Abstract:A series of difluoromethyl-containing a-acyloxycarboxamide derivatives were synthesized through a Passerini reaction of acids, aldehydes, and phenylsulfanyl-protected difluorinated isocyanide followed by meta-chloroperoxybenzoic acid mediated oxidation and the removal of the phenylsulfonyl protecting group. The unexpected formation of deacylation products from the reaction between the Passerini products and Bu3SnH/azobis(isobutyronitrile) was also reported.

Abstract:Due to the uniqueness of fluorine atom and C—F bond,difluoromethylene has special properties.As a bioisostere of an oxygen or a carbonyl group,it plays an important role in medicines,pesticides and materials.Difluoromethyl heteroaryl sulfones,represented by 2-PySO2CF2H (Hu reagent),have been developed recently as difluoromethylation reagents,and widely recognized by synthetic chemists for their ease of preparation,good functional group tolerance and universal applicability to a wide range of carbonyl compounds.Through different types of reactions such as nucleophilic substitution,nucleophilic addition,Julia-Kocienski olefination reaction,and radical-mediated difunctionalization,they introduced difluoromethyl,difluoromethylene,gem-difluoroalkene and other fluorine-containing groups into aldehydes,ketones,and heterocyclic compounds.For the first time,the synthesis of fluorine-containing organic compounds involved in various difluoromethyl heteroaryl sulfones in the past decade is reviewed from the perspective of reaction types and their application studies.

Abstract:The nucleophilic difluoroalkylation of isocyanates with difluoromethyl 2-pyridyl sulfone furnished 2,2-difluoro-2-pyridinylsulfonylacetamides with good to excellent yield. These products can be converted to 2,2-difluoroacetamide sulfinate, iodo-difluoroacetamide and 2-aminopyridine derivatives easily.

Abstract:Under mild conditions, a series of alpha,alpha-difluoro-beta-ketoamides were synthesized with Selectfluor (R) as the F+ source in the presence of K2CO3 in the mixed-solvent of water and PEG-400 (V-H2O : VPEG-400 = 3 : 1), without additional phase-transfer catalyst. By using this approach, most cases examined in this study proceeded in nearly quantitative conversions regardless of the electronic nature of the substituent pattern. (c) 2013 Elsevier B.V. Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:Novel acylamino-directed relay disubstitutions realize the sequential difunctionalizations of anilides (1) under mild and metal-free conditions for the first time. This [bis(trifluoroacetoxy)iodo]benzene (PIFA) and BF3·Et2O promoted straightforward reaction produces a series of p-acetoxyl- or p-alkoxyl- o-nitro- N-arylamides (2), which are key scaffolds of various drugs, functional materials, and bioactive molecules. The flexibility with respect to the functional groups in these products affords this novel protocol excellent versatility for synthetic applications.

Abstract:Fluorinated small molecules are commonly utilized in the realm of functional small-molecule chemistry, and N-difluoromethyl (N-CF2H) compounds are especially fascinating because of their unique and unexplored physiochemical properties. However, despite limited progress, a general applicable methodological approach to the synthesis of N-CF2H compounds remains elusive. Herein, guided by computation, we present an operationally simple and practical protocol to access N-CF2H amides and related carbonyl derivatives. The protocol utilizes a one-pot conversion of thioformamides via desulfurization-fluorination and acylation, providing N-difluoromethylcarbamoyl fluoride building blocks that can be further diversified to a diversity of unexplored N-CF2H carbonyl compounds with rich functionality. Furthermore, preliminary studies on their properties and stability showcased their application in pharmaceuticals and agrochemicals.

Abstract:Asymmetric detrifluoroacetylative Mannich reactions between α‐fluorinated arylsulfonyl gem ‐diols and fluoroalkyl imines have been developed under operationally convenient conditions. Varieties of α‐fluorinated arylsulfonyl gem ‐diols and fluoroalkyl imines are well tolerated with the corresponding α‐difluorinated β‐amino sulfone products generated in moderate to excellent yields as well as high diastereoselectivity. The reaction can be reproduced on gram‐scale with uncompromised yield and stereochemical outcome. The synthetic value of the products is demonstrated by two transformations by means of desulfonylation and removal of the sulfinyl protecting‐group.

Abstract:N-Arenesulfonyl α-hydroxyamides are conveniently synthesized from N-arylethynylsulfonamides (ynamides) via oxidation with dimethyl sulfoxide (DMSO) in the presence of equivalent of trifluoromethanesulfonic acid (TfOH) followed by treatment with water. In this reaction, ynamides are activated by protonation with TfOH and then undergo double DMSO nucleophilic addition and subsequent hydrolysis. Investigations on the substrate scope indicate N-arenesulfonyl and arylethynyl substituted ynamides proceed the oxidation well. Compared to the previous methods, the current method realizes the direct conversion from N-arylethynylsulfonamides to N-arenesulfonyl α-hydroxyamides with mild DMSO as a nucleophilic oxidant in the presence of strong acid trifluoromethanesulfonic acid.

Abstract:A new synthetic approach for the preparation of amides and peptides containing amino acids as well as aryl acids and amines by employing [4-(acetylamino)phenyl]imidodisulfuryl difluoride (AISF) as a carboxylic acid activator under mild conditions is delineated. The use of AISF as an acid activator allows the reaction to be performed efficiently. This operationally simple amidation is amenable to a wide variety of carboxylic acids and N α -protected amino acids. Further, racemization did not occur during the coupling. Abstract:The synthesis of N‐(2‐SF5‐ethyl)amides is described via an amidation reaction between 2‐(pentafluoro‐λ6‐sulfanyl)acetic acid, the SF5‐containing building block, and various amines. A total of 27 examples of SF5‐containing amides were synthesized, with yields going from 43% to quantitative isolated yield. We showed that a N‐(2‐SF5‐ethyl)amide could be converted to a N‐(2‐SF5‐ethyl)amine and a SF5‐containing oxadiazoles. Finally, the effect of the SF5 substituent on the lipophilicity of the amide of a model compound was evaluated.
