using a brain injection of amyloid β-protein 25-35 (Aβ 25-35 , 200 μM), followed by continuous administration of SDC-1-8 and HT for 4 weeks, and found that they improved cognitive dysfunction; ameliorated neuronal damage and apoptosis; decreased oxidative stress, and mitochondrial fission protein levels; and increased mitochondrial fusion protein levels in AD mice. Moreover, SDC-1-8 and HT inhibited mitochondrial membrane depolarization, reduced intracellular stored Ca 2+ levels, enhanced mitochondrial respiration, increased mitochondrial fusion, and decreased mitochondrial division in Aβ 25-35 -induced PC12 cells even in the presence of mdivi-1. Furthermore, molecular docking simulations showed that SDC-1-8 and HT interacted with dynamin-related protein 1 with higher affinity than mitofusin 1. Thus, it is summarized that SDC-1-8 and HT may have neuroprotective effects by balancing the abnormalities of mitochondrial fission and fusion, and SDC-1-8 and HT are the components providing the therapeutic basis of CF.

Abstract:Long-term consumption of phytochemicals has been associated with a decreased risk of dementia. The modes of action of two flavonols (morin and isoquercitrin) and two flavanones (hesperidin and neohesperidin) were characterized as single-compound drugs in several Alzheimer's disease (AD)-related assays. First, these phytochemicals were assayed in an amyloid toxicity model (MC65 cells). Second, we examined the activity of the flavonoids in cell-free assays against β- and γ-secretases and acetylcholinesterase activities, as well as agents able to modify the fibrillogenesis of the amyloid β-peptide. Additionally, they were assayed against glutamate-induced oxytosis, as scavengers of reactive oxygen species (ROS), as inhibitors of caspase-3, -8 and -9 activation and as modulators of the chymotrypsin-like activity of the ubiquitin-proteasome system. Morin and isoquercitrin, unlike flavanones, exhibited significant activities as β- and γ-secretase inhibitors, as well as capacity to inhibit Aβ aggregation and favor its disaggregation. Flavonols and flavanones showed ROS scavenger activity (P < 0.05), attenuation of caspase-3 and -9 activation (P < 0.05) and restoration of the reduced chymotrypsin-like activity of proteasome 20S (P < 0.05) upon H2O2 exposure of APPswe cells. Flavanones failed to protect against glutamate-induced oxytosis. These findings provide new insight into the anti-amyloidogenic effects of morin and isoquercitrin.

Abstract:Nine undescribed compounds, along with eight known compounds, were isolated from the stipes of Lentinus edodes . Their structures were established by extensive spectroscopic and circular dichroism analyses. The protective effects against A β 25-35 -induced N9 microglia cells injury of these compounds were tested by MTT method, and the levels of apoptosis and ROS were detected by flow cytometry. In addition, the binding sites and interactions of compound with amyloid precursor protein were revealed using molecular docking simulations. These findings further establish the structural diversity and bioactivity of stipes of L . edodes , and provide an experimental basis for targeting Alzheimer's disease as a potential strategy.

Abstract:Eubacterium limosum ZL-II is an anaerobic bacterium with demethylated activity, which was isolated from human intestinal bacteria in our previous work. In this study, the flavonolignan constituents of Silybi Fructus were biotransformed by E. limosum(1) ZL-II, producing four new transformation products - demethylisosilybin B (T1), demethylisosilybin A (T2), demethylsilybin B (T3) and demethylsilybin A (T4), among which T1 and T2 were new compounds. Their chemical structures were identified by ESI-TOF/MS, (1)H NMR, (13)C NMR, HMBC and CD spectroscopic data. The bioassay results showed that the transformation products T1-T4 exhibited significant inhibitory activities on Alzheimer's amyloid-β 42 (Aβ42(2)) aggregation with IC50 values at 7.49 μM-10.46 μM, which were comparable with that of the positive control (epigallocatechin gallate, EGCG(3), at 9.01 μM) and much lower than those of their parent compounds (at not less than 145.10 μM). The method of biotransformation by E. limosum ZL-II explored a way to develop the new and active lead compounds in Alzheimer's disease from Silybi Fructus. However, the transformation products T1-T4 exhibited decreased inhibitory activities against human tumor cell lines comparing with their parent compounds.

Abstract:The fungus strain DZ-3 was isolated from twigs of the well-known medicinal plant Eucommia ulmoides Oliver and identified as Aspergillus flavipes. Two new alkaloids, named asperflaloids A and B (1 and 2), together with 10 known compounds (3–12) were obtained from the EtOAc extract of the strain. Interestingly, the alkaloids 1–4 with different frameworks are characterized by the presence of the same anthranilic acid residue. The structures were established by detailed analyses of the spectroscopic data. The absolute configuration of asperflaloids A and B was resolved by quantum chemistry calculation. All compounds were screened for their inhibitions against α-glucosidase and the antioxidant capacities. The results were that compound 3 had an IC50 value of 750.8 μM toward α-glucosidase, and the phenol compounds 7 and 8 exhibited potent antioxidant capacities with IC50 values 14.4 and 27.1 μM respectively.

Abstract:Abstract: Alzheimer’s disease is one of the neurodegenerative diseases which causes cognition deficit. There are currently few medications available to treat Alzheimer's disease, even though researchers have devoted a great deal of time studying the condition and offering many benefits. Thus, only a few drugs are available for the treatment of Alzheimer’s disease. Amentoflavone is a dietary component found in many plants and herbs that has several health advantages. Amentoflavone has demonstrated strong protective benefits against a range of brain illnesses in preclinical trials, most frequently in Alzheimer's disease. Amentoflavone, a biflavonoid, can be identified in a variety of herbs upon isolation. Considering the beneficial properties of this compound, this review emphasizes the pharmacological effects and botanical sources of amentoflavone, as well as the compound's benefits and possible applications in the treatment of Alzheimer's disorders.
