Abstract:OBJECTIVE:To study the chemical constituents of Abelmoschus manihot.METHOD:Chromatographic methods were used to isolate compounds from A. manihot, and spectroscopic methods were used to identify the structures.RESULT:Thirteen compounds, myricetin (1), cannabiscitrin (2), myricetin-3-O-beta-D-glucopyranoside (3), glycerolmonopalmitate (4), 2, 4-dihydroxy benzoic acid (5), guanosine (6), adenosine (7), maleic acid (8), heptatriacontanoic acid (9), 1-triacontanol (10) , tetracosane (11), beta-sitosterol (12), beta-sitosterol-3-O-beta-D-glucoside (13) were obtained.CONCLUSION:2-11 were obtained from the genus for the first time.

Abstract:Flower of Abelmoschus manihot (FAM) is clinically effective to treat chronic kidney disease (CKD) with a relatively high dosage. To improve the efficacy and the compliance of patients, macroporous resins were adopted to enrich and purify flavonoids from FAM, which are thought to be the major renal protective constituents in FAM. After screening six different kinds of macroporous resins, HPD-100 was selected for its great adsorption and desorption capacity. Then, orthogonal design tests were used to optimize parameters in the processes of impurity removal and flavonoids of FAM desorption on column chromatogram. Moreover, process scale-up was performed, and purification effects maintained after amplification. After purification, the content of seven main flavonoids in the product increased from 8.29% to 51.43%. Protective and anti-inflammatory effects of crude extract and the flavonoid component of FAM after purification were investigated on the adriamycin-damaged HK-2 cells and lipopolysaccharide-stimulated Raw 264.7 cells models. Both bioactivities were improved greatly after purification for these two cell models. Therefore, the purification process had enriched the main bioactive constituents with potential alleviating kidney injury activities. The flavonoid component of FAM is worthy of being developed as an improved remedy for CKD with better patients' compliance.

Abstract:A new flavonoid glucoside was isolated from the aerial parts of Alhagi pseudoalhaki(M.B.) Desv..On the basis of spectral data and chemical reaction,it was elucidated to be syringetin-3-O-beta-D-glucoside.Moreover,fourteen known compounds were isolated and identified as beta-sitosterol,stigmasterol,kaempferol,rhamnetin,ombuine,isorhamnetin,tamarixetin,kaempferol-3-O-beta-D-(6-O-p-coumaroyl)-glucosiole, isoquercitrin, D-3-O-methylinositol, 1-O-betalmethyl-glucoside,isoswertianolin,isorhamnetin-3-O-beta-D-rutinoside,and tyramine.These compounds were isolated for the first time from the aerial parts of A.pseudoalhagi.
Simultaneous Determination Of Seven Active Flavonols In The Flowers Of Abelmoschus Manihot By Hplc

Abstract:A high-performance liquid chromatography method is developed for the simultaneous quantification of seven flavonols, namely quercetin-3-O-robinobioside, hyperin, isoquercetin, hibifolin, myricetin, quercetin-3'-O-glucoside, and quercetin, in the flower of Abelmoschus manihot. These seven flavonols are selected as chemical markers because they are the major pharmacologically active constituents in the flower. The method involves the use of a Thermo ODS-2HYEPRSIL reversed-phase column (5 pm, 250 x 4.6 mm) at 25 degrees C with a mixture of acetonitrile and aqueous H3PO4 as the mobile phase and detection at 370 nm. The recovery of the method is 94.31-107.08% with an RSD <= 3.14% and the linearity (r(2) > 0.9996) is obtained for all the flavonoids. The current assay method can be readily utilized for the determination of the flavonols present in the flower and is considered to be suitable for the quality control of A. manihot samples. The comparison of flowers collected from nine locations shows that flavonoid glucoside is more stable than aglycon in the flower. This is the first study that analyzes the stability of flavonoids in the flower of A. manihot. This research also provides important evidence that the flower is a potentially abundant resource for obtaining hibifolin.

Abstract:Flavonol glycosides are health-promoting phytochemicals important for human nutrition and plant defense against environmental stresses. Glycosylation modification greatly enriches the diversity of flavonols. Morella rubra, a member of the Myricaceae, contains high amounts of myricetin 3-0-rhamnoside (M3Rha), quercetin 3-0-rhamnoside (Q3Rha), and quercetin 3-0-galactoside (Q3Ga1). In the present study, MrUGT78R1 and MrUGT78R2 were identified as two functional UDP-rhamnosyltransferases, while MrUGT78W1 was identified as a UDP-galactosyltransferase. Site-directed mutagenesis identified Pro143 and Asn386 as important residues for rhamnosyl transfer activity of MrUGT78R1, while the two corresponding positions in MrUGT78W1 (i.e. Ser147 and Asn370) also play important roles in galactosyl transfer activity. Transient expression data for these three MrUGTs in Nicotiana benthamiana tested the function of MrUGT78R1 and MrUGT78R2 as rhamnosyltransferases and MrUGT78W1 as a galactosyltransferase in glycosylation of flavonols. This work enriches knowledge of the diversity of UDP-rhamnosyltransferase in planta and identifies two amino acid positions important for both rhamnosyltransferase and galactosyltransferase.

Abstract:Introduction: Huangkui capsule (HKC) is made from the ethanol extract of Abelmoschus manihot (L.) Medik [Malvaceae; abelmoschi corolla] and received approval from the China Food and Drug Administration (Z19990040) in 1999. Currently, HKC is used for treatment of the patients with diabetic nephropathy (DN) in China. The bioactive chemical constituents in HKC are total flavonoids of A. manihot (L.) Medik (TFA). The present study aims to identify the primary flavonoid metabolites in HKC and TFA and their metabolism fates in db/db mice, the animal model for the study of type 2 diabetes and DN. Methods: HKC (0.84 g/kg/d) and TFA (0.076 g/kg/d) or vehicle were respectively administered daily via oral gavage in db/db mice for 4 weeks. The metabolism fate of the main metabolites of HKC in serum, liver, kidney, heart, jejunum, colon, jejunal contents, colonic contents, and urine of db/db mice were analyzed with a comprehensive metabolite identification strategy. Results and Discussion: In db/db mice administered with HKC and TFA, 7 flavonoid prototypes and 38 metabolites were identified. The related metabolic pathways at Phases I and II reactions included dehydroxylation, deglycosylation, hydrogenation, methylation, glucuronidation, sulphation, and corresponding recombined reactions. Quercetin, isorhamnetin, quercetin sulphate, quercetin monoglucuronide, and isorhamnetin monoglucuronide presented a high exposure in the serum and kidney of db/db mice. Thereby, the present study provides a pharmacodynamic substance basis for better understanding the mechanism of A. manihot (L.) Medik for medication of DN.

Abstract:Flavonoids, natural products derived from plants, have shown promising potential in food and medicine. These compounds are commonly found in the form of glycosides, which exhibit increased solubility and improved bioavailability. The glycosylation of flavonoid is mainly catalyzed by UDP-glycosyltransferase (UGT). In this study, a new UDP-glucosyltransferase UGT73AC11 from Glycyrrhiza uralensis was identified which can efficiently transfer a glucose or xylose moiety to the 7-OH or 4'-OH position of flavonoids to yield glycosides. The biochemical properties of UGT73AC11 were extensively characterized. The substrate binding mechanism was also explored through molecular dynamics simulation. Finally, UGT73AC11 was successfully coupled with a sucrose synthase to synthesize the monoglucosides of liquiritigenin with sucrose as sugar donor, with 2.1 mM neoliquiritin produced, which was the highest level reported so far. Therefore, UGT73AC11 held great potential as a broad-spectrum flavonoid biocatalyst for large-scale synthesis of flavonoid glycosides.

[Antidepressant activity of flavonoid ethanol extract of Abelmoschus manihot corolla with BDNF up-regulation in the hippocampus]

Abstract:Abelmoschus manihot (L.) Medic., a folk herbal medicine in China, is a flowering plant belonging to Abelmoschus L. genus and Malvaceae family, which has been reported with an antidepressant activity. The study was designed to isolate flavonoids from Abelmoschus manihot corolla and explore the action mechanism of antidepressant activities. The flavonoids were isolated and purified by D101 macroporous resin column, polyamide column and Sephadex LH-20 sequentially and identified as myricetin-3-O-β-D-glucoside (1), gossypetin-8-O-β-D-glucuronide (2, G-8-G), gossypetin-3'-O-β-D-glucoside (3), quercetin-3'-glucoside (4, Q-3-G), isoquercitrin (5, IQT), hyperoside (6, HY), myricetin (7), quercetin (8, QT). Compounds 2, 4, 5, 6 and 8 (15, 30 and 60 mg·kg−1) were orally administered to mice and the reaction was observed in tail suspension test (TST) and forced swimming test (FST). Western blot analysis was used in determination of the protein expressions of brain-derived neurotrophic factor (BDNF), tyrosine receptor kinase B (TrkB) and phosphorylation eukaryotic elongation factor 2 (p-eEF2). The results revealed that only Q-3-G and G-8-G (15, 30, 60 mg ·kg−1) significantly reduced the immobility time in FST and TST. Furthermore, Q-3-G and G-8-G remarkably increased the expression of BDNF and TrkB, and decreased the expression of p-eEF2. These results suggest that Q-3-G and G-8-G had an obvious antidepressant activity via up-regulation of BDNF expression. The new observation will provide a new direction in the development of antidepressant in the treatment of major depressive disorder (MDD).

Abstract:BACKGROUND: This research will establish the ultraviolet colorimetric method to determine the total flavonoid content in different species and different parts of Abelmoschus esculentus L.MATERIALS AND METHODS: We establish the reversed-phase high-performance liquid chromatograph (RP-HPLC) method to determine the content of the three flavonoid glycosides in different species and different parts of the A. esculentus. Adopt the NaNO2-Al (NO3)3-NaOH colorimetric method to determine the total flavonoid content; at the same time, adopt the RP-HPLC method to determine the contents of the three flavonoid glycosides. Using the methods of ultraviolet colorimetry and RP-HPLC, we determined and analyzed the total flavonoid content and the content of the three flavonoid glycosides in different species and different parts of A. esculentus.RESULTS: There are great distribution differences of the total flavonoids and the three flavonoid glycosides in different species and parts of A. esculentus. Among them, the content of the effective constituents in the flower is relatively high, next is in the fruit. In the different species of A. esculentus, the content of the flavonoids of finger relatively high. The HPLC method established in this research is simple and convenient and its results are accurate and reliable. In addition, it has a very good repeatability.CONCLUSION: The results provided the reference data for the medicinal use of A. esculentus and it can be used in quality analyzing of its effective constituents.

Abstract:The fruits of Alpinia oxyphylla has been used a famous traditional Chinese medicine. A chemical investigation on the fruits of A. oxyphylla resulted in the isolation of one new acetylated flavanol glucuronide (1) and two known compounds (2-3). Compound 1 displayed a weak inhibitory effect on U251 cells with the IC50 value 128.51 μM while the positive control drug temozolomide exhibited the IC50 value 35.37 μM.

Abstract:Several kinds of column chromatography method were used to investigate the chemical constituents of the leaves of Lophatherum gracile. The structures of the isolated compounds were identified based on their physicochemical properties and spectral data. A new flavone C-glycoside was isolated and its structure was identified as 3'-methoxyl-luteolin 6-C-beta-D-galactopyranosiduronic acid (1 --> 2) -alpha-L-arabinopyranoside (1).
Conjugated metabolites represent the major circulating forms of Abelmoschus manihot in vivo and show an altered pharmacokinetic profile in renal pathology

Abstract:Context: The nephron-protective efficacy of Abelmoschus manihot (Linn.) Medicus (Malvaceae) has been proved by randomized controlled clinical trial. Objective: Flavonoids are main active components of A. manihot, which can be transformed into glucuronide/sulfate conjugates in vivo. Exploring the pharmacokinetic profile of these conjugates is necessary to further elucidate the mechanism of action. Material and method: Flavonoid fraction of A. manihot (FFA) was extracted from A. manihot flower with ethanol. FFA (400 mg/kg) was orally given to normal rats and chronic kidney disease (CKD) model rats. Blood samples were collected at 5, 15, 30, 45, 60, 90, 120, 240, 360, and 720 min after administration. The plasma concentrations of quercetin and isorhamnetin glucuronide/sulfate conjugates were analyzed by UPLC-MS/MS. Results: In normal rats, AUC of quercetin-glucuronide conjugates, isorhamnetin-glucuronide conjugates, quercetin-sulfate conjugates, and isorhamnetin-sulfate conjugates was 459.45 ± 192.70, 1153.01 ± 697.04, 417.81 ± 220.31, and 2475.19 ± 1085.22 μmol h/L, respectively. While AUC of quercetin and isorhamnetin was 5.47 ± 2.54 and 30.73 ± 25.95 μmol h/L. AUC of the glucuronide-sulfate conjugates of quercetin and isorhamnetin is 125-times higher than that of aglycone (quercetin and isorhamnetin), showing that glucuronide/sulfate conjugates represent the major circulating forms of A. manihot flavonoid in vivo. AUC of isorhamnetin-glucuronide conjugates and quercetin-sulfate conjugates was 719.65 ± 619.22 and 275.49 ± 1 60.95 μmol h/L, indicating that less conjugated metabolites were formed in CKD rats compared with normal rats. The ratio of AUCglucuronide/sulfate/AUCaglycone decreased from 125 to 104, which implied the impaired phase II metabolism ability in CKD rat. Discussion and conclusion: Glucuronide-sulfate conjugates provide an important clue for further elucidating the activity of conjugated metabolites and their relationship with the nephroprotective efficacy of A. manihot. It is necessary to take caution when extrapolating pharmacokinetics parameters from healthy animals in designing pharmacological studies.

Abstract:Hibifolin, the highest-content bioactive flavonoid of the flowers of Abelmoschus manihot, was incubated with human intestinal bacteria, and four metabolites (1-4) were obtained from the incubated solution by chromatographic methods. The structures of the four metabolites were elucidated as gossypetin 8-O-beta-D-4"-deoxy-Delta 4"-glucuropyranoside (1), gossypetin (2), quercetin (3), and 8-methoxy-quercetin (4), respectively, on the basis of UV, NMR, and MS data. Metabolite 1 was obtained as a new compound with a specific beta-D-4"-deoxy-Delta 4"-glucuropyranosyl moiety, which was formed through a unique and novel metabolic pathway that has not been reported previously.
Efficient Production of Flavonoid Glucuronides in Escherichia coli Using Flavonoid O -Glucuronosyltransferases Characterized from Marchantia polymorpha

Abstract:As a model liverwort, Marchantia polymorpha contains various flavone glucuronides with cardiovascular-promoting effects and anti-inflammatory properties. However, the related glucuronosyltransferases have not yet been reported. In this study, two bifunctional UDP-glucuronic acid/UDP-glucose:flavonoid glucuronosyltransferases/glucosyltransferases, MpUGT742A1 and MpUGT736B1, were identified from M. polymorpha. Extensive enzymatic assays found that MpUGT742A1 and MpUGT736B1 exhibited efficient glucuronidation activity for flavones, flavonols, and flavanones and showed promiscuous regioselectivity at positions 3, 6, 7, 3', and 4'. These enzymes catalyzed the production of a variety of flavonoid glucuronides with medicinal value, including apigenin-7-O-glucuronide and scutellarein-7-O-glucuronide. With the use of MpUGT736B1, apigenin-4'-O-glucuronide and apigenin-7,4'-di-O-glucuronide were prepared by scaled-up enzymatic catalysis and structurally identified by NMR spectroscopy. MpUGT742A1 also displayed glucosyltransferase activity on the 7-OH position of the flavanones using UDP-glucose as the sugar donor. Furthermore, we constructed four recombinant strains by combining the pathway for increasing the UDP-glucuronic acid supply with the two novel UGTs MpUGT742A1 and MpUGT736B1. When apigenin was used as a substrate, the extracellular apigenin-4'-O-glucuronide and apigenin-7,4'-di-O-glucuronide production obtained from the Escherichia coli strain BB2 reached 598 and 81 mg/L, respectively. Our study provides new candidate genes and strategies for the biosynthesis of flavonoid glucuronides.

Purification of Total Flavonoids of Abelmoschus Manihot (L.) Medic and Its Antioxidant Properties in Vivo

Abstract:In order to optimize the purification conditions and the antioxidant properties in vivo of total flavonoids from Abelmoschus manihot (L.) Medic, AB-8 macroporous resin is used for static adorption and elution. The optimum purification conditions are as follows: Sample solution concerntration of 0.1 g/mL, pH 3.5, 70% ethanol as eluant, 20∶1(mL/g) of eluant volum to resin weight ratio. The results in the optimum purification conditions show that the purity of TFA reaches 48.50%. There are no significant difference in indexes of mice organs between experimental groups and normal control group (P＞005). MDA content exhibits a significant decrease both in serum and liver of mice (P＜0.05). While the level of CAT, GSH-Px and SOD activities in serum and liver are higher in experimental groups than those in normal control group showing dose-response relationship. These results indicate that TFA has good antioxidant properties in vivo.

Abstract:A SPE-HPLC method was developed and validated for the simultaneous determination of flavonols, isoquercitrin (1), hibifolin (2), myricetin (3), quercetin-3′-O-d-glucoside (4) and quercetin (5) in rat plasma and urine after oral administration of the total flavonoids from Abelmoschus manihot (TFA). The astragalin (6) and kaempferol (7) were used as internal standards (IS). Plasma and urine samples were pretreated by solid-phase extraction using Winchem™ C18 reversed-phase cartridges. Analysis of the plasma and urinary extract was performed on YMC-Pack ODS-A C18 and Thermo ODS-2HYEPRSIL C18 reversed-phase column, respectively and a mobile phase of acetonitrile–0.1% phosphoric acid was employed. HPLC analysis was conducted with different elution gradients. The flow rate was 1.0mL/min and the detection wavelength was set at 370nm. Calibration ranges in plasma for flavonols 2–5 were at 0.011–2.220, 0.014–2.856, 0.022–4.320, and 0.028–5.600μg/mL, respectively. In urine calibration ranges for flavonols 1, 2, 4 and 5 were at 2.00–16.00, 8.56–102.72, 2.70–21.60, and 3.00–24.00μg/mL, respectively. The RSD of intra- and inter-day was less than 5.40% and 4.89% in plasma, and less than 3.96% and 6.85% in urine for all the analyses. A preliminary experiment to investigate the plasma concentration and urinary excretion of the flavonols after oral administration of TFA to rats demonstrated that the present method was suitable for determining the flavonols in rat plasma and urine.

Fries es Nym. The structure of 1 was elucidated by analysis of its spectroscopic data, including UV, IR, HR-ESI-MS and extensive 1 D and 2 D NMR techniques. In vitro test, compound 1 behaved similarity to swertianolin against α‑glucosidase and more potent inhibitory effects than the positive control, acarbose.

Abstract:Flavonol synthase (FLS) is a key enzyme of the flavonoid biosynthetic pathway, which catalyzes the conversion of dihydroflavonols into flavonols. In this study, the FLS gene IbFLS1 was cloned and characterized from sweet potato. The resulting IbFLS1 protein showed a high similarity with other plant FLSs. The conserved amino acids (HxDxnH motifs) binding ferrous iron and residues (RxS motifs) binding 2-oxoglutarate were found in IbFLS1 at conserved positions, as in other FLSs, suggesting that IbFLS1 belongs to the 2-oxoglutarate-dependent dioxygenases (2-ODD) superfamily. qRT-PCR analysis showed an organ-specific pattern of expression of the IbFLS1 gene, which was predominantly expressed in young leaves. The recombinant IbFLS1 protein could catalyze the conversion of dihydrokaempferol and dihydroquercetin to kaempferol and quercetin, respectively. The results of subcellular localization studies indicated that IbFLS1 was found mainly in the nucleus and cytomembrane. Furthermore, silencing the IbFLS gene in sweet potato changed the color of the leaves to purple, substantially inhibiting the expression of IbFLS1 and upregulating the expression of genes involved in the downstream pathway of anthocyanin biosynthesis (i.e., DFR , ANS , and UFGT ). The total anthocyanin content in the leaves of the transgenic plants was dramatically increased, whereas the total flavonol content was significantly reduced. Thus, we conclude that IbFLS1 is involved in the flavonol biosynthetic pathway and is a potential candidate gene of color modification in sweet potato.
