Abstract:OBJECTIVE To study the flavonoids of Artemisia anomala S. Moore. METHODS The compounds were isolated by various column chromatography, and their structures were elucidated on the basis of spectral analysis and physical-chemical properties.RESULTS Eleven flavonoids were obtained and identified as isorhamnetin(Ⅰ), kaempferol(2), eupatorin(3), luteolin(4), apige-nin (5), quercetin (6) , cosmosiin (7), kaempferol-3-ο-α-L-rhamnopyranoside (8), luteolin-7-ο-β-D-glucopyranoside (9), quercetin-3-ο-α-L-rhamnopyranoside(10) and avicularin(11). CONCLUSION All the compounds were obtained from this plant for the first time, in addition, compound 8 and 11 were obtained from this genus for the first time.

Abstract:Thirty-four reported compounds were obtained from Anemone vitifolia Buch.-Ham., and were identified as triterpenoid saponins (1–28), flavonoid glycosides (29–32) and steroidal saponins (33–34). The structure of these compounds was determined by physicochemical constants and spectral analyses (NMR, MS, IR). Twelve compounds (7, 8, 10, 14, 20, 21, 23, 26–28, 30 and 32) were firstly identified in genus Anemone. Compounds 2, 4 and 14 can be considered as characteristic components of A. vitifolia. Finally, the chemotaxonomic significance of these compounds is discussed.

Abstract:In the last decades interest in the group of flavonoids has markedly increased due to their presumed beneficial effects in humans. Many flavonoids act as radical scavengers and are suggested to prevent chronic diseases such as atherosclerosis or even cancer [1]. For a critical evaluation of the effects of flavonoids with regard to human health, quantitative (daily intake as part of diet or as food supplements) as well as qualitative (structural) aspects have to be taken into consideration [2]. As part of a broader Sino-German project on flavonoids from plants used for human consumption, we analyzed four different cultivars of Vigna angularis (Willd.) Ohwi & H. Ohashi, two of them growing in China (Hangzhou) and the other two cultivated in Germany (Düsseldorf). All analyzed plants yielded catechin, myricetin–3-O–rutinoside, and rutin as main compounds but differed with regard to their total flavonoid concentrations which were lower in the Chinese cultivars. We analyzed pharmacological effects of these compounds focussing mainly on their antioxidative properties. All flavonoids exhibited a good radical scavenger activity in a cell free system (DPPH assay). In different cellular systems (rat H4IIE hepatoma and C6 glioma cells) we further analyzed protective effects of these substances against oxidative stress using the fluorescent probe DCF. Beside these antioxidative effects, cytotoxic and pro-apoptotic activities of these compounds were also determined. In consideration of these data it can be emphasized that a known composition of flavonoids is necessary in nutrient uptake.

Abstract:Anemone vitifolia is a small herb found in Asia that is used to treat a range of diseases in Chinese traditional medicine. GNPS-based molecular networking of an Anemone vitifolia specimen revealed the presence of a network containing numerous ions indicating the presence of lignans, several of which suggested that there might be previously undescribed compounds in the extract. Fractionation of the organic extract yielded five undescribed lignans, the vitifolignans, together with one known. The structures were identified based on extensive spectroscopic data analysis (NMR, HR-ESI-MS, and UV), coupling constant calculation and comparison with reported data. Their absolute configurations were determined by comparison of experimental ECD spectra with calculated spectra. Compounds 4/5 showed weak inhibition of LPS-induced NO production in mouse mononuclear macrophages.

Abstract:Phytochemical investigation from the roots of Anemone vitifolia Buch.-Ham. led to the isolation of eight compounds, including one triterpenoid saponin, two sugars, one coumarin, one amide, one saturated alkane, one olefin and one fatty acid. The structures of these metabolites were elucidated by spectroscopic data and comparisons with the data available in the literature. Among them, compound 7 ((6Z,9Z,12Z)-6,9,12-Eicosatriene) was isolated for the first time as a natural product. Furthermore, compounds 2 (D-(+)-raffinose), 3 (mixture of beta-D-fructopyranose and beta-D-fructofuranose) and 5 (bonaroside) were obtained from the Ranunculaceae family for the first time. Compounds 4 (siderin) and 6 (n-hexadecane) were isolated from A. vitifolia for the first time. The chemotaxonomic significance of the isolated compounds was discussed.

Abstract:Flavonoids and volatiles in a traditional herbal medicine Chrysanthemum morifolium Ramat flower were determined by HPLC and GC/MS, respectively. Eight flavonoids and fifty eight volatiles were identified. Luteolin-7-glucoside and quercitrin were the most abundant flavonoids and they amounted for 85.7% of the total detected flavonoids. -Humulene was the most abundant volatile and ledene oxide-(I) the next abundant one and the two volatiles were 16.3 and 9.0% of total volatiles, respectively. The bioactivities of the partial components in C. morifolium Ramat flower were discussed. It is considered that the health benefits of C. morifolium Ramat might be related to its abundant flavonoids and volatiles.
Flavonoid Metabolism in Tetrastigma hemsleyanum Diels et Gilg Based on Metabolome Analysis and Transcriptome Sequencing

Abstract:Tetrastigma hemsleyanum Diels et Gilg, known as a "plant antibiotic", possesses several attractive properties including anti-inflammatory, anti-tumor, and antioxidant effects, with its efficacy being attributed to flavonoids. However, the flavonoid biosynthesis of T. hemsleyanum has rarely been studied. In this study, we investigated the flavonoid metabolism of T. hemsleyanum through metabolome analysis and transcriptome sequencing. The metabolomic results showed differences in the flavonoids of the leaves and root tubers of T. hemsleyanum. A total of 22 flavonoids was detected, and the concentrations of most flavonoids in the leaves were higher than those in the root tubers. Transcriptome analysis revealed that differentially expressed genes (DEGs) in the leaves and root tubers were enriched in photosynthesis-antenna proteins. Pearson correlation analysis indicated that the expression levels of chalcone isomerase (CHI) and UDP-glycose flavonoid glycosyltransferase (UFGT) were highly correlated with the concentrations of most flavonoids. Further, this study found that the photosynthesis-antenna proteins essentially contributed to the difference in the flavonoids in T. hemsleyanum. The gene expressions and concentrations of the total flavonoids of leaves and root tubers in Hangzhou, Jinhua, Lishui, and Taizhou in Zhejiang Province, China, showed that CHI (CL6715.Contig1_All, Unigene19431_All, CL921.Contig4_All) and UFGT (CL11556.Contig3_All, CL11775.Contig1_All) were the potential key genes of accumulation of most flavonoids in T. hemsleyanum.

Uncovering nutritional metabolites and candidate genes involved in flavonoid metabolism in Houttuynia cordata through combined metabolomic and transcriptomic analyses

Abstract:The perennial herb Houttuynia cordata has long been cultivated and used as medicinal and edible plant in Asia. Nowadays, increasing attention is attracted due to its numerous health benefits. Flavonoids are the main chemical constituents exerting pharmacological activities. In the present study, we investigated both metabolome and transcriptome of two H. cordata accessions (6# and 7#) with distinct flavonoids contents. In total 397 metabolites, i.e., 220 flavonoids, 92 amino acids and derivatives, 20 vitamins, and 65 saccharides were abundant in aboveground part. Cyanidin-3-O-rutinoside and quercetin-3-O-galactoside were the most abundant flavonoids, which can be categorized into seven classes, namely anthocyanidins, chalcones, flavanols, flavanones, flavanonols, flavones, and flavonols. Flavonols was the most abundant group. Contents of 112 flavonoids differed significantly between the two accessions, with catechin-(7,8-bc)-4α-(3,4-dihydroxyphenyl)-dihydro-2-(3H)-one, cinchonain Id, and cinchonain Ic being the dominant flavonoid metabolites among them. Pinocembrin-7-O-neohesperidoside, pinocembrin-7-O-rutinoside, and kaempferol-3-O-galactoside-4'-O-glucoside were uniquely abundant in accession 7. Transcriptome data revealed a total of 110 different expressed genes related to flavonoid metabolism, with more highly expressed genes observed in 7#. We annotated a total of 19 differential flavonoid metabolites and 34 differentially expressed genes that are associated with the flavonoid metabolic network. Based on the transcriptome and qPCR data a total of 8 key candidate genes involved in flavonoid metabolism were identified. The ANS gene were found to play an important role in the synthesis of cyanidin-3-O-glucoside, while the CHI, F3'H and FLS genes were mainly responsible for controlling the levels of flavanones, flavones, and flavonols, respectively. Collectively, the present study provides important insights into the molecular mechanism underlying flavonoid metabolism in H. cordata.

Metabolomics and transcriptomics provide insights into the flavonoid biosynthesis pathway in the roots of developing Aster tataricus

Abstract:Aster tataricus (L.) is an important medicinal plant in China. Its roots are rich in flavonoids, the main medicinal components. However, the molecular basis of flavonoid biosynthesis in the roots of A. tataricus remains unclear. In this study, the content of total flavonoid of A. tataricus roots at different developmental stages was measured first, and the results showed that the content of total flavonoid gradually decreased from September to November, which may be caused by the stagnation of A. tataricus growth due to the decrease in temperature after September. Then, an integrated analysis of transcriptome and metabolome was conducted on five developing stages of A. tataricus roots to identify flavonoid compositions and potential genes involved in flavonoid biosynthesis. A total of 80 flavonoid metabolites, of which 75% were flavonols and flavonoids, were identified in metabolomic analyses, among which isorhamnetin, kaempferol, quercetin, and myricetin were the main skeletons of these flavonoids. Cluster analysis divided these 80 flavonoids into 3 clusters. The compounds in cluster I mainly accumulated in S1, S3, and S5. In cluster II, the relative content of the flavonoid metabolites showed an upward trend from S2 to S4. In cluster III, the flavonoids decreased from S1 to S5. A total of 129 structural genes, including 43 PAL , 23 4CL , 9 C4H , 4 CHS , 18 CHI , 3 F3H , 5 F3'H , 1 F3′5′H , 21 FLS , and 2 FSII , and 65 transcription factors, including 22 AP2/ERF , 7 bHLH , 5 bZIP , 8 MYB , 11 NAC , and 12 WRKY , showed significant correlation with total flavonoid content. Eighteen genes (7 4CL , 5 C4H , 2 CHI , 1 F3H , and 3 FLS ) and 30 genes (5 PAL , 9 4CL , 1 C4H , 2 CHI , 1 F3H , 1 DFR , 7 3AT , 1 BZ1 , and 3 UGT79B1 ) were identified as key structural genes for kaempferol and anthocyanins biosynthesis, respectively. Our study provides valuable information for understanding the mechanism of flavonoid biosynthesis in A. tataricus root.

Abstract:Tetrastigma hemsleyanum Diels et Gilg is a folk herb in Zhejiang Province with anti-inflammatory, antineoplastic, and anti-oxidation effects. Given its pharmacological activity, T. hemsleyanum is known as New "Zhebawei" and included in the medical insurance system of Zhejiang and other provinces. Flavonoids are the most important components of T. hemsleyanum, and their contents are mainly regulated by ultraviolet (UV) radiation. In this study, the total flavonoid contents, flavonoid monomer contents, and flavonoid synthesis related enzyme activities (phenylalanine ammonia-lyase, chalcone synthase, and chalcone isomerase), anti-oxidant enzyme activities (catalase, peroxidase, and superoxide dismutase), and biochemical indicators (malondialdehyde, free amino acid, soluble protein, and soluble sugar) in the leaves (L) and root tubers (R) of T. hemsleyanum with UV treatments were determined. Three kinds of UV radiation (UV-A, UV-B, and UV-C) and six kinds of radiation durations (15 and 30 min, 1, 2, 3, and 5 h) were used. Appropriate doses of UV-B and UV-C radiation (30 min to 3 h) induced eustress, which contributed to the accumulation of flavonoids and improve protective enzyme system activities and bioactive compound contents. Especially, certain results were observed in several special structures of the flavonoid monomer: quercetin contents in L increased by nearly 20 times, isoquercitrin contents in R increased by nearly 34 times; most of flavonoids with glycoside content, such as quercitrin (19 times), baicalin (16 times), and apigenin-7G (13 times), increased multiple times. Compared with the CK group, the flavonoid synthase activities, anti-oxidant enzyme activities, and biochemical substance contents in L and R all increased with UV treatments. This study provides a theoretical foundation for regulating flavonoids by light factors and improving the quality of T. hemsleyanum in production and medical industries.

Abstract:Chemical constituents in the leaves of Sabina vulgaris Antoine were studied.Nine flavonoids were isolated and purified by extraction and different kinds of column chromatography,and their structures were elucidated as cupressuflavone(Ⅰ),cupressuflavone 4',4-dimethyl ether(Ⅱ),amentoflavone(Ⅲ),podocarpusflavone A(Ⅳ),catechin(Ⅴ),quercetin(Ⅵ),isoscutellarein 7-O-β-D-xylopyranoside(Ⅶ),myricetin 3-O-β-D-glucoside(Ⅷ),rutin(Ⅸ).CompoundsⅠ,Ⅱ,Ⅴ,Ⅶ,Ⅷ and Ⅸ were obtained from this plant for the first time.Compounds Ⅷ and Ⅸ were isolated from Sabina genus for the first time.All of isolated compounds were tested for their antioxidative activities by 1,1-diphenyl-2-picrylhydrazyl free radical(DPPH·)scavenging assay.It was found that compounds Ⅴ and Ⅵ showed antioxidative activity close to that of positive control,L-ascorbic acid,and the flavonoids glycosides(Ⅶ,Ⅷ and Ⅸ)are more active than the biflavones(Ⅰ,Ⅱ,Ⅲ and Ⅳ).

Abstract:AIM: To study the chemical constituents from the stems of Aquilaria sinensis (Lour.) Gilg. METHODS: Chromatographic separations of silica gel, Sephadex LH-20 and prep-HPLC were used. All the structures were elucidated on the basis of chemical and spectroscopic analysis. RESULTS: Twelve flavonoids were isolated and their structures were identified as aquilarinoside AI (1), lethedioside A (2), 7, 4'-dimethylapigenin-5-O-xylosylglucoside (3), lethedoside A (4), 7-hydroxyl-4'-methyl-5-O- glucosideflavonoid (5), 7, 3'-dimethyl-4'-hydroxyl-5-O-glucosideflavonoide (6), 7, 4'-dimethyl-5-O-glucosideflavonoide (7), 5-hydroxyl-7, 4'-dimethoxyflavone (8), luteolin-7, 3', 4'-trimethyl (9), hydroxylgenkwanin (10), 5, 7-dihydroxyl-4'-methoxyflavone (11), and formononetin (12). CONCLUSION: Compound 1 is a new flavone glycoside, and compounds 2, 4-7 were isolated from genus Aquilaria sinensis for the first time. 5-O-glycosylflavones 1-7 showed inhibitory activity for nitric oxide (NO) production by activated RAW 264.7 cells.

The structures of the new compounds were elucidated as 3- O -β- d -ribopyranosyl-(1→3)-α- l -rhamnopyranosyl-(1→2)-[β- d -glucopyranosyl-(1→4)]-α- l -arabinopyranosyl hederagenin 28- O -α- l -rhamnopyranosyl-(1→4)-β- d -glucopyranosyl-(1→6)-β- d -glucopyranoside ( 1 ), 3- O -β- d -ribopyranosyl-(1→3)-α- l -rhamnopyranosyl-(1→2)-[β- d -glucopyranosyl-(1→4)]-β- d -xylopyranosyl hederagenin 28- O -α- l -rhamnopyranosyl-(1→4)-β- d -glucopyranosyl-(1→6)-β- d -glucopyranoside ( 2 ) and 3- O -β- d -galactopyranosyl-(1→3)-α- l -rhamnopyranosyl-(1→2)-β- d -xylopyranosyl oleanolic acid 28- O -α- l -rhamnopyranosyl-(1→4)-β- d -glucopyranosyl-(1→6)-β- d -glucopyranoside ( 3 ) on the basis of chemical and spectral evidence. In the oligosaccharide chains of compound 3 , the characteristic d -galactose residue is a rare structural feature and secondly encountered among triterpenoid saponins from Anemone.

Biochemical Systematics and Ecology

Abstract:A phytochemical investigation of Anemone vitifolia (Buch.-Ham.) led to the isolation of twelve compounds, including six lignans (1-6), four benzoic acid esters (7-10), one norsesquiterpenoid (11) and one lactone (12). The structures of these metabolites were established on the basis of detailed spectroscopic analysis, as well as comparisons with the data available in the literature. Among them, compounds 10 (dimethyl 2-(benzoyloxy) succinate) and 12 (4-hydroxy-5-methyl-gamma-butyrolactone) were isolated for the first time as natural products. Compounds 1 and 11, compounds 2, 3, 5-7 and 9, and compounds 4 and 8 were isolated for the first time in family Ranunculaceae, genus Anemone and A. vitifolia respectively. The chemotaxonomic significance of the isolated compounds was discussed.
